Branched-chain Amino Acids are Beneficial to Maintain Growth Performance and Intestinal Immune-related Function in Weaned Piglets Fed Protein Restricted Diet

  • As a novel approach for disease control and prevention, nutritional modulation of the intestinal health has been proved. However, It is still unknown whether branched-chain amino acid (BCAA) is needed to maintain intestinal immune-related function. The objective of this study was to determine whether BCAA supplementation in protein restricted diet affects growth performance, intestinal barrier function and modulates post-weaning gut disorders. One hundred and eight weaned piglets ($7.96{\pm}0.26kg$) were randomly fed one of the three diets including a control diet (21% crude protein [CP], CON), a protein restricted diet (17% CP, PR) and a BCAA diet (BCAA supplementation in the PR diet) for 14 d. The growth performance, plasma amino acid concentrations, small intestinal morphology and intestinal immunoglobulins were tested. First, average daily gain (ADG) (p<0.05) and average daily feed intake (ADFI) (p<0.05) of weaned pigs in PR group were lower, while gain:feed ratio was lower than the CON group (p<0.05). Compared with PR group, BCAA group improved ADG (p<0.05), ADFI (p<0.05) and feed:gain ratio (p<0.05) of piglets. The growth performance data between CON and BCAA groups was not different (p>0.05). The PR and BCAA treatments had a higher (p<0.05) plasma concentration of methionine and threonine than the CON treatment. The level of some essential and functional amino acids (such as arginine, phenylalanine, histidine, glutamine etc.) in plasma of the PR group was lower (p<0.05) than that of the CON group. Compared with CON group, BCAA supplementation significantly increased BCAA concentrations (p<0.01) and decreased urea concentration (p<0.01) in pig plasma indicating that the efficiency of dietary nitrogen utilization was increased. Compared with CON group, the small intestine of piglets fed PR diet showed villous atrophy, increasing of intra-epithelial lymphocytes (IELs) number (p<0.05) and declining of the immunoglobulin concentration, including jejunal immunoglobulin A (IgA) (p = 0.04), secreted IgA (sIgA) (p = 0.03) and immunoglobulin M (p = 0.08), and ileal IgA (p = 0.01) and immunoglobulin G (p = 0.08). The BCAA supplementation increased villous height in the duodenum (p<0.01), reversed the trend of an increasing IELs number. Notably, BCAA supplementation increased levels of jejunal and ileal immunoglobulin mentioned above. In conclusion, BCAA supplementation to protein restricted diet improved intestinal immune defense function by protecting villous morphology and by increasing levels of intestinal immunoglobulins in weaned piglets. Our finding has the important implication that BCAA may be used to reduce the negative effects of a protein restricted diet on growth performance and intestinal immunity in weaned piglets.

Effects of transforming growth factor .betha.1 and interleukin-2 on IgA isotype switching at the clonal level

  • IgA is the predominant immunoglobulin isotype in mucosal secretions(1). It has been reported that a population of Peyer's patch T cells can selectively induce IgM bearing B cells to switch to surface IgA bearing B cells(2,3). Further, IL-4, IL-5, and IL-6 alone and in combination, can significantly influence murine IgA B cell differentiation in vitro(4-7). However, it remains an open question which cytokines have a major role in class switching to the IgA isotype. Recently, it has been reported that transforming growth factor .betha.1(TGF .betha.1) alone, or in combination with IL-2 increases IgA secretion by LPS-activated surface IgA negative (sIgA$\^$-/) murine spleen B cells while concurrently downregulating IgM and IgG secretion by such cells(8-11). In the present study, limiting dilution analysis was used to demonstrate, at the clonal level, that TGF .betha.1 has siginificant activity as an IgA isotype switch factor.

  • 본 연구는 고등학교 남자태권도 선수 21명을 대상으로 옥타코사놀 섭취에 따른 단기간 체중감량 시 혈중 피로물질면역글로불린에 미치는 영향을 구명하기 위해 6일간 동일한 합숙훈련을 하였으며 옥타코사놀 섭취감량군에 비섭취 감량군은 점차적으로 칼로리를 줄이는 식사제한을 통해 자기체중의 5% 이상 감량하였다. 옥타코사놀은 오전 ${\cdot}$ 오후로 나누어 식후 30분에 20 mg씩 섭취시켰으며 비섭취 감량군과 통제군은 같은 방법으로 위약을 섭취시켜 다음과 같은 결론을 얻었다. 1. 젖산과 암모니아는 옥타코사놀 섭취감량군이 유의하게 감소하였으며 집단 간 비교에서는 비섭취 감량군과 통제군보다 유의하게 감소하였다. 2. 젖산탈수소효소는 비섭취 감량군과 통제군이 감량 후 유의하게 증가하였다. 3. 면역글로불린 IgA, IgG는 모두 집단에서 증가한 것으로 나타났다.

