The purpose of this study was to evaluate the Influence of oil concentration and inoculum size on petroleum biodegradation in soil by Nocardia sp. H17-1, isolated from oil-contaminated soil. To investigate the effect of initial oil concentration on total petroleum hydrocarbon (TPH) degradation, the soil was artificially contaminated with 10, 50 or 100 g of Arabian light oil per kg of soil, respectively. After 50 days, Nocardia sp. H17-1 degraded 78,94 and 53% of the each initial TPH concentration, respectively. Also, it produced 1.35, 4.21, and 5.91 mmol of $CO_2$ per g of soil, respectively. The degradation rate constant (k) of TPH was decreased in proportion to the initial oil concentrations while $CO_2$ production was increased with the concentration. The growth of Nocardia sp. H17-1 was remarkably inhibited when it was inoculated into soil containing 100 g of oil per kg of soil. To evaluate the effect of the inoculum size, the soil was artificially contaminated with 50 g of Arabian light oil per kg of soil, and inoculated with $3${\times}$10^{6}$ , $5${\times}$10^{7}$ , $2${\times}$10^{8}$ cells per g of soil, respectively. After 50 days, the degradation of TPH was remained with similar in all treatment but degradation rate constant (k) and evolved $CO_2$ was increased with increasing the inoculum size.
Applicability of bioleaching techniques using a sulfur-oxidizing bacteria, Acidithiobacillus thiooxidans, for remediation of shooting range soil contaminated with toxic heavy metals was investigated. The effects of sulfur concentration, the amount of bacterial inoculum and operation temperature on the efficiency of heavy metal solubilization were examined as well. As sulfur concentration and the amount of bacterial inoculum increased, the solubilization efficiency slightly increased; however, significant decrease of heavy metal extraction was observed with no addition of sulfur or bacterial inoculum. Bacteria solubilized the higher amount of heavy metals at $26^{\circ}C$ than $4^{\circ}C$. Lead showed the highest removal amount from the contaminated soil but the lowest removal efficiency when compared with Zn, Cu and Cr. It was likely due to formation of insoluble $PbSO_{4(s)}$ as precipitate or colloidal suspension. Sequential extraction of the microbially treated soil revealed that the proportion of readily extractable phases of Zn, Cu and Cr increased by bacterial leaching, and thus additional treatment or optimization of operation conditions such as leaching time were required for safe reuse of the soil. Bioleaching appeared to be a useful strategy for remediation of shooting range soil contaminated with heavy metals, and various operating conditions including concentration of sulfur input, inoculum volume of bacteria, and operation temperature exerted significant influence on bioleaching efficiency.
The establishment of a laboratory assay and a greenhouse assay was conducted for evaluating the pathogenicity of Macrophomina phaseolina causing soybean charcoal rot established. In the laboratory assay, microsclerotia and hyphae were used as inoculum. In the laboratory assays using microsclerotia as an inoculum, disease incidences of M. phaseolina NSW17-108 and HSM17-034 were higher at 35℃ than 25℃. Of the two isolates NSW17-108 and HSM17-034, the disease incidence of HSM17-034 isolated from diseased sesame is higher than that of NSW17-108 isolated from diseased soybean. When the mycelia of M. phaseolina were used as an inoculum, the disease incidence of NSW17-108 and HSM17-034 at 35℃ exceeded 80% even after only 5 days of inoculation. Even at 25℃, furthermore, that of HSM17-034 exceeded 80% 5 days later. In the pathogenicity assays at a greenhouse, toothpicks where microsclerotia were produced or microsclerotia harvested from potato dextrose agar medium were used as an inoculum. In all greenhouse assays, M. phaseolina NSW17-108 and HSM17-034 showed 40-60% of disease incidences 35-65 days after inoculation with the pathogen, depending on the inoculation method. Between the two isolates, the pathogenicity of HSM17-034 was stronger than that of NSW17-108, and this result was consistent with laboratory assay results. Since the laboratory and greenhouse test methods tested in this study have different advantages and disadvantages depending on each test method, it is thought that the test method that can meet the purpose of the study should be selected and used.
