Effects of Nitrifying Bacterial Communities with Different HRTs and Backwashing Periods in Modified BAF Process (Modified BAF 공정에서 HRT 및 역세주기가 질산화 미생물의 군집에 미치는 영향)

  • The upflow Biobead$^{(R)}$ process, one of biological aerated filters (BAF), which was used commercially, invented for removal of organic materials and nitrification. This process was modified to enhance the ability of denitrification through the induction of pre-anoxic tank. In this research, we investigated the effects of hydraulic retention time (HRT) and backwashing period in aerobic tank. The characteristics of nitrifying bacteria, which are composed of ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (NOB), also investigated using fluorescence in situ hybridization (FISH). Even though the HRT was shortened, the efficiency of nitrification was not decreased when the organic loading rate and ammonium-nitrogen loading rate were $2.10kg/m^3/day$ and $0.25kg/m^3/day$, respectively. And then the distribution ratios of AOB and NOB showed the similar patterns. However, when the backwashing period was lengthened from 12 hours to 24 hours in aerobic 1 tank, the nitrification efficiency was decreased to 63.9% from 89.2%. The results of FISH explained that this decrease of nitrification efficiency was caused by the decrease of distribution ratio of AOB in aerobic 1 tank. The nitrification efficiencies of aerobic 1 and aerobic 2 tank were increased when the backwashing period was lengthened because of relative high distribution ratios of nitrifying bacteria.

  • The aim of this study was to provide basic guidelines for conservation and management of endangered plants in the national parks of Korea. Iris dichotoma Pall. (Iridaceae), which is a popular garden plant, is considered a second-class endangered species by Korean government and it is listed as a EN (Endangered) species in Red Data Book of Korea. We analyzed ecological conditions of I. dichotoma habitats based on vegetation properties and soil characteristics. This species which is known to inhabit in grassland adjacent to the ocean of lowlands slope and its population was located at an elevation of 8 m to 11 m. In the study sites, the mean of soil organic matter, total nitrogen and soil pH were 6.16%, 0.234% and 5.39 respectively. Additionally, the genetic variation and structure of three populations were assessed using ISSR (Inter Simple Sequence Repeat) markers. The genetic diversity of I. dichotoma (P = 59.46%, H = 0.206, S = 0.310) at the species level was relatively high. Analysis of molecular variance (AMOVA) showed 82.1% of the total genetic diversity was occurred in within populations and 17.9% variation among populations. Lastly, we developed predicted distribution model based on climate and topographic factors by applying SDMs (Species Distribution Models). Consequently, current status of I. dichotoma habitats is limited with natural factors such as the increase of the coverage rate of the herbs due to ecological succession. Therefore, it is essential to establish in situ and ex situ conservation strategies for protecting natural habitats and to require exploring potential and alternative habitats for reintroduction.

Characterization of Polyurethane and Soil Layers for In-situ Treatment of Landfill Leachate (매립지 침출수 현장 처리를 위한 폴리우레탄과 개질토의 특성 분석 실험에 관한 연구)

  • A chemical and biological permeable barrier with economic feasibility is suggested to treat landfill leachate in this study. The proposed composite layers consist of bentonite, and polyurethane (PU) foam that is mixed with powdered activated carbon (PAC) and inoculated with microorganisms from local wastewater treatment plant. Each layer is mixed with local sand, and yellow brown soil. Batch tests were conducted to investigate the sorptions of nitrate on the PU foam and PAC, and nitrification/denitrification rate of each layer material. Nitrification occurred in 30 minutes with initial ammonia concentration of 100 mg/L, and the concentration of nitrate attached in the PU foam increased after 270 minutes. Results of denitrification batch tests showed 76.6%, 87.3% and 88% of nitrate removal efficiency at 10%, 20% and 30% of the volume ratio of PU foam, respectively. The pH increased from 7 to 9.42, and alkalinity increased from 980 mg/L to 1720 mg/L during the denitrification batch tests. In the column experiments using the proposed composite layers with 20% of the volume ratio of the PU foam, about 96% of BOD, 63% of COD, 58.1~79.5% of total nitrogen were removed.

In situ Structural Investigation of Iron Phthalocyanine Monolayer Adsorbed on Electrode Surface by X-ray Absorption Fine Structure

