Effect of Replacing Corn and Wheat Bran With Soyhulls in Lactation Cow Diets on In Situ Digestion Characteristics of Dietary Dry Matter and Fiber and Lactation Performance

  • An in situ digestion trial (Experiment 1) and a lactation trial (Experiment 2) were conducted to determine the effects of replacing corn and wheat bran with soyhulls (SH) in lactating dairy cow diets on the extent and kinetics of digestion of DM and NDF, and lactation performance. In experiment 1, five mixed feeds consisting of mixed concentrate and roughages (50:50 on a DM basis) were formulated on isonitrogenous and isoenergetic bases to produce five levels (0, 25, 50, 75 and 100%) of SH replacement for corn and wheat bran. SH had high in situ digestion (92 and 89% for potentially digestible DM and NDF) and fairly fast digestion rate (7.2 and 6.3 %/h for DM and NDF). Increasing level of SH replacement resulted in increased NDF digestibility (linear, p=0.001-0.04) and similar DM digestibility (beyond 12 h incubation, p=0.10-0.41). As level of SH replacement increased, percentage of slowly digestible fraction (b) of DM increased (linear, p=0.03), percentage of rapidly digestible fraction (a) of DM tended to decrease (linear, p=0.14), and DM digestion lag time tended to be longer (linear, p=0.13). Percentage of potentially digestible fraction (a+b) and digestion rate (c) of slowly digestible fraction of dietary DM remained unaltered (p=0.36-0.90) with increasing SH in the diet. Increasing level of SH for replacing corn and wheat bran in the diet resulted in increases in percentages of b (quadratic, p<0.001), a (linear, p=0.08), a+b (quadratic, p=0.001) and a tendency to increase in c for NDF (linear, p<0.19). It was also observed that there was a satisfactory fit of a non-linear regression model to NDF digestion data ($R^2=0.986-0.998$), but a relatively poor fit of the model to DM digestion data ($R^2=0.915-0.968$). In experiment 2, 42 lactating Holstein cows were used in a randomized complete block design. SH replaced corn and wheat bran in mixed concentrates at 0, 25, and 50%, respectively. These mixed concentrates were mixed with roughages and fed ad libitum as complete diets. Replacing corn and wheat bran with SH at 0, 25 and 50% levels did not influence (p=0.56-0.95) DM intakes (18.4, 18.6, and 18.5 kg/d), milk yields (27.7, 28.4 and 27.6 kg/d), 4% fat-corrected-milk (FCM) yields (26.2, 27.6, and 27.3 kg/d) and percentages of milk protein (3.12, 3.17 and 3.18%), milk lactose (4.69, 4.76 and 4.68%) and SNF (8.50, 8.64, and 8.54%). On the other hand, milk fat percentges linearly increased (3.63, 3.85 and 3.90% for SH replacement rates of 0, 25 and 50% in the diet, p=0.08), while feed costs per kg FCM production were reduced.

  • Anti-reflection properties of post diffusion doped spin-on source (in-situ AR coating) have been investigated in some detail. A simple experiment for reflectivity study using oblique incidence of light and necessary modification of the theory of minimum reflectivity at oblique incidence has been established. The comparative study of the in-situ AR coating with available spin-on AR film on silicon Solar Cell Surface have been investigated.

  • In this presented study, we established a method for diagnosis of porcine epidemic diarrhea(PED) by in situ hybridization(ISH), which made distinct progress in diagnostic pathology. We also carried out the retrospective diagnosis through ISH to assume the exact time of the first outbreak and incidence of PED in Korea. The outbreak of PED in Korea reported in 1992. However, since the end of 1980's, some researches of pig-industry have already suspected the outbreak of PED, not transmissible gastroenteritis(TGE). In this experiment, we performed the ISH using 80 formalin-fixed and paraffin-embedded tissues of porcine intestine which were requests for pathological diagnosis from 63 farms whose primary sign was diarrhea from 1984 to 1991. We prepared biotinylated cDNA probe(492base pairs) for ISH by nick translation method and carried out the ISH, using $Microprobe^{TM}$ capillary action system(Fisher $Biotech^R$). We detected PED virus in intestinal mucosa of 2 cases in 1992, 1 case in 1988, and 1 case in 1987. As a result, we assume that the outbreak of PED in Korea have already started since 1987.

Multi-dimensional analyses of plant chromosomes and genomes.

  • Genome and chromosome analyses in plants using fluorescence in situ hybridization (FISH) and immuno-staining (IMS) methods are reviewed by presenting the recent results obtained by the Chromosome Link, a group of chromosome and genome researchers. FISH is now effective to detect unique nucleotide sequences with 153 bp on the extended DNA fibers. Genomic in situ hybridization (GISH) also allows painting plant chromosomes of different genomes. GISH is quite effective to detect the genomic differentiation in the individual chromosomes within a nucleus. Three dimensional (3D) analyses are now available by confocal microscopy and a deconvolution system. These techniques are invaluable to visualize both the structural and functional dynamics within a nucleus. 3D-FISH revealed the spatial differentiation of different genomees within a nucleus. 3D-FISH also proved structural partition of centromeric and telomeric domains within a barely nucleus. The dynamic acetylation of histone H4 at the specific regions of a genome during a cell cycle is also analyzed using 3D-IMS. It is anticipated that these methods will provide us powerful tools to understand the structural and functional significance of plant chromosomes and genomes.

  • Fracture analysis of coating cracking on a substrate system described in a companion paper was applied and verified by four-point bending tests. The multiple cracking of coating was predicted using a fracture mechanics approach. The strain energy release rate (G) due to the formation of a new crack in a coating was obtained. A crack density vs. strain data of metallic and polymeric substrate was used to get the in-situ fracture toughness of coating with respect to various baking time and temperature. The $G_c$ was decreased as the baking temperature and time was increased. This paper gave insight about usefulness of four-point bending test for fracture toughness evaluation of coating and it gave a new method for in-situ coating toughness.

  • The widening project on Freeway No.1 in Taiwan has a total length of roughly 14 kilometers, and includes three special bridges, namely a 216 m long-span bridge crossing the original freeway, an F-bent double decked bridge in a co-constructed section, and a steel and prestressed concrete composite bridge. This study employed in-situ monitoring in conjunction with numerical modeling to establish a real-time monitoring system for the three bridges. In order to determine the initial static and dynamic behavior of the real bridges, forced vibration experiments, in-situ static load experiments, and dynamic load experiments were first carried out on the newly-constructed bridges before they went into use. Structural models of the bridges were then established using the finite element method, and in-situ vehicle load weight, arrangement, and speed were taken into consideration when performing comparisons employing data obtained from experimental measurements. The results showed consistency between the analytical simulations and experimental data. After determining a bridge's initial state, the proposed in-situ monitoring system, which is employed in conjunction with the established finite element model, can be utilized to assess the safety of a bridge's members, providing useful reference information to bridge management agencies.

  • In order to acquire a realistic forecast for the lifetime and post-closure period of the LILW (Low- and Intermediate-Level Radioactive Waste) repository and to establish the overall management plan associated gas issues. it is essential to carry out the long-term experimental research in a similar condition to actual disposal environment. Regarding this, as a part of the following-up actions on a construction and operation license for the first stage of the LILW repository at Gyeongju city, a large-scale in-situ experiment is being planned. For securing basic data on the experiment, the experimental researches related to gas generation previously performed in foreign countries are reviewed in detail. Consequently, it is judged that data on the gas generation experiment in Finland could be practically applied as the benchmark for our large-scale in-situ experiment because the same disposal concept as the Korean repository is adopted and the experiment is performed in a scale large enough to allow the use of regular waste packages.
