Phosphatase Activity in Cheonho Reservoir

  • Phosphatase activity was measured with other environmental factors in Cheonho reservoir in 1994. It ranged form 95 to 1,685 nM/1/h and was correlated significantly with chlorophyll-a. Such a close relation well matched the fact that over 90% of phosphatase activity was detected in > 3 $\mu\textrm{m}$ fraction. The phosphatase activity also correlated negatively with dissolved inorganic phosphate concentration, which implies derepression of phosphatase production by phosphate limitation. Significant correlation was analyzed between phosphatase activity and BOD, which also appeared to be closely correlated with chlorophyll-a. A great percentage of organic materials seems to be generated autochthonously by algae and extracellular enzyme even though allochthonous influence was thought to be stronger in Cheonho reservoir.

A Study on the Influence of Release Characteristics of Phosphorus Fraction in the Sediment (연안 퇴적물에서 인의 존재형태가 용출 특성에 미치는 영향에 대한 연구)

  • This study investigated the effect of the sediment phosphorus fraction sampled from the southern coast of Korea on the release characteristics of sediments by environmental changes of water quality. We conducted the release experiment in the laboratory for 20 days and measured the phosphorus fraction properties, the environmental factors of water quality, and the release rate of total phosphorus. The results showed a decrease in dissolved oxygen by the growth of microorganisms in the water layer, leading to the anaerobic condition in which the redox potential of the sediments decreased. As such, the decreasing variability of phosphates bonded to iron oxide in the sediment phosphorus was higher after 20 days of the release experiment than the first day. It means that the metal ions and the separated inorganic phosphorus transfer into the water when the iron oxide is reduced. The separated inorganic phosphorus is easily absorbed by the plankton. The analysis of total phosphorus in the water layer showed that it continuously increased to up to 0.304 mg/L for 20 days, and the release rate had a high correlation with the decrease of dissolved oxygen after 5 days of culture. Therefore, it is necessary to pay attention to the characteristics of iron bonded to phosphorus in the phosphorus fraction and dissolved oxygen to manage the eutrophication of the system.

Ammonium Behavior and Nitrogen Isotope Characteristics of 2:1 Clay Minerals from Submarine Hydrothermal System in the Wakamiko Crater of Kagoshima Bay, Southwestern Japan (일본 서남부 가고시마 와카미코 해저 열수환경에서 형성된 2:1 점토광물 내 암모늄 거동 및 질소동위원소 특성)

  • 2:1 clay minerals such as smectite incorporating ammonium were extracted to investigate the ammonium behavior and nitrogen isotope characteristics for two different sediment cores which were collected from shimmering sites on seafloor of the Wakamiko crater, southwestern Japan. Inorganic nitrogen contents in clay fraction were estimated by calibration curve based on consistently decreasing carbon and nitrogen ratio during the treatment to decompose organic materials, after removing inorganic carbon. The results show that the proportions of inorganic nitrogen for total nitrogen in clay fraction of SWS site(Core#1094MR: av. 18.2%) are higher than those in SES site(Core#1093MG: av. 11.5%). Relatively good crystallinity of the former suggests that exchangeable ammonium was transformed to non-exchangeable ammonium during more evolving diagenetic process. Nitrogen isotope variance of clay fraction(SES site: Core#1093MG: -4.4 ~ +0.2 ‰, av. -2.4 ‰; SWS site: Core#1094MR: -0.7 ~ +3.0 ‰, av. +1.5 ‰) during sequential decomposition of exchangeable ammonium suggests that heat flow derived from deep magma led to nitrogen isotope fractionation between dissolved ammonium and ammonia in the fluids involved in the formation of 2:1 clay mineral incorporating ammonium with local temperature variation.