  • After a detailed review of the literature availabe on the problems of immunoglobulin level in colostrum and blood serum of calves and after a discussion of its impact on health condition, the following investigations on this subject are presented. 1. Calves to fail ill in the rearing period show significantly lower serum Ig levels even during the first two days of life, when they are still clinically healthy, as well as up to the end of the first week of life than those of calves remaining in good health. 2. Ig values sharply rise during the disease period, IgA and IgM responding more rapidly than IgG. 3. In calves suffering from a disease of the gastrointestinal tract serum IgM titer is lowest, while in calves suffering from a disease of the respiratory duct serum IgA titer is lowest and in those suffering or dying from both disease complexes all Ig titers are reduced. 4. Calves showing high serum Ig levels during the first three days of life reveal higher daily gains in weight than those with lower serum Ig levels during that period.

한우 송아지의 초유섭취에 의한 수동면역 획득 상태 (A quantitative study of the transfer of colostral immunoglobulins to the newborn Korean native calf)

  • The levels of the serum and colostral total protein and immunoglobulins of the Korean native cows immediately after parturition and colostrum-conferred passive immune status of the Korean native calves were studied, and the results obtained were summerized as follows: The mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in sera of 10 Korean native cows immediately after parturition were $6.8{\pm}0.4g/dl$, $24.39{\pm}3.41mg/ml$, $19.49{\pm}2.70mg/ml$, $4.56{\pm}1.14mh/ml$, and $0.35{\pm}0.08mg/ml$ respectively. And the mean total protein, total immunoglobulin, IgG, IgM, and IgA concentrations in colostrum were $15.0{\pm}1.1g/dl$, $116.13{\pm}23.07mg/ml$, $101.51{\pm}22.59mg/ml$, $9.46{\pm}1.99mg/ml$, and $5.17{\pm}1.59mg/ml$ respectively. The mean concentrations of total protein and immunoglobulins in colostrum were 2.1 times to 14.8 times higher than those in serum. The mean concentrations of total protein, total immunoglobuln, IgG, IgM, and IgA in sera from 211 Korean native calves at 2 days old were $6.7{\pm}0.9g/dl$, $35.74{\pm}9.56mg/ml$, $29.06{\pm}8.07mg/ml$, $4.91{\pm}2.99mg/ml$, and $1,9{\pm}0.99mg/ml$ respectively. Although the calves had been suckled the dam's colostrum, each of calves was considerably various in levels of serum total protein and immunoglobulins and 16.1 percent of the calves were in a state of hypoglobulinemia.

광섬유 소산파를 이용한 면역 센서 제조 및 그 특성 (Fabrication of fiber-optic evanescent wave immunosensor and its measuring characteristics)

  • 광섬유 소산파 센서(fiber-optic evanescent wave sensor)를 제작하였다. 클래드층을 제거한 광섬유 코아 표면에 anti-mouse immunoglobulin G(IgG)를 결합시키고, 형광이 표지된 mouse IgG와의 반응을 직접적인 방법과 경쟁적인 방법을 통하여 측정하였다. 직접적인 방법과 경쟁적인 방법 모두 $1{\mu}g/m{\ell}$이하의 mouse IgG를 측정할 수 있는 감도를 얻을 수 있었다. Anti-mouse IgG는 단순 흡착 방법에 의하여 광섬유 코아 표면의 93.9%에 고정되었고 비특이적 결합반응을 제거하기 위하여 실시한 소혈청 알부민(bovine serum albumin : BSA)을 이용한 표면 코팅 효과는 없었다. Mouse IgG에 결합된 fluorescein의 비율이 높을수록 형광 발생량이 많았으나 관계는 직선적이지 않았다. 본 연구에서 제작된 광섬유 소산파 센서는 $1{\mu}g/m{\ell}$ 이하의 항원 항체 반응을 소산파 여기에 의한 형광량으로 측정할 수 있어 면역센서로의 응용이 가능할 것으로 판단된다.

