Rust Fungus Diferentiation on Host Leaves (기주식물엽에 있어서 녹병균의 분화)

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4$ 1.0g, sucrose 30.0g, $MgSo_4$ $7H_2$O 0.5g, $FeCl_36H_2$O 1.0mg thiamine 200.gamma.g, and distilled water 1000ml, pH6.5.

Weekday/weekend Chemical Characteristics of Water-Soluble Components of PM10 at Busan in Springtime (부산지역 봄철 주중/주말의 PM10 중 이온성분의 화학적 조성)

  • This study investigates weekday/weekend characteristics of $PM_{10}$ concentration and chemical composition of water-soluble ions in Busan in the spring of 2013. Contribution rate of water-soluble ions to PM10 concentration in weekday/weekend were 41.5% and 38.5%, respectively. Contribution rate of SO_4{^{2-}}$ to total ion mass in weekday/weekend were 30.4% and 33.8%, respectively. Contribution rate of total inorganic water-soluble ions in PM10 in weekday/weekend were 42.2% and 39.1% (mean 41.4%), respectively. $[NO_3{^-}/SO_4{^{2-}}]$ ratio in weekday/weekend were 1.01 and 0.97(mean 0.99), respectively, which indicated that weekday ratio was higher. Contribution rate of sea salts and $Cl^-/Na^+$ ratio in PM10 in weekday/weekend were 8.1% and 7.6%, 0.37% and 0.41%, respectively. This research will help understand chemical composition of water-soluble ions during the weekday/weekend and will be able to measure the contribution level of artificial anthropogenic source on urban air.

Synthesis, Characterization, DFT Modeling and Antimicrobial Studies on the Ti(IV), Y(III) and Ce(IV) Ofloxacin Solid Complexes

  • A new solid complexes of Ti(IV), Y(III) and Ce(IV) have been synthesized with ofloxacin. The formulae and structure of the complexes have been proposed in the light of analytical, spectral ($^1H$ NMR, IR and UV-Visible), magnetic, molar conductivities and thermal studies. The complexes are soluble in DMSO-$d_6$ and DMF. The measured molar conductance values indicate that, the three complexes are electrolyte in nature. The results support the formation of the complexes and indicated that ofloxacin reacts as a bidentate ligand chelate to the metal ion through the pyridone oxygen and one carboxylato oxygen. The kinetic parameters of thermogravimetric and its differential have been evaluated by using Coats Redfern (CR) and Horowitz-Metzeger (HM) methods. The thermodynamic data reflect the thermal stability for all complexes. The metal- ligand binding of the Ti(IV), Y(III) and Ce(IV) complexes is predicted using density funcational theory at the B3LYP-CEP-31G level of theory and total energy, dipole moment estimation of different Ti(IV), Y(III) and Ce(IV) ofloxacin structures. The biological activities of the ofloxacin, inorganic salts and their metal complexes were assayed against different bacterial species.

Pulverization and Densification Behavior of YAG Powder Synthesized by PVA Polymer Solution Method

  • YAG (Yttrium Aluminum Garnet, Y3Al5O12) has excellent plasma resistance and recently has been used as an alternative to Y2O3 as a chamber coating material in the semiconductor process. However, due to the presence of an impurity phase and difficulties in synthesis and densification, many studies on YAG are being conducted. In this study, YAG powder is synthesized by an organic-inorganic complex solution synthesis method using PVA polymer. The PVA solution is added to the sol in which the metal nitrate salts are dissolved, and the precursor is calcined into a porous and soft YAG powder. By controlling the molecular weight and the amount of PVA polymer, the effect on the particle size and particle shape of the synthesized YAG powder is evaluated. The sintering behavior of the YAG powder compact according to PVA type and grinding time is studied through an examination of its microstructure. Single phase YAG is synthesized at relatively low temperature of 1,000 ℃ and can be pulverized to sub-micron size by ball milling. In addition, sintered YAG with a relative density of about 98 % is obtained by sintering at 1,650 ℃.

