산소농도 및 Superoxide Dismutase가 돼지 난포란의 체외성숙과 배발달에 미치는 영향 (Effect of Oxygen Concentrations with Superoxide Dismutase on In Vitro Maturation of Porcine Follicular Oocytes and In Vitro Development of Porcine IVM/IVF Embryos)

  • 본 연구는 돼지 난포란의 체외성숙배양액에 superoxide dismutase(SOD)를 첨가배양하여 체외성숙과 수성이후 배발달에 미치는 영향과 산소농도 및 SOD가 체외생산된 돼지초기수정란치 배발달에 미치는 영향을 규명하고자 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1. 돼지 난포란을 체외성숙배양액인 NCSU-23에 SOD를 각각 0, 100,500 및 1,000uni1s/m1 첨가하여 성숙시킨 다음, 체외수정을 실시한 결과, 핵성숙률, 정자침입율, 다정자침입률 및 평균침입정자수는 처리구와 대조구간 유의적(P<0.05)인 차이가 인정되지 않았고, 웅성전핵형성률은 처리구(90.9∼81.2%)보다 오히려 대조구(95.0%)가 유의적으로 높은 결과를 나타냈다(P<0.05). 2. 체외수정 후, 배발달배양액인 NCSU-23에 7일 동안 배양한 결과, 배반포형성률과 총세포수는 처리구가 대조구보다 유의적(P>0.05) 으고 낮은 결과를 나타냈다. 3. 배발달배양액인 NCSU-23에 SOD를 각각 0, 100, 500 및 1,000uni1s/m1을 첨가하고 5% 및 20% 산소조건하에서 7일동안 배발달을 유기한 결과, 배양 7일째 산소조건에 따른 처리구간 배반포형성률 및 총세포수는 유의차가 없었으며, SOD농도별 배반포형성률은 100 units/ml 처리구에서 0, 500 및 1,000uni1s/m1 처리구 보다 유의적으로 높은 배반포형성률을 나타냈으나, 총세포수에서는 처리구(27.6∼37.3개)가 대조구(45.6개)보다 유의적(P<0.05) 로 낮은 결과를 나타내었다. 따라서 NCSU-23배양액에 SOD의 첨가는 돼지난포란의 체외성숙과 초기배발달에 영향을 미치지 않는 것으로 조사되었으며, 5% 및 20% 산소농도에 따른 영향도 없는 것으로 사료된다.

체외성숙 조건이 소 체외수정란의 체외발달에 미치는 영향 (Effects of In vitro Maturation Condition on Bovine IVF Embryos Development)

  • This study was performed to improve the development of the in vitro fertilized bovine embryos by the condition of in vitro maturation. COCs were matured in TCM 199 supplemented with 0.1% PVA, 10ng/ml EGF, Hormones (5$\mu\textrm{g}$/ml FSH, 10 IU hCG, 1 $\mu\textrm{g}$/ml estradiol 17-$\beta$) or granulsa cell+Hormones atmosphere 39$^{\circ}C$, 5% CO2, 95% air for 24hrs. Matured oocytes were fertilized with frozen-thawed semen capacitated with 5mM caffein in BO medium for 20 hrs. IVF embryos were cultured in TCM 199 containing with hormones(same as matured medium), 10% FBS and co-culture with bovine oviduct epitherial cells. Maturation rates of COCs were showed 73.8%, 78.5%, 83.2% and 87.6% respectively, and were significant differences between PVA, EGF, and Hormones, GC+Hormones(p<0.05). The cleavage rates of IVF embryos were revealed 72.5%, 78.4%, 82.3% and 84.2% and showed same tendency as maturation rates(p<0.05). The blastocysts matured by above maturation condition and cultured for 7~10 days after fertilization had 34.4, 43.6, 52.3 and 59.3 cells had no differences among the treatments. These results suggest that high molecules as a substitutes of serum and growth factor may induce nuclear resumption of COCs but we need more study to produce transferable IVF blastocysts by use of that agents.

