Perfusion배양시 세포성장 및 항체생산 향상을 위한 아미노산의 보강 (Fortification of Amino Acids to Improve Hybridoma Cell Growth and Monoclonal Antibody Production in Perfusion Culture)

  • 본 실험에서는 하이브리도마세포 배양 중 아미노산 량의 변화를 조사하여 세포성장, 세포대사 및 항체생산성에 미치는 아미노산의 영향을 조사하였다. 실험결과에 의하면 vR8 하이브리도 마세포 배양 중 세포성장이 급속히 이루어지는 시점과 최대 세포농도에 도달한 시점에서의 glutamine, arginine, leucine 등 다수 아미노산의 고갈이 관찰되었고, 이후 세포활성이 급속히 저하되었다. 이때 glucose 농도, lactate, armmonia 등의 대사산물 농도 및 pH, DO, 교반속도 등은 적절하게 유지되었기 때문에 이들 요소에 의한 세포성장의 저해는 없었다. 따라서 이들 아미노산의 고갈이 세포활성의 저하에 커다란 영향을 주었다고 생각된다. 한편, 고갈된 아미노산을 첨가한 배지(GC-HY-S2)로 배양을 해본 결과 세포농도를 $2.91\times10^7$cell/mL까지 증가시킬 수 있었다. 즉, 변형배지에서는 세포활성의 저하를 가져오는 세포성장구간을 지나 계속적인 세포성장을 보여 27배 정도 더 높은 최대 세포농도를 보였다. 세포농도의 증가로 specific productivity, volumetric productivity도 각각 1.75배, 56배 증가하였다. 최대 세포농도에서의 아미노산 량을 조사한 결과를 보면 충분한 양의 아미노산이 공급되었음을 알 수 있다. 고농도 하이브리도마세포 배양에서의 일부 아미노산의 고갈은 일반적으로 관찰되는 현상이다. 그러므로 세포를 고농도로 배양하기 위해서는 아미노산 분석을 통해 고갈된 아미노산을 첨가해 주어야 하는데, 아미노산의 요구는 세포주마다 다르므로 배지 최적화과정에서 아미노산 분석에 의한 배지의 개선이 필요하다고 생각된다.

  • Due to the increase of the number of farmers who have entrepreneurial intentions and the expansion of governmental support to invigorate the regional economy, the interest in manufacture of high-quality agricultural processed food is increasing, and the study of the method to realize it efficiently is continuing. Especially, the method to visualize a manufacturing process by simulation and analyze it to extract an improved scheme has advantages of saving time and cost in comparison with trial and error method. In this study, the current manufacturing environment was analyzed by applying simulation on Korean traditional sweets, which is typical agricultural processed food. By extending this kind of method into small businesses processing other agricultural products, as well as Korean traditional sweets, it can draw strengthening of comparativeness through the improvement of productivity and product quality.

A STUDY ON SELECTING OPTIMAL HAUL ROUTES OF EARTHMOVING MACHINE

  • Earthmoving equipment's haul-route has a great influence on the productivity of the earth work operation. Haul-route grade is a critical factor in selecting the haul-route. The route that has low grade resistance contributes to increase machine travel speed and production. This study presents a mathematical model called "Hauling-Unit Optimal Routes Selecting system" (HUORS). The system identifies optimal path that maximize the earth-work productivity. It consists of 3 modules, i.e., (1) Module 1 which inputs site characteristic data and computes site location and elevation using GIS(Geographical Information System); (2) Module 2 which calculates haul time; (3) Module 3 which displays an optimum haul-route by considering the haul-route's gradient resistances (i.e., from the departure to the destination) and hauling time. This paper presents the system prototype in detail. A case study is presented to demonstrate the system and verifies the validity of the model.

생육속도 및 배양온도가 효모 RNA 축적과 autolysis 효율에 미치는 영향 (Effect of Growth Rate and Cultivation Temperature on the Yeast RNA Accumulation and Autolysis Efficiency)

  • 배양환경이 효모 RNA 축적과 autolysis 효율에 미치는 영향을 알아보기 위하여 연속배양을 실시하였다. Dilution rate의 증가에 따라 균체내의 RNA 함량은 증가하여 dilution rate=0.35 $h^{-1}$에서 14.8%를 나타내었고, 더불어 RNA productivity와 specific RNA productivity도 증가하였다. 고정된 D=0.2 $h^{-1}$에서 최적 균체량은 배양온도 $30^{\circ}C$ 에서 얻어졌으나, RNA 함량은 배양온도가 낮아질수록 높은 값을 보여 $24^{\circ}C$에서 12.7%로 나타났다. 한편, autolysis 효율 측면에서 균체 생육속도가 증가할수록 효모 extract의 추출율은 감소하였고, 효모 extract의 분해율은 증가하는 경향을 보였으나, 배양온도는 효모 autolysis 효율에 큰 영향을 미치지 않았다.

