Effects of Egg Storage Material and Storage Period on Hatchability in Japanese Quail

  • The present study aimed to determine the effects of different storage materials and storage period on some hatchability traits of Japanese quail (Coturnix coturnix japonica) eggs. A total of 32 male and 102 female quail (twelve weeks of ages) were housed in multiple-bird cages. Eggs laid were divided into three groups with respect to the period of storage (I = 1st, 2nd and 3rd days, II = 6th, 7th and 8th days, III = 12th, 13th and 14th days) prior to incubation. A total of 816 eggs was stored in 3 different storage environments during each storage period (B = no use of any storage material, P = use of perlite, H = use of hay) and kept in environmental conditions, where the temperature was $21^{\circ}C$ and relative humidity was 75%, prior to incubation. Statistical analyses were performed after the exclusion of values pertaining to non-fertile eggs (190 eggs) from the data set. The fertility rate of the eggs in the experiment was 76.7%. In the present study, the influence of storage material and different storage periods on egg weight loss were found to be statistically significant (p<0.01). Upon the comparison of hatchability of fertile eggs values, the influence of storage material was determined to be significant (p<0.05), and the influence of storage period was demonstrated to be significant (p<0.01). The storage materials used were determined not to have any influence on early and late embryonic death rates. Perlite was concluded to be safe for use in the storage of hatching eggs. The extension of the storage period (more than 8 days) resulted in decreased hatchability values of fertile eggs in each group.

Studies on Lytic, Tailed Bacillus cereus-specific Phage for Use in a Ferromagnetoelastic Biosensor as a Novel Recognition Element

  • This study investigated the feasibility of the lytic, tailed Bacillus cereus-specific phage for use in a ferromagnetoelastic (FME) biosensor as a novel recognition element. The phage was immobilized at various concentrations through either direct adsorption or a combination of 11-mercapto-1-undecanoic acid (11-MUA) and [N-(3-dimethylaminopropyl)-N'-carbodiimide hydrochloride and N-hydroxysuccinimide (EDC/NHS)]. The effects of time and temperature on its lytic properties were investigated through the exposure of B. cereus (4 and 8 logCFU/ml) to the phage (8 logPFU/ml) for various incubation periods at $22^{\circ}C$ and at various temperatures for 30 and 60 min. As the phage concentration increased, both immobilization methods also significantly increased the phage density (p < 0.05). SEM images confirmed that the phage density on the FME platform corresponded to the increased phage concentration. As the combination of 11-MUA and EDC/NHS enhanced the phage density and orientation by up to 4.3-fold, it was selected for use. When various incubation was conducted, no significant differences were observed in the survival rate of B. cereus within 30 min, which was in contrast to the significant decreases observed at 45 and 60 min (p < 0.05). In addition, temperature exerted no significant effects on the survival rate across the entire temperature range. This study demonstrated the feasibility of the lytic, tailed B. cereus-specific phage as a novel recognition element for use in an FME biosensor. Thus, the phage could be placed on the surface of foods for at least 30 min without any significant loss of B. cereus, as a result of the inherent lytic activity of the B. cereus-specific phage as a novel recognition element.

소 부루세라병 잠복기간과 방역대책;-제주도 소 부루세라병 근절대책을 중심으로- (The latent period and anti-epidemic measure of bovine brucellosis)