A Study on the Optimal Mixture Ratio for Stabilization of Surface Layer on Ultra-soft Marine Clay (초연약 해성점토의 표층고화처리를 위한 최적배합에 관한 연구)

  • Recently, as large constructions on the coast are performed frequently, surface layer stabilization method which Is one of the improvement methods for dredged soft clay has been applied. However, there have been few studies about the surface layer stabilization method. The purpose of this study is to clarify characteristics of ultra-soft marine clay and hardening agent. Also, optimal mixture ratio of hardening agent was verified through the laboratory tests such as statistical analysis and pilot tests. Laboratory tests were performed with proper hardening agent and test soil and standard mixing tables of hardening agent were determined according to ground conditions through statistical analysis. Also, applicability of surface layer stabilization method to field was verified by pilot tests. From the results of the tests, it was found that hardening agent materials such as cement, slag, fly-ash, inorganic salts, arwin, gypsum etc. affect on the appearing compressive strength. It was defined optimal mixture ratio which satisfies the required compressive strength from the statistical analysis. Also, It was compared the effect of ground improvement by cements and hardening agents through the pilot tests. This study will serve as data for design or construction criteria of stabilization of surface layer on ultra-soft marine clay.

Characteristics of Metallic and Ionic Elements Concentration in PM10 at Guducsan in Busan (부산 구덕산 미세먼지의 금속성분 및 이온성분 농도 특성)

  • This study investigates the characteristics of metallic and ionic elements concentration, concentration according to transport path, and factor analysis in $PM_{10}$ at Guducsan in Busan in the springtime of 2015. $PM_{10}$ concentration in Guducsan and Gwaebeopdong were $59.5{\pm}9.04{\mu}g/m^3$ and $87.5{\pm}20.2{\mu}g/m^3$, respectively. Contribution rate of water-soluble ions and secondary ion in $PM_{10}$ concentration in Guducsan were 37.0% and 27.8% respectively. [$NO_3{^-}/SO{_4}^{2-}$] ratio and contribution rate of sea salt of $PM_{10}$ in Guducsan and Gwaebeopdong were 0.91 and 1.12, 7.0% and 5.3%, respectively. The results of the backward trajectory analysis indicates that $PM_{10}$ concentration, total inorganic water-soluble ions and total secondary ions were high when the air parcels moved from Sandong region in China than non-Sandong and northen China to Busan area. The results of the factor analysis at Guducsan indicates that factor 1 was anthropogenic source effects such as automobile emissions and industrial combustion processes, factor 2 was marine sources such as sea salts from sea, and factor 3 was soil component sources.

Selection of Cell Source and the Effect of pH and MS Macronutrients on Biomass Production in Cell Cultures of Tongkat Ali (Eurycoma longifolia Jack)

  • Callus and cell suspension cultures of Eurycoma longifolia Jack were initiated from leaves of different trees. The leaf explant of tree Eu9 produced the most calli and also induced high cell biomass in the cell suspension culture. Optimum production of cell biomass could be initiated in proliferating culture medium with a pH of 5.75 prior to autoclaving. The effects of macronutrient inorganic salts of Murashige and Skoog (MS) liquid medium supplemented with X on production of cell biomass of Eurycoma longifolia were also investigated. The highest cell biomass was produced in MS medium containing macronutrients of $21\;mM\;NH_4NO_3,\;12.25\;mM\;KNO_3,\;3.00\;mM\;CaCl_2.2H_2O,\;0.575\;mM\;MgSO_4.7H_2O$, and $1.83\;mM\;KH_2PO_4$. A new medium labeled as TAM was formulated for the production of Eurycoma longifolia cell biomass in the cell suspension culture.

