Protective effect of 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride on hypoxia-induced toxicity by suppressing microglial activation in BV-2 cells

  • We recently reported the anti-inflammatory effects of 3-(naphthalen-2-yl(propoxy)methyl)azetidine hydrochloride (KHG26792) on the ATP-induced activation of the NFAT and MAPK pathways through the P2X7 receptor in microglia. To further investigate the underlying mechanism of KHG26792, we studied its protective effects on hypoxia-induced toxicity in microglia. The administration of KHG26792 significantly reduced the hypoxia-induced expression and activity of caspase-3 in BV-2 microglial cells. KHG26792 also reduced hypoxia-induced inducible nitric oxide synthase protein expression, which correlated with reduced nitric oxide accumulation. In addition, KHG26792 attenuated hypoxia-induced protein nitration, reactive oxygen species production, and NADPH oxidase activity. These effects were accompanied by the suppression of hypoxia-induced protein expression of hypoxia-inducible factor 1-alpha and NADPH oxidase-2. Although the clinical relevance of our findings remains to be determined, these data results suggest that KHG26792 prevents hypoxia-induced toxicity by suppressing microglial activation.

Inhibitory Effects of Glycine on Morphine-Induced Hyperactivity, Reverse Tolerance and Postsynaptic Dopamine Receptor Supersensitivity in Mice

  • The effects of glycine on morphine-induced hyperactivity, reverse tolerance and postsynaptic dopamine receptor supersensitivity in mice was examined. A single administration of morphine (10 mg/kg, s.c.) induced hyperactivity as measured in mice. The morphine-induced hyperactivity was inhibited by pretreatment with glycine (100, 200 and 400 mg/kg, i.p.). In addition, it was found repeated administration of morphine (10 mg/kg, s.c.) to mice daily for 6 days caused an increase in motor activity which could be induced by a subsequent morphine dose, an effect known as reverse tolerance or sensitization. Glycine (100, 200 and 400 rng/kg, i.p.) also inhibited morphine-induced reverse tolerance. Mice that had received 7 daily repeated administrations of morphine also developed postsynaptic dopamine receptor supersensitivity, as shown by enhanced ambulatory activity after administration of apomorphine (2 mg/kg, s.c.). Glycine inhibited the development of postsynaptic dopamine receptor supersensitivity induced by repeated administration of morphine. It is suggested that the inhibitory effects of glycine might be mediated by dopaminergic (DAergic) transmission. Accordingly, the inhibition by glycine of the morphine-induced hyperactivity, reverse tolerance and dopamine receptor supersensitivity suggests that glycine might be useful for the treatment of morphine addiction.

지성보명단(至聖保命丹)의 항경련(抗痙攣) 작용에 관한 실험적 연구 (An Experimental Study on the Anticonvulsive Effects of Jisungbomuyngtan)

  • Objectives In order to investigate the anticonvulsive effects of Jisungbomuyngtan, the experiments were done in mice. Methods After solid extracts of Jisungbomuyngtan was orally administered to mice, four types of convulsion were induced in mice by ECT, strychnine, picrotoxin, caffein. Each of the time to convulsion and death was measured. Results 1. The solid extracts of Jisungbomuyngtan showed a remarkable effect in delaying the onset of convulsion induced by ECT 2. In case of convulsion induced by strychnine, the solid extracts of Jisungbomuyngtan revealed a slight, but not statistically significant effect. 3. Considerable prolongation of time to death was observed in convulsion induced by picrotoxin due to the anticonvulsive effect of Jisungbomuyngtan. 4. The anticonvulsive effect of Jisungbomuyngtan was not found in convulsion induced by caffein. Conclusions Jisungbomuyngtan effect remarkably in delaying the onset of convulsion induced by ECT. But slightly effect by strychnine. And Jisungbomuyngtan effect in prolonging time to death in convulsion induced by picrotoxin, but not found in convulsion induced by caffein.

