Effect of $SO_2$ on Leaf Tip Blight of Ginkgo biloba caused by Coniothyrium sp. (은행나무의 잎끝마름병에 미치는 $SO_2$ 의 영향)

  • To examine the relationship between air pollution and occurrence of disease in plants, we investigated the alteration of physiology and biochemistry of Ginkgo biloba by inoculating with Coniothyrium sp. and fumigating with 0.1 ${\mu}l/l\;SO_{2-}$ Visual damage did not appear but photosynthesis, $CO_2$ use efficiency(CUE) and water use efficiency(WUE) were reduced when G. biloba was exposed to $SO_{2-}$for 7days (8 hours a day). When inoculated with Coniothyrium sp., the seventies of damage in G. biloba showed a threefold difference between $SO_{2-}$ treatment (SI) and $SO_{2-}$ free treatment (SFI) at day 42 from initial inoculation. Little difference was observed in sugar contents that may be used pathogens feed, among control, SFI and SI. In spite of the reduction in photosynthetic rate, sugar contents and CUE were maintained. WUE was enhanced 13% more at SI than SFI. The $CO_2$fixation boosted because of enhanced WUE, and thus sugar synthesis was not affected. In addition, sugar transport seems to be retarded for some internal alteration. Consequently, the severity of SI was more serious than that of SFI because Coniothyrium sp. easily invades into the physical texture of G. biloba weakened by $SO_2$ fumigation and because sugar was accumulated in leaves of G. biloba.

Quality Characteristics of Functional Fermented Sausages Added with Encapsulated Probiotic Bifidobacterium longum KACC 91563

  • The present study aimed at evaluating the utilization possibility of encapsulated probiotic Bifidobacterium longum for production of functional fermented sausages. The B. longum isolated from the feces samples of healthy Korean infants encapsulated with glycerol as a cryprotectant was used for fermented sausages production as a functional bacterial ingredient, and its effect was also compared with those inoculated with commercial starter culture (CSC). Results showed that most inoculated encapsulated B. longum (initial count, 5.88 Log CFU/g) could survive after 4 days fermentation (5.40 Log CFU/g), and approximately a half (2.83 Log CFU/g) of them survived in the products after 22 days of ripening. The products inoculated with encapsulated B. longum presented the lowest lipid oxidation level, while had higher total unsaturated fatty acid content and more desirable n-6/n-3 fatty acids than those inoculated with CSC or non-inoculated control. Moreover, the odor and taste scores in the samples made with B. longum were comparable to those in the treatment with CSC. The inoculation with the B. longum had no effects on the biogenic amine contents as well as did not cause defects in color or texture of the final products. Thus, the encapsulation could preserve the probiotic B. longum in the meat mixture, and the encapsulated B. longum could be used as a functional ingredient for production of healthier fermented meat products.

Inhibition of Bacillus cereus in Doenjang Fermented with Multiple Starters Showing Inhibitory Activity against Pathogens (유해균 억제능을 지닌 복합종균을 접종하여 제조한 된장에서 Bacillus cereus 억제)

  • Doenjang samples were prepared by inoculation of multiple starters consisting of two Bacillus spp., one yeast, and one fungus. Doenjang A was fermented with Bacillus amyloliquefaciens EMD17, B. amyloliquefaciens MJ1-4, Pichia farinosa SY80, and Rhizopus oryzae. Doenjang B and C were fermented with the same yeast and fungus but different Bacillus strains; namely, B. amyloliquefaciens EMD17 and B. subtilis CH3-5 for doenjang B, and B. amyloliquefaciens MJ1-4 and B. subtilis CH3-5 for doenjang C. Doenjang D was fermented with microorganisms present in rice straw (control). The doenjang samples were spiked with B. cereus ATCC14579 at two different levels, 104 CFU/g doenjang (I) and 107 CFU/g doenjang (II). All eight doenjang samples were fermented for 70 days at 25℃. Growth of B. cereus was inhibited in doenjang A, B, and C, with the bacterial cell count after 70 days being less than the initial 104 CFU/g added, whereas B. cereus was not inhibited in doenjang D. Doenjang B showed the strongest inhibitory activity against B. cereus, with a cell count of less than 103 CFU/g after 42 days, even when B. cereus was initially added at 107 CFU/g. Some properties of the doenjang samples, such as pH, TA, and amino-type nitrogen content, were similar to those of doenjang fermented with starters only. The results indicate that carefully selected starters can effectively prevent the growth of B. cereus during doenjang fermentation.