Volatile Flavor Compounds from Pear Juice (Pyrus pyrifolia cv. Niitaka) (배주스의 휘발성 향기성분)

  • The aim of this study was to determine volatile flavor compounds in Shingo pear juice. Volatile flavor compounds were analyzed using solid-phase micro-extraction (SPME) - gas chromatography-mass spectrometry (GC-MS). The effect of inorganic salts solution on the extraction ability of the SPME fiber was treated by adding saturated $CaCl_2$ solution at the ratio of 1:20 (v/v) after 0, 60, 120 min of preparing pear juice, respectively. As a result, a total of 22 volatile compounds were identified in Shingo pear juice. Ethyl acetate was found to be the most abundant volatile compound ($13.36{\sim}19.61{\mu}g/kg$), followed in order by hexanal, ethyl hexanoate, ethyl 3-(methylthio)-2-propenoate, ethyl octanoate and 2-hexenal. Total contents of volatile flavor compounds were $31.07{\mu}g/kg$ (control), $40.93{\mu}g/kg$ (0 min), $27.62{\mu}g/kg$ (60 min) and $26.32{\mu}g/kg$ (120 min). This result indicated that the addition of saline solutions could inhibit the enzymatic reaction of volatile flavor compounds effectively when treated as soon as juice preparation.

Direct Detection of Water-dissolved Ammonia Using Paper-based Analytical Devices

  • A microfluidic paper-based analytical device (µPAD) is proposed for the selective detection of ammonia in water by using the modified Berthelot reagent and a fluidic channel consisting of hollow paper. The modified Berthelot reagents were uniformly dispersed in cyclohexane and then immobilized in a detection zone of the µPAD. The loading position of the reagents and the type of a sample flow channel were optimized to achieve a sensitive ammonia detection within a short analytical time. The NH3 µPAD exhibits a linear colorimetric response to the concentration of ammonia dissolved in water in the range of 1-100 mg L-1, and its limit-of-detection is 1.75 mg L-1. In addition, the colorimetric response was not influenced by the addition of 100 mg L-1 nitrogen containing compounds (sodium nitrate, sodium nitrite, uric acid, hydroxylamine, butylamine, diethylamine) or inorganic salts (NaCl, Na2HPO4), presenting the enough selectivity in the detection of water-dissolved ammonia against possible interferents.

Influence of a Single Dose of Fe Dextran Administration with Organic Trace Mineral Supplementation on the Performance of Piglets

  • This study was designed to evaluate the influence of a single or double dose of Fe dextran with organic trace mineral supplementation on the performance of piglets from dams fed diets with either inorganic (ITM) or organic trace minerals (OTM). It also determined the effect of the source of the trace minerals on the reproductive performance of sows. The trace mineral premixes were prepared using metal proteinates and the corresponding inorganic salts for the OTM and the ITM, respectively. Each mineral premix provided 100 ppm Fe/175 ppm Fe, 35 ppm Cu/170 ppm Cu, 90 ppm Zn/120 ppm Zn, and 40 ppm Mn/35 ppm Mn when added at 0.20% in sows /weaned pigs' diets, respectively. The first dose of Fe dextran was administered to piglets at 3 d and the second dose at 10 d after birth. One dose of Fe dextran supplied 100 mg of Fe. A total of 16 gestating sows (Landrace${\times}$Yorkshire) in parities 2 to 4 were randomly allocated to four treatments: 1) diet with ITM/one dose of Fe dextran to piglets, 2) diet with ITM/two doses of Fe dextran to piglets, 3) diet with OTM/one dose of Fe dextran to piglets, and 4) diet with OTM/two doses of Fe dextran to piglets. The total born alive, weaned, body weight at birth and at weaning were not affected by the sow's dietary treatment. Although organic trace mineral supplementation tended to increase the milk Fe content (p<0.10) at 7 d postpartum, piglets in all treatments performed equally from birth to weaning. The double doses of Fe dextran neither improved the average daily gain (ADG) nor influenced the survival of piglets from birth to weaning (21 d). Results suggest that a single dose of Fe dextran given to suckling pigs is adequate to sustain their needs for growth throughout the lactation period (21 d). Furthermore, there was a 21% improvement in both the ADG and the average daily feed intake (ADFI) (p<0.05) in weaned pigs fed diets with OTM. Cu and Fe in the liver (p<0.01), and Zn in both the bone (p<0.01) and the serum (p<0.01) were higher in piglets fed OTM than in those fed ITM. It would be concluded that single dose of Fe dextran administration with organic trace mineral supplementation show similar growth performance compared to 2 dose Fe dextran administration with inorganic mineral supplementation in young pigs.