  • The 37,000 holstein which had been raised in Jeju island were examined for the purpose of brucellosis detection and so on. We investigated the relationship between the farms showing positive outbreak and the number of examination, incubation period, and the outbreak rate of positive cattle. We endeavored to separate germs from brucellosis positive cattles blood and negative bulls semen against brucellosis serum test and concluded as follows. We intend to offer you institutional supplements and preventions of brucellosis for the future. 1. Outbreak of brucellosis could not lead the outbreak rate of positive cattle to decrease through 1-2 examination per year, For the past 4 years, the outbreak rate of brucellosis positive cattle had a tendency to show a gradual decrease through at least 5 examinations per year. 2. As we examined live-in cattle with positive cattle in mass outbreak farms for 9 months, we found out positive cattle every month. In a grazing land, the group of cattle producd enormous positive cattle in 9-12 months. 3. Annual brucellosis positive outbreak rate was 20-25% among live-in cattle with brucellosis positive cattle, but the rate might be 50-100% depending on extent of pollution. 4. 94's brucellosis examination showed that 200 positive cattle of 71,153 cattle and most of them were live-in cattle with confirmed brucellosis positive cattle. 5. 1 head of 200 positive cattle showed positivity against serum of bulls test and the rate of bulls was about 1%. 6. Brucellosis germ was separated from 2 cattle's semen among 52 negative bulls against brucellosis serum test and there was the one brucellosis positive cattle against tube agglutination of semen test, so the positive rate of brucellosis was 5.8%. 7. Brucellosis germ was not separated from 15 brucellosis positive cattle's blood.

장내 병원균의 하수중 생존 상태에 관한 연구 (Survival Studies of Some Enteric Pathogens in Sewage)

  • In order to study the growth and survival of enteric pathogens causing water-borne infections in sewage, the filter-sterilized and autoclaved sewages of Dae Gu City were inoculated with Salmonella typhimuriuim, Shigella flexneri 2a, Sh. sonnei I, Vibrio eltor and V. parahaemolyticus, as test series and Escherichia coli as control. After varying periods of incubation up to 15 days at $4^{\circ}$, $15^{\circ}$, $25^{\circ}$ and $37^{\circ}C$, viable cells in the inoculated sewages were counted by colony count technique. Distilled water and 0.9% saline were subjected to inoculation of the organisms was observed in the filter-sterilized and autoclaved sewages at $4^{\circ}$ and the sewages became sterile within a few days. At $15^{\circ}$, no growth and rapid inactivation of the organisms in the filter-sterilized sewage and slight or no growth in the autoclaved sewage was noted. Some viable cells were found in the autoclaved sewage after 15 days. A considerable growth was observed in the filter-sterilized and autoclayed sewages, at $25^{\circ}$ and $37^{\circ}$, and large numbers of viable cells were found even after 15days of incubation. In general, the autoclaved sewage supproted the growth more noticeably than the filter-sterilized, except for V.parahaemolyticus which grew well in filter-sterilized sewage. No marked difference was noted between incubations at $25^{\circ}$ and $37^{\circ}$, but V. parahaemolyticus showed a slightly more active growth at $25^{\circ}$ than at $37^{\circ}$. Distilled water inactivated the organisms within a few days, but saline supported the growth at $25^{\circ}$ and $37^{\circ}$. Marked differences were noted in the survival test of sewages pathogens of different origins.

미생물제(微生物劑)의 퇴비부숙(堆肥腐熟) 효과(效果) (Effects of Decomposition of Organic Substances as Rice Straw and Chicken Manure mixed with Saw Dust treated with Commercial Inoculums)

  • 미생물제의 퇴비부숙효과를 검토하기 위하여 국내에 유통되고있는 Lacto외 4종 미생물제에 대한 퇴비부숙 촉진 효과를 보면 볏짚에서는 락토 및 천보, 생력등 일부 미생물(微生物)제의 처리에 의하여 초기에 부숙촉진경향이 관찰되었으나 초기에 부숙이 촉진되었던 처리는 후기에 부숙속도가 늦어지고 초기에 부숙속도가 늦었던 처리는 후기에 부숙속도가 빨라짐으로서 결국 미생물제처리 40일경에는 미생물제 사이에 뚜렷한 차이가 인정되지 않았다. 또한 미생물제를 시용하면 볏짚부숙중에 사상균수의 증가가 현저하였으며 볏짚의 부숙과 사상균수의 변동과 일치하는 경향을 보였으나, 계분톱밥부숙에 미치는 미생물제의 영향을 보면 퇴적초기에는 세균, 방선균, 사상균 모두 균수가 극히 적었으며 세균수만이 시간이 경과함에 따라 균수의 증가가 현저한 점으로 미루어 볼 때 세균이 계분톱밥퇴비의 부숙에 밀접한 관련이 있는 것으로 판단되었다.

