Studies on the Fermentative Production of Guanosine -5′-monophosphate by Microorganism (Part 1) Derivation of XMP-aminase Producing Mutants (미생물에 의한 5′-GMP의 생산에 관한 연구 (제1보) XMP-aminase 생산균주의 분리)

  • By the treatment of several mutagens, a number of 5'-guanylic acid producing mutants from 5'-xan-thylic acid were obtained from Brevibacterium ammoniagenes ATCC 6871. The indispensensable genetic-characters of the mutants were adenine requirement, lack of GMP-reductase and mutation to adenosine resistance from adenosine sensitiveness. Main product from 5'-xanthylic acirl by strain BA-17-2 was 5'-guanulic acid, and was isolated in a crystalline form by the use of anion exchange resin, Duolite 102 D. The isolated crystalline was identified as 5'-guanylic acid by means of paper chromatography, ultrav-iolet absorption spectra, and infrared absorption spectrum.

AKARI near-infrared spectral observations on the shocked H2 gas of a supernova remnant IC 443

  • IC 443 is famous for its interaction with nearby molecular clouds and intense H2 emission lines in infrared. Therefore, it has been studied extensively for the understanding of molecular shocks. We observed H2 mission lines toward the shock-cloud interaction regions of IC 443, known as clumps B, C, and G. The observations were performed with the InfraRed Camera (IRC) onboard a satellite AKARI over 2.5-5.0 um, where previous space observations, e.g. Infrared Space Observatory (ISO) and Spitzer, do not cover. Our AKARI observations provide spectra of sequential pure-rotational and ro-vibrational H2 emission lines. For the clumps C and G, combining with previous mid-infrared observational results, we found that the H2 level populations show a significant separation between v=0 and v=1 levels; v=1 levels are under-populated than v=0 levels, therefore, the population cannot be described by two temperature LTE model, as many people have analyzed for the shocked H2 gas. We also applied the thermal admixture model, dN(H2; T)~T^(-b) dT, with varying ortho-to-para ratios according to the temperature, to describe the level population, and obtained plausible ranges of the H2 gas density and power-law index b.

Shocked $H_2$ Gas with Non-equilibrium Ortho-to-Para Ratios Observed from Two Supernova Remnants IC 443 and HB 21

  • We present the near-infrared spectra (2.5-5.0 um) of shocked $H_2$ gas, observed with the InfraRed Camera onboard the satellite AKARI. Two supernova remnants, IC 443 and HB 21, were observed, and they all showed the ortho-to-para ratios (OPRs) of less than 3.0: 2.1-2.2 for IC 443 and 1.6-1.8 for HB 21. These non-equilibrium OPRs are first reported at E(v,J) > 7000 K, as far as we are aware of. Based on our previous study, we try to interpret that the non-equilibrium OPRs originate from dissociative J-shocks. Dissociative J-shocks mainly generate infrared H2 emissions from their $H_2$ reformation zone, and the OPR of 3.0 are expected for the reformed $H_2$ from the theoretical study. This is contradictory to our observational results. We propose other possible origins of the non-equilibrium OPRs, such as, abnormal $H_2$ reformation, partially dissociative J-shocks, etc.

Lithium intercalation into a plasma-enhanced-chemical-vapour-deposited carbon film electrode

  • Electrochemical lithium intercalation into a PECVD (plasma enhanced chemical vapour deposited) carbon film electrode was investigated in 1 M $LiPF_6-EC$ (ethylene carbonate) and DEC (diethyl carbonate) solution during lithium intercalation and deintercalation, by using cyclic voltammetry supplemented with ac-impedance spectroscopy. The size of the graphitic crystallite in the a- and c-axis directions obtained from the carbon film electrode was much smaller than those of the graphite one, indicating less-developed crystalline structure with hydrogen bonded to carbon, from the results of AES (Auger electron spectroscopy), powder XRD (X-ray diffraction) method, and FTIR(Fourier transform infra-red) spectroscopy. It was shown from the cyclic voltammograms and ac-impedance spectra of carbon film electrode that a threshold overpotential was needed to overcome an activation barrier to entrance of lithium into the carbon film electrode, such as the poor crystalline structure of the carbon film electrode showing disordered carbon and the presence of residual hydrogen in its structure. The experimental results were discussed in terms of the effect of host carbon structure on the lithium intercalation capability.

Fabrication of CNT/PVDF Composite Film and Its Electrical Properties (CNT/PVDF 압전 복합막의 제작과 전기적 특성)

  • The carbon nanotube / poly-vinylidene fluoride (CNT/PVDF) composite films for the nano-generator devices were fabricated by spray coating method using the CNT/PVDF solution, which was prepared by adding PVDF pellets into the CNT dispersed N-Methyl-2-pyrroli-done (NMP) solution. The flexible CNT/PVDF composite films were investigated by the scanning electron microscopy, which revealed that the CNTs were uniformly dispersed in the PVDF matrix and thickness of the films was approximately $20{\mu}m$. Fourier transform infra-red spectra were used to investigate crystal structure of the as-spray-coated CNT/PVDF films, and we found that they revealed extremely large portion of the ${\beta}$ phase PVDF. The capacitance of the CNT/PVDF films increased by adding CNTs into the PVDF matrix, and finally saturated. However, the resistance didn't show any saturation effect in the CNT concentration range of 0~4 wt%. Finally, the resulting nano-generator devices revealed reasonable current output after given mechanical stress.

