Objectives: The purpose of this study was to evaluate the concentrations of particulate matter generated during coffee roasting and to study various factors affecting the concentrations. Methods: Differences in concentration levels were investigated based on various factors to understand the emission rates of particulate matter over time and to compare the mass and number concentrations according to their size. Sampling was performed in closed laboratories without the operation of air conditioning or ventilation. Optical Particle Sizer(OPS) was used as a measuring device. An OPS measures using a light-scattering method. Sampling was performed for sixty minutes at one-minute intervals. The background concentration was measured for about 30 minutes before starting of coffee roasting. The concentrations of particulate matter generated during coffee roasting were monitored until roasted coffee beans were removed from the roaster and cooled down. Several factors affecting the concentrations of particulate matter were investigated, which includes the origins of green beans, the roasting level, and the input amount of green beans. Results: The results of this study may be summarized as follows: 1) There was no difference in particulate matter concentration levels by the origin of the green beans, but a statistically significant difference in concentration levels by roasting level and the input amount of green beans; The higher the roasting level, the higher was the particulate matter concentration. The more green beans we put in the roaster, the higher were the concentrations; 2) The PM10 mass concentrations increased over time. The average concentration after roasting was higher than the average concentration during roasting; 3) In the distribution of mass and number concentration by particle diameter, the majority of particles was below 2.5 ㎛. Conclusions: Persons who work in roastery cafes can be exposed to high concentrations of particulate matter. Therefore, personal exposure and risk assessment should be conducted for roastery cafe workers.
Oxidative stress due to reactive oxygen species (ROS) can cause oxidative damage to cells. Cells have a number of defense mechanisms to protect themselves from the toxicity of ROS. Mitochondria are especially important in the oxidative stress as ROS have been found to be constantly generated as an endogen threat. Mitochondrial defense depends mainly on super-oxide dismutase (SOD) and glutathione peroxidase (GPx), whereas microsomal defense depends on catalase (CAT), which is an enzyme abundant in microsomes. SOD removes superoxide anions by converting them to $H_2O$$_2$, which can be rapidly converted to water by CAT and GPx. Also, GPx converts hydroperoxide (ROOH) into oxidized-glutathione (GSSG). Ovariectomized (OVX) rats are used as an oxidative stress model. An ovariectomy increased the levels of MDA, one of the end-products in the lipid peroxidative process, and decreased levels of the antioxidative enzymes; SOD, CAT and GPx. However, Chondroitin sulfate (CS) decreased the levels of MDA, but increased the levels of SOD, CAT and GPx in a dose-depen-dent manner. Moreover, inflammation and cirrhosis of liver tissue in CS- treated rats were sig-nificantly decreased. These results suggest that CS might be a potential candidate as an anti oxidative reagent.

Background: Ammonia and hydrogen sulfide are harmful gases generated during aerobic/anaerobic bacterial decomposition of livestock manure. We evaluated ammonia and hydrogen sulfide concentrations generated from workplaces at livestock farms and determined environmental factors influencing the gas concentrations. Methods: Five commercial swine farms and five poultry farms were selected for monitoring. Real-time monitors were used to measure the ammonia and hydrogen sulfide concentrations and environmental conditions during the manure-handling processes. Monitoring was conducted in the manure storage facility and composting facility. Information on the farm conditions was also collected through interview and walk-through survey. Results: The ammonia concentrations were significantly higher at the swine composting facilities (9.5-43.2 ppm) than at other manure-handling facilities at the swine and poultry farms, and high concentrations of hydrogen sulfide were identified during the manure agitation and mixing process at the swine manure storage facilities (6.9-19.5 ppm). At the poultry manure-handling facilities, the ammonia concentration was higher during the manure-handling processes (2.6-57.9 ppm), and very low hydrogen sulfide concentrations (0-3.4 ppm) were detected. The air temperature and relative humidity, volume of the facility, duration of manure storage, and the number of animals influenced the gas concentrations. Conclusion: A high level of hazardous gases was generated during manure handling, and some levels increased up to risk levels that can threaten workers' health and safety. Some of the farm operational factors were also found to influence the gas levels. By controlling and improving these factors, it would be possible to protect workers' safety and health from occupational risks.
Oxidative stress has been associated with a number of diseases in human. Among the sources that can generate oxidative stress, it has been reported that iron can generate reactive oxygen species (ROS)with thiol. In iron overload state, increased thiol levels in plasma appeared to be associated with human mortality. In this study we examined whether iron could interact with thiols in plasma, generating ROS. In human plasma, unlike with Fe(III), Fe(II) increased lucigenin-enhanced chemiluminescence in concentration-dependent manner, and this was inhibited by SOD. Boiling of plasma did not affect chemiluminescence induced by Fe(II). Hovever, thiol depletion in plasma by pretreatment with N-ethylmaleimide (NEM)decreased Fe(II)-induced chemiluminescence significantly, suggesting that Fe(II) generated superoxide anion by the nonenzymatic reaction with plasma thiol. Consistent with this findings, albumin, the major thiol contributor in plasma, also generated ROS with Fe(II) and this generation was inhibited by pretreatment with NEM. Treatment with Fe(II) to plasma resulted un significant reduction of oxygen radical absorbance capacity (ORAC) value, suggest that total antioxidant capacity could diminished in iron overload state. In conclusion, In iron overload state, plasma may be affected by oxidative stress mediated by nonenzymatic reaction of Fe (II)with plasma thiol.
