Degradation of Fat, Oil, and Grease (FOGs) by Lipase-Producing Bacterium Pseudomonas sp. Strain D2D3

  • Biodegradation of fat, oil, and grease (FOGs) plays an Important role in wastewater management and water pollution control. However, many industrial food-processing and food restaurants generate FOG-containing waste waters for which there Is no acceptable technology for their pretreatment. To solve these problems, this study evaluated the feasibility of effective FOG-degrading microorganisms on the biodegradation of olive oil and FOG-containing wastewater. Twenty-two strains capable of degrading FOGs were isolated from five FOG-contaminated sites for the evaluation of their FOG degradation capabilities. Among twenty-two strains tested, the lipase-producing Pseudomonas sp. strain D2D3 was selected for actual FOG wastewater treatment. Its biodegradability was performed at 3$0^{\circ}C$ and pH 8. The extent of FOG removal efficiency was varied for each FOG tested, being the highest for olive oil and animal fat (94.5% and 94.4%), and the lowest for safflower oil (62%). The addition of organic nitrogen sources such as yeast extract, soytone, and peptone enhanced the removal efficiency of FOGs, but the addition of the inorganic nitrogen nutrients such as $NH_4$Cl and $(NH_4)_2SO_4$ did not increase. The $KH_2PO_4$ sources in 0.25% to 0.5% concentrations showed more than 90% degradability. As a result, the main pathway for the oxidation of fatty acids results in the removal of two carbon atoms as acetyl-CoA with each reaction sequence: $\beta$-oxidation. Its lipase activity showed 38.5 U/g DCW using the optimal media after 9 h. Real wastewater and FOGs were used for determining the removal efficiency by using Pseudomonas sp. strain D2D3 bioadditive. The degradation by Pseudomonas sp. strain D2D3 was 41% higher than that of the naturally occurring bacteria. This result indicated that the use of isolated Pseudomonas sp. strain D2D3 in a bioaugmentating grease trap or other processes might possibly be sufficient to acclimate biological processes for degrading FOGs.

Biomass Production and Cell Wall Lysis of Rhizopus oryzae (Rhizopus oryzae의 균체생산 및 세포벽제거)

  • Several kinds of organic acids, alcohols, aromatic compounds and sugars as carbon sources were tested in order to produce the cell mass of Rhizopus oryzae which is used in part of food processing or organic acid fermentation. Sodium acetate among them was good enough for carbon source as well as glucose under the concentration of one percent. All nitrogenous substances tested such as ammonium, nitrate or organic nitrogen compounds were well used by this strain of Rhizopus oryzae as nitrogen source. Ammonium sulfate among inorganic nitrogen compounds was most utilized as a nitrogen source in glucose or acetate medium. This strain did not require any growth factors such as yeast extract. The following composition of medium was therefore determined in order to produce the cell mass of Rhizopus oryzae: Na-acetate 1 %, (NH$_4$)$_2$SO$_4$ 0.2%, $K_2$HPO$_4$ 0.05%, MgSO$_4$.7$H_2O$ 0.01%, NaCl 0.01% (PH 5.5). The cell wall of mycelium grown in above medium was lysed optimally at pH 6.5 and 5$0^{\circ}C$ by the action of Strepzyme 115-5. On producing protoplast from mycelium by enzymatic action, almost all of the mycelium was damaged after 4hrs of treatment.

  • Effects of vitamins, metabolic intermediates and several inorganic mineral salts on the biomass and lipid accumulation of Mucor plumbeus were investigated after 15 days of incubation at $37^{\circ}C$ under static culture condition. The optimum concentrations of various vitamins were ${\gamma}/l$ for biotin, and 0.01 g/l for nicotinic acid, pyridoxine, thiamine and riboflavin. Among them pyridoxine was the most stimulatory. The maximum felt weight and lipid content per 50ml medium were $2.82{\pm}0.14\;g$ and 62.8%. Triglyceride content of neutral lipid produced under this condition was 64.9%. The major fatty acids were oleic acid (50.0%), linoleic (23.8%) and palmitic acid (13.9%). Malonic acid was considered not to be desirable even though it stimulated the biomass and lipid accumulation because triglyceride content was lowered considerably comparing with control. $MgSO_4{\cdot}7H_2O$ was the most stimulatory among the various magnesium salts and its optimum concentration was 5 g/l. Mucor plumbeus did not require $NaH_2PO_4$ for the stimulation of felt and lipid production. However, the addition of $MnCl_2$ at the concentration of 2 g/l was stimulatory to show $2.76{\pm}0.28\;g$ of felt/50 ml and 56.4% lipid content, and 73.9% triglyceride in the neutral lipid.