A STUDY ON OXIDATION TREATMENT OF URANIUM METAL CHIP UNDER CONTROLLING ATMOSPHERE FOR SAFE STORAGE

  • The U metal chips generated in developing nuclear fuel and a gamma radioisotope shield have been stored under immersion of water in KAERI. When the water of the storing vessels vaporizes or drains due to unexpected leaking, the U metal chips are able to open to air. A new oxidation treatment process was raised for a long time safe storage with concepts of drying under vacuum, evaporating the containing water and organic material with elevating temperature, and oxidizing the uranium metal chips at an appropriate high temperature under conditions of controlling the feeding rate of oxygen gas. In order to optimize the oxidation process the uranium metal chips were completely dried at higher temperature than $300^{\circ}C$ and tested for oxidation at various temperatures, which are $300^{\circ}C$, $400^{\circ}C$, and $500^{\circ}C$. When the oxidation temperature was $400^{\circ}C$, the oxidized sample for 7 hours showed a temperature rise of $60^{\circ}C$ in the self-ignition test. But the oxidized sample for 14 hours revealed a slight temperature rise of $7^{\circ}C$ representing a stable behavior in the self-ignition test. When the temperature was $500^{\circ}C$, the shorter oxidation for 7 hours appeared to be enough because the self-ignition test represented no temperature rise. By using several chemical analyses such as carbon content determination, X-ray deflection (XRD), Infrared spectra (IR) and Thermal gravimetric analysis (TGA) on the oxidation treated samples, the results of self-ignition test of new oxidation treatment process for U metal chip were interpreted and supported.

Structural Charateristics of Silk Fibroin Gel on The Preparation Conditions (Silk Fibroin Gel의 제조조건에 따른 구조특성)

  • Silk fibroin dissolved in highly concentrated calcium chloride and ethanol mixture aqueous solution turned into gel under suitable conditions. Preparation conditions and properties of gel were investigated as a function of parameters such as pH of solution, fibroin concentration, glycerol concentration and molecular weight. When pH of silk fibroin aqueous solution was near the isoelectronic point(pH 3.9~4.0), gelation occurred rapidly and strength of gel was stonger than that of pH-unadjusted due to electrostatic repulsion decrease between silk fibroin macromolecules. As concentration of silk fibroin and glycerol was higher, gelation occurred more rapid. FT Infra-red spectra of freeze-dried fibroin gel showed that gelation was derived by intermolecular anti-parallel ${\beta}$-sheet structure formation. In addition to, it was found that white-precipitate occurred instead of gelation when aqueous silk fibroin was treated by enzyme(flavouzyme), however, after flavouzyme-treated silk fibroin aqueous solution was centrifugated gelation occurred instantly. The results of differential scanning thermal analysis and infra-red spectroscopy showed that thermal stability and crystallinity of enzyme-hydrolyzed fibroin are superior to those of unhydrolyzed fibroin.

Characteristics of Polysaccharide Extracted from the Cultured Mycelia of Schizophyllum commune (치마버섯 균사체 배양물로부터 분리한 다당류의 이화학적 특성)

  • To examine physico-chemical properties of the polysaccharide extracted from liquid-cultured mycelia of Schizophyllum commune, each the polysaccharide was extracted with hot water treatment and then fractionated with ethanol, alkaline solution and ultrafiltration. And we determined carbohydrate contents, composition of amino acids, infra-red spectrum and viscosity. Carbohydrate contents of polysaccharide treated with ethanol and ultrafiltration were 72.0% and 62.3%, and proteins content were 15.3% and 32.0% respectively. The carbohydrate consisted of four monosaccharides and the protein contained 16 amino acids. The polysaccharide obtained from ultrafiltration was shown an absorption band characteristic of the ${\beta}$-glycosidic linkage by infra red spectra. These results suggest that the polysaccharide extracted from Schizophyllum commune showed the characteristics of proteinbounded polysaccharide, and it was ${\beta}$-glycosidic linkage with strong viscosity.

  • Product distribution and consumption in the military is difficult due to unique contracts and supply systems. It is difficult to change suppliers immediately when quality problem is encountered. Due to these special circumstances, the quality of products must be thoroughly controlled. Sesame oil is used to increase the taste and nutrition of food, but it is more expensive than other cooking oils. Oil producers may blend other cooking oils with sesame oil to make higher profits, so it has become important to identify good and bad products. In this study, pure sesame oil and blend oils were compared by analyzing their smell, taste, chemical components, and near infra-red spectra to determine quality differences between them.

Evaluation of Chemical Composition in Reconstituted Tobacco Leaf using Near Infrared Spectroscopy (근적외선 분광분석법을 이용한 판상엽 화학성분 평가)

  • Near InfraRed Spectroscopy(NIRS) is a quick and accurate analytical method to measure multiple components in tobacco manufacturing process. This study was carried out to develop calibration equation of near infrared spectroscopy for the prediction of the amount of chemical components and hot water solubles(HWS) of reconstituted tobacco leaf. Calibration samples of reconstituted tobacco leaf were collected from every lot produced during one year. The calibration equation was formulated as modified partial least square regression method (MPLS) by analyzing laboratory actual values and mathematically pre-treated spectra. The accuracy of the acquired equation was confirmed with the standard error of prediction(SEP) of chemical components in reconstituted tobacco leaf samples, indicated as coefficient of determination($R^2$) and prediction error of sample unacquainted, followed by the verification of model equation of laboratory actual values and these predicted results. As a result of monitoring, the standard error of prediction(SEP) were 0.25 % for total sugar, 0.03 % for nicotine, 0.03 % for chlorine, 0.16 % for nitrate, and 0.38 % for hot water solubles. The coefficient of determination($R^2$) were 0.98 for total sugar, 0.97 for nicotine, 0.96 for chlorine, 0.98 for nitrate and 0.92 for hot water solubles. Therefore, the NIRS calibration equation can be applicable and reliable for determination of chemical components of reconstituted tobacco leaf, and NIRS analytical method could be used as a rapid and accurate quality control method.
