Real-Time Force Sensing in the Envelope of Zebrafish Egg during Micropipette Penetration

  • In biological cell manipulation, manual thrust or penetration of an injection pipette into an egg is currently performed by a skilled operator, relying only on visual feedback information. Massive load of various micro injection of either genes, fluid or cells in the postgenomic era calls a more reliable and automatic micro injection system that can test hundreds of genes or cell types at a single experiment. We initiated to study cellular force sensing in zebrafish eggs as the first step for the development of a more controllable micro injection system by any inexperienced operator. Zebrafish eggs at different developmental stages were collected and an integrated biomanipulation system was employed to measure cellular force during penetrating the egg envelope, the chorion. First of all, the biomanipulation system integrated with cellular force sensing instrument is implemented to measure the penetration force of cell membranes and characterize mechanical properties of zebrafish embryo cells. Furthermore, implementation of cellular force sensing system and calibration are presented. Finally, the cellular force sensing of penetrating cell membranes at each developmental stages was experimentally performed. The results demonstrated that the biomanipulation system with force sensing capability can measure cellular force at real-time while the injection operation is undergoing. The magnitude of the measured force was in the range of several hundreds of uN. The precise real-time measurement should provide the first step forwards for the development of an automatic and reliable injection system of various materials into biological cells.

경계요소법을 이용한 사출성형금형 냉각시스템의 최적설계 (Optimum design of injection molding cooling system via boundary element method)

  • The cooling stage is the very critical and most time consuming stage of the injection molding process, thus it cleary affects both the productivity and the part quality. Even through there are several commercialized package programs available in the injection molding industry to analyze the cooling performance of the injection molding coling stage, optimization of the cooling system has npt yet been accomplished in the literature due to the difficulty in the sensitivity analysis. However, it would be greatly desirable for the mold cooling system designers to have a computer aided design system for the cooling stage. With this in mind, the present study has successfully developed an interated computer aided design system for the injection molding cooling system. The CAD system utilizes the sensitivity analysis via a Boundary Element Method, which we recently developed, and the well-known CONMIN alforuthm as an optimization technique to minimize a weighted combination (objective function) of the temperature non-uniformity over the part surface and the cooling time related to the productivity with side constranits for the design reality. In the proposed objective function , the weighting parameter between the temperature non-uniiformity abd the cooling time can be adjusted according to user's interest. In this cooling system optimization, various design variable are considered as follows : (i) (design variables related to processing conditions) inlet coolant bulk temperature and volumetric flow rate of each cooling channel, and (ii) (design variables related to mold cooling system design) radius and location of each cooling channel. For this optimum design problem, three different radius and location of each cooling channel. For this optimum design problem, three different strategies are suffested based upon the nature of design variables. Three sample problems were successfully solved to demonstrated the efficiency and the usefulness of the CAD system.

  • Frozen shoulder is known as a self-limited disease. But, its long duration and pain nature can make the patients debilitative. And most patients cannot tolerate a chronically painful extremity and are concerned about the possibility of developing permanent dysfunction. In painful phase of frozen shoulder, some aggressive mordalties as like trigger point injection or suprascapular nerve block can beneficial to: reduce discomfort and pain. In order to document clinical results, we evaluated the results of 134 frozen shoulders treated with trigger point injection and/or suprascapular nerve block at Kyungpook National University Hospital, from January 1995 to April 1997. The treatment group was divided into 3 modalities: 17 cases in trigger point injection(TPI), 39 cases in suprascapular nerve block(SSB), and 78 cases in both methods. The supportive treatment including oral medication, heat and stretching exercise was also applied. The average age at the time of diagnosis was 57 years old and average follow-up time was 18 months. The results were as follows: Average time of significant improvement in pain was 9 days. Eighty-eight percent (119 cases) was improved in pain and range of motion after injecllion treatments; 82%(14/17) with TPI, 85%(33/39) with SSB, and 92%(72/78) with both. Early improvement of paih within 1 week was 72% in the treatment-responsive group, in which TPI group has 100% response(14/14) and sse has 94% response(31/33)

WDM-PON용 주입 잠금 패브리-페롯 레이저 다이오드의 시영역 대신호 모델링 (Time-domain Large-signal Modeling of Injection-locked Fabry-Perot Laser Diode for WDM-PON)