Protective effects of Hizikia fusiforme and Chlorella sp. extracts against lead acetate-induced hepatotoxicity in rats

  • In the present study, the protective effects of Hizikia fusiforme and Chlorella sp. extracts on lead acetate-induced hepatotoxicity were investigated. Hepatic damage was induced in rats by intraperitoneal (i.p.) injection of lead acetate and the protective effects of H. fusiforme (HZK) and Chlorella sp. (CHL) extracts on lead acetate-induced hepatic damage in rat liver were examined. The results revealed significantly increased glutamic oxaloacetate and glutamic pyruvic transaminase levels in the group treated with lead acetate only (Pb group); oral administration of HZK and CHL extracts tended to decrease the enzyme levels similar to those observed in the control group. Regarding antioxidant enzymes, superoxide dismutase activity was increased in the Pb group and decreased in a concentration-dependent manner in the HZK- and CHL-treated groups. Glutathione levels were increased in a concentration-dependent manner in the HZK- and CHL-treated groups. There was no significant difference in catalase activity. Western blot analysis showed inflammation-related protein expression in mitogen-activated protein kinase and Nrf2 pathways was affected in the HZK- and CHL-treated groups. Therefore, HZK and CHL extracts exerted antioxidant and anti-inflammatory effects against lead acetate-induced hepatotoxicity. Development of functional health foods containing HZK and CHL extracts, which have hepatoprotective effects against inhaled lead acetate, should be considered.

Protective effects and mechanism of coenzyme Q10 and vitamin C on doxorubicin-induced gastric mucosal injury and effects of intestinal flora

  • Doxorubicin (Dox) is widely used to the treatment of cancer, however, it could cause damage to gastric mucosa. To investigate the protective effects and related mechanisms of coenzyme Q10 (CoQ10) and vitamin C (VC) on Dox-induced gastric mucosal injury, we presented the survey of the 4 groups of the rats with different conditions. The results showed Dox treatment significantly induced GES-1 apoptosis, but preconditioning in GES-1 cells with VC or CoQ10 significantly inhibited the Dox-induced decrease and other harm effects, including the expression and of IκKβ, IκBα, NF-κB/p65 and tumor necrosis factor (TNF-α) in GES-1 cells. Moreover, high-throughput sequencing results showed Dox treatment increased the number of harmful gut microbes, and CoQ10 and VC treatment inhibited this effect. CoQ10 and VC treatment inhibits Dox-induced gastric mucosal injury by inhibiting the activation of the IkKB/IκBα/NF-κB/p65/TNF-α pathway, promoting anti-inflammatory effects of gastric tissue and regulating the composition of the intestinal flora.

  • This article presents the change characteristics of induced noise whether the sheath layer of the cable is grounded or not. As what affects the induced noise, there are power influence or longitudinal transverse voltages and its weighted filtered voltage. The sheath ground is basicaly predicted to have the effects of alleviation on the power influence. But practically the effects may not happen in the case of common cable's sheath layer. Rather there are cases that the ground of sheath affects so that the noise level could increase. So we need to scrutinize the effects of the sheath gorund in the measures for the protection against electromagnetic induction by powerline or traction line system. And the evaluation of using the designated shielding purpose cable is needed.

A correlation-based analysis on wind-induced interference effects between two tall buildings

  • Wind-induced mean and dynamic interference effects of tall buildings are studied in detail by a series of wind tunnel tests in this paper. Interference excitations of several types of upwind structures of different sizes in different upwind terrains are considered. Comprehensive interference characteristics are investigated by artificial neural networks and correlation analysis. Mechanism of the wakes vortex-induced resonance is discussed, too. Measured results show significant correlations exist in the distributions of the interference factors of different configurations and upwind terrains and, therefore, a series of relevant regression equations are proposed to simplify the complexity of the multi-parameter wind induced interference effects between two tall buildings.

보장지사탕의 항사하작용에 관한 연구 (Experimental Studies on the Anti-cathartic Effects of Bojangjisa-tang)

  • Objectives : The aim of this experimental study was to investigate the effects of Bojangjisa-tang on anticatharsis by using mice and Guinea pigs. Methods : We observed the actions on gastrointestinal smooth muscles, on the transportability of small and large intestines and on diarrhea induced by castor oil, philocarpine, and barium chloride. Results : Bojangjisa-tang showed alleviation of the contraction of gastrointestinal smooth muscles induced by acetylcholine chloride and barium chloride. The transportability of the large intestine was constrained by Bojangjisa-tang, which also showed substantial anti-cathartic effects on diarrhea induced by castor oil, philocarpine and barium chloride. Conclusion : These results demostrate that Bojangjisa-tang has advantageous effects on drug-induced diarrhea and will contribute to the development of diarrhea treatment through further related studies.
