비구조 단백질 유전자 primer를 사용한 RT-PCR에 의한 인플루엔자 A형 바이러스의 검출 (Detection of influenza A viruses by RT-PCR with single primer of nonstructural gene)

  • Influeza type A virus have been worldwide problematic in animals as well as in humans. In this study, the use of reverse-transcriptase polymerase chain reaction (RT-PCR) was described for detecting influenza virus type A. The primer of RT-PCR was designed from an nonstructural (NS) gene of Influenza A virus. By RT-PCR, a product with the size of 189 bp was detected only when influenza virus type A was used as template. No products could be detected with Influenza virus type B as well as other respiratory pathogens. The detection limit of the RT-PCR was up to $10^{0.3}TCID_{50}$ which is comparable to the sensitivity of cell culture method. The RT-PCR could detect the influenza A virus from nasal turbinates of the ferrets infected with influenza virus type A not type B.

Comparison study of SARIMA and ARGO models for in influenza epidemics prediction

  • The big data analysis has received much attention from the researchers working in various fields because the big data has a great potential in detecting or predicting future events such as epidemic outbreaks and changes in stock prices. Reflecting the current popularity of big data analysis, many authors have proposed methods tracking influenza epidemics based on internet-based information. The recently proposed 'autoregressive model using Google (ARGO) model' (Yang et al., 2015) is one of those influenza tracking models that harness search queries from Google as well as the reports from the Centers for Disease Control (CDC), and appears to outperform the existing method such as 'Google Flu Trends (GFT)'. Although the ARGO predicts well the outbreaks of influenza, this study demonstrates that a classical seasonal autoregressive integrated moving average (SARIMA) model can outperform the ARGO. The SARIMA model incorporates more accurate seasonality of the past influenza activities and takes less input variables into account. Our findings show that the SARIMA model is a functional tool for monitoring influenza epidemics.

인플루엔자 예방접종 권유와 예방 가능성 인식이 대학생의 예방접종 실천에 미치는 영향에 대한 융합연구 (Convergence Study on the Influence of Vaccination Encouragement and Perception of Influenza Preventability on Influenza Vaccination Practice Among University Students)

  • 본 연구는 인플루엔자 예방접종에 대한 권유와 인플루엔자 예방 가능성에 대한 인식이 대학생의 예방접종 실천에 미치는 영향을 파악하기 위해 시행하였다. 2018년 4월 30일부터 11월 30일까지 설문지를 배부하고, 239명의 자료를 분석하였다. 연구결과 36.8%의 대학생이 지난 1년간 인플루엔자 예방접종을 시행하였다. 57.6%가 예방접종 권유를 받았으며, 인지한 예방 가능성은 78.29%였다. 인플루엔자 예방접종 실천 영향요인으로 예방접종 권유는 유의하였으나(p<.001) 예방 가능성에 대한 인식은 유의하지 않았다(p=.853). 이에 대학생 인플루엔자 예방접종 프로그램 개발 시 예방접종에 대한 직접 권유뿐 아니라 다양한 매체를 활용한 간접적인 권유 전략을 포함하면 실천율을 높이는데 효과적일 것이다.

Current situation and control strategies of H9N2 avian influenza in South Korea

  • The H9N2 avian influenza (AI) has become endemic in poultry in many countries since the 1990s, which has caused considerable economic losses in the poultry industry. Considering the long history of the low pathogenicity H9N2 AI in many countries, once H9N2 AI is introduced, it is more difficult to eradicate than high pathogenicity AI. Various preventive measures and strategies, including vaccination and active national surveillance, have been used to control the Y439 lineage of H9N2 AI in South Korea, but it took a long time for the H9N2 virus to disappear from the fields. By contrast, the novel Y280 lineage of H9N2 AI was introduced in June 2020 and has spread nationwide. This study reviews the history, genetic and pathogenic characteristics, and control strategies for Korean H9N2 AI. This review may provide some clues for establishing control strategies for endemic AIV and a newly introduced Y280 lineage of H9N2 AI in South Korea.

