심부 화강암반의 수리특성 평가를 위한 현장수리시험 비교 및 해석 연구 (Comparison and Analysis of Field Hydraulic Tests to Evaluate Hydraulic Characteristics in Deep Granite Rockmass)

  • 고준위방사성폐기물 처분부지 선정에 있어 부지의 수리지질학적 연구는 매우 중요하며, 수리전도도와 저류계수 등이 대표적 수리지질학적 인자이다. 본 연구는 원주지역 심부 화강암반을 대상으로 두 가지 다른 종류의 현장수리시험 장비와 방법으로 획득한 수리전도도를 비교 분석하여 심부 화강암반의 수리특성을 파악하였다. 하나는 지하수 관정 자동 주입 시스템 장비를 활용하여 최대 심도 602.0 m에 대해 루전시험, 정압주입시험, 슬러그시험을 수행하였고, 산정된 수리전도도는1.26E-9 ~ 4.16E-8 m/s의 범위를 보였다. 전체 심도에서 수리전도도의 최대와 최소 차이는 약 33배로 나타났고, 동일 시험 구간에서 시험 방법이나 해석방법에 의한 차이는 1.13 ~ 8.25배로 나타났다. 다른 하나는 대심도 수리특성 조사 시스템을 활용하여 최대 심도 705.1 m에 대해 정압주입시험, 펄스시험을 수행하여 산정된 수리전도도는 1.60E-10 ~ 2.05E-8 m/s으로 최대와 최소 차이는 약 130배 정도로 나타났다. 정압주입시험에서 해석방법에 따른 차이는 1.02 ~ 2.8 배로 나타났다. 두 시험 장치와 방법에서 산정한 수리전도도는 대체로 E-9와 E-8 m/s로 유사한 범위를 보였으며, 심도에 따른 뚜렷한 경향은 관찰되지 않았다. 현장수리시험이 수행된 원주지역 화강암반은 낮은 또는 매우 낮은 암반투수성을 보임을 알 수 있었고, 적용된 시험 장치와 시험 방법에 따라 측정할 수 있는 수리전도도 범위나 적용 심도 등의 차이가 존재하나 대체로 신뢰할 수 있는 결과를 제시한 것으로 판단된다.

K-1 기지 주변 지질 구조와 지하수위 변동 특성 (Geological Structures and Their Relation to Groundwater System around K-1 Oil Stockpile)

  • K-1 유류비축기지는 인위적 지하 공동을 이용하기 때문에 주변 지하수계의 안정이 중요하다. 기지 주변의 지하수계 특성을 이해하기 위해 광역적 지하수 유동특성, 기지내 공간적 지하수위 분포 양상을 파악하였으며, 지하수 유동에 영향을 미치는 지질 요소를 검토하였다. 연구지역에서 지히수 유동에 크게 영향을 미칠 수 있는 지질구조적 특성은 2가지로서, 편마암-화강암 관입경계를 따라 재활성된 부분과 산성 암맥을 따라 발달한 단열 및 단층이다. 기지 내 4개 관측공들의 수위변동 양상을 검토한 결과, 대부분 강우와 지표수 유입 혹은 공동내 주입수에 의해 지하수위 변동이 영향을 받고 있는 것으로 나타났다. 이 연구에서는 기지 남쪽에 위치한 관정으로부터의 양수가 기지의 지하수위 변동에 미치는 영향에 대해 검토하였다. 산성 암맥을 따라 기지로부터 1.2km 남쪽에 위치한 지점에서의 양수작업은 관측 당시에 기지내 지하수위 변동에 영향을 미치지 않았으나, 모델링 결과에 의하면 $250\;m^3$/day 이상의 양수량으로 지속적인 양수작업이 진행되는 경우 지하수 시스템 안정을 위해 주의가 요구된다.