Comparative Study on the Characteristics of Microalgae as Standard Species for Marine Ecotoxicity Tests (Skeletonema sp., Dunaliella tertiolecta) (해양생태독성시험 표준생물로서 미세조류의 특성 비교 연구(Skeletonema sp., Dunaliella tertiolecta))

  • To understand the ecotoxicological differences between representative Skeletonema sp. and Dunaliella tertiolecta, both producers as international standard test species for marine ecotoxicity testing, we compared each standard test method, and comparatively analyzed the suitability of the species for environmental assessment and their sensitivity to various test substances. Although most of the test conditions were the same in each method, there were differences in limitation of pH changing and the initial inoculation density in the validation criteria, which is supposed to originate from the low growth rate of D. tertiolecta. In terms of suitability, both species showed consistency in test performance by repeatedly meeting the validation criteria required by the standard test methods. The salinity ranges available for testing were 20 and 10 psu for Skeletonema sp. and D. tertioelecta, respectively. Finally, regarding sensitivity, the toxicity sensitivity of Skeletonema sp. was relatively higher than that of D. tertiolecta for the reference toxicant, actual polluted water discharged (ballast water), and other chemicals. This implies that using at least two species of microalgae from different classification groups could help increase the reliability and objectivity of test results in the performance of marine ecotoxicity tests using producers.

The Growth of Vibrio vulnificus in Meat Homogenates of Fish and Shellfish (어패육에서의 Vibrio vulnificus의 증식에 관한 연구)

  • The change of cell counts of Vibrio vulnificus in meat homogenates of fish and shellfish by the storage time and temperature was examined to get basic information for precautionary steps against septicemia from slices of raw fish (sashimi). Therefore, we inoculated raw and cooked meat homogenates of fish and shellfish with Vibrio vulificus M-8 (isolated from shellfish ) and stored them at $-20^{\circ}C,\;4^{\circ}C\;and\;30^{\circ}C$ for 72 hours. Vibrio vulnificus M-8 was not detected in 32 hours when it was frozen and stored at $-20^{\circ}C$ after inoculating them into phosphate buffer solution at concentration of $10^5\;cell/ml$, while the existance of Vibrio vulnificus was identified after 72 hours of storage at the same temperature in case of inoculation into the meat homogenate of yellow tail. The cell count of Vibrio vulnificus was decreased as about $20\%$ of initial count after 2 hours storage at $4^{\circ}C$ in phosphate buffer solution with fish and shellfish homgenates. From the experimental results it was recognized that Vibrio vulnificus was labile to the cold stress. In comparison to the growth of growth of Vibrio M-8 at $30^{\circ}C$ in the raw and cooked meat of the yellow tail(Seriola guingueradita), snapper(Chrysophrys major), ark shell(Anadra brouhgtonii), and oyster(Crassostrea gigas), the raw meat homogenates were more excellent than the cooked ones though all fish and shellfish meat homogenates were proves to be good for the growth of the microbe.

Large-scale Culture of Plant Cell and Tissue by Bioreactor System

  • Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

Control Efficacy of Entomopathogenic Microagent against Spodoptera exigua on Organic Chinese Cabbage (유기재배 배추포장 내 파밤나방에 대한 곤충병원성 미생물의 방제 효과)

  • This study was carried out to identify the control effect of entomopathogenic microagent against Spodoptera exigua on organic chinese cabbage. In laboratory condition, insecticidal activity of 4 commercial BT pesticides against S. exigua were lower than 10% against second instar S. exigua. The insecticidal activity of entomopathogenic nematode were 33.3%, 83.3% and 100% at the concentration of $1{\times}10^2$, $3{\times}10^2$, $1{\times}10^3nematodes/ml$, respectively. Mixture of BT and nematode showed growth inhibition against S. exigua larvae. S. exigua nucleopolyhedrovirus (SeNPV) of $10^5PIBs/ml$ showed more than 70% insecticial activity. The yield of SeNPV was increased as in higher initial inoculation concentration of NPV, food supply, and growth temperature increased. In greenhouse experiment, the control value of BT and nematode mixture treatment was higher than BT and nematode treatment alone against S. exigua. In treatment of $10^7PIBs/ml$ of SeNPV, S. exigua was controlled completely. In farm condition, mixture of microbial agent and organic agricultural material showed higher control value against lepidopteran pest including S. exigua than BT single treatment.

