A New Cyclophilin Inhibitor from Ganoderma lucidum: Purification and Characterization

  • A new inhibitor for peptidylprolyl cis-trans isomerase (PPIase) has been isolated from Ganoderma lucidum and purified to homogeneous state by organic solvent extraction. The purified PPIase inhibitor (GPI) is assumed to be a membrane-associated glycoprotein. GPI inhibits specifically the bovine brain PPIase, a cyclophilin, and has no effect on the FKBP activity. The results of our chemical modification study of GPI indicate the presence of Lys residue(s) at or near its binding site. Like CsA-cyclophilin complex, GPI-bovine brain PPIase complex strongly inhibits the calcineurin activity in vitro, suggesting the possible involvement of GPI in immunomodulating pathway by the formation of PPIase-inhibitor-calcineurin complex.

Immunomodulating Activity of Alginate (Alginate의 면역조절작용)

  • Alginates are polysaccharides with gel-forming properties composed of 1,4-linked $\beta$-D-mannuronic acid (M), $\alpha$-L-guluronic acid (G), and alternating (MG) blocks. The M-and the MG-blocks, but not the G-blocks, have been known to be the active components of the alginates in experimental models. In this study, we have examined the ability of high M-alginate to activate immune cells. Alginate induced the macrophage anti-viral activity and the lymphocyte blastogenesis, and enhanced cytotoxicity of natural killer cell. In addition, alginates stimulated the macrophages to induce the production of $H_{2}O_{2}$, whereas alginates had no effect on NO production and suppressed the production of TNF-$\alpha$. These findings suggest that high M-alginate may be modulating various elements of the host immune response.

Studies on Anti-Mcirobial and Anti-Cancer Functions of Polysaccharide Extracted from Ganoderma lucidum (영지버섯 다당체의 항미생물작용 및 항암작용에 관한 연구)

  • This study was carried out to elucidate the immunomodulating activity of protein bound polysac charide extracted from Ganoderma lucidum(PSG). Macrophage drived reactive radicals were known as an effector for antimicrobial and anticancer functions. The promising immune response molecule, reactive oxygen intermediates(ROIs), was determined in TIB 71 cells with PSG at various experimental conditions. Treatment with 0.5mg PSG significantly increased the production of ROIs, superoxide anion as well as hydrogen peroxide, from TIB 71 cells(p<0.001). Under the same concentration, con siderable results were obtained from 24 hour cultivation with 106 cells at 5% CO2 incubator. The cells were trigged with PMA(5.3 M) after primed BCG(100 M) or IFN (100U) alone could not induce the production of ROIs, but it had a significant potentiating effect on ROIs secretion when the cells were treated with PSG.

  • PALMIWON is composed of 8 oriental herbs which has been known to show some pharmacological effects in kidney, blood vessels and immune systems, and used for the treatment of kid ney disease, hypertension, nervous disease and diabetic mellitus in the Orient for a long time. Based on our previous report that PALMIWON showed different effects on immune cells and ${\beta}$-cells, the immunoreactivity of ICSA (Islet Cell Surface Antibody) with ${\beta}$-cell (RINm5F) and the cell proliferation and function of interleukin-1${\beta}$ damaged ${\beta}$-cells in the presence of PALMIWON were examined. It was observed that PALMIWON significantly inhibited the immunoreactivity of ICSA with ${\beta}$-cell, and markedly increased cell proliferation and insulin release of interleukin-1${\beta}$ damaged ${\beta}$-cells.

  • This work was performed to investigate the effects of brazilin in vitro on mitrogen-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant IL-2 using splenocytes from C57BL/6 female mice. Brazilin (29-80 ng/ml) caused a noticeable increase in TCGF production of splenocytes, but did not affect responsivness to recombinant IL-2, the expression of ConA-induced high affinity IL-2 receptor and mitogen-induced proliferation of splenocytes.

  • Experiments were undertaken to evaluate the effects of brazilin and hematoxylin on immune functions in normal young mice. Brazilin and hematoxylin decreased the circulating leucocyte counts. Brazilin and hematoxylin decreased the cirulating leucocyte counts. Both compounds did not produce a significant change of immunoorgan weights in comparison with those in the control group. Brazilin decreased significantly IgM plaque forming cells. Brazilin and hematoxylin decreased significantly Arthus reaction at 3 hours-post challenge with respect to aggregated bovine serum albumin.

  • Fruiting bodies of Phellinus linteus were extracted by hot water and alkali methods. Sugar contents of PL-H (hot water extract) and PL-A (alkali water extract) were 81.1%, 37.4% and protein contents were 6.2%, 21.8%, respectively. Amino acid pattern showed that two extracts contained large amount of aspartic acid and alanine. Two extracts showed characteristic IR absorption pattern for glycosidic bond at $890\;cm^{-1}$. PL-H was divided two fractions by gel filtration chromatography and the molecular weights of each fraction were estimated to be about 10 kD and 225 kD, respectively and also PL-A was estimated 10 kD. Two extracts showed strong antitumor, immunomodulating and antioxidant activities, and were compared with commercialized glycopeptide anticancer drugs.

  • The first original inducer of interferon had been synthesized in Russia in 1965-1967. It was 2-car-boxiethyl-germanysesquioxan (Lx-13) and other-1 hydroxigermanytrans-monohydral (Lx-5), These compounds have very high induct$\circledcirc$on of interferon. The strain of Panax ginseng C. A. Meyer was recultivated on medium with different concentiation of (Lx-13) and (Lx-5) many times. The strain was stabilized when biomass of cells contain of the Ge form 10${\times}$10-3mg% to 2.3x10-3mg% to ashes, Biomass are hard physical work (swimming test) and also had stress-protective, immunomodulating action and anticarcinogenic effect hampered the sarcoma-180 by 33-80% and suppressed quantity of metanes on 43-46%.

  • Isolation, purification and characterization of biophysicochemical properties of the three new lectins, MLA-I, MLA-II, MLA-III from the hemolymph of Meretrix lusoria have been reported previously. A series of immunochemical studies were investigated in this work. The three lectins were revealed as having partial identity each other by immunodiffusim and immunoelectrophoresis. These results suggest that MLA lectins are isolectins having similar biophysicochemical properties. Particularly, MLA-I proved to be a potent mitogen for murine splenic as well as human peripheral lymphocytes, and the optimum mitogenic dose were 62.5 and 1.95 $\mu\textrm{g}$/ml, respectively.