Antihepatotoxic effect of Heat-treated Allium victorialis var. platyphyllum in $CCl_4-induced$ Rats and the Gas Chromatographic Analysis of Volatile Sulfur Substances

  • The ethanolic extracts of the leaves and bulbs of Allium victorialis var. platyphyllum (Liliaceae) collected from Daegwallyoung (D) and Ullung Island (U) in Korea were obtained using three different extracting methods. The first extracts, DL-1 DB-1, UL-1 and UB-1, were obtained from leaves (L) and bulbs (B) dried at $90^{\circ}C$, respectively, and the second extracts, DL-2, DB-2, UL-2 and UB-2, were obtained by extracting the leaves and bulbs of fresh plant parts. The third extracts DL-3, DB-3, UL-3 and UB-3 were obtained by incubating leaves and bulbs at $36^{\circ}C$. The six extracts obtained from A. victorialis var. platyphyllum at Daegwanllyoung (cultivated site) were orally administered to examine for a possible antihepatotoxic effect in $CCl_4-induced$ rats. DL-1 exhibited the most pronounced effect. The extracts inhibited serum ALT, AST, SDH, ${\gamma}-GT$, ALP and LDH activities elevated by $CCl_4$ injection and attenuated decreased glutathione S-transferase, glutatione reductase and ${\gamma}-glutamylcysteine$ synthetase activities and a decreased hepatic glutathione. However, the extracts obtained from Ullung Is. (native site) were less active than the extracts from Daegwallyoung, suggesting that A. victorialis var. platyphyllum from the cultivated site is more useful for functional food than of native site. These results also suggest that the antihepatotoxic effect is due to a higher content of hepatic glutathione. Gas chromatography of the twelve extracts showed significantly different sulfides, disulfides or trisulfides contents belonging to volatile sulfur substances (VSS). Nine components were identified on the basis of their mass spectra, namely, dimethyl disulfide, dimethyl trisulfide, diallyl disulfide, dipropyl disulfide, allyl methyl sulfide, allyl methyl trisulfide, 2-vinyl-4H-1,3-dithiin, 3,4-dihydro-3-vinyl-1,2-dithiin, and allithiamine. Extract DL-1 had the highest VSS content. Dried plant materials contained larger amounts of the VSSs than other extracts, and the leaves contained larger amount than the bulbs. These results suggest that heat treatment increases the antiheaptotoxic ability of A. victorialis var. platyphyllum by increasing the proportion of VSSs.

해성점토층에서 SRC 지반보강에 관한 현장적용성 평가 (Estimation of Application on the Site of SRC Method for the Ground Reinforcement in Marine Clay)

  • 현재 서해안 지역에는 대규모의 항만공사 및 간척사업 등 대규모의 투자와 개발이 집중되고 있는 실정에 있어 연약지반 처리라는 문제점이 큰 이슈가 되고 있다. 그리고 연약지반 보강 시 지반보강을 위한 공법은 사용목적과 지반조건 및 시공여건, 공사비 등을 고려하여 주입공법을 선정해야 하며, 고압분사주입공법 중 SRC공법은 강제 압송 충전시키는 방법으로서 지반의 특성과 무관하게 균질한 강도를 가지는 개량체 조성이 가능하며, Slime 발생량이 적어 환경에 대한 영향을 적게 받는 이점을 가진다. 이에 본 연구에서는 경기도 화성시 전곡리 주변 조석 차가 비교적 큰 서해안 지역의 해성점토층 지반을 대상으로 하여 SRC공법을 시공하여 지반보강 및 현장적용성에 대해 평가하고자 한다. SRC공법 현장적용 결과, 시추작업에 의해 측정된 일축압축강도시험 결과 재령 28일 강도 값이 $14,700{\sim}31,800kN/m^2$으로 $5,333kN/m^2$ 이상의 설계기준을 모두 만족하였다. 따라서 서해안 해성점토층 지반에 SRC공법을 시공하여 지반보강에 관한 현장적용성의 평가 결과 우수한 강도 값을 나타내었으며, 이는 곧 내구성 또한 뛰어난 것으로 입증된다고 사료된다.

