Bovine viral diarrhea (BVD) is very important disease in cattle industry with a worldwide distribution. Detection and elimination of persistently infected calves with bovine viral diarrhea virus (BVDV) was valuable strategy for BVD eradication because those calves were main source for transmission. We surveyed persistent infection with BVDV by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry (IHC) using whole blood and skin. Five hundred thirty nine Korean native calves were tested. Four calves (0.7%) were positive for BVDV antigen for both tests. Those calves remained positive for follow-up by RT-PCR and IHC. Therefore they were identified as persistently infected with BVDV. We confirmed that immunohistochemistry using skin biopsy samples was very useful tool to detect persistently infected calves with BVDV. As far as we know, this would be first report on persistent infection with BVDV in Korea.
Primary leptomeningeal glioblastomatosis is a rare and fatal tumor of the central nervous system, the condition is characterized by diffuse infiltration of the tumor in the meninges without evidence of primary tumor within the brain or spinal cord. We reported an unusual case of leptomengial glioblastomatosis, which was detected by the consecutive cerebrospinal fluid (CSF) cytology with application of immunohistochemistry, in addition to its cytologic findings. A healthy 21 year old man, who was enlisted in the army, presented with a stuporous mental state and diffuse enhancement of meninges without evidence of primary mass lesion in the brain and spinal cord on magnetic resonance imaging(MRI). CSF cytology showed small loose clusters of tumor cells with single cells and lymphocytes. The tumor showed variable pleomorphism with coarse chromatin, irregular nuclear membranes and multi lobated nuclei. On immunohistochemical staining, the tumor cells were founded to be positive for GFAP. In conjunction with radiologic findings, brain biopsy confirmed the diagnosis of leptomenigeal glioblastomatosis. The use of immunohistochemistry is helpful in confirming CSF cytologic diagnosis in patients with primary leptomeningeal glioblastomatosis.
In an outbreak of acute porcine diarrhea in newborn piglets, an etiological study was carried out using piglets submitted in Cheju Province Institute for Livestock Promotion(Cheju Veterinary Service for the disease diagnosis). Sixteen piglets(2-7 days old) were collected from 4 farms during outbreaks of diarrhea disease(from January to April 2000). Specimens were taken after necropsy and examined by immunohistochemistry using of monoclonal antibodies for porcine epidemic diarrhea(PED) virus, transmissible gastroenteritis(TGE) virus, and porcine rotavirus. Immunohistochemistry showed that PED virus antigens, but both TGE virus and rota virus antigens not, were localized in the some epithelial cells of the intestines of 14 animals among 16 piglets examined. PEB virus antigens were mainly detected in the cytoplasm of enterocytes. Infected cells, which were most abundant in the villous epithelial cells of the jejunum and ileum, were uncommon in the crypt, epithelial cells, the lamina propria and Peyer's patches of piglets examined. The results suggest that PED virus is one of the most prevailing agents in an outbreak of fatal diarrhea in newborn piglets on Cheju island and PED virus was need to further study to prevent this disease.
To elucidate pathologic change of skin in porcine exudative epidermitis, immunohistochemical and electron microscopical observations were carried out in the skin of the suckling pigs inoculated with Staphylococcus hyicus subsp hyicus which were isolated from natural case. In immunohistochemistry, ATPase-positive dendritic cells were more populated in epidermo-dermal junctional areas and perivascular area in dermis than in epidermal area as the disease was proceeded. These dendritic cells were identified as Langerhans cell by immunoperoxidase staining and these cells were populated granulomatous bodies. Electron microscopical study showed various retrogressive degeneration and vacuolation of epidermal cell organelles with retention of amorphorous exudates in intercellular space, and cellular seperation. Langerhans cells present in intercellular space of epidermis were populated in epidermo-dermal junctional areas, in dermis, and around granulomatous bodies. Langerhans cells contained decreased Birbeck granules in number but increased lysosome and ribosome. These cells were in contact with lymphocytes. This study was discussed relation between the various immunocytes and the formation of granulomatous bodies, and this inflammation was considered as delayed type hypersensitivity.
WooHwangChungSimWon is a traditional prescription for treating with hypertension, arteriosclerosis, coma, and stroke in China, Korea, and Japan. In the new presciption of WooHwangChungSimWon, the covet is substituted for the musk, the major component of WooHwangChungSimwon, because of the prohibition law about the musk. We have made a comparative study of the effects on the ischemic damage between the musk containing and civet containing WooHwangChungSimWon. In order to investigate the effects of WooHwangChungSimWon on the ischemic damage, each samples were administrated for 12 days, ischemia was induced for 10 minutes at 7th day, and immunohistochemistry was performed in the region of hippocampus of mongolian gerbils. According to the result of immunohistochemistry, the survival rates of neuroal cells in the hippocampal CA1 region are 37.8% in the high dose of musk containing WooHwangChungSimWon (HM-WHCSW) administrated group, 27.8% in low dose of civet containing WooHwangChungSimWon (LC-WHCSW), and 35.5% in high dose of civet containing WooHwangChungSimWon (HC-WHCSW) administrated group. These survial rates were significantly different from the survival rate of sham control group (14.4%). The results suggest that all the samples except the low dose of musk containing WooHwangChungSimWon (LM-WHCSW) have protective or preventive effects on cerebral ischemia.