  • For the research of the natural marine antioxidant, several bacteria were isolated from the coast of jin-Hae in Korea. Among the marine bacteria studied, strain HJ-14, a gram-negative, motile, strait rod, aerobic, and $Na^{+}$ required bacterium showed high activity of 1,1-diphenyl-2-picrylhydrazyl radical scav- enging. The morphological, physiological, and biochemical characteristics of the strain HJ-14 were similar to those of the Alteromonas macleodii ATCC $27126^{T}$ . Thus, it was tentatively identified as Alteromonas sp. HJ-14. The compositions of major fatty acids in cell membrane of Alteromonas sp. HJ-14 were $C_{ 14:0}$ , $ C_{15:0}$ , $C_{16:0}$ and $C_{17:1}$ $_{w8c}$ , which also suggest that it is affiliated with Alteromonas sp. The optimum culture conditions for production of antioxidant materials with Alteromonas sp. HJ-14 were at $25$~$37^{\circ}C$ and pH 6~8. The optimum conditions for the production of antioxidant for carbon, inorganic nitrogen, and sodium chloride sources were 2.5%(w/v) dextrin, 0.5%(w/v) ammonium sulfate, and 2~6%(w/v) sodium chloride, respectively. The hydroxyl radical scavenging ability of Alteromonas sp. HJ-14 broth was 90.03%, which is higher than ascor-bic acid(83.28%) and lower than butylated hydroxyanisole(95.46%) and $\alpha$-tocopherol(97.17%).

INFLUENCE OF DIETARY PROTEIN ON THE FRACTIONATION OF SELENIUM IN THE RUMEN OF SHEEP

  • The effects of diets differing in protein content through soybean meal supplementation on ruminal fractionation of selenium (Se) were studied. A $3{\times}3$ Latin square design was used with three Japanese Corriedale wethers (45 kg average body weight), three periods, and three dietary treatment. The three dietary treatments were : Diet 1, without soybean meal supplementation (14% crude protein, CP); Diet 2, with 10% soybean meal supplementation (16.5% CP); and Diet 3, with 20% soybean meal supplementation (19% CP). All the diets had a Se supplementation in the form of sodium selenite at 0.2 mg Se/kg dietary DM. The Se supplement and the concentrate mixture were fed only in the morning before the hay was given. Daily feeding schedule for gay was set at 09:00 and 17:00 h. On the final day of collection period, ruminal fluid samples were obtained at 0.5, 2, 6, 12 and 24 h post-feeding starting at 09:00 h. Total ruminal fluid Se was markedly higher (p<0.05) in Diet 3 than those in Diets 1 and 2 at almost all sampling time except at 24 h. The proportion of Se in soluble protein to the total ruminal Se was higher (p< 0.05) in Diet 3 (40%) followed by Diet 2 (28%) and Diet 1 (21%). The proportion of free inorganic Se to the total ruminal Se was the reverse, especially after two hours where Diet 1 (p<0.05) was higher than the other diets. Bacterial Se was lower (p < 0.05) in Diet 1 than those in Diets 2 and 3 at any sampling time. The highest was observed at 2 h postprandially in all diets with a value of 421, 556, $655{\mu}g/kg$ bacterial DM for Diet 1, 2 and 3, respectively. No differences (p>0.05) were observed on ruminal pH, ammonia and total nolatile fatty acids although increasing protein supplementation tended to decline the ruminal pH and increase ruminal ammonia. This study concludes that increasing dietary protein content by soybean meal supplementation can affect the ruminal Se metabolism.

Effect of Continuous use of Inorganic Fertilizer on the Soil Organisms and Food Chain (무기질비료의 장기연용이 토양생물 및 먹이연쇄에 미치는 영향)

  • BACKGROUND: This study aimed to evaluate the combined effects of three components (NPK) of chemical fertilizers with basal application of compost on soil organisms.METHODS AND RESULTS: The soil was treated with five treatments continuously for 15 years: control, PK, NK, NP and NPK. The application of N increased plant growth or biomass, and enhanced organic matter content in the soils. Levels of microbial phospholipid fatty acids (PLFAs) in the soils did not show marked differences among the soils treated with different treatments. However, the principal component analysis showed the changes in the structure of the microbial community in the soil, depending on treatments added. Nitrogen application caused a decrease of pH and an increase of EC in the soils, and these environmental stresses appeared to offset the promoting effect of increased organic matter content on microbial abundance. The abundance of bacterivorous nematodes was the highest in the soils after treating NPK; however, the abundance of fungivorous nematodes was unaffected. There was no significant correlation between the abundances of microbial groups and their feeders. Organic matter content was significantly correlated with the abundance of nematodes in the soils.CONCLUSION: Our results showed that chemical fertilizers affect the soil food chains through both biotic and abiotic factors, and a trophic cascade in the soils may not occur in response to long-term fertilization.