MTBE(Methyl Tertiary Butyl Ether)의 토양내 이동특성에 관한 연구 (A Study on Transport Characteristics of MTBE(Methyl Tertiary Butyl Ether) in Soil)

  • 본 연구에서는 주유소 등의 지하 유류저장탱크나 파이프의 제한된 수명으로 인해 발생할 수 있는 가솔린의 누출에 의해 MTBE로 토양이 오염되었을 경우를 가정하여, 칼럼실험을 수행하고, CXTFIT 기법을 이용하여 토양 내 MTBE의 이동특성을 살펴 보았다. 칼럼실험에서는 토성, 수분함량, 유기물함량, 주입유속을 달리하여, 주입액과 유출액의 MTBE의 농도 측정값을 비교하고, CXTFIT기법을 이용하여 two-site 비평형 흡착모델에 사용된 매개변수(D, R, $\beta$, $\omega$)를 구하였다. 이들 매개변수와 파과곡선을 이용하여 MTBE의 토양 내 이동특성을 살펴보았다. 토양 내 미세입자와 유기물함량이 많을수록 이류에 의한 영향이 감소하는 것으로 나타났으며, 수분함량과 유속의 증가는 이류에 의한 MTBE의 이동을 더욱 가속시키는 것으로 나타났다.

Approaches to Improving Production Efficiencies of Transgenic Animals

  • Transgenic animals are very useful for scientific, pharmaceutical, and agricultural purposes. In livestock, transgenic technology has been used forthe genetic alteration of farm animals, the production of human proteins inlarge quantities in the milk of transgenic farm animals, and the generation of animals with organs suitable for xenotransplantation. To date, the transfer of foreign genes into farm animals has been performed mainly by microinjection of DNA into the pronuclei of fertilized eggs. However, the overall success rate of transgenic animals in livestock so far has been disappointingly low, eg., the efficiency is 0∼5% in swine, and less than 1% in sheep and cattle, compared with the rate in mice where 5% microinjected develop into transgenic animals. Recently, McGreath et al. (2000) have succeeded in producing the gene targeted sheep by the use of nuclear transfer from cultured somatic cells transfected with a foreign gene in vitro. However, we may need plenty of time until currently employ this method for gene transfer to farm animals. We have been studying to exploit the method for improving production efficiencies of transgenic animals with emphasis of its application to farm animals. The present paper describes three approaches that we have made in our laboratory to improve production efficiencies of transgenic animals, based on the DNA microinjection method. 1. Co-injection of restriction enzyme with foreign DNA into the pronucleus for elevating production efficiencies of transgenic animals. 2. Efficient selection of transgenic mouse embryos using EGFP as a marker gene. 3. Phenotypes of tansgenic mice expressing WAP/hGH-CAG/EGFP fusion gene produced by selecting transgenic embryos. 4. Efficient site-specific integration of the transgene targeting an endogenous lox like site in early mouse embryos.

대수층 함양관리에 있어서 지질매질에 의한 비소 자연저감 가능성 평가 (Assessment of Potential Natural Attenuation of Arsenic by Geological Media During Managed Aquifer Recharge)

  • Managed aquifer recharge (MAR) is a promising water management strategy for securing stable water resources to overcome water shortage and water quality deterioration caused by global environmental changes. A MAR demonstration site was selected at Imgok-ri, Sangju-si, Korea, based on screening for the frequency of drought events and local water supply situations. The abundant groundwater discharging from a nearby abandoned coal mine is one of the potential recharge water sources for the MAR implementation. However, it has elevated levels of arsenic (~12 ㎍/L). In this study, the potential of the natural attenuation of arsenic by the field geological media was investigated using batch and column experiments. The adsorption and desorption parameters were obtained for two drill core samples (GM1; 21.8~22.8 m and GM2; 26.0~27.8 m depth) recovered from the potentially water-conducting fracture-zones in the injection well. The effluent arsenic concentrations were monitored during the continuous flow of the mine drainage water through the columns packed with the core samples. GM2 removed about 60% of arsenic in the influent (0.1 mg-As/L) while GM1 removed about 20%. The results suggest that natural attenuation is an acitive process occurring during the MAR operation, potentially lowering the arsenic level in the mine drainage water below the regulatory standard for drinking water. This study hence demonstrates that using the mine drainage water as the recharge water source is a viable option at the MAR demonstration site.