Background: Millions of people in the world are suffering from atopic dermatitis (AD), which is a chronic inflammatory skin disease triggered by Th2 immune responses. The NC/Nga mouse is the most extensively studied animal model of AD. Like human AD, NC/Nga mice demonstrate increased levels of IgE, a hallmark of Th2 immune responses. Adaptive immunity cannot be generated without help of innate immunity. Especially natural killer T (NKT) cells and marginal zone B (MZB) cells have been known to play important roles in linking innate immunity to adaptive immunity. Methods: Through flow cytometric analysis and ELISA assay, we investigated whether these lymphocytes might be altered in number in NC/Nga mice. Results: Our data demonstrated that the number of NKT cells was reduced in NC/Nga mice and IFN${\gamma}$ production by NKT cells upon ${\alpha}-GalCer$ stimulation decreased to the levels of CD1d KO mice lacking in NKT cells. However, reduction of NKT cells in NC/Nga mice was not due to CD1d expression, which was normal in the thymus. Interestingly, there was a significant increase of $CD1d^{high}B220^+$ cells in the spleen of NC/Nga mice. Further, we confirmed that $CD1d^{high}B220^+$ cells are B cells, not dendritic cells. These $CD1d^{high}B220^+$ B cells show $IgM^{high}CD21^{high}CD23^{low}$, a characteristic phenotype of MZB cells. Conclusion: We provide the evidence that there are decreased activities of NKT cells and increased number of MZB cells in the NC/Nga mice. Our findings may thus explain why NC/Nga mice are susceptible to AD.
Objective : This study was carried out to understand effects of ginseng(hearinafter ; GS, Panax Ginseng) extract on regeneration responses on injured sciatic nerves in rats. Methods :Using white mouse, we damaged sciatic nerve & central nerve, and then applied GS to the lesion. Then we observed regeneration of axon and non-neuron. Results : 1. NF-200 protein immunostaining for the visualization of axons showed more distal elongation of sciatic nerve axons in GS-treated group than saline-treated control 3 and 7 days after crush injury. 2. GAP-43 protein was increased in the injured sciatic nerve and further increased by GS treatment. Enhanced GAP-43 protein signals were also observed in DRG prepared from the rats given nerve injury and GS treatment. 3. GS treatment in vivo induced enhanced neurite outgrowth in preconditioned DRG sensory neurons. In vitro treatment of GS on sensory neurons from intact DRG also caused increased neurite outgrowth. 4. Phospho-Erk1/2 protein levels were higher in the injured nerve treated with GS than saline. Phospho-Erk1/2 protein signals were mostly found in the axons in the injured nerve. 5. NGF and Cdc2 protein levels showed slight increases in the injured nerves of GS-treated group compared to saline-treated group. 6. The number of Schwann cell population was significantly increased by GS treatment in the injured sciatic nerve. GS treatment with cultured Schwann cells increased proliferation and Cdc2 protein signals. 7. GS pretreatment into the injured spinal cord generated increased astrocyte proliferation and oligodendrocytes in culture. In vitro treatment of GS resulted in more differentiated pericytoplasmic processes compared with saline treatment. 8. More arborization around the injury cavity and the occurrence at the caudal region of CST axons were observed in GS-treated group than in saline-treated group. Conclusion :GS extract may have the growth-promoting activity on regenerating axons in both peripheral and central nervous systems.

This paper deals with the design and implementation of an efficient topology for cascaded multilevel inverters with reduced part counts. In the proposed design, a well-established basic unit is first developed. The series extension of this unit results in the formation of the proposed multilevel inverter. The proposed design minimizes the number of power electronic components including insulated-gate bipolar transistors and gate driver circuits, which in turn cuts down the size of the inverter assembly and reduces the operating power losses. An explicit control strategy with enhanced device efficiency is also acquired. Thus, the part count reductions enhance not only the economical merits but also the technical features of the entire system. In order to accomplish the desired operational aspects, three algorithms are considered to determine the magnitudes of the dc voltage sources effectively. The proposed topology is compared with the conventional cascaded H-bridge multilevel inverter topology, to reflect the merits of the presented structure. In continue, both the analytical and experimental results of a cascaded 31-level structure are analyzed. The obtained results are discussed in depth, and the exemplary performance of the proposed structure is corroborated.