난구, 난관 상피세포 및 자궁 내막세포와의 공동배양이 돼지 난포란의 체외수정 및 분할율에 미치는 영향에 관한 연구 (Studies on the Effects of Co-Culture with Cumulus Cells, Oviduct Epithelial Cells and Uterine Endometrial Cells on in-vitro Fertilization and Cleavage Rate of Porcine Oocytes)

  • The studies were carried out to investigate the effects of co-culture with cumulus cell, oviduct epithelial cells and uterine endometrial cells on the in-vitro fertilization and cleavage rate of porcine follicular oocytes. The ovaries were obtained from slaughtered swine. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluids from the visible follicles of diameter 3~5 mm. The follicular oocytes were cultured in TCM-199 medium containing hormones and 10% FCS for 24~48 hrs in a incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18 hrs with motile capacitated sperm by preincubation. The results obtained in these experiments were summarized as follows : 1. The in-vitro maturation and fertilization rate of porcine oocytes co-cultured with cumulus cells in TCM-199 meidum were 64.6%~74.5% and 37.5%~55.3%, respectively. And in-vitro fertilization rate of cumulus-enclosed oocytes(51.5%) were significantly(p<0.05) higher than cumulus-denuded oocytes(21.7%). 2. The in-vitro maturation and fertilization rate of porcine oocytes co-cultured with 1$\times$104 cells/ml, 1$\times$106 cells/ml, 1$\times$108 cells/ml and 1$\times$1015 cells/ml oviduct epithelial cells in TCM-199 medium were 53.5% and 37.2%, 61.7% and 46.8%, 54.5% and 31.8%, 42.2% and 26.7%, respectively. 3. The in-vintro maturation and fertilization rate of porcine oocytes co-cultured with 1$\times$106/ml, 1$\times$108/ml, 1$\times$1015/ml uterine endometrial cells in TCM-199 medium were 54.3% and 39.1%, 58.3% and 43.8%, 55.5% and 33.3%, and 45.7% and 30.4%, respectively. 4. When the in-vitro fertilized oocytes were co-cultured with porcine cumulus cells, ovdiduct epithelial cells and uterine endometrial cells, the development rate to the blastocyst stage was 9.5%, 10.7% and 11.8%, respectively and the rates were higher than that of control, 2.1%(p<0.05).

Participation of Protein Synthesis in in vitro Oocyte Maturation and Fertilization in Cattle

  • Bovine oocytes with compact and complete cumulus cells were cultured for up to 24h in TCM199 buffered with 25mmol/l hepes and supplemented with 10% FBS (fetal bovine serum), 1mg/ml $17{\beta}$-estradiol, 20IU/ml hCG(human chorionic gonadotropin). All of the oocytes were divided into at 6 groups depending upon incubation times (control, 0 hour, 6 hours, 12 hours, 16 hours, 18 hours). To all experimental media, $200{\mu}g/ml$ puromycin was added at different incubation times mentioned above. Following these culture times, in vitro insemination was conducted with frozen-thawed bovine spermatozoa in medium BO (Brackett and Oliphant medium for in vitro insemination) with $10{\mu}g/ml$ BSA(bovine serum albumin) and 10 mg/ml heparin added. After 22h culture, the oocytes were fixed with acetic alcohol solution and stained with orcein acetic solution to evaluate sperm nuclear progression. Addition of puromycin after 0, 6 and 12 h of culture resulted in near of oocyte maturation at the M1 stage. Contrariwise, puromycin addition after 12 h of culture led to restoration of nuclear progression to M2 stage. On the one hand, puromycin affected the synthesis of Cyclin B protein that may be involved in the oocyte maturation and sperm capacitation for in vitro fertilization. The present study suggests the participation of protein synthesis, cyclin B, in the oocyte development from M1 to M2 stages in vitro.

