Studies on the Risk Class and Morphological Characteristics of Injured Microorganisms in Wooden Cultural Properties (목조문화재 가해 미생물의 형태학적 특성 및 가해위험도 연구)

  • In the study, we confirmed morphological traits and calculated the Risk Class of 25 mould species, including Tyromyces palustris, through decomposition rate of substrate for characteristic confirmation of injured microorganisms. We received the injured microorganisms in wood and it was pure cultivated. We observed growth traits and decomposition rate of substrate. Also, it allows the calculation of Risk Class to be worried in the case of growth in wooden cultural properties. As a result, Tyromyces palustris, representative of brown-rot, has the characteristics about decomposition rate of substrate - Cellulose ; more active(+++), Lignin ; more active(+++), Xylan ; more active(+++), Starch ; active(++). We classified Tyromcyes palustris as a first Risk Class and as a second Risk Class about Schizophyllum commune et al(9 species, See table 1. We look forward to offering standard data base for evaluating Risk Class as a matter of applying preventive conservation in wooden cultural properties through our study.

Physico-Chemical and Microbiological Changes during Storage of Fresh Ginseng (수삼저장중 이화학적 및 기생물학적 변화)

  • Physical, chemical and microbiological changes were periodically studied during six-month storage of fresh ginseng under N2, CO2 gas or subatmospheric pressure condition. The results were summarized as follows. 1. The moisture contents of fresh ginseng gradually decreased during the first 2-month storage and thereafter generally reached at equillibrium. 2. There was no significant change in the reducing sugar content in 1-month storage, followed by$.$a decrease in between 2-and 3-month storage. Thereafter, the reducing sugar content increased at the end of 4-month storage. 3. The total sugar content increased significantly during the first 3-month storage. Under CO2 and Nr gas storage, the total sugar content gradually decreased after 3-month storage, while no significant change was observed in the samples stored under subatmospheric Pressure. Amylase activity gradually decreased as storage period increased 4. The content of saponin decreased as storage period increased, but ginsengoide Rf, Rd, Rc and Rb2 increased significantly in 1-month storage. 5. Regardless of storage methods, sprouting of ginseng and growth of microorganisms were inhibited in all samples during the first 4-month storage. However, growth of microorganisms was observed in the rhizome and injured areas of ginseng after 5-month storage in the N2 and CO2 gas atmosphere.

Lettuce leaves에 접종된 Foodborne Pathogen에 대한 Aerosolized Peroxyacetic Acid의 살균 효과

  • As an alternative to gaseous sanitizer which having penetrating and diffusing capacities, aerosolsanitizer’s effect on killing pathogens was investigated. To test the efficacy of aerosolized sanitizer, peroxyacetic acid was aerosolized($5.42-11.42\mum$) by nebulizer(Royal-G Enterprise, ShenZhen,China) in a model cabinet against artificially inoculated target microorganisms on lettuce. Lettuceleaves were inoculated with a cocktail of three strains each of Escherichia coli O157:H7, Listeriamonocytogenes, and Salmonella Typhimurium and treated with sanitizer aerosol for 10 min, 30 and60 min in a model aerosol cabinet at room temperature($22\pm2^\circC$). After treatment, surviving cellsincluding injured cells were enumerated on appropriate selective agar or using the over-lay agar method, respectively. Inoculated lettuce leaves exposed to antimicrobial aerosol for 10 min experienceda 0.8 log reduction in E. coli O157:H7, a 0.3 log reduction in Salmonella Typhimurium and a 2.5 logreduction in L. monocytogenes when compared to the control. After 30 min treatment, the threepathogens were reduced in number of CFU by 2.2, 3.3, and 2.7-log and after 60 min, the reductions were 3.4, 4.5, and 3.8-log, respectively. Aerosolization can be new antimicrobial agents deliverysystem in food sanitizing.

A Microbiological Study on the Frozen Beef and Pork in Seoul Area (서울시 일부지역 冷凍牛肉 및 豚肉의 細菌學的 調査硏究)

  • This study was carried out to investigate the bacterial contamination of retailed beef and pork in Seoul area. For this study 44 samples were collected at butcheries and department stores from March to April, 1987 and were examined on the following items. 1. Comparison of bacterial contamination between butcheries and department stores. 2. Degree of bacterial enumeration by repair method of injured microorganisms in the frozen beef and pork. 3. Degree of bacterial contamination in the frozen beef and pork. As the result of this study, the following conclusions were obtained. 1. Difference of bacterial contamination between butcheries and department stores was not significant. 2. Enumeration of total bacterial count by solid repair procedure resulted in about 1.5-fold increase in beef and about 1.8-fold increase in port. 3. Enumeration of total coliform count by liquid repair procedure resulted in about 32.1-fold increase in beef and about 12.0-fold in pork. 4. Salmonella species were not detected, but Staphylococcus species were detected 8 cases (18.2%) in beef, 7 cases (15.9%) in pork. 5. In total bacterial count, all beef samples were lower than 10$^6$/g, but 2 pork samples were higher than 10$^6$/g. 6. The ratio of total coliform count to total bacterial count was 0.07 in beef, 0.05 in pork by standard method and 0.48 in beef, 0.25 in pork by repair method.

Selective Sterilization of Vibro parahaemolyticus from a Bacterial Mixture by Low-Amperage Electric Current

  • The objective of this study was to investigate the possibility of using low-amperage electrical treatment (LAET) as a selective bacteriocide. Mixtures containing Escherichia coli, Staphylococcus aureus, and Vibrio parahaemolyticus were treated with different electric current intensities and for different times. The results showed that at 263 mA, treating bacteria for 100 ms eliminated all V. parahaemolyticus colonies. Although LAET reduced the populations of the three microorganisms, V. parahaemolyticus was more injured by LAET than S. aureus and E. coli when treated at the same processing conditions.
