Construction of Luminescence- and Fluorescence-Tagged Burkholderia pseudomallei for Pathogen Tracking in a Mouse Model

  • Molecular imaging is a powerful method for tracking various infectious disease-causing pathogens in host organisms. Currently, a dual molecular imaging method that can provide temporal and spatial information on infected hosts at the organism, organ, tissue, and cellular levels simultaneously has not been reported for Burkholderia pseudomallei, a high-risk pathogen that causes melioidosis. In this study, we have established an experimental method that provides spatiotemporal information on infected hosts using luminescent and fluorescent dual-labeled B. pseudomallei. Using this method, we visualized B. pseudomallei infection at the organism, organ, and tissue levels in a BALB/c mouse model by detecting its luminescence and fluorescence. The infection of B. pseudomallei at the cellular level was also visualized by its emitted fluorescence in infected macrophage cells. This method could be an extremely useful and applicable tool to study the pathogenesis of B. pseudomallei-related infectious diseases.

A Study on the Zoning of Wards and Intensive Care Units (ICUs) in the Infectious Disease Hospital (감염병전문병원의 병동부 및 중환자부 영역 구성에 관한 연구)

  • Purpose: This study aims to provide basic data for the future construction plans of the Infectious Disease Hospitals by analyzing the area composition and required room ratios in the wards and ICU of currently under-construction infectious disease hospitals. Methods: 3 Methods have been used in this paper. 1) This study conducted a literature review on major considerations and related guidelines for hospitals specializing in infectious diseases using existing data. 2) Based on the objects and activities of the hospital space, zones and areas were set for each department according to infection control. 3) Based on the established zones and areas, basic plan drawings of three hospitals specializing in infectious diseases currently under construction were collected and architectural drawing analysis was performed. Results: 1) Infectious Diseases Hospital must have a spatial organization that can accommodate patient isolation, infection control, efficiency of medical service, and changes. 2) Zones for infection control are divided into negative pressure and non-negative pressure zones based on airborne precaution isolation. It is divided into clean and contaminated zone according to class of cleanliness by Aseptic technique. Areas are classified by objects (patients, healthcare workers, supplies) and activities (access, medical treatment, support), and a system for organizing space is established based on this. 3) By analyzing the area composition of each departmental area, each required room, and each required space in the wards and intensive care units, it provides basic data for the spatial organization for architectural planning of the infectious disease hospital. Implication: It can be used as basic data when planning related facilities by analyzing the characteristics of the space plan of the required room according to the relationship between activities, movement lines, and operation plans based on user behavior.

Status of national notifiable infectious diseases in Jeju Province, Korea: 2001~2019 (제주도 법정감염병 신고 현황: 2001~2019)

  • Jeju Province has unique geographical and climatologic characteristics compared with other areas in Korea. Thus it needs to evaluate the status of nationwide notifiable infectious diseases(NND) continuously. The aim was to conduct a descriptive study for NNDs in Jeju Province, 2001~2019. The source of data was on the Infectious Disease Statistics System operated by Centers for Diseases Control & Prevention in Korea between 1 September 2001 and 31 August 2019. When calculating the proportion of the sum of years showing a higher incidence in Jeju province than nationwide divided by the sum of comparing years, the NDD with the proportion of the sum of years over 75% in 2001~2019 included varicella and syphilis. The results support significant evidence to plan strategies and distribute community resources for the control and prevention of NND in Jeju Province, Korea.

Plasmid Profile and ${\beta}$-Lactamase Type of Multidrug-Resistant Salmonella typhi Isolated from Korea, 1997 (국내 분리 다제내성 장티프스균의 Plasmid 및 ${\beta}$-lactamase의 특성에 관한 연구)

  • Eight strains of multidrug-resistant (MDR) Salmonella typhi were isolated from Kyonggi area during January-February, 1997. They were resistant to ampicillin, amoxicillin, carbenicillin, tetracycline, chloramphenicol, trimethoprim/sulfamethoxazole, trimethoprim. Eight strains had one plasmid respectively which size was approximately M.W 220 kb and showed same restriction pattern by endonuclease HindIII. The plasmid was similar to the plasmid in size that was related to multidrug resistant S. typhi isolated from southeast Asia. It were transferred by conjugation to recipient E. coli K-12 in frequency of $2.43{\times}10^4-1.73{\times}10^{-2}$ and transconjugant showed same drug-resistant pattern with donor cells. All of 8 strains produced ${\beta}$-lactamase that was assummed to TEM-l type by isoelectric focusing and PCR.

Scarless Genomic Point Mutation to Construct a Bacillus subtilis Strain Displaying Increased Antibiotic Plipastatin Production

  • Bacillus strains produce various types of antibiotics, and random mutagenesis has traditionally been used to overproduce these natural metabolites. However, this method leads to the accumulation of unwanted mutations in the genome. Here, we rationally designed a single nucleotide substitution in the degU gene to generate a B. subtilis strain displaying increased plipastatin production in a foreign DNA-free manner. The mutant strain (BS1028u) showed improved antifungal activity against Pythium ultimum. Notably, pps operon deletion in BS1028u resulted in complete loss of antifungal activity, suggesting that the antifungal activity strongly depends on the expression of the pps operon. Quantitative real-time PCR and lacZ assays showed that the point mutation resulted in 2-fold increased pps operon expression, which caused the increase in antifungal activity. Likewise, commercial Bacillus strains can be improved to display higher antifungal activity by rationally designed simple modifications of their genome, rendering them more efficient biocontrol agents.

