Polymerase chain reaction(PCR)-based inter-simple sequence repeats(I-SSR) markers were analyzed in 48 megagametophytes of a single tree of Abies koreana $W_{ILS}$. Nineteen of the 35 primers, screened with 6 megagametophyte DNA and produced the clearest amplification products in the preliminary experiment, were used for PCR with 48 megagametophyte DNAs sampled from a single tree. On the basis of the chi-square test, a total of 51 amplicons, amplified by the 19 primers, were revealed to be segregated according to the Mendelian ratio(i.e., 1 : 1 segregation ratio) in the 48 megagametophytes at 5% significance level. Based on the linkage analysis, the observed 51 Mendelian loci turned out to be unlinked each other, which suggested that they are evenly distributed in the genome. However, majority of RAPD markers are known to belong to the independent linkage blocks, which frequently results in the amplification of RAPD markers from the restricted regions of the genome. Owing to the nature of even distribution of the 51 loci observed in this study, the I-SSR markers could give better resolution of estimating genetic diversity from the whole genome than RAPD markers. And I-SSR markers are also more suitable than RAPD markers for reconstructing phylogenetic relationship by a cladistic method which requires to fulfil the assumption of independent evolution of the different characters.
The objectives of this study was to identify genetic variations of Tricholoma matsutake (S. Ito et Imai) Sing. growing in different geographic ranges in South Korea. Mushrooms were collected during fruiting seasons from 1994 to 1997 from 11 major sites which included four sites (Bonghwa, UIjin, Goryoung, and Chungdo) in Kyongbuk Province, three sites (Changnyung, Hadong, and Hamyang) in Kyongnam Province, two sites (Yangyang and Inje) in Kangwon Province, one site (Goisan) in Choongbuk Province, and one site (Namwon) in Chonbuk Province. Two mushrooms each from three to eight shiros in each sites were collected. Genetic characteristics were analyzed by Inter-Simple Sequence Repeat Polymerase Chain Reaction (I-SSR PCR) method using six primers. With a total of 131 DNA bands identified, Nei's genetic distance and UPGMA tree were constructed. It was estimated that genetic variations between sites amounted to 12.9%, while 87.1% of total variation was explained by variations among individuals within sites. The cluster analysis indicated that the eleven major sites were clustered into four groups, group I (Yangyang, Hamyang, Inje, Hadong and UIjin), group II (Changnyung, Namwon and Chungdo), group III (Goryoung), and group IV (Bonghwa and Goisan). It is concluded that matsutake mushrooms in South Korea have a considerable degree of genetic variations between major sites.
Inter-simple sequence repeat (I-SSR) markers were analyzed from diploid genomes of 95 nutmeg trees (Torreya nucifera Siev. et Zucc.) in 5 populations. A total of 62 I-SSR amplicons were observed and 7 of them were monomorphic in 95 individuals. DNA fingerprint of each tree was verified by pooling the observed I-SSR amplicons. Most of the genetic diversity was allocated within population (90.65%) and all the populations revealed similar level of I-SSR amplicon diversity within population. Degree of population differentiation (${\phi}_{ST}=9.35%$) was moderate on the basis of criteria obtained from isozyme analysis. Based on the results of the cluster analysis of UPGMA, genetic relationships among 5 populations were not coincided with the pattern of geographic distribution. Non-significant confidence interval at each node also suggests that all the nutmeg populations are genetically not much differentiated.

We investigated the genetic variation in Stewartia koreana Nakai by examining 61 I-SSR amplicons in 120 individuals distributed among six natural populations in Korea. The overall percentage of polymorphic I-SSR amplicons was 81.9% and mean number of amplicons per I-SSR primer was 12.2. Levels of genetic diversity within 6 populations were similar each other[Shannon's Index $0.358{\sim}0.467$(mean: 0.407)]. The Mt. Obong population had the highest level of genetic diversity and was most distinctive from the other populations. Most variation existed among individuals within population(88.2%). Genetic differentiation among populations(${\phi}_{ST}$) was 0.118. The UPGMA dendrogram based on the genetic distance failed in showing decisive geographic relationships.