활성슬러지 반응탱크의 풍량제어지표인 NADH에 관한 연구 (Study on NADH which is the Air Volume Sensor in the Activated Sludge Reaction Tank)

  • Domestic sewerage treatment plant is operated by activated sludge method and its modified method by using microorganism. In most cases, a method of using microorganism is directly controlled by the operator based on individual judgment through factors of DO, pH and ORP. In addition, under aerobic condition in bioreactor, energy consumption including excessive air injection is learned to be somewhat plenty. In order to solve this problem, in most of the process, improvement of internal recycling and activated environmental factor of microorganism were researched extensively. However, as factors are changed depending on various conditions, it is not sufficient as an indicator of judgment. As such, a research on operation of bioreactor that measures metabolic change in short time by directly measuring activated condition of microorganism using NADH fluorometer is required in reality.It is considered that the method like this could supplement problem of energy consumption being occurred in the existing treatment method and operational optimization of bioreactor would be enabled by controlling optimal air volume. Therefore, in this study, in order to obtain optimal operational indicator of bioreactor, proper air volume control test was performed and through batch test and site evaluation, possibility of NADH sensor being utilized as operational control indicator of bioreactor is intended to be analyzed. In order to compare with measured value, DO, ORP that are operational control indicator of existing bioreactor were used. As MLSS concentration was increased through batch test, NADH value was increased and site evaluation also showed similar tendency to batch test. Resultantly, it could be confirmed that changing level of NADH fluorometer was a sensor that could measure bioreactor condition effectively and optimized scale of bioreactor is considered to be utilized.

Targeted Base Editing via RNA-Guided Cytidine Deaminases in Xenopus laevis Embryos

  • Genome editing using programmable nucleases such as CRISPR/Cas9 or Cpf1 has emerged as powerful tools for gene knock-out or knock-in in various organisms. While most genetic diseases are caused by point mutations, these genome-editing approaches are inefficient in inducing single-nucleotide substitutions. Recently, Cas9-linked cytidine deaminases, named base editors (BEs), have been shown to convert cytidine to uridine efficiently, leading to targeted single-base pair substitutions in human cells and organisms. Here, we first report on the generation of Xenopus laevis mutants with targeted single-base pair substitutions using this RNA-guided programmable deaminase. Injection of base editor 3 (BE3) ribonucleoprotein targeting the tyrosinase (tyr) gene in early embryos can induce site-specific base conversions with the rates of up to 20.5%, resulting in oculocutaneous albinism phenotypes without off-target mutations. We further test this base-editing system by targeting the tp53 gene with the result that the expected single-base pair substitutions are observed at the target site. Collectively, these data establish that the programmable deaminases are efficient tools for creating targeted point mutations for human disease modeling in Xenopus.

A Simple, Reliable, and Inexpensive Intraoperative External Expansion System for Enhanced Autologous Structural Fat Grafting

  • External volume expansion of the recipient site by suction has been proposed as a way of improving fat graft survival. The objective of this study was to present an innovative and simple intraoperative external expansion system to enhance small-volume autologous fat grafting (40-80 mL) and to discuss its background and its mechanism of action. In this system, expansion is performed using a complete vacuum delivery system known as the Kiwi VAC-6000M with a PalmPump (Clinical Innovations). The recipient site is rapidly expanded intraoperatively 10 times for 30 seconds each with a negative pressure of up to 550 mm Hg before autologous fat injection. During this repetitive stimulation, the tissues become grossly expanded, developing macroscopic swelling that regresses slowly over the course of hours following the cessation of the stimulus. The system sets various mechanisms in motion, including scar release, mechanical stimulation, edema, ischemia, and inflammation, which provide an environment conducive for cell proliferation and angiogenesis. In order to maintain the graft construct in its expansive state, all patients are encouraged postoperatively to use the Kiwi three times daily for one minute per session over the course of three days. The handling of this system is simple for both the patients and the surgeon. Satisfactory clinical outcomes have been achieved without significant complications.
