청조구폐탕의 항암효과에 관한 연구 (Effect of Chungjokupye-tang on the Anti-cancer Property)

  • The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-carcinogenic action. The cell viability of mouse spienocytes and thymocytes were enhanced by the addition of CJKPT. CJKPT were increased of splenic and thymic T lymphocytes, such as T/sub H/ cells were markedly increased by the treatment of CJKPT in vivo. CJKPT treatment induced the apoptotic cell death of Jurkat and HL60 leukemia cells. CJKPT reduced mitochondrial transmembrane potential and increased the expression of ICE, c-myc and p53 gene in Molt-4cells dose dependant manner. These results suggest that CJKPT have an anti-carcinogenic action via immunoregulatory mechanism.

Chestnut Extracts as new Anti-aging agent

  • Inner nutshell of Castanea crenata has been used as an anti-aging and anti-wrinkle agent from the ancient time in east Asia. In order to develop new anti-aging and anti-wrinkle, ethanolic extract of inner nutshell of Castanea crenata was prepared and various biological activities were evaluated. Cor-285 possessed potent tree radical scavenging activity in vitro compared to gallic acid. Cor-285 showed the preventive effect against UV-induced cytotoxicity of fibroblast at concentration of 25-250mg/ml. When Cor-285 was evaluated for its anti-allergic activity, it effectively inhibited histamine release from mast cells induced by compound 48/80. The inhibitory activity was stronger than that of glycyrrhizinate. Cor-285 also showed on vivo inhibition against delayed hypersensitivity as well as croton-oil induced ear edema in mice when topically applied. These results strongly suggest that Cor-285 may reduce immunoregulatory/inflammatory skin trouble. From the attempts to isolate the scavenger, were isolated. In a clinical trial of twenty healthy volunteers with aged skin, 6 weeks application of Cor-285 decreased wrinkle about 26% and increased moisturizing 20% on the skin. All of these results indicate that Cor-285 may be an effective anti-aging and anti-wrinkle agent.

맥문동탕의 면역조절 효과 (Effect of Maekmoondong-tang on the Immunomodulatory action)

  • The purpose of this research was to investigate the effect of Maekmoondong-tang(MMDT) on the activity of immune cell. The addition of MMDT(10 ㎍/㎖) enhanced the proliferation of cultured-splenocytes and thymocytes. And administration of MMDT(250, 500 ㎎/㎏) accelerated subpopulation of splenic T lymphocytes in BALB/c mice. In contrast, the treatment of the high concentration (500 ㎎/㎏)of MMDT were decreased thymic T lymphocytes. Administration of MMDT(250, 500 ㎎/㎏) eminently enhanced the production of IFN-γ. And MMDT did not affect the cell viability of Jurkat leukemia cells. These results suggest that MMDT have a immunoregulatory effect via enhanced cell mediated immunity

소음인(少陰人) 승양익기탕(升陽益氣湯)의 면역조절작용(免疫調節作用) (Immunoregulatory Action of Soeumin Seungyangikkitang)

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

LPS로 유도된 염증 상태 내에서 MAPKs 세포신호 전달체계를 저해하는 올방개의 항염증 효과 (Eleocharis kuroguwai Ohwi Ameliorates LPS-mediated Inflammation by Suppressing MAPKs Signaling)

  • 벼 농사에서 마릅으로 불리는 올방개는 문제의 잡초 종류 중 하나로서 제초 작용의 대상이다. 하지만, 올방개는 임상적으로 황달, 해열 및 통경을 위한 전통 의학에 쓰여왔다. 올방개는 오랫동안 임상적으로 사용되어왔음에도 불구하고, 분자생물학 기반의 생물학적 효과는 아직 밝혀지지 않았다. 때문에, 본 연구진은 LPS로 유도된 대식세포 내에서 올방개 추출물 처리에 의한 NO 생산량을 스크리닝 함으로써 항염증 효과를 조사했다. 가장 효과적인 항염증 fraction을 찾기 위해, 본 연구진은 HP20 column chromatography를 이용하여 5 종류의 EtOH sub-fraction을 동정하였다; 20%, 40%, 60%, 80%, 및 100% EtOH. 동정한 sub-fraction 중에서, 60%와 80% 성분에서 가장 유의성 있는 NO 생산의 저해를 나타냈으며, 세포독성 효과도 나타내지 않는 결과를 나타냈다. 추가적으로 80% sub-fraction은 유의성 있는 iNOS 저해와 전염증성 매개체의 mRNA; IL-6, TNF-α, 그리고 IL-1β를 저해하였으며, 이 결과는 MAPKs 세포신호전달과 관련 있는 JNK 및 ERK 단백질의 인산화 저해가 원인임을 밝혔다. 본 연구진의 결과는 올방개 추출물 80% sub-fraction EtOH가 항염증 활성을 가장 효과적으로 저해하는 성분 임을 밝혔으며, 이는 MAPKs 세포신호전달의 과 발현에 의한 염증성 질병의 치료제 후보 성분 일 수 있음을 시사한다.