Faba bean (Vicia faba L.) is one of the most important legume crops in Egypt. However, production of faba bean is affected by several diseases including fungal diseases. Fusarium wilt incited by Fusarium oxysporum Schlecht. was shown to be the most common wilt disease of faba bean in Assiut Governorate. Evaluation of 16 faba bean genotypes for the resistance to Fusarium wilt was carried out under greenhouse conditions. Three molecular marker systems (inter-simple sequence repeat [ISSR], sequence related amplified polymorphism [SRAP], and simple sequence repeat [SSR]) and a biochemical marker (protein profiles) were used to study the genetic diversity and detect molecular and biochemical markers associated with Fusarium wilt resistance in the tested genotypes. The results showed that certain genotypes of faba bean were resistant to Fusarium wilt, while most of the genotypes were highly susceptible. The percentage of disease severity ranged from 32.83% in Assiut-215 to 64.17% in Misr-3. The genotypes Assiut-215, Roomy-3, Marut-2, and Giza2 were the most resistant, and the genotypes Misr-3, Misr-1, Assiut-143, Giza-40, and Roomy-80 performed as highly susceptible. The genotypes Assiut-215 and Roomy-3 were considered as promising sources of the resistance to Fusarium wilt. SRAP markers showed higher polymorphism (82.53%) compared with SSR (76.85%), ISSR markers (62.24%), and protein profile (31.82%). Specific molecular and biochemical markers associated with Fusarium wilt resistance were identified. The dendrogram based on combined data of molecular and biochemical markers grouped the 16 faba bean genotypes into three clusters. Cluster I included resistant genotypes, cluster II comprised all moderate genotypes and cluster III contained highly susceptible genotypes.
The jujube is an important fruit tree species in Korea. Traditionally, classifications of jujube cultivars have been based on morphological characters; however, morphological identification can be problematic because morphological traits are affected by environmental conditions. Therefore, DNA markers are now being used for the rapid and accurate identification of plant species. Inter-simple sequence repeat (I-SSR) is one of the best DNA-based molecular marker techniques, which is useful for studying genetic relations and for the identification of closely related cultivars. In this study, 5 Korean jujube trees and 1 jujube tree imported from China were analyzed for 16 I-SSR primers. Amplification of the genomic DNA of jujube cultivars by using I-SSR analysis generated 100 bands, with an average of 6.25 bands per primer, of which 45 bands (45%) were polymorphic. The number of amplified fragments with I-SSR primers ranged from 2 to 13. The percentage of polymorphism ranged from 10% to 100%. I-SSR finger printing profiles showed that 'Boeun jujube' and 'Daeri jujube' had characteristic DNA patterns, indicating unequivocal cultivar identification at molecular level. According to the results of clustering analysis, the genetic similarity coefficient ranged from 0.68 to 0.92. 'Boeun jujube' and 'Daeri jujube' were divided into independent groups, and 'Bokjo jujube', 'Geumseong jujube', 'Wolchul jujube', and 'Mudeung jujube' were placed in the same group. Therefore, I-SSR markers are suitable for the discrimination of 'Boeun jujube' and 'Daeri jujube' cultivars.
Winter bud explants from 15 individual angelica tree (Aralia elata) were cultured in vitro to find out optimal conditions for somatic embryo induction as well as plant regeneration. Calli are induced and grown on MS medium supplemented with 1.0 mg/L 2,4-D for 4 weeks and subcultured on a half-strength MS medium without phytohormones to induce somatic embryos. Inter-simple sequence repeat (I-SSR) markers were analyzed with total DNAs extracted from the trees. Genotype effects on somatic embryo induction were examined by cluster analysis. Callus induction rate varied from 58.5 to 100% among the genotypes. Somatic embryo induction rate also greatly varied from 0 to 100% among the genotypes. There was a significant difference in somatic embryo induction rate even among the individual trees that showed close genetic relationships each other. This suggested that somatic embryo induction rate in Aralia elata be influenced by a few major specific genes rather than whole genomic similarity among individual trees. Four individuals of Ulneong-7, Cheju-1, Shingu and China, which are recalcitrant to somatic embryo induction, turned out to have a close genetic relationship, suggesting that both physiological and genetic factors affect somatic embryo induction. The results suggest that genotype selection be the most important factor to achieve an efficient propagation, although cultural optimization through medium and explant manipulation may also play crucial roles in somatic embryogensis as well as plant regeneration of these species.
Level and distribution of genetic diversity in seven populations of Albizia lucida Benth. in eastern region of the Indian sub-continent were estimated using ISSR markers. Relatively higher level of genetic diversity within populations was observed in seven populations of A. lucida (mean of 0.38). From the result of AMOVA, majority of genetic diversity was allocated within populations (96.2%) resulting in a moderate degree of population differentiation. The observed distribution pattern of I-SSR variant among the populations was coincided with the typical pattern of long-lived woody tree species. Genetic relationships among the populations, reconstructed by UPGMA method, revealed two genetic groups. The population of Anugul and Bargarh turned out to be the most closely related despite a distance location between them. These formations will be of great value in the development of conservation plans for species exhibiting high levels of genetic differentiation due to fragmentation, such as indication of conservation unit size, which populations should be chosen as priority in conservation plans and which samples should be introduced in areas with a low number of individuals of A. lucida.