Cavities are inevitable structures in automobile configuration. The flow-induced noise is generated from the wheel housing section by the interaction between a rotating wheel and the unsteady flows in the cavity. In this research the wheel housing was assumed by a rectangular cavity for simplification. We measured the radiated sound from the 2-D cavity without cylinder and from the rotating cylinder in the cavity by using the sound source localization method with an acoustic mirror system. In the 2-D cavity case of low Mach number(Ma=0.029), the sound sources were found to be located near the leading edge of cavity due to the shear layer instabilities. Comparing the cases of the rotating and the non-rotating cylinder, it is observed that the sound Pressure levels around the rotating cylinder in the cavity increased and the main acoustic sources were located at the rear section of the rotating wheel.
Physical activity is a primary cancer control strategy that has received little attention to date. However, an Increasing number of epidemiological studies have proposed that physical exercise may be beneficial by enhancing anticancer immune system responses. We investigated the effects of acute exercise on changes in nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression. The amounts of NO generated by abdominal macrophages in mice were measured after exercise. Thirty-two mice, which were challenged with thioglycollate broth to activate peritoneal macrophages, were randomly assigned to control, exercise and recovery groups. The mice exercised on a motor-driven treadmill for 3 consecutive days, either moderately (18m/min, 30 min/day, 5% grade) or severely (18-35m/min, 60 min/day, 5% grade). The mice were killed immediately after exercise or after 6 hrs of recovery. Nitric oxide was quantified by the Griess assay. The exercised mice showed higher levels of NO generation than those of the control mice, but the intensity of exercise had no significant effect on NO generation. Mice allowed six hours of recovery after exercise showed higher levels of NO generation than that of animals sacrificed immediately after exercise, but there were no significant differences in NO generation with variations in the intensity of exercise. Increased levels of iNOS were found in the exercised groups, and this was greatest in the groups allowed six hours of recovery compared to those groups sacrificed immediately after exercise. The results of this study suggest that acute exercise may enhance an immune response by inducing macrophage-derived NO generation; these results support the epidemiological findings which support the benefits of exercise in the prevention and control of cancer. Further study is needed to determine the physiological significance of these findings, which could be applied to the use of therapeutic exercises to assist in the prevention and control of cancer.
양파 재배 시 국내 토양에 절대적으로 부족한 황을 처리 횟수를 달리하여 재배 생산한 후, 황 함유 성분(sulfur-containing compounds)을 포함한 양파의 화학적 조성과 물리적 관능적 특성이 개선되었는지를 평가하였다. 선행연구를 토대로, 파종 시 토양에 직접 MSM 식이 유황을 1회 공통 처리하고 수확 2달 전황을 1회만 추가 처리한 군(Sulfur-1)과 수확 3달 전부터 수확 1달 전 까지 일정 간격으로 황을 총 4회 처리한 군(Sulfur-4)으로 구별하였으며, 황을 전혀 처리하지 않은 군(Control-S)과 타 지역 양파(Control-M)를 비교를 위한 대조군으로 선정하였다. 황 처리 횟수가 증가함에 따라, 수분은 감소하는 경향을, 총 질소로 측정된 조단백질과 무기질 총량인 회분(p<0.01), 그리고 식이섬유는 증가하는 경향을 나타내었으며, Mg, K, Fe, Zn 등이 증가하여(p<0.05), 전반적으로 무기질을 포함한 화학적 조성이 개선되었음을 시사하였다. 또한, 항산화력의 지표인 총 환원물질의 양도 2배 가량 유의적으로 증가하는 결과가 관찰되었다(p<0.05). 그러나, 황 함유 성분인 thiosulfinate와 함황 아미노산에서는 황 처리 효과가 관찰되지 않았고, 전자코로 측정된 향미 프로파일 역시 향미 성분의 차이를 시사하지 않았다. 이는, 본 실험에서 처리된 황 수준은 질소대사 및 토양의 pH를 변화시키는 간접적 경로를 통한 양파의 이화학적 조성 변화는 가능하게 할 수 있었으나, 황 함유 성분들을 직접적으로 증가시키기에는 충분하지 못했음을 시사하였다. 그럼에도 불구하고, 황 처리 횟수 증가와 함께, 함황 성분들에 의해 주로 인지되는 매운 맛과 매운 향 특성은 관능적으로 강해지는 것으로 평가되었다. 이러한 차이는, 양파 특유의 냄새 성분들이 지닌 낮은 역치에서 기인한 것으로 해석하였다. 한편, Control-S, Sulfur-1, Sulfur-4 양파들은 $4^{\circ}C$에서 4개월 동안 저장한 후, 모두 수분의 큰 증가 없이 총 환원력이 2배 가량 증가하였으며(p<0.01), 황 처리군이 무처리군에 비해 여전히 유의적으로 높았다(p<0.001). 반면, 동일조건에서 Control-M은 유의적 변화가 관찰되지 않았다. 이러한 차이는, 황 처리, 호흡작용으로 인한 환원당의 증가 이외에 지역적 영향이 관여했을 가능성을 시사하였다.
