  • Although the cone type venturi flow meter is superior to another differential pressure type flow meter in precision, the venturi is installed at the pipe as an L beam, so that the dynamic load due to drag force and flow-induced force is generated in the flow meter. In this paper we propose a methodology to evaluate this dynamic load directly by using stain-gages attached on the venturi and we discuss about the dynamic characteristics on the basis of flow-induced vibration theory.

곡판의 용접변형 예측을 위한 간이 해석법 (A Simplified Method to Predict the Weld-induced Deformation of Curved Plates)

  • A three-dimensional finite element model has been used to simulate the bead on plate welding of curved steel plates having curvature in the welding direction. By using traditional method such as thermal-elastic-plastic(TEP) finite element analysis. the weld-induced deformation can be accurately predicted. However, three-dimensional finite element analysis is not practical in analyzing the weld-induced deformation of large and complex structures such as ship structures in view of computing time and cost. In this study, used is the equivalent loading method based on inherent strain to illustrate the effect of the longitudinal curvature upon the weld-induced deformation of curved plates.

파랑작용에 의한 해저파이프라인 주변지반의 응답특성 (Wave-Induced Soil Response around Submarine Pipeline)

  • Recently, the nonlinear dynamic responses among waves, submarine pipeline and seabed have become a target of analyses for marine geotechnical and coastal engineers. Specifically, the velocity field around the submarine pipeline and the wave-induced responses of soil, such as stress and strain inside seabed, have been recognized as dominant factors in discussing the stability of submarine pipeline. The aim of this paper is to investigate nonlinear dynamic responses of soil in seabed, around submarine pipeline, under wave loading. In order to examine wave-induced soil responses, first, the calculation is conducted in the whole domain, including wave field and the seabed, using the VOF-FDM method. Then, velocities and pressures, which are obtained on the boundary between the wave field and the seabed, are used as the boundary condition to compute the wave-induced stress and strain inside seabed, using the poro-elastic FEM model, which is based on the approximation of the Biot's equations. Based on the numerical results, the characteristics of wave-induced soil responses around submarine pipeline are investigated, in detail, inrelation to relative separate distance of the submarine pipeline from seabed. Also, the velocity field around the submarine pipeline is discussed.

Establishment of inflammatory model induced by Pseudorabies virus infection in mice

  • Background: Pseudorabies virus (PRV) infection leads to high mortality in swine. Despite extensive efforts, effective treatments against PRV infection are limited. Furthermore, the inflammatory response induced by PRV strain GXLB-2013 is unclear. Objectives: Our study aimed to investigate the inflammatory response induced by PRV strain GXLB-2013, establish an inflammation model to elucidate the pathogenesis of PRV infection further, and develop effective drugs against PRV infection. Methods: Kunming mice were infected intramuscularly with medium, LPS, and different doses of PRV-GXLB-2013. Viral spread and histopathological damage to brain, spleen, and lung were determined at 7 days post-infection (dpi). Immune organ indices, levels of reactive oxygen species (ROS), nitric oxide (NO), and inflammatory cytokines, as well as levels of activity of COX-2 and iNOS were determined at 4, 7, and 14 dpi. Results: At 105-106 TCID50 PRV produced obviously neurological symptoms and 100% mortality in mice. Viral antigens were detectable in kidney, heart, lung, liver, spleen, and brain. In addition, inflammatory injuries were apparent in brain, spleen, and lung of PRV-infected mice. Moreover, PRV induced increases in immune organ indices, ROS and NO levels, activity of COX-2 and iNOS, and the content of key pro-inflammatory cytokines, including interleukin (IL)-1β, IL-6, tumor necrosis factor-α, interferon-γ and MCP-1. Among the tested doses, 102 TCID50 of PRV produced a significant inflammatory mediator increase. Conclusions: An inflammatory model induced by PRV infection was established in mice, and 102 TCID50 PRV was considered as the best concentration for the establishment of the model.