Distribution of phosphorus in particle-size separates and specific gravity separates of soils (입경 및 비중별(比重別) 토양분화과 인산분포(燐酸分布))

  • 1) Soils (volcanic ash and muck) were fractionated into particle-size separates (200 - 20, 20 - 2, 2 - 0.5 and finer than $0.5{\mu}$ in diamter), and of which the silt fraction was further fractionated into specific gravity separates (more than 2.0, 2.0 - 1.7, 1.7 - 1.4 and less than 1.4 in $g/cc^3$). And total organic and inorganic phosphorus in the separates were determined. 2) The amounts of total, organic and inorganic phosphorus distributed in the particle-size separates were as follows fine clay > coarse clay > silt > fine sand fraction. The increase rate in the amounts of phosphorus was great in the separates finer than $20{\mu}$, and greatest in the fine clay fraction. 3) The amounts of total, oganic and inorganic phosphorus distributed in the specific gravity separates were as follows: 2.0 - 1.7 > 1.7 - 1.4 > heavier than 2.0 fraction. The increase rate in the amounts of phosphorus was in the following order 2.0 - 1.7 > 1.7 - 1.4 > heavier than 2.0 fraction. 4) Distribution of carbon, amorphous aluminum and free iron oxides in the particle-size separates and the specific gravity separates were examined, and the distribution and the formes of organo-minera1 complexes in the separates were discussed to shed light on the factors affecting the distribution of phosphorous into the separates. And it was estimated that there was close relation among the distribution of organic and inorganic phosphorus, and the distribution and the formes of organo-minera1 complexes.

Retention of Benzoic Acids in RPLC (역상액체크로마토그래피에서 벤조산류의 머무름)

  • Chromatographic retention mechanism of seven benzoic acids in RPLC were investigated. Inorganic salt is pertinent for the measurement of dead time in benzoic acid. Logarthims of capacity factor (log k') and Hildebrand solubility parameter (${\delta}$) showed proportional relationship which imply the hydrophobic effect of the retention mechanism in RPLC. Enthalpies of solute transfer showed inverse proportion to temperature and organic modifier ratio of the mobile phase. It was found that the S value showed positive slope in plot of log k' vs. volume fraction of water in mobile phase. Free energy change increases with increasing organic volume fraction. The hydrophobicity index, ${\varphi}_0$(organic volume fraction) is inversely proportional to column temperature.

The Effect of Ginseng Saponin Fraction on Several Glycolytic Enzymes of Yeast Cell (인삼 사포닌이 효모의 몇 가지 해당 효소에 미치는 영향)

  • It was attempted in this study to investigate the effect of ginseng saponin on several glycolytic enzymes of yeast cell and the following results were obtained. The amount of $CO_2$formed during the incubation of yeast cells in medium containing saponin fraction of Panax ginseng C.A. Meyer was greater than that of control cells and found that the $CO_2$ formation was greatest when the cells were grown in the medium containing 10$^{-3}$% of the saponin fraction, at which the uptake of inorganic phosphate and glucose consumption were also increased. Radioactivity study of several glycolytic intermediates of yeast cells cultured in the medium containing [U-$^{14}$C]-glucose showed that the radioactivity of fructose 6-phosphate of test cells was as much as 1.6times that of control group. On the other hand, the radioactivity of pyruvate of test cells was considerably decreased compared to control. Investigation of the effect of ginseng saponin on yeast hexokinase, phosphoglucose isomers, pyruvate kinase and perverted decarboxylase in vitro showed that the maximum activities of the above enzymes were observed when the concentration of ginseng saponin was 10-$^{-5}$% in the reaction mixture. It seemed that the ginseng saponin stimulated both glycolytic enzymes such as hexokinase, phosphoglucose isomers and perverted decarboxylase significantly.