MICROBIAL COLONISATION AND DEGRADATION OF SOME FIBROUS CROP RESIDUES IN THE RUMEN OF GOATS

  • An investigation was carried out to study the microbial colonization and degradation of five crop residues, viz., sago waste, rice straw, oil palm trunk shavings, untreated palm press fibre and palm press fibre teated with 3% ammonium hydroxide in the rumen of goats. Colonisation by rumen bacteria and fungi was already established on all the five crop residues 8 h after incubation. However, the extent of colonization varied among the crop residues. Microbial colonization was poor on palm press fibre (treated and untreated) but more extensive on sago waste, oil palm trunk shavings and rice straw. By 24 h, most of the soft-walled tissues in sago waste, rice straw and oil palm trunk shavings were degraded leaving the thick-walled tissues extensively colonized by bacteria and fungi. Degradation on palm press fibre was still limited. At 48 h, the thick-walled tissues of sago waste, oil palm trunk shavings and rice straw showed various degrees of degradation - from small erosion zones to large digested areas. Bacterial growth was similar to that at 24 h but fungal growth was less. On palm press fibre, microbial colonization was more extensive than at 24 h but degradation of the fibres was still limited. Degradation of all the five crop residues at 72 h was somewhat similar to that at 48 h. Overall, microbial colonization and degradation were the most extensive on sago waste, followed by rice straw and oil palm trunk shavings, and the least on palm press fibre (treated and untreated). Dry matter loss of the five crop residues at the various incubation periods also showed the same order of degradation.

  • 갈대(Phragmites communis Trin) 유아(幼芽)를 $27^{\circ}C$의 암소에서 110시간 동안 사경(砂耕) 배양(培養)하였다. 유아(幼芽)가 자라는 동안 일정(一定)한 시간마다 시료(試料)를 채취하여 methanol로 처리(處理)하여 그 추출물(抽出物)에 대하여 thin layer chromatography를 실시(實施)하였다. 그 결과 배양(培養)하지 않은 유아(幼芽)에서는 indole화합물(化合物) 중에서 serotonin, tryptophan, tryptamine이 다량(多量)으로 나타났으나, 배양(培養) 과정(過程) 중에 이들 화합물(化合物)의 감소(減少)와 더불어 다른 여러 가지 indoleamine류(類)의 생합성(生合成)이 현저(顯著)하였다. 즉 배양(培養) 과정(過程) 중에 tryptophan의 양(量)에는 큰 변화(變化)가 없으나, serotonin 및 tryptamine이 감소하면서, 이들의 methyl화(化) 유도체들이 증가(增加) 하였다. 그 증감(增感)의 상호관계(相互關係)를 관찰하여 본 결과, bufotenine은 tryptophan에서 tryptamine을 거쳐 serotonin, N-methylserotonin의 경로(經路)로 합성(合成)될 것임을 시사하였다.

  • Degrees of hydrolysis by alkaline protease produced from Serratia marcescens S3-R1 is 3.95-6.30% of whey proteins during 5, 15, 30, 60, 90, 120,180, 240 min incubation at $40^{\circ}C$. Proteolytic pattern of the whey proteins showed that various low molecular weight peptides were generated during the incubation periods. The biological function of in Raw 264.7 cells treated with whey protein hydrolytic peptides, anti-inflammatory effect showed exhibit in the expression of pro-inflammatory cytokines such as TNF-${\alpha}$, IL-6, COX-2 and iNOS by PCR analysis. COX-2 and iNOS gene expression inhibited in Raw 264.7 cells on whey protein hydrolysates below 3,000 dalton. The protease from Serratia marcescens S3-R1 showed a potential in production of low molecular weight whey protein hydrolysates which could be used for industrial application.