Proteomic Analysis of Serum Samples from Natural Herb and Immunoglobulin Yolksac (Ig Y) treated Porcine

  • Purpose. Natural herb has been used for traditional medicine for thousands of years. For this reason, alternative medicine methods affect to domestic animal field. The main purpose of this study was to confirm the regulated proteins after feed additive treatment. Methods. We used total thirty male pigs were used for this experiment. E. coli and Salmonella typhimurium, were used for promote enteritis. Animals were divided into a negative control group, positive control group and test group to determine the effect of an additive mixture on the changes in protein expression in serum. The pigs were given a food supplemented with a natural herbal additive containing immunoglobulin yolksac (IgY) at concentrations 1%. On the 1st week and after eight weeks of feeding, the serums were collected from each group. The changes in the serum proteome as a response to the herbal additive were examined using two-dimensional polyacrylamide gel electrophoresis. Results. A total of 14 differentially expressed protein spots were identified by comparing the protein profiles of the control and additive treated porcine serums. Finally, 7 proteins were detected by MALDI-TOF/MS. Moreover, the proteins detected are involved in a cholesterol control factor, inflammation regulator, Conclusion. These results support of the hypothesis that a natural herbal additive containing IgY can affect the immune regulation system and reduce the pathological process by microbial infections.

  • The stabilizing effect of fructooligosanharide (FO) on hen's egg yolk immunoglobulin (yIgG) by heat and acid was investigated. The heat stability of yIgG at 70∼80$^{\circ}C$ was enhanced in a concentration-dependent manner by adding 0∼50% (w/v) FO to a yIgG solution. Acid-induced inactivation of yIgG was also suppressed in a concentration-dependent relationship by addition of FO. Addition of 50% FO almost completely stabilized yIgG at pH 3. The remarkable stablizing effect of FO on yIgG may enhance the use of yIgG as functional food ingredients.

난황항체제제(IgY) 첨가가 육용오리의 생산성, 혈액성상, 장내 미생물 및 면역글로불린 함량에 미치는 영향 (Effects of Egg Yolk Antibody Powder (IgY) Supplementation on Growth Performance, Blood Component Profile, Intestinal Microflora, and Immunoglobulin G in Meat Ducks)

  • 본 시험은 오리 사료 내 난황항체제제 첨가 시 생산성, 혈액 특성, 장내 미생물 및 면역글로불린에 미치는 영향을 조사하기 위하여 수행하였다. 1일령 오리(Cherry valley) 총 300수를 공시하여 5처리 3반복, 반복당 20수씩 공시하여 6주간 시험을 실시하였다. 처리구는 항생제를 첨가하지 않은 처리구(NC, negative control), 항생제 처리구(PC, positive control), 난황항체제제 0.1% 첨가구(T1), 난황항체제제 0.5% 첨가구(T2) 그리고 난황항체제제 1.0% 첨가구(T3)로 나누어 실험을 실시하였다. 종료 시 체중은 난황항체제제 첨가구와 항생제첨가구가무항생제처리구보다 유의적으로 높았고(P<0.05), 증체량 역시 난황항체제제 첨가구와 항생제 첨가구가 무항생제 처리구에 비해 높게 나타났다(P<0.05). 하지만 전 기간에 대한 사료 섭취량과 사료 요구율은 처리구간에 통계적인 유의차가 나타나지 않았다. 6주간 사양 T실험 후 혈액 생화학 및 혈구 분석 결과, 난황항체제제 0.1, 1.0% 첨가구에서는 total cholesterol 함량이 무항생제 처리구에 비해 다소 감소하는 경향을 보였으나, 통계적인 유의차는 없었다. 또한 AST와 ALT 함량 역시 처리구 간에 유의적인 차이를 보이지 않았다. 오리의 맹장 내 유산균 수는 난황항체제제 0.1와 0.5% 급여 시 다른 처리구에 비해 가장 높았으며(P<0.05), 대장균 수는 난황항체제제 수준이 증가함에 따라 감소하는 경향을 보였으나, 통계적 유의차는 없었다. 또한 혈청 내 면역글로불린 함량에도 처리구간에 유의적인 차이는 나타나지 않았다. 결과적으로 오리 사료 내 난황항체첨가 시 오리의 생산성을 개선시키고, 장내 미생물 균총에 효과적인 것으로 나타났으나, 아직까지 국내에서는 난황항체제제 첨가에 따른 사양 시험과 관련된 연구가 부족하다는 점을 감안할 때 향후 이에 대한 많은 연구가 필요할 것으로 사료된다.