  • WDM-PON 용 저가형 광원으로 유망한 주입 잠금 패브리-페롯 레이저 다이오드의 특성 해석을 위한 모델링 방법을 제시하였다. 해석 방법으로는 시영역 대신호 모델을 이용하였으며, 계산 결과는 기존의 실험 결과와 유사한 경향을 보임을 확인하였다. 이를 토대로 특정한 파라미터를 가지는 레이저 다이오드 구조에서의 단면 반사율에 따른 동작 특성과 디튜닝, 아이 다이어그램등을 모델링 하였다. 특히 단면 반사율의 값에 따른 부모드 억제율 특성 및 디튜닝의 영향을 살펴보았으며, 주입되는 단면의 반사율이 1% 미만으로 유지되어야 안정된 특성을 얻을 수 있음을 확인하였다. 아이 다이어그램의 경우 155 Mbps 급에서는 손쉽게 아이 열림을 얻을 수 있지만, 1.25 Gbps 급에서는 레이저 다이오드의 활성층 두께 등의 파라미터를 적정화해야 적정한 아이특성을 얻을 수 있음을 보였다.

Measurement of the Rate of Protein Synthesis in Chickens by HPLC/MS

  • The fractional synthesis rates(FSR) were measured with 2l-wk and 3l-wk-old broiler breeder pullets and hens to investigate the effect of sexual maturity on FSR. The FSR were obtained from chicken tissues and blood samples using High-Performance Liquid Chromatography/Mass Spectrometry(HPLC/MS). A L-l-13C, 15N -leucine saline solution was infused by bolus injection as a tracer into broiler breeder pullets in the experiment. A rapid HPLC/MS method was developed to measure the isotopic enrichments of leucine in plasma, tissue samples, and eggs. The enrichments of stable isotope leucine incorporated into protein and the enrichments of the stable isotope free leucine were measured in liver, breast muscle and blood samples. Two sets of experiments were conducted. In experiment one, 2l-wk-old, sexually immature broiler breeder pullets were divided into groups of three and blood samples were collected at 20 or 30 min intervals until 1.5 h from initial injection. The pullets were sacrificed in groups of three at varying time intervals for 7 h after injection. The liver, breast muscle and blood samples were removed for analysis. The FSR were estimated to be 8.7l%/day for liver, 4.06%/day for breast muscle, and 5.08%/day for blood samples in 30 minutes after injection from the enrichment ratios. In experiment two, sexually matured 3l-wk-old broiler breeder hens were assorted into groups of three and blood samples were obtained at 20 or 30 min intervals for 2 h. The FSR for blood samples were determined. The broiler breeder hens were sacrificed in groups of three at various time intervals until 7 h after injection and liver, breast muscle and blood samples were removed for analysis. The FSR were calculated to be 5.96%/day for liver. Eggs were collected from five chickens daily for 10 days after large bolus injection. The average of total enrichments of stable isotope in egg albumin was increased by 0.064% at 4 days after injection and was back to normal in 7 days.

  • Nationally, flatfish vaccination has been performed manually, and is a laborious and time-consuming procedure with low accuracy. The handling requirement also makes it prone to contamination. With a view to eliminating these drawbacks, we designed an automatic vaccine system in which the injection is delivered by a Cartesian coordinate robot guided by a vision system. The automatic vaccine injection system is driven by an injection site location algorithm that uses a template-matching technique. The proposed algorithm was designed to derive the time and possible angles of injection by comparing a search area with a template. The algorithm is able to vaccinate various sizes of flatfish, even when they are loaded at different angles. We validated the performance of the proposed algorithm by analyzing the injection error under randomly generated loading angles. The proposed algorithm allowed an injection rate of 2000 per hour on average. Vaccination of flatfish with a body length of up to 500mm was possible, even when the orientation of the fish was random. The injection errors in various sizes of flatfish were very small, ranging from 0 to 0.6mm.