Background: Vitiligo is a chronic autoimmune disease in which the destruction of melanocytes causes white spots on the affected skin. Janus kinase (JAK) is a family of intracellular, non-receptor tyrosine kinases that transduce cytokine-mediated signals via the JAK-signal transducer and activator of transcription pathway. The aim of the present study is to explore the possible role of JAK1 in the pathogenesis of vitiligo using immunohistochemical methods. Methods: The current study was conducted in a sample of 39 patients who presented with vitiligo and 22 healthy individuals who were age and sex matched as a control group. We used immunohistochemistry to evaluate JAK1 status (intensity and distribution) and assess the percentage of residual melanocytes using human melanoma black 45 (HMB45). Results: Intense and diffuse JAK1 expression was significantly more likely to indicate vitiliginous skin compared to normal skin (p<.001). Strong and diffuse JAK1 expression was associated with short disease duration, female sex, and lower percentage of melanocytes (detected by HMB45) (p<.05). Conclusions: JAK1 may be involved in the pathogenesis of vitiligo, as indicated by intense and diffuse expression compared to control and association with lower percentage of melanocytes detected by HMB45 immunostaining.

The carotid sinus nerve (CSN) is well known as mediating baroreflexes. However, studies of its detailed histological analysis are scant in the literature. Therefore, the current anatomical study sought to better elucidate the microanatomy of the CSN. Ten fresh frozen adult cadavers underwent dissection of the CSN. Then, it was harvested and submitted for histological and immunohistochemical staining. Specimens were all shown to be nerve fibers on histology and immunohistochemistry. We identified tyrosine hydroxylase positive fibers in all CSN specimens. These fibers were always found to be within the CSN and not on its surface i.e., epineurium. Based on our findings, the majority of fibers contained in the CSN are tyrosine positive in nature. Further studies are necessary to understand the true function of this autonomic nerve fibers.
The distribution of glucagon-immunoreactive cells in the pancreas during various developmental stages (fetus, neonate, 1-month-old, 6-month-old and adult) of the Korean native goat was investigated by immunohistochemical methods. The varying distribution and frequency of glucagon-immunoreactive cells in the pancreas of the Korean native goat were observed. The glucagon-immunoreactive cells were detected in both exocrine and endocrine portions (pancreatic islets) at all developmental stages and also in ducts of the 6-month-old and adult. The relative frequencies of glucagon-immunoreactive cells increased in the pancreatic islets and ducts with age, but decreased in the exocrine portions. Generally, they were distributed in the interacinar spaces or marginal zone of the pancreatic islets during all stages of development. However, the cell distributions of the pancreatic islets in the neonate divided into two types: 1) ones which were distributed in the inner zone, and 2) others in the peripheral zone.

Infections involving Echinostoma hortense (E. hortense) are considered to more severe than infections caused by other heterophyids. Although parasite expulsion by host immune responses attenuates the symptoms of infection, the detailed mechanisms of the host immune response need to be determined, especially in local immune responses involving cytokine and immunoglobulin expressions. We infected BALB/c mice with E. hortense and examined recovery rates together with expressions of multiple cytokines and immunoglobulins in the villi and crypts of the small intestine using immunohistochemistry. We observed a close correlation between worm expulsion rates and cytokine/immunoglobulin expressions in E. hortense infected mice. This study contributes to an understanding of the relationship between the immune response and parasite expulsion in hosts.
An 8-year-old, female lionhead rabbit with clinical sign of hematuria and vaginal discharge with/without blood was submitted to a local animal hospital. On exploratory laparotomy, three round to oval masses were observed in both uterine horns. The lumen of uterus was severely obstructed and distorted because of massive neoplastic proliferation. Histopathologically, the uterine masses revealed papillary projections along with irregular glandular structures into the lumen. The neoplastic foci were composed of numerous irregular sized neoplastic glands originated from uterine glands. These neoplastic cells showed very strong invasive tendency to muscle layer, therefore emboli of neoplastic cells were located in lymphatics. According to immunohistochemistry, the tumor cells in uterine masses demonstrated strong positive signals for cytokeratin, but negative for vimentin. Based on the gross, histopathologic and immunohistochemical features, this case was diagnosed as uterine adenocarcinoma in lionhead rabbit.