Biology of a predatory bug Eocanthecona furcellata Wolff (Hemiptera : Pentatomidae) on Vapourer tussock moth larvae: a major pest of tasar silkworm food plants

  • Tropical Tasar culture is forest and agro-based activity that covers agricultural and industrial activity. Tasar silk is produced by the silkworm, Antheraea mylitta which is polyphagous in nature. A large number of pests are reported to attack both the host plant as well as silkworm. As the rearing is conducted outdoor silkworms are exposed to attack of various pests during their life cycle. Eocanthecona furcellata (Hemiptera: Pentatomidae) is the major predator of tasar silkworm during young age rearing. Both nymphs and adults of the stink bug suck the heamolymph of silkworm leading to death. The present work was undertaken to study the life cycle of E. furcellata on larvae of vapourer tussock moth which is a major pest of tasar host plants. The incubation period, nymphal duration and total developmental period from egg to adult was $8.0{\pm}0.45d$, $16.0{\pm}3.24$ and $22.0{\pm}4.20d$, respectively. Pre-oviposition, oviposition and longevity of male and female stink bug was $3.20{\pm}0.38$, $17.60{\pm}1.86$, $42.40{\pm}1.94$, $37.00{\pm}3.18d$, respectively. Analysis of the data revealed no significant difference in life cycle of the predator when provided silkworm and vapourer tussock moth larvae as hosts. Which clearly indicates that E. furcellata can multiply on vapourer tussock moth larvae during non rearing periods. Adoption of recommended pest control measures for defoliators of tasar host plants during non-rearing periods will help in reducing the incidence of host plant pests and also in reducing the incidence of stink bug during rearing season. During the study it was also observed that larvae semilooper, boll headed caterpillar and nymphs of leaf hopper serve as alternate/secondary hosts to E. furcellata during non rearing seasons.

Epidemiological Investigations to Optimize the Management of Pepper Anthracnose

  • An understanding of anthracnose (Colletotrichum acutatum) infections, including the infection of flowers and latent infection early in the season, is necessary to achieve successful control by means of properly timed spraying with a curative fungicide. In the present study, latent anthracnose infection of chili was investigated under greenhouse and field conditions in 2007-2008. Flowers on greenhouse-grown seedlings were infected and 11% of the young fruits subsequently showed symptoms of anthracnose. Apparently healthy-looking green peppers obtained from unsprayed fields or an organic market also exhibited symptoms of anthracnose after 4 days of incubation under high moisture conditions at $25^{\circ}C$; less than 1% of the peppers were found to be latently infected. To determine the natural timing of infection in the field, 3,200 fruits were wrapped in paper bags and then selectively unwrapped and examined for signs of infection. Field experiments were conducted at Suwon (cvs. Yokkang, Manitta, Olympic) and Asan (cv. Chunhasangsa) in 2008. The 7- to 10-day wrapping periods were July 25-31, July 31-August 7, August 7-15, August 15-24, and August 24-September 3. The 1-to 2-month wrapping periods were from July 4, July 31, and August 15 until harvest (Sept. 3). The controls consisted of 1,712 field-grown non-wrapped fruits. The rates of infection on the various cultivars were Yokkang 55%, Manitta 37%, Olympic 55%, and Chunhasangsa 20%. A distinct period in which anthracnose infection suddenly increased could not be identified; however, attempts to guess the approximate timing of field infection showed that 0-39% of the plants had latent infections, while depending on the cultivar, 8-14% of the plants examined in August and 4-13.5% of the those examined during May-July showed symptoms of infection. Delaying fungicide spraying by 24 and 48 h after artificial infection decreased the rates of infection by 10% and 25-30%, respectively. Chemical control of anthracnose based on a forecasting model should be considered starting from the transplanting stage, with spraying within a day after warning and care being taken not to latently infect apparently healthy pepper fruits.