  • Structural changes of an iron phthalocyanine (FePC) monolayer induced by adsorption and externally applied potential on high area carbon surface have been investigated in situ by iron K-edge X-ray absorption fine structure (XAFS) in 0.5 M $H_2S0_4.$ Fine structures shown in the X-ray absorption near edge structure (XANES) for microcrystalline FePC decreased upon adsorption and further diminished under electrochemical conditions. Fe(II)PC(-2) showed a 1s ${\rightarrow}$ 4p transition as poorly resolved shoulder to the main absorption edge rather than a distinct peak and a weak 1s ${\rightarrow}$ 3d transition. The absorption edge position measured at half maximum was shifted from 7121.8 eV for Fe(lI)PC(-2) to 7124.8 eV for $[Fe(III)PC(-2)]^+$ as well as the 1s ${\rightarrow}$ 3d pre-edge peak being slightly enhanced. However, essentially no absorption edge shift was observed by the 1-electron reduction of Fe(Il)PC(-2), indicating that the species formed is $[Fe(II)PC(-3)]^-$. Structural parameters were obtained by analyzing extended X-ray absorption fine structure (EXAFS) oscillations with theoretical phases and amplitudes calculated from FEFF 6.01 using multiple-scattering theory. When applied to the powder FePC, the average iron-to-phthalocyanine nitrogen distance, d(Fe-$N_p$) and the coordination number were found to be 1.933 $\AA$ and 3.2, respectively, and these values are the same, within experimental error, as those reported ( $1.927\AA$ and 4). Virtually no structural changes were found upon adsorption except for the increased Debye-Wailer factor of $0.005\AA^2$ from $0.003\AA^2.$ Oxidation of Fe(II)PC(-2) to $[Fe(III)PC(-2)]^+$ yielded an increased d(Fe-Np) (1 $.98\AA)$ and Debye-Wailer factor $(0.005\AA^2).$ The formation of $[Fe(II)PC(-3)]^-$, however, produced a shorter d(Fe-$N_p$) of $1.91\AA$ the same as that of crystalline FePC within experimental error, and about the same DebyeWaller $factor(0.006\AA^2)$.

  • Thermal behavior of residue and damaged layer formed by reactive ion etching (RIE) in $CHF_3/C_2F_6$ were investigated using X-ray photoelectron spectroscopy(XPS) and secondary ion mass spec-trometry(SIMS) techniques. Decomposition of polymer residue film begins at $200^{\circ}C$ and above $400^{\circ}C$ carbon compound as graphite mainly forms by in-situ resistive heating. It reveals that thermal decomposition of residue can be completed by rapid thermal anneal treatment above $800^{\circ}C$ under nitrogen atmosphere and out-diffusion of carbon and fluorine of damaged layer is observed.

Analysis of Microbial Community Structure for Effective Removal of Mixed Wastewater in Biological Wastewater Treatment (혼합폐수의 효율적인 처리를 위한 생물학적 처리공정 내의 미생물 군집 특성 분석)

  • Depending on season, mixed wastewater can show great deviations in terms of the influent ratios of tannery and seafood-wastewater. Increases in the ratio of tannery wastewater in influent water also result in increases in the concentration of chromium, which decreases the ratio of BOD/T-N so that the removal efficiency of organic and nitrogen pollutants in biological wastewater treatment deteriorates. No substantial differences occur in the ratios of Eubacteria/total bacteria as the ratio between tannery wastewater and seafood wastewater changes in the influent water. In contrast, the cell numbers and activities of Eubacteria and total bacteria significantly decline with increasing ratios of tannery wastewater in the influent water. Stable removal of organic and nitrogen pollutants by biological wastewater treatments leads to dominance of Proteobacteria groups in all biological treatment basins. In aeration and oxic basins, ${\gamma}$-Proteobacteria account for approximately 21% of the Eubacteria groups, at $1.9{\times}10^9{\sim}2.0{\times}10^9$ cells/mL, while in an anoxic basin, ${\beta}$-Proteobacteria account for approximately 19% of the Eubacteria groups, at $1.3{\times}10^9$ cells/mL. However, a substantial decline in dominance of approximately 11% occurs for ${\gamma}$-Proteobacteria in aeration and oxic basins and about 1% for ${\beta}$-Proteobacteria in an anoxic basin. Mixed wastewater that undergoes extensive property changes of the influent water shows an efficiency of biological treatment that is greatly influenced by the ratio of dominant Proteobacteria groups.

Seasonal Variations of Marine Water Quality and Eutorphication Index in Mokpo Harbour (목포항의 수질 및 부영양도의 계절 변화)

  • The in situ observations and the seawater analyses were conducted for 3 years from August 1996 to August 1999 in order to elucidate and evaluate seasonal variations of marine water quality and trophic state in Mokpo harbour of Korea. Compared the seasonal seawater qualities of Mokpo harbour with the OECD standards of trophic classification in parameters such as Secchi depth, dissolved inorganic nitrogen, phosphate phosphorus and chlorophyll-a, the trophic level of seawater in Mokpo harbour was evaluated to be in mesotrophic or eutrophic state through all 4 seasons and to be in eutrophic state, particularly In summer. The estimation of pollution index by eutrophication showed the seawater quality of Mokpo harbour to deteriorate and fall under the regular grades through all 4 seasons, although the seawater quality of Mokpo harbour was evaluated to be equivalent to the second or third class of the Korean seawater quality standards in view of COD values. The results of eutrophication index estimation showed the high potentiality of red tide occurrence In Mokpo harbour, particularly in summer or fall. In the light of the average atomic ratio of N/P in seawater, the limiting nutrient factor against the growth of phytoplankton was concluded to be phosphorus rather than nitrogen in Mokpo Harbour.