과잉 PbO에 의한 (Pb,Y) $(Zr,Sn,Ti)O_3$세라믹스의 유전 및 전기장유기변형 특성 (Dielectric and Field-induced Strain Behaviors due to Excess PbO in Lead Yttrium Zirconate Stannate Titanate Ceramics)

  • 디지털형 압전/전왜 액츄에이터 응용을 위하여 상경계(반강유전상/강유전상) 조성인 $Pb_{0.94}Y_{0.04}[(Zr_0.6Sn_0.4)_{0.915}Ti_0.085]O_3$ (PYZST) 계를 택하여 과잉 PbO의 첨가량 및 소결 조건 변화에 따른 상전이 특성, 유전 특성 및 전기장 유기변형 특성을 연구하였다. 사방정 구조를 갖는 PYZST 세라믹스에서 과잉 PbO 첨가에 따른 결정구조의 변화는 거의 확인되지 않았으나, 소결 후 입자가 약간 작아지며 둥근 형태로 변화하였고 첨가량 증가에 따라 적정 소결온도는 감소하였다. 과잉 PbO의 첨가량이 증가함에 따라, 분극측정시 반강유전상이 보다 안정되는 경향을 보였고, 전계유도변형 측정시 인가전기장 제거상태에서의 변형의 형상기억성이 감소하고 디지털형 변형곡선 특성이 강화되었다. 또한 최대 유전상수와 전계 유기변형량은 감소하였으나 반면 상전이(반강유전상$\leftrightarrow$강유전상) 전기장 및 비저항은 증가하는 경향을 나타냈다. 이러한 결과는 과잉으로 첨가된 PbO에 의한 격자 결함반응 및 분역벽 이동 거동 가능성과 연관시켜 설명하였다.

  • Lysine의 analogue인 thialysine은 야생주 Candida utilis NCYC-359의 생육을 강력하게 저해했으며 0.1%에서는 18시간까지 9.5%에서는 24시간까지 본 효모의 유도기를 지연시켰다. thialyssine에 의한 야생주의 생육저해의 회복에 미치는 아미노산 및 비타민의 영향을 조사한 결과 glycine, mthionine, arginine, histidine 그리고 trypphan등은 어느정도 효과가 있었으며 비타민은 거의 효과가 없었다. 특히 최소배지에 L-lysine을 thialysine의 1/8량만 첨가해도 thialysine에 의한 생육저해는 거의 일어나지 많았다. NTC 1회 처리시 thialysine에 대한 내성을 나타내는 빈도를 보면 최소배지에 thialysine 0.1% 첨가시 그 빈도가 0.11%, 0.5%시 0.03%로 나타났으며 이 내성맥리주에 의한 lysine 생산능을 검토한 결과 최소배지에 첨가된 thialyslne의 농도와 lysine생산능과는 뚜렷한 관계가 성립되지 않았으며 NTC를 회복처리하여 균체외로 lysine을 다양 분필 하는(450$\mu\textrm{g}$/$m\ell$) thialysine 내성맥리주 Candida utilis TRN-4006을 최종적으로 분리선별하였다.

출아효모에서 재조합 neoagarobiose hydrolyase의 생산을 위한 최적 발현시스템 (Optimal Expression System for Production of Recombinant Neoagarobiose Hydrolyase in Saccharomyces cerevisiae)

  • 본 연구에서는 Saccharomyces cerevisiae를 이용해서 neoagarobiose hydrolase (NABH)를 효율적으로 생산하기 위한 NABH558 유전자 발현시스템을 구축하였다. ADH1 promoter와 GAL10 promoter 하류에 NABH558 유전자를 가진 pAMFα-NABH plasmid와 pGMFα-NABH plasmid는 S. cerevisiae 2805 균주에 형질전환되었다. 2805/pAMFα-NABH 균주는 YPD (2% dextrose) 배지에서 가장 높은 NABH 효소 활성(0.069 unit/ml/DCW)을 보였고, 2805/pGMFα-NABH 균주의 경우는 배지의 조성과 상관없이 비슷한 수준의 NABH 활성(0.02-0.027 unit/ml/DCW)을 보였다. RT-PCR을 통한 NABH558 유전자의 transcription level은 NABH 활성 증가에 따라 비슷한 수준으로 증가되었음을 확인할 수 있었다. 또한 재조합균주에서 생산된 NABH는 agarose를 galactose와 AHG로 분해하였다. 따라서 NABH558 유전자의 발현에는 ADH1 promoter를 사용하는 것이 더 효율적이며 GAL10 promoter와 비교해서 최대 3배정도 높은 활성의 재조합 NABH를 생산할 수 있음을 알 수 있었다.