Hanwoo and Jeju Black cattle (Jeju Black) are native breeds of Korean cattle. Jeju Black cattle are recognized as natural monuments and are known to exhibit slower growth rates compared to Hanwoo. While several studies have analyzed the genetic characteristics of these cattle, there has been limited research on the differences in their microbiome. In this study, rumen fluid was obtained from three Hanwoo steers and three Jeju Black steers, and three different diets (total mixed rations [TMRs] for growing, early fattening, and late fattening periods) were used as substrates for in vitro fermentation. The in vitro incubation was conducted for 3 h and 24 h following a 2 × 3 factorial arrangement. After both incubation periods, fermentation characteristics were analyzed, and ruminal microbiome analysis was performed using 16S rRNA gene sequencing, employing both QIIME2 and PICRUSt2. The results revealed significant differences in the ruminal microbiota due to the inoculum effect. At the phylum level, Patescibacteria and Synergistota were found to be enriched in the Jeju Black inoculum-treated group. Additionally, using different inocula also affected the relative abundance of major taxa, including Ruminococcus, Pseudoramibacter, Ruminococcaceae CAG-352, and the [Eubacterium] ruminantium group. These microbial differences induced by the inoculum may have originated from varying levels of domestication between the two subspecies of donor animals, which mainly influenced the fermentation and microbiome features in the early incubation stages, although this was only partially offset afterward. Furthermore, predicted commission numbers of microbial enzymes, some of which are involved in the biosynthesis of secondary metabolites, fatty acids, and alpha amylase, differed based on the inoculum effect. However, these differences may account for only a small proportion of the overall metabolic pathway. Conversely, diets were found to affect protein biosynthesis and its related metabolism, which showed differential abundance in the growing diet and were potentially linked to the growth-promoting effects in beef cattle during the growing period. In conclusion, this study demonstrated that using different inocula significantly affected in vitro fermentation characteristics and microbiome features, mainly in the early stages of incubation, with some effects persisting up to 24 h of incubation.
Korean food waste, one of the abundantly available but environmentally problematic organic wastes in Korea, was utilized as solid-substrate by fungal strain Aspergillus niger ATcC 6275 for the production of enzymemixture containing amylase, cellulase and xylanase. The enzyme mixture can be used as high value-added animal feed. Solid-state fermentation method yielded a 84-fold enhancement in xylanase activity compared with submerged fermentation method. The effect of incubation period, incubation temperature, pH of medium, moisture content, inoculum size and enrichment of the medium with nitrogen and carbon sources were observed for optimal production of these enzymes The optimal amylase activity of 33.10 U/g, cellulase activity of 24.41 U/g, xylanase activity of 328.84 U/g were obtained at 8 days incubation with 50%(w/w) soy bean flake, with incubation temperature of $25^{\circ}C$, pH of 6.38, optimal moisture content of 55% and with inoculum size of $3.8{\times}10^6$spore/g. Enzyme activities were enhanced when ImM $CaSO_4$, 2% Malt extract and 2% galactose were added as mineral, nitrogen and carbon enrichment respectively.
This study was carried out to determine factors affecting on the mass propagation of Rehmannia glutinosa seedlings in bioreactor culture. Air-lift type bioreactor was more compatible to shoot formation than stirrer type. Fifty grams(90 stem explants) of inoculum in 1.5L medium was placed into 2.5L bioreactor with aeration rate of 0.5 v.v.m., which was proper for effective shoot formation. Adding MES as pH buffer to culture medium increased the numbers of shoot formation. Adding 5g/l of anti-vitrifying agent into culture medium was highly effective for diminishing the rate of vitrification in shoots formed.
A study was undertaken to determine the thermal inactivation of Escherichia coli O157:H7 as influenced by the effects of temperature, time, suspension medium and ascorbate. Tryptic soy broth was more heat resistant than pfosphate buffer (pH 7.1), with D values of 1.52~1.68 min at 6$0^{\circ}C$ and 1.51~1.63 min at 7$0^{\circ}C$ compared with 1.52~1.65 min at 6$0^{\circ}C$ and 1.26~1.61 min at 7$0^{\circ}C$ for phosphate buffer as suspension medium. E. coli O157:H7 was completely inhibited within 30 min when small inoculum (106 CFU/$m\ell$) was heated at 7$0^{\circ}C$. When E. coli O157:H7 was preheated at 48$^{\circ}C$ for 60 min in phosphate buffer before heating, D values were 1.28~1.60 min at 6$0^{\circ}C$, and 1.13~1.56 min at 7$0^{\circ}C$, showing that preheating increases the heat resistance of the strain. Phosphate buffer containing ascorbate (0.001 M) was enhanced the thermal inactivation of the strain when inoculated as large inoculum (109 CFU/$m\ell$), while ascorbic acid was no effect at low cell concentrations (109 CFU/$m\ell$).

Mechanical transmission of barley seedlings with barley yellow mosaic virus (BaYMV) is generally inefficient and is the major constraint for testing cultivar resistance to the virus. To explore mechanical transmission, BaYMV-infected barley plants were grown at different conditions and used as inoculum sources to seedlings of susceptible barley cultivar Baegdong. Extracts prepared from BaYMV-infected Baegdong plants at 47, 53, 74, and 90 days after symptom appearance (DASA) and grown at 10 and $12^{\circ}C$ gave 10, 30, 68 and 76% infection, respectively on inoculated susceptible barley cv. Baegdong seedlings. While Jinyangbori, another susceptible cultivar obtained 95% infection rate inoculated with extracts from 90 DASA disease source and grown at $10/12^{\circ}C$. However, low infection rates were obtained when the virus sources were grown in a greenhouse at $15-18^{\circ}C$. Our results indicate that longer incubation period and lower temperature are required for virus accumulation and stability.