Effects of Cryopreservation on the Seed Germination and Growth Properties of Seedlings of Maackia amurensis (초저온 저장이 다릅나무 종자의 발아와 유묘의 생장특성에 미치는 영향)

  • This study was conducted to investigate effects of cryopreservation by vitrification on the seed germination rate and growth and physiological properties of seedlings of Maackia amurensis. Cryopreservation significantly decreased the germination rate of seeds of M. amurensis, but the reduction of germination rate was mitigated by the treatment of cryoprotectant (plant vitrification solution, PVS2) before plugging into liquid nitrogen and fast thawing rate after cryopreservation. Long-term PVS2 exposure decreased seed germination rate, whereas cryopreservation time didn't have influence on seed germination rate. In addition, growth and physiological properties of seedlings were not affected by PVS2 exposing time and cryopreservation time. Therefore cryopreservation could be widely used as a technique of long-term ex situ conservation without any damage and deterioration of cells or tissues of the forest seeds. However, in order to increase the effect of cryopreservation, we have to develope the lower toxic cryoprotectant and suitable techniques to the structural or chemical properties of a variety of seeds.

Distributions and Sources of Dissolved Organic Matter in Seawaters Surrounding Aqua Farms on the Haengwon-ri in Jeju-Island in Summer 2015 (2015년 하계 제주 행원리 일대 양식장주변 해수 중 용존유기물 분포와 기원)

  • Concentrations of dissolved organic carbon (DOC), dissolved organic nitrogen (DON), dissolved organic phosphate (DOP), and colored dissolved organic matter (CDOM) were measured in the coastal sea off inland aqua farms in northeastern Jeju Island in summer 2015. The highest concentrations of DOC, DON, and DOP were revealed in the surface water off Hado-ri where the lowest salinity conditions prevailed (31.6). The concentrations of DOC, DON, and DOP in the surface water were lower in the inner stations (SH1-1, 1-2, and 1-3) near the aqua farms of the Haengwon-ri than in the outer stations. The concentrations of DOC, DON, and DOP negatively correlated with salinity. These results indicate that the contribution of dissolved organic matter (DOM) from the aqua farms seems to be not significant. On the other hand, the higher concentrations of DON and DOP in the inner stations of Hado-ri (HD 1-1) seem to be attributed to excrement of migrating birds. The three components of CDOM (T, M, and C peaks) showed no relationship with salinity, perhaps due to various in situ productions by marine organisms and decomposition by ultraviolet radiation. The observed lower C:M ratio, an indicator of terrestrial source, and the higher biological index (BIX) of CDOM in the station off Hado-ri indicate that DOM is produced mainly by biological activity. Based on the higher humification index (HIX) of CDOM and the higher DOC:DON ratio off Haengwon-ri, refractory DOM in the inland aqua farms is likely transported to the coastal sea.

Kimchi cabbage (Brassica rapa L.) by-products treated with calcium oxide and alkaline hydrogen peroxide as feed ingredient for Holstein steers

  • This study aimed to investigate the effects of Kimchi cabbage by-products either treated or untreated with calcium oxide (CaO) and alkaline hydrogen peroxide (AHP) as substitutional ingredient of total mixed ration (TMR) on in vitro fermentation, in situ disappearance and growth performance of Holstein steers. Cannulated Holstein (600 ± 47 kg) was used for both the in vitro and in situ experiments. The treatments used were TMR only (CON), TMR + 30% Kimchi cabbage by-products fresh matter (FM) basis (TC), TMR + 30% Kimchi cabbage by-products FM basis + 5% CaO FM basis (TCC), and TMR + 30% Kimchi cabbage by-products FM basis + 5% CaO FM basis + 3.22% AHP FM basis (TCCA). For in vivo experiment, thirty-four Holstein steers (273 ± 45 kg) were subjected to a 150-day feeding trial, divided into two groups: CON and TC. In the in vitro experiment, pH of TCCA was greatest (p < 0.05) among other treatments at all incubation times. Ammonia nitrogen and volatile fatty acid concentrations were not significantly different for each treatment. However, butyrate was greater (p < 0.05) in TCC and CON than in both TC and TCCA. During in situ experiment, the dry matter (DM) disappearance was greatest (p < 0.05) in TCCA among other treatments. Also, disappearance of neutral detergent fiber (NDF) and acid detergent fiber (ADF) were observed greatest (p > 0.05) in TCCA treatment. In the in vivo experiment, average daily gain (ADG) did not differ between CON and TC. In blood profile analysis, alanine aminotransferase, aspartate aminotransferase, glucose, total cholesterol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and total protein concentration were not significantly different between treatments. But, creatinine concentration was greater (p < 0.05) in TC than in CON. Overall results suggest that Kimchi cabbage by-products either treated or untreated with CaO and AHP can be used as substitutional ingredient in TMR for Holstein steers.