Inactivated Sendai Virus Strain Tianjin Induces Apoptosis in Human Breast Cancer MDA-MB-231 Cells

  • Sendai virus strain Tianjin is a novel genotype. Here, we investigate the antitumor and proapoptotic effects of ultraviolet-inactivated Sendai virus strain Tianjin (UV-Tianjin) on human breast cancer MDA-MB-231 cells in vitro, as well as the involvement of the apoptotic pathway in the mechanism of UV-Tianjin-induced antitumor effects. MTT assays showed that treatment with UV-Tianjin dose-dependently inhibited the proliferation of MDA-MB-231 cells but not normal MCF 10A breast epithelium cells. Hoechst staining and flow cytometric analysis revealed that UV-Tianjin induced apoptosis of MDA-MB-231 cells in a dose-dependent manner. Moreover, UV-Tianjin treatment resulted in reduction in the mitochondria membrane potential (MMP) and release of cytochrome complex (cyt c) via regulation of Bax and Bcl-2, as well as activation of caspase-9, caspase-3, Fas, FasL and caspase-8 in MDA-MB-231 cells. In summary, our study suggests that UV-Tianjin exhibits anticancer activity in human breast cancer MDA-MB-231 cells through inducing apoptosis, which may involve both the endogenous mitochondrial and exogenous death receptor pathways.

Induced Systemic Resistance by Bacillus vallismortis EXTN-1 Suppressed Bacterial Wilt in Tomato Caused by Ralstonia solanacearum

  • Biocontrol activity of five strains of selected rhizo-bacteria were tested in tomato against bacterial wilt caused by Ralstonia solanacearum. After root bacterization the plants were grown in a perlite-hydroponic system. Upon challenge inoculation with the pathogen, all of the rhizobacterial strains efficiently suppressed the bacterial wilt in tomato in various rates, at maximum by the strain, Bacillus vallismortis strain EXTN-1. While the percent of infected plants in the non-bacterized control plants were 95%, it was only 65% in plants pre-treated with EXTN-1. It was also demonstrated that the movement of R. solanacearum within the stem was significantly hampered when the plants were root bacterized. As EXTN-1 has no antagonistic properties against R. solanacearum, the bacterial wilt was probably suppressed by a mechanism other than antibiosis. Previously, the strain had been proven to produce an efficient elicitor for inducing systemic resistance in many crops. As the present study confirmed that EXTN-1 has the ability for reducing the pathogen spread in tomato, the strain could be effectively used as a potential biocontrol agent against bacterial wilt.

Comparative study on antioxidant activity of Gold 1, a new strain of Pyropia yezoensis

  • The global output of Pyropia yezoensis (dried seaweed or laver, also called 'Gim' in Korea) has been reduced over the half-decade due to the wide spread of red rot disease, a serious algal disease affecting P. yezoensis. Recently, Gold 1 (G1), which is a resistant strain of P. yezoensis to red rot disease, was developed and commercialized in South Korea, yet its physiological activity has not been investigated. In this study, a comparative study was performed on G1 and commercially available strain of P. yezoensis (CP) for their antioxidative activities. Aqueous extract of G1 showed more marked 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity compared to that of CP. In 293T cells, antioxidant activity against H2O2-induced reactive oxygen species (ROS) formation was only observed in G1 extract. In addition, G1 extract showed more potent inhibitory effect on H2O2-induced apoptotic cell death than CP extract, as examined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and fluorescence microscopy. Expression levels of various apoptosis-related genes, including B-cell lymphoma 2-associated X protein, p53, capase-3, and inflammatory cytokines, in H2O2-treated cells were significantly decreased by the treatment of G1. Taken together, the present study suggests that a new strain of red seaweed G1 can recover oxidative stress effectively by improving the imbalance of ROS generation and has a potential to be used a functional ingredient as an antioxidant source.