Assessment of New Algicide Thiazolidinedione (TD49) for the Control of Marine Red Tide Organisms (해양적조생물제어를 위한 살조물질 Thiazolidinedione 유도체(TD49) 평가)

  • Worldwide development of harmful algal blooms causes serious problem for public health and fisheries industries. To evaluate the algicidal impact on the harmful algae bloom species in aquatic ecosystems of coast, a new algicide thiazolidinedione derivative (TD49) were tentatively examined in the growth stages (i.e., lag, logarithmic and stationary phase) of rapidophyceae $Heterosigma$ $akashiwo$, $Chattonella$ $marina$ and $Chattonella$ sp..Three strains could easily destroy in the lag phase due to relatively weak cell walls than those of the logarithmic and stationary phase. It is thought that inoculation of TD49 substances into initial or developmental natural blooms with a threshold concentration ($2{\mu}M$) can maximize the algicidal activity. Also, bio-chemical assays revealed that the algicidal substances from all culture strains were likely to be extracellular substances because those cells have easily destroyed in cell walls. On the other hand, natural zooplankton communities were influenced within the exposure experiments of $2{\mu}M$, which is showed the maximum algcidal activity of tested organisms. These results indicate that although the TD49 substance is potential agents for the control of $H.$ $akashiwo$, $C.$ $marina$ and $Chattonella$ sp. in the enclosed eutrophic bay and coastal water, more detailed research of acute toxicity effect on high trophic organism in marine ecosystems need to be conducted.

Growth Characteristics and Optimal Culture Conditions of Bacterial Strains Degrading Ethylene Glycol and Terephthalic Acid in Polyester Weight Loss Wastewater (Polyester 감량폐수 중에 존재하는 Ethylene Glycol과 Terephthalic Acid를 분해하는 Bacteria 균주들의 성장특성과 최적 배양조건)

  • Strains degrading ethylene glycol(EG) and terephthalic acid(TPA) were isolated from water systems, and identified as Pseudomonas sp. They were named as Pseudomonas sp. EAW for EG and as Pseudomonas sp. TS2 for TPA. The optimal culture conditions of temperature, pH and nitrogen source were found to be $35^{\circ}C$, 7.5 and ammonium sulfate, respectively. The growth of strains and removal efficiency was slightly promoted by trace elements such as niacin and biotin in case of EG, and by trace elements such as $Na_2MoO_4{\cdot}2H_2O$ and thiamin i case of TPA. With increasing inoculation sloe for batch culture, the removal efficiency of EG by the strain EAW was conspicuously increased, while the removal efficiency of TPA by the strain TS2 was not changed as much as that of EG. The growth rate of the strain EAW was much more decreased than that of the strain TS2 in the enrichment medium, as the frequency of repeated-batch culture in the rich-medium increased. in case of real wastewater, growth rate and removal efficiencies of EG and TPA were lower than those in the enrichment medium. $COD_{Mn}\;and\;COD_{Cr}$ removal efficiencies after 48 hrs batch culture in real wastewater were 89% and 93%, respectively. The specific growth rate was inhibited when the initial concentration of EG or TPA was more than 25g/L.

The Optimal Condition and Enzyme Activity of Entomopathogenic Fungus Beauveria bassiana Using Extracted Rice Bran (미강추출물을 이용한 곤충병원성 곰팡이 Beauveria bassiana의 최적 배양조건 및 효소활성)

  • The greenhouse whitefly, Bemisia tabaci, is considered one of the most destructive pests of crops. In this study, we aimed to determine the optimal liquid culture conditions in shake flasks for maximal sporulation of Beauveria bassiana M130 using rice bran. The optimal initial pH for the spore production of B. bassiana using extracted rice bran medium was 5.2 and $28^{\circ}C$. The screening in shake flasks of carbon and nitrogen sources resulted in the identification of an optimal medium based on 0.5% $(NH_4)_2SO_4$, with extracted rice bran 8:1. Using this medium, a production level of $2.15{\times}10^9$ spores per ml was obtained after six days from culture inoculation at $28^{\circ}C$ in a rotary shaking incubator at 130 rpm. In addition, the specific activities of extracellular enzymes of chitinase and protease were $4,296{\mu}mol$ and $375{\mu}mol$, respectively. These results suggest that Beauveria bassiana M130 could be a bio-controller for the greenhouse whitefly.