  • In this work, the evaluation of swirl nozzle injector performance was conducted by investigating effective area ($A_{eff}$), injection mass ($m_{inj}$), injection rate ($Q_{inj}$), and injection delay ($t_{delay}$) under various test conditions. To achieve these, fuel injection analysis system which was composed of fuel supply system, injection system, and control system was installed. At the same time, the swirl nozzle that had 12 orifice hole with $120^{\circ}$ injection angle was used in this work. It was revealed that the difference of injection mass ($m_{inj}$) between base and swirl nozzle injector increased as the injection pressure ($P_{inj}$) and energizing duration ($t_{eng}$) decreased under the same test conditions. The maximum injection rate ($Q_{inj}$) of swirl nozzle injector was higher than base nozzle injector about 2~5%. The injection performance of swirl nozzle was better than base nozzle at low injection pressure ($P_{inj}$) and short energizing duration ($t_{eng}$) conditions.

TPE를 적용한 자동차 윈도우 모터커버의 개발 (Development of Automobile Windows Motor Cover by Thermoplastic Elastomer(TPE))

  • It was attempted to develop an auto part by over molding injection mold that produces precision products in high productivity with use of an eco-friendly TPE substitute material for NBR. NBR is currently used in motor gear cover, one of the key parts in motor module for auto doors. Gear cover is composed of plastics and rubber mostly today, which requires a two (2) step process for production using two presses of different types. A hot press is used at this time for forming the rubber, which has drawback of requiring a rather long forming time of 400 seconds for one forming process. Even though this difficulty is overcome by reducing production time through employment of multi-cavity molds, time for forming process must be shortened for improvement of the productivity eventually, and the existing method of insert injection for products that have been formed with plastic material must be outgrown. In this point of view, over molding injection using TPE has a big advantage. Forming time is shortened to 54 seconds, and working the two (2) processes in series by one (1) press could solve the durability problem caused by deflection of the plastics, not to mention shortening the process time. Enhancement of productivity by almost 80% and improvement in the accuracy of the product could thus be achieved.

PET 병용 프리폼 사출성형에서 잔류응력과 수축 최소화를 위한 성형조건의 연구 (Investigation of the Molding Conditions to Minimize Residual Stress and Shrinkage in Injection Molded Preform of PET Bottle)

  • PET병은 프리폼(preform)을 사출성형한 후 이를 블로우 성형기에 이송시켜 블로잉하여 성형된다. 내열을 요구하는 병 즉, 주스나 곡류음료용 PET병은 목 부분(neck 또는 thread 부분)에 내열성을 부여하기 위해 프리폼 성형 후 블로잉하기 전에 목 부분을 결정화시킨다. 그러나 사출성형품에 존재하는 잔류응력이 결정화를 방해하기 때문에 프리폼 목 부분의 충분한 결정화를 위해 사출 후 프리폼을 열처리(annealing)한다. 이 열처리는 잔류응력을 해소시키기 위해서 수행하는데 사출 시 성형조건의 최적화를 통하여 잔류응력을 최소화한다면 열처리 시간을 단축시킬 수 있다. 본 논문에서는 사출 시 프리폼에 형성되는 잔류응력을 최소화하고 치수정밀도를 유지하기 위한 연구로 CAE 해석을 통하여 최적 사출조건을 제시하였다. 성형조건별 잔류응력 및 수축률의 변화를 관찰하고 이를 최소화시키는 성형조건을 찾기 위해 실험계획법을 적용하였다. 사출온도, 보압크기, 그리고 사출시간을 인자로 하여 최적의 성형조건을 결정하였다. 잔류응력에 영향을 주는 인자는 사출온도와 사출시간 순으로 나타났고 수축률에 영향을 주는 인자는 사출온도로 나타났다. 본 연구에서 결정한 최적 조건에서 최대 잔류응력, 잔류응력의 분포, 그리고 수축률이 기존 조건에 비해 각각 22%, 40%, 그리고 25% 감소하였다.

모니터링과 제어를 위한 사출성형 파라미터 인터페이스에 관한 연구 (A Study on the Interface of Injection Molding Parameter for Monitoring and Control)

  • Recently, monitoring systems, such as POP, take a core role in scheduling or planning of manufacturing facilities for production, maintenance, and so on. Such monitoring systems require functionalities for real-time parameter monitoring and controlling to maximize efficiency of facilities. However, vendors usually do not provide internal communication protocols or interface to access the machine controller. Therefore, the values of parameters related to machine operations and controls cannot be easily accessed from external devices. In this paper, we propose an interface methodology for a real-time monitoring and controlling of injection molding machine parameters such as user input parameters, embedded sensor data and injection molding status information.