INORGANIC SELENIUM FOR SHEEP I. SELENIUM BALANCE AND SELENIUM LEVELS IN THE DIFFERENT RUMINAL FLUID FRACTIONS

  • The effects of inorganic selenium (Se), selenate and selenite on Se balance levels in the different ruminal fluid fractions were studied using Japanese Corriedale wethers with an average body weight of 47 kg. A $3{\times}3$ Latin square design was used with three animal, three periods and three treatments. In each period, there was 7 d dietary adjustment followed by 5 d total collection of urine and feces. Ruminal fluid samples were obtained at 0, 1, 3, 5 and 7 h postprandially on the final day of the collection period. The three dietary treatments were: (1) without Se supplementation (control); (2) with Se supplement as sodium selenate; and (3) sodium selenite at a rate of 0.2 mg Se/kg dietary DM. The basal diet was timothy hay (Phleum pratense L.) fed 2% of body weight/d. Results indicated that Se balance were higher (p < 0.05) for those animals under supplementation than those animals under control. Overall data gathered showed a similar digestion balance of selenate and selenite in sheep. Inorganic Se, both selenate and selenite produced positive Se contents of the ruminal feed particles and protozoa. Bacterial Se increased (p < 0.05) on the first three hours post-prandially in Se supplemented diets. Gross ruminal fluid fraction, although there was improvement on their Se content under the supplemented diets, the changes were insignificant over the control. free inorganic Se and Se in soluble protein of the ruminal fluid were not significantly different for selenate and selenite. Most of the Se in the ruminal fluids of the animals under supplementation were insoluble, indicating the influence of rumen environments on Se bioavaliability.

Daily Concentration Measurements of Water-soluble Inorganic Ions in the Atmospheric Fine Particulate for Respiratory Deposition Region (호흡기 침착부위에 따른 미세먼지 중 수용성 이온성분의 일별 농도 측정)

  • In oder to understand the deposition possibility of water-soluble inorganic ions in the atmospheric fine particulates for the human respiratory tract, the mass size distribution of ion species was measured using an Anderson sampler in the Iksan during fall, 2004. Samples were analyzed for major water-soluble ions using Dionex DX-100 ion chromatograph. The size distribution of water-soluble inorganic ions in the atmospheric particulates appeared bimodal distribution, which were divided around $1-2{\mu}m$ into two groups. Mass site distribution of total ion in the coarse mode was found to be almost similar level during the sampling period, but fluctuations of mass size distribution in the fine mode were observed. Considering the mass size distribution of total ion concentrations for the respiratory deposition region, it was found that about 77.1% of total tons could be deposited in the alveolar region, and which dominated the daily variation of total ion concentrations. The concentration of total ions, which could be deposited in both the head region and the tracheobronchial region, was $3.95{\mu}g/m^3$, whereas that in the alveolar rerion was $13.28{\mu}g/m^3$. Dominant ions which could be deposited in the alveolar region were ${NO_3}{^-},\;{SO_4}^{2-}\;and\;{NH_4{^+}$, accounting for about 40%, 27% and 22% of the total ions, respectively. Although $K^+$ was approximately 3% of total ions, it was shown that most of this could be deposited in the alveolar region due to its high fraction of small size distribution originated from anthropogenic source of biomass burning. The presence of these ions in the fine mode may be of public health significance as they are very biologically harmful to health and have a high probability of being deposited in human lung tissue.

Contrast Effect of Citric Acid and Ethylenediaminetetraacetic Acid on Cadmium Extractability in Arable Soil

  • Chelating agents have been proposed to improve the efficiency of phytoextraction of heavy metal hyperaccumulator. However, little studies to elucidate mechanism of chelating agents to increase cadmium (Cd) extractability have been conducted. The objectives of this study were to evaluate effect of different chelating agents on Cd extractability and to determine mechanism of Cd mobilization affected by these agents. An arable soil was spiked with inorganic Cd ($CdCl_2$) to give a total Cd concentration of $20mgCdkg^{-1}$. Ethylenediaminetetraacetic acid (EDTA) and citric acid (CA) were selected and mixed with the arable soil at the rates of 0 and $5mmolkg^{-1}$. The mixture soils were incubated at $25^{\circ}C$ for 4 weeks in dark condition. Concentration of F1 Cd fractions (water soluble) significantly increased with addition of EDTA but did not changed with addition of CA. Especially; concentration of F5 Cd fractions (residual) significantly increased with addition of CA. Increase in water soluble with EDTA might be attributed to complexation of Cd and EDTA. Dissolved organic carbon concentration significantly increased with EDTA addition, but did not with CA implying that considerable amount of CA was decomposed to inorganic carbon by microorganism. Log activity of carbonate ($CO_3{^{2-}}$) which might be generated from CA increased with addition of CA. Increase in residual Cd fraction might be due to precipitation of Cd as $CdCO_3$. As a result, EDTA was effective in increasing Cd extractability, by contrast CA had significant effect in reducing Cd extractability.