Heat waves impair cytoplasmic maturation of oocytes and preimplantation development in Korean native cattle (Hanwoo)

  • There has been widespread warming and a general increase in summer temperatures over the Korean peninsula ($0.5^{\circ}C$/10 years from 2001 to 2010). South Korea is transforming into a subtropical region, and the productivity of livestock is affected by the climatic changes. In this study, we investigated whether the summer heat waves affect the developmental competency of Korean native cattle (Hanwoo), a taurine type of cattle with a small portion of indicine varieties. We collected oocytes during the summer (heat stress, HS) and autumn (non-HS condition) and examined the developmental competencies including in vitro maturation and preimplantation embryo development. No significant differences were observed between the HS and non-HS oocytes in nuclear maturation (extrusion of the polar body); however, the cleavage and blastocyst rates were significantly lower in the HS group than those in the non-HS group. The lower developmental competence of the HS oocytes compared to the non-HS is, in part, due to insufficient cytoplasmic maturation because of a higher production of Reactive oxygen species (ROS) levels as well as peri/cortical distributed mitochondria in the HS oocytes after in vitro maturation. Next, we examined the ROS and mitochondria distribution and found a significant increase in the levels of ROS in the HS oocytes and a polarized distribution (pericortical cytoplasm) of mitochondria in the HS oocytes. In summary, impaired cytoplasmic maturation of oocytes from exposure to HS affects the preimplantation embryo development by dysfunction of mitochondria. To improve reproductive performance, embryo transfer using cryopreserved embryos/oocytes is recommended in the hot summer season of South Korea.

배양중인 생쥐 난자의 성숙에 미치는 dbcAMP 및 Progesterone의 동시영향에 관한 연구 (Studies on Synergistic Effect of dbcAMP and Progesterone on the Maturation of Mouse Oocytes in vitro)

  • 생쥐난자의 성숙을 억제하는 것으로 알려진 dbcAMP와 progesterone을 동시에 처리하였을 때의 영향을 알아보기 위하여 본 실험을 행한 결과는 다음과 같다. 1) 10 $\\muM$/ml의 dbcAMP 혹은 2 $\\muM$/ml의 progesterone을 단독처리하였을 때에는 난자의 성숙을 억제하지 못했다. 2) 그러나 dbcAMP(10 $\\mug$/ml)와 progesterone(2 \\mug$/ml)을 동시에 배양액에 처리하였을 때에는 난자의 성숙이 억제되었다. 3) 난자를 먼저 4시간동안 위의 두 억제제가 들어있는 배양액에서 배양한 뒤 다시 정상배양액에 옮겨 계속 배양하면 나자들은 대조군에서와 같은 비율로 성숙을 하였다. 위의 결과로 보아 dbcAMP와 progesterone이 동시에 배양액에 존재할 때에는 각각 단독 존재하였을 때 영향이 없는 농도의 dbcAMP와 progesterone으로도 능히 난자의 성숙을 억제 시킬 수 있었으며 이 같은 억제효과는 가역적임을 알게되었다.

Epidermal Growth Factor가 돼지 미성숙난포란의 체외성숙에 미치는 영향 III. 체외 수정에 미치는 Epidermal Grwoth Factor의 효과 (Effect of Epidermal Growth Factor on In Vitro Maturation in Pig Immature Oocytes III. Effect of Epidermal Growth Factor on In Vitro Fertilization)

  • 본 연구는 epidermal growth factor(EGF)가 처리된 돼지 체외성숙란의 수정능력을 조사하기 위하여 실시하였다. 돼지 미성숙란의 성숙을 위해서 배양액내에 EGF(10ng/ml)와 FSH(10$\mu\textrm{g}$/ml) 또는 FBS(10%)를 단독 혹은 공동 첨가함으로서 4처리군으로 처리하였다(1:무처리군, 2:EGF 단독 처리군, 3: FSH와 FBS 공동 처리군, 4: EGF와 FSH 그리고 FBS 공동 처리군). 그 결과 2, 3, 4 처리군의 핵성숙율은 무처리군보다 현저하게 높았다(P<0.001). 수정율에 있어서 EGF 단독 처리군의 경우 3과 4 처리군보다 낮았지만, 무처리군보다는 현저하게 높았다(P<0.05). 더욱이 EGF와 FSH 그리고 FBS의 공동 처리군의 경우 다른 모든 처리군에 비하여 정자 침입뿐만 아니라 웅성 전핵 형성율이 높았다(P<0.05). 따라서 돼지 난포란의 체외성숙시 EGF 단독 처리만으로는 핵성숙에 수반된 세포질 성숙률은 낮았지만, EGF와 FSH 및 FBS를 공동 처리했을 때 돼지 체외성숙란의 세포질 성숙도 효과적으로 유도된다는 것을 알 수 있다.