Effects of Bring Your Own Device (BYOD) Attributes on Work-to-life Conflict

  • The rapid adoption of smartphones and tablets among employees has recently forced organizations to proactively embrace bring your own device (BYOD). The situation of COVID-19 makes the concept of BYOD even paramount. Allowing employees to bring their own mobile devices to the workplace has helped companies realize productivity gains and cost benefits. However, BYOD has also blurred the boundaries between work and personal life, thereby creating a stressful environment for employees (Doargajudbur and Hosanoo, 2023). This study explores the relationships between several BYOD attributes and investigates the effects of these attributes on work-to-life conflict. It contributes by introducing workplace connectivity after hours as a new dimension of BYOD attribute to influence work-to-life conflict. Based on boundary theory, a theoretical model is developed and tested with an online survey. The results reveal that flexibility and workplace connectivity after hours positively influence productivity, and flexibility helps to relieve instead of increase work overload. Meanwhile, a higher level of flexibility and productivity can help reduce work-to-life conflict, and a higher level of work overload induces a higher level of work-to-life conflict.

Micromonospora purpurea에 의한 gentamicin생성 (Production of Gentamicin by Micromonospora purpurea)

  • Using Micromonospora strain, gentamicin was produced by fermentation. The increase in gentamicin productivity was studied by strain improvement and systematic optimization of fermentation process variables. The productivity of parent strain of M.purpurea (ATCC15835) was improved by selection of superior mutant after U.V. irradiation and induction of genetamicin resistance. Potato starch and soy bean meal were the best carbon and nitrogen sources for gentamicin fermentation, respectively. The optimum stimulating concentration of Co ion for gentamicin production was 0.006g $CoCl_2$ per liter of broth. Oxygen ws found to be an important factor for gentamicin yield. The optimum pH for the cell growth and gentamicin production were 7.2 and 6.8 respectively. By controlling the pH, oxygen, and other conditions found in this study at the optimal conditions for cell growth and gentamicin production, the total productivity of gentamicin was increased significantly.

  • For the merit of a container in transportation and cargo handling the amount of container freight has been continuously on the increase. On the other hand container crane had got the bottle neck in cargo handling due to low productivity so that freight congestion had been often occurred at ports. in this paper A mathematical model for container crane system is represented a method for designing a fuzzy controller of container crane system for high productivity in cargo handling is presented. The fuzzy controller is compared with other optimal controller at the same condition. in the computer simulation the fuzzy controller obtained an excellent response to reference change better than the optimal controller. For disturbance such as a strong storm and parameter change due to change of cargo weight the result was also stable and robust than the optimal controller.

  • Traditional CVP (Cost-Volume-Profit) analysis employs linear cost and revenue functions within some specified time period and range of operations. Therefore CVP analysis is assumption of constant labor productivity. The use of linear cost functions implicity assumes, among other things, that firm's labor force is either a homogenous group or a collection homogenous subgroups in a constant mix, and that total production changes in a linear fashion through appropriate increase or decrease of seemingly interchangeable labor unit. But productivity rates in many firms are known to change with additional manufacturing experience in employee skill. Learning curve is intended to subsume the effects of all these resources of productivity. This learning phenomenon is quantifiable in the form of a learning curve, or manufacturing progress function. The purpose d this study is to show how alternative assumptions regarding a firm's labor force may be utilize by integrating conventional CVP analysis with learning curve theory, Explicit consideration of the effect of learning should substantially enrich CVP analysis and improve its use as a tool for planning and control of industry.

  • To determine the effect of ERp57 expression on thrombopoiein (TPO) productivity in recombinant Chinese hamster ovary (rCHO) cells, TPO producing rCHO cell line with doxycycline-regulated ERp57 expression \Vas developed. The Erp57 expression level could be regulated by addition of different concentrations of doxycycline to culture medium. The doxycycline concentration of I ${\mu}g/mL$ was high enough to suppress the ERp57 expression. Up to 5 ${\mu}g/mL$ doxycycline concentration used in culture medium, no observable cytotoxic effect of doxycycline was detected during culture. Overexpression of ERp57 、 vas found to increase the specific TPO productivity ($q_{Tpo}$) without growth inhibition, probably due to the chaperone-like activity of ERp57 in CHO cells.

  • Many researches and projects are actively implemented in the academic world and industrial world in order to increase the productivity through changing utility layout. Effective layout makes the flow of parts and products go on wheels and helps labor forces and utilities to be used efficiently. In this study, we looked at effect on utility layout change through a case of a company making a fuel tank of vehicle in Korea. Due to the layout improvement, this company could have flowshop system on welding line, so the stock in line was decreased, checking the number of stock became easier than before and it was possible to evaluate exact cycle time per unit. Through not only confirming qualitative effect, but also checking quantitative effect such as UPH(Unit per Hour), we obtained the conclusion that the layout improvement increased the productivity.