Small Metal Ion Effect on The Harvest of Perilla Leaves in Aquiculture (수경재배 들깻잎의 수확량에 미치는 미량금속 이온 효과)

  • The perillas were cultivated to investigate for the small metal ion effect on the harvest of perilla loaves in aquicul-ture system in the constant flow rate. The perillas were und-ergrown at the condition of low concentration of KNO$_3$and NH$_4$ H$_2$ PO$_4$as below 270ppm and 152ppm respectively. The high concentration of the metal and non-metal ca-tions of $Ca^{2+}$, $K^+,NH^{4+},Mn^{2+},Zn^{2+},Cu^{2+},Na^+,Mo^{+6}$ are influenced to the growth of plant length of perillas at the earlier time. The low concentration of the metal cations of $Ca^{2+}, K^+, NH^{4+}, NO_3^-, H_2PO_4^-$ are influenced to lower growth of perillas. The concentration of the cations of $Ca^{2+}, K^+$and $NH_4^+$ and the anions of $NO_3^-$and $H_2PO_4^-$ are affected the growth of leaf length and width of leaf of perillas. The spectoscopic analytical results showed that the perillas were growing rapidly in the period of 6 days from June 7 to June 12 by high amount of $Mg^{2+}$ ion with accumulation inside perillas. The crude protein, the crude fat and the hydrocarbon are accumulated in the leaves of perillas by binding the inorganic with amino acids to provide the nutritions needed for growth of perillas.

  • The growth responses of Chinese Cabbage, lettuce, and spinach were evaluated to investigate the feasibility of carbonized rice-hulls as a sanitary cultivation media instead of soil. The carbonized rice-hulls were pretreated with several inorganic acid such as nitric, sulfuric, hydrochloric, and phosphoric acid for neutralization. The neutralization of the carbonated material using nitric, sulfuric, and phosphoric acid did not alter the growth responses of these vegetables compared to those of control plants grown on the normal cultivation soil. Especially, the nitric acid pretreatment resulted in the better growth of these plants than the mixture of soil and compost. This is probably due to the additional supply of the nitrate-nitrogens. However the growth responses of lettuce and spinach on the carbonated materials neutralized with hydrochloric acid were different and appeared to be having symptom which is analogous to the chloride toxicity during the late stage of growing. Although no difference was observed in chemical composition, the contents of protein-nitrogen were higher in the plants grown on the carbonated material compared to those of plant grown on the mixture of soil and compost. Based on our preliminary results, the carbonized and neutralized rice-hulls are very useful bed material for the sanitary cultivation under the controlled environmental condition.

Inhibitory Effect of Lactic Acid Bacteria and its Metabolites on the Growth of Staphylococcus aureus (젖산균과 그 대사산물이 Staphylococcus aureus의 생육에 미치는 억제효과)

  • Lactic acid bacteria(LAB) and its metabolites were tested for their inhibitory effect on the growth of Staphylococcus aureus in vitro. When S. aureus and LAB were incubated simultaneously in MRS broth, the growth of S. aureus began to be suppressed after 12 hour of incubation, completely inactivated within 24 hour of incubation by L. lactis and 48 hour by L. casei, L. acidophilus, and Sc. thermophilus. The pH values of media incubated by S. aureus and LAB were about 4.5 at 12 hour of incubation and 3.5 at 48 hour. The metabolites of the four LAB all exerted antibacterial activity on the growth of S. aureus in TS broth, but is got lost the antibacterial activity by heating ($100^{\circ}C,\;20\;min$). On treating metabolites with catalase, only L. lactis were lost its antibacterial activity. Organic acids (acetate, lactate) showed more active inhibition than inorganic acid (HCl) at pH 3.5, 4.5 but there is no significant differance at pH 5.5, 6.5.

Physicochemical Properties of Hallabong Tangor(Citrus Kiyomi ${\times}$ ponkan) Cultivated with Heating (가온재배한 한라봉 감귤의 성분분석)

  • Physicochemical properties of Hallabong Tangor(Citrus Kiyomi ${\times}$ ponkan) cultivated in Heated greenhouse were investigated. Moisture contents of Hallabong and M16A (a variant species of Hallabong) were 87.42% and 88.12% total sugar were 8.01% and 7.81% and acid content were 1.09% and 0.99% respectively. Vitamin C content of Hallabong was 72.01 mg/100 g that was higher than Citrus unshiu. Potassium content of M16A was 938.33 mg/kg, while Hallabong was 1369.33 mg/kg. The contents of inorganic element in a decreasing order were K > Ca > P > Mg > Na in Hallabong, and K > P > Ca > Mg > Na in Ml6A. Sucrose in Hallabong and M16A were 3.60% and 4.36%, respectively, which is half of total free sugars. Fructose and glucose Hallabong and M16A were 2.22% and 1.90%, 1.94% and 1.65% respectively. Citric acid in Hallabong and M16A was 82.32% and 69.88%, respectively among total organic acids. The content of malic acid was higher in M16A, compared to Citrus unshiu. Hesperidin and narirutin were identified main flavonoids.