Vaccine Strategy That Enhances the Protective Efficacy of Systemic Immunization by Establishing Lung-Resident Memory CD8 T Cells Against Influenza Infection

  • Most influenza vaccines currently in use target the highly variable hemagglutinin protein to induce neutralizing antibodies and therefore require yearly reformulation. T cell-based universal influenza vaccines focus on eliciting broadly cross-reactive T-cell responses, especially the tissue-resident memory T cell (TRM) population in the respiratory tract, providing superior protection to circulating memory T cells. This study demonstrated that intramuscular (i.m.) administration of the adenovirus-based vaccine expressing influenza virus nucleoprotein (rAd/NP) elicited weak CD8 TRM responses in the lungs and airways, and yielded poor protection against lethal influenza virus challenge. However, a novel "prime-and-deploy" strategy that combines i.m. vaccination of rAd/NP with subsequent intranasal administration of an empty adenovector induced strong NP-specific CD8+ TRM cells and provided complete protection against influenza virus challenge. Overall, our results demonstrate that this "prime-and-deploy" vaccination strategy is potentially applicable to the development of universal influenza vaccines.

서울지역 급성호흡기 환자들로부터 분리한 인플루엔자 바이러스의 유행양상 (2010-2012) (Epidemiological Characterization of Influenza Viruses detected from Acute Respiratory Patients in Seoul during 2010-2012)

  • Objectives: For our survey of the incidence of influenza viruses among respiratory viral infections in Seoul, we evaluated their prevalence among infectious acute respiratory viral patients in Seoul from 2010 to 2012 through regular surveillance. Methods: For influenza virus detection, we conducted real-time PCR analyses on 2,544 throat specimens collected from patients with respiratory viral infections in Seoul between 2010 and 2012. They were collected and then tested for the presence of influenza viruses through reverse transcription (RT) - polymerase chain reaction (PCR). Results: 19.1% (486/2,544) of the throat specimens were determined to be positive for influenza viruses. The incidences of influenza viral infection in the case of respiratory viral infections through regular surveillance in Seoul were 23.0% (212/923) in 2010, 6.4% (47/738) in 2011, and 25.7% (227/883) in 2012, and 10.8% (275/2,544) of type A, and 8.3% (211/2,544) type B influenza viruses. In addition, the greatest prevalence was in the 20-49 age group (51.6% ), which shows that influenza viruses constituted a major causative agent of acute respiratory viral infections. Conclusions: The distributions of influenza viruses and the epidemiologic patterns of the viral pathogen in acute respiratory viral infectious patients may provide potentially effective data for epidemiological studies in Seoul, Korea.

당뇨병 환자의 인플루엔자 예방접종률과 관련 요인 (The Estimated Proportion for Influenza Vaccination and Related Factors in Korean Diabetics)

  • Background: The purpose of this study was to investigate the estimated proportion for influenza vaccination and to identify factors associated with influenza vaccination in Korean diabetic patients. Methods: Data from the fourth, fifth, and sixth (except for 2013) Korea National Health and Nutrition Examination Survey (n=3,726) was used. A chi-square test was performed to investigate the estimated proportion for influenza vaccination, and a multiple logistic regression analysis was used to identify the factors associated with self-reported influenza vaccination. Results: In men, 28.8% of diabetes patients 30-64 years of age, and 76.1% of elderly (over 65 years of age) diabetes patients received influenza vaccination. In women, 37.7% of diabetes patients 30-64 years of age, and 78.4% of elderly diabetes patients received influenza vaccination (p<0.0001). The determinants of influenza vaccination were marriage, hypertension (odds ratio [OR], 1.55; 95% confidence interval [CI], 1.07-2.24), residence within a mega city, unemployment (OR, 3.94; 95% CI, 1.24-12.54), and exercise via (weekly) walking for diabetic men; and hypertension (OR, 1.71; 95% CI, 1.16-2.52), chronic disease (OR, 1.81; 95% CI, 1.08-3.02), and exercise via walking (OR, 2.65; 95% CI, 1.49-4.73) for diabetic women. Conclusion: Influenza vaccination remains relatively low in young diabetic patients. It is necessary to recommend vaccination to young diabetic patients, and to devise other strategies to improve vaccination.