The Distribution of Mismatches in Government Policy Response against COVID-19 in Terms of Risk Communication and its Implications

  • Purpose: This study aims to explore the effects of government's value intervention messages on the prevention and control of infectious disease in the risk communication process in terms of goveremnt policy response to COVID-19 from the mismatch message perspective, and draws the relevant implicaitons. Due to the infeciton spread, some people point out that depression caused by COVID-19 is because of wrong signals of the government and infectious disease prevention and control authorities, namely value intervention messsages. Research design, data and methodology: This study examined the epidcmic situation through message deails regarding the effects of government's mismatch messages on prevention and control of infectious disease and the resulting phenomena. Results: People's lives are under serious threat overall, so the declaration of the end of COVID-19 is almost impossible unlike MERS. Economic downturn due to foreced prevention and control regulaitons of COVID-19, mistruct of social distancing, fatigue on mismatch messages, and moral hazard on the awareness of prevention and control of infectious disease are negative phenomena to risk communiaiton on COVID-19. Conclusions: This study investigated the government authorities' policy sending wrong signals due to mismatching of the reality at this point in time for infectious disease prevention and control from the risk communicaiton perspectrive.

  • Several studies have suggested the possibility of airborne transmission of infectious diseases such as tuberculosis, pandemic influenza. because the number of patients increases explosively, if infectious disease had a high basic reproduction number, pharmaceutical interventions such as vaccination, chemoprophylaxis in the early stage of epidemic. Thus, non-pharmaceutical interventions such as mask-wearing, installing air cleaners, school closure are important to control and prevent the infectious diseases. However, the current technology on the mask, air cleaning, ventilation, and etc., seems to be not originated from the understanding of infection via airborne transmission. It is important to estimate the aerodynamic behavior of saliva droplets by coughing or speaking in order to understand the phenomena of airborne infection. In addition, the prediction of transmission of infectious diseases through the air is critical to prevent or minimize the damage of infection. In this review, we reviewed the recent studies on the airborne infection by focusing on the aerodynamic characteristics of saliva droplets and modeling of airborne transmission.

Retrospective Survey on the Mortality of Gorals at overland Zoological Gardens (1976-1999) (애버랜드 동물원에서 사육중인 산양의 폐사 원인 분석(1976-1999))

  • The mortalities of the gorals (Nemorhaedus goral raddeanus) that have been raised in Everland Zoological Gardens were retrospectively analyzed based on the clinical charts and autopsy reports from 1976 to 1999. During that Period a total of 17 gorals were died. Infectious factos (35.2%), digestive (23.5%) and respiratory disorders (17.6%) account feta the majority of cause of death. The The mortality of newborn gorals (50%) were significantly higher than that of adult (20%) and juvenile gorals (0%) due to infectious disease. 83.5% of the infectious disease were found in the garals younger than 5 months old. The cause of death of 5 newborn gorals died by infectious disease were as follows: 1aryngeal diphtheria, 2 (40%); actinobacillosis,2 (40%): coccidiosis. 1 (20%). It was considered that proper hygenic epic precautions in goral Pens ir feeding Places together with avoidance of rough feed are needed for the prevention of death of infectious cause at the goral raising in zoo-logical gardens.

Current Status of General Thoracic Surgery in Korea (한국의 일반 흉부수술 현황)

  • Overall 25,095 cases of general thoracic surgery were analysed, which were performed by 48 institutes in Korea during recent 6 years[242 hospital-years]. The proportions of tumorous disease and infectious disease to be operated were 6,864 cases[27.4%] and 6,775 cases [27.0%], The most common organ involved for operation was lung-bronchus 16,542 cases [69.5%], and remainders were pleura 2,500 [10.0%], esophagus 2,433[9.7%], mediastinum 1,902[7.6%], chest wall 1,297 [5.2%], and diaphragm 421 [1.7%] in order. Among 6,864 cases of tumorous diseases, the most common causes for operation were lung-bronchus tumor 3132 cases [45.6%] and most of them were lung cancer 2,731 cases [88.7%]. In the 2,019 cases of primary lung cancer with known cell type, squamous cell carcinoma 1,296 cases [64.2%] and adenocarcinoma 460 cases [22.8%] were the most. The common types in the 1,207 cases of mediastinal tumor with known cell type were neurogenic tumor 348 cases [28.8%], thymoma 311 [25.8%], and teratoma 252[20.9%]. The annual cases of operation for tumorous disease including malignant tumor were increased steadily. Operation for infectious lung diseases [including bronchiectasis and tuberculosis] were about twice common than infectious pleural disease [i.e. empyema], and operations for tuberculous disease occupied about half cases of infectious lung disease. In 11,456 cases of other disease entities, excluding tumorous and infectious disease, there were bullous lung disease 9,074 cases[79.2%], benign esophageal disease 484[4.2%], myasthenia gravis 356[3.1%], chest wall deformity 483[4.2%], and diaphragmatic lesion 421[3. 7%] in order. We propose that above results for inquiry can be used as the basic data of general thoracic surgery in Korea.