Studies on the Cellulase produced by Myriococcum of albomyces (Myriococcum albomyces가 생산하는 Cellulase에 관한 연구)

  • As a study on the cellulase of Myriococcum albomyces the culture media for enzyme formation and properties of its crude preparation were investigated and the crude enzyme preparation was further fractionated. The results are summarized as follows: 1. Wheat bran solid culture produced stronger activities of cellulase than rice bran or defatted soy bean meal solid culture. 2. Shaking culture with wheat bran, rice bran or defatted soy bean meal produced higher cellulase activities than solid culture with the corresponding media. 3. The enzyme formation was higher at $45^{\circ}C$ than at $37^{\circ}C$ or $50^{\circ}C$ regardless of the kind of culture medium. 4. The formation of CMCase activity was more promoted by organic nitrogen source than inorganic nitrogen source. 5. The formation of cellulase activities were increased 1.5 to 3.0-fold by adding CMC, Avicel or cellulose powder as an inducer into 5% wheat bran basal medium. 6. Cellulase production using a tank culture procedure with addition of CMC or Avicel as an inducer was the highest at fifth day and thereafter decreased slightly. 7. The crude enzyme preparation showed pH optimum in 4.0 to 4.5, and pH stability in the range of 3.5 to 8.0. Optimum temperature for the activity was $65^{\circ}C$ which was higher than among other cellulases and it was stable at $60^{\circ}C$ for 120 minutes. 8. Dialyzed crude enzyme was activated by $Ca^{++}$ and $Mg^{++}$, but inhibited by $Hg^{++}$, $Cu^{++}$ and $Ag^{+}$. 9. Four different types of cellulase, i. e., fraction I, fraction II-a, fraction II-b, and fraction III were purified from the culture filtrate of Myriococcum albomyces through a sequence of ammonium sulfate fractionation, and elution chromatography on DEAE-Sephadex A-25, Amberlite CG-25 type 2 and hydroxyapatite columns. 10. These four cellulase fractions were showed to be homogenous by electrophoresis and ultracentrifugation and also gave a typical ultraviolet absorption spectrum of protein. 11. Four purified fraction showed different specificity toward substrates, fraction I has a stronger activity toward Avicel, cellulose powder, and gauze than that of other cellulase fractions. Fraction II-a had a powerful activity toward cellobiose but it was almost inactive agaisnt fibrous cellulose contrary to fraction I. On the contrary, the main component fraction II-b had a fairly higher activity on CMC and Avicel. Activity of fraction II-b toward cellobiose was about one-third of that of fraction II-a and activity on Avicel was lower than that of fraction I. Fraction III had a more powerful activity in decreasing viscosity of CMC. 12. Final hydrolysis products of fibrous cellulose by each fraction were cellobiose and glucose. Whereas oligosaccharides were predominant in the early stage of hydrolysis, prolonged reaction produced more glucose than cellobiose. Fraction I and fraction II-a acted synergically on Avicel. 13. Optimum pH for the activities of cellulase fraction I, fraction II-a, fraction II-b and fraction III were found to be 5.5, 5.0, 4.0 and $4.0{\sim}4.5$, respectively. These fractions were found to be stable in the range of pH $3.0{\sim}7.5$. 14. Optimum temperature for the activities of fraction I, fraction II-a, fraction II-b, and fraction III were $50^{\circ}C$, $55^{\circ}C$, $60^{\circ}C$ and $55^{\circ}C$, respectively. No less of activity was found by heating 120 minutes at $55^{\circ}C$ and fraction II-a was more stable than the others at $60^{\circ}C$. 15. Fraction I and fraction II-b were activated by $Ca^{++}$ and $Mg^{++}$ but inhibited by $Hg^{++}$ and $Ag^{+}$.