한국 청소년 인플루엔자 예방접종에 영향을 미치는 개인요인과 부모요인에 관한 융합적 연구 : 제 6기 국민건강영양조사를 바탕으로 (Personal and Parental Factors Influencing Influenza Vaccination in Adolescents: Based on the 6th Korea National Health and Nutrition Examination Survey)

  • 청소년의 인플루엔자 예방접종은 지역사회의 인플루엔자 확산 방지에 중요하다. 따라서 본 연구에서는 국민건강영양조사 제 6기 자료를 이용하여 청소년의 인플루엔자 예방접종에 영향을 미치는 개인 요인과 부모 요인을 알아보고자 하였다. 연구대상은 만 12-18세 청소년 1,020명과 그 어머니 884명, 아버지 557명이었다. 분석을 위해 Rao-Scott $x^2$, 다중회귀분석을 실시하였다. 청소년의 인플루엔자 예방접종률은 26.2%로 매우 낮았고, 청소년의 연령이 적을수록, 최근 2주간 몸이 불편했던 경험과 폐렴 과거력이 있으며, 자동차 앞좌석 안전벨트를 착용하는 경우 인플루엔자 예방접종률이 높았다. 또한 부모의 연령이 적을수록, 부모 본인이 인플루엔자 예방접종을 한 경우 청소년의 인플루엔자 예방접종률이 높았다. 우리나라 청소년의 인플루엔자 예방접종을 확대하기 위해서는 미디어와 의료인들은 청소년과 부모에게 인플루엔자 예방접종에 대한 교육이나 홍보를 통해 인플루엔자 예방접종에 관한 인식을 개선하고 다양한 융합적 중재를 제공해야 한다.

돼지 인플루엔자 바이러스의 혈청학적 역학조사 및 유전학적 분석 (Sero-epidemiology and genetic characterization of swine influenza virus)

  • Total of 1085 swine sera (1996-1997) from nation-wide were tested for the presence of antibodies to influenza A virus. Fifty nine percent of the tested sera showed seropositive by HI test. Positive sera consisted of 24--- of H3, 15--- of H1, and 20--- of the sample had both antibodies, respectively. Sera collected from various region represented 7~27--- seropositivity to H1N1, 15~25--- to H3N2, respectively. Swine influenza field isolate from nasal swab was characterized antigenically and genetically to elucidate its relatedness with other known strains of influenza A virus. The study was focused on the HA gene which is related to pathogenecity and antigenic variability of the influenza virus. By RT-PCR using influenza A/H1N1 specific primers, influenza virus H1N1 specific DNA fragment was amplified from A/Swine/Iowa/15/30(H1N1), US field isolate but not in H3N2 strain. PCR products were sequenced by dideoxy chain termination method to determine nucleotide homology with other strains of influenza A virus. The US field isolate and A/Swine/Indiana/1726/88 strain had 97--- of nucleotide homology and 98--- of amino acid homology. Based on the results obtained from this experiment, the field isolate was genetically related to A/Swine/Indiana/1726/88 and had higher homology with A/Swine/Indiana/1726/88 than with classical swine influenza virus, A/Swine/Iowa/15/30. The field isolate had no amino acid changes at the antigenic site compare to that of the A/Swine/Indiana/1726/88. The proteolytic enzyme cleavage site between HA1 and HA2 had no alteration and the amino acid arginine was intact. There is no evidence has been found that the field isolate has genetic shift or genetic drift which might altered antigenic determinant.

경남지역 내 돼지에서의 swine influenza virus (H1N1, H3N2) 감염률 조사 (Seroprevalence survey of swine influenza virus (H1N1, H3N2) in pigs in Gyeongnam area)

  • Swine influenza is an acute respiratory disease prevalent in pig-growing areas all around the world and plays the roles of an intermediate host to be transmitted to mammals including human beings through a genetic recombination with the avian influenza virus. Recognizing that people could be contracted with swine influenza, this study set out to investigate the seroprevalence of individual and multiple infections with two subtypes (H1N1 and H3N2) of the swine influenza virus in pig farms in the Gyeongnam region according to age, area, and season, as well as to provide basic data for the prevention and control of swine influenza. Used in the study were total 904 swine sera that were not vaccinated against the influenza gathered from the pig farms in the Gyeongnam region from November, 2009 to October, 2010. HerdChek SIV (H1N1, H3N2) ELISA kit (IDEXX Laboratories, USA) was used for antibody testing against swine influenza. The test results show that 370 sera (40.9%) were infected with either H1N1 or H3N2 with 37.3% (337 sera) being contracted with H1N1, 13.1% (118 sera) with H3N2, and 9.4% (85) with both H1N1 and H3N2.
