보리(Hordeum vulgare)추출물의 항보체 및 면역증강 효과 (Anticomplementary Activity and Immune-Stimulating Effect of the Extracts from Barley (Hordeum Vulgare))

  • By using several solvents, barley extracts containing the anticomplementary activities in classical pathway were prepared (250 $\mu\textrm{g}$/ml): methanol (83.1%), ethanol (71.9%), water extract (25.4%), M-1 (250 $\mu\textrm{g}$/ml), and the soluble part of methanol extract which showed the highest activity (83.4%) and the yield. Anticomplementary activity of methanol extract as well as protease digestion in classical pathway showed 82.4% and 78.4% in the concentration of 250 $\mu\textrm{g}$/ml, respectively. It was found that protein was not involved in anticomplementary activity in the classical pathway and the methanol extract made an impact on classical pathway, but not on alternative pathway. For the immune-stimulating effect, the T cell proliferation effect of the protease digestion displayed little effect irrespective of the dose. In addition, the T cell proliferation effect of methanol extract showed 13-fold higher proliferation effect compared with positive control. It was revealed that the substance containing protein serves as an important factor for the immune proliferation. Therefore, the anticomplementary activity ${\beta}$-glucan in classical pathway and alternative pathway displayed the lowest activity, showing 2.2%, 22.3% respectively. However, the immune-stimulating effect of ${\beta}$-glucan showed the T cell stimulating effect 13 times higher than positive control.

A comparative study on immune-stimulatory and antioxidant activities of various types of ginseng extracts in murine and rodent models

  • Background: Ginseng (Panax ginseng) is a widely used traditional herbal supplement that possesses various health-enhancing efficacies. Various ginseng products are available in market, especially in the Korean peninsula, in the form of drinks, tablets, and capsules. The different ginseng types include the traditional red ginseng extract (RGE), white ginseng, and black red ginseng extract (BRGE). Their fermented and enzyme-treated products are also available. Different treatment regimens alter the bioavailability of certain compounds present in the respective ginseng extracts. Therefore, in this study, we aimed to compare the antioxidant and immune-stimulating activities of RGE, BRGE, and fermented red ginseng extract (FRGE). Methods: We used an acetaminophen-induced oxidative stress model for investigating the reduction of oxidative stress by RGE, BRGE, and FRGE in Sprague Dawley rats. A cyclophosphamide-induced immunosuppression model was used to evaluate the immune-stimulating activities of these ginseng extracts in BALB/c mice. Results: Our results showed that most prominently, RGE (in almost all experiments) exhibited excellent antioxidant effects via increasing superoxide dismutase, catalase, and glutathione peroxidase activities in the liver and decreasing serum 8-hydroxy-2'-deoxyguanosine, aspartate aminotransferase, and lactate dehydrogenase levels compared with the groups treated with FRGE and BRGE. Moreover, RGE significantly increased the number of white blood cells, especially T and B lymphocytes, and antibody-forming cells in the spleen and thymus, and it also activated a number of immune cell subtypes. Conclusion: Taken together, these results indicate that RGE is the best supplement for consumption in everyday life for overall health-enhancing properties.

  • In this study we demonstrated whether the extract of Polygonum aviculare L. (PAE) can be applied to the immune-stimulating responses in macrophages (Raw 264.7 cells). Cell viability was determined by WST-8 assay, and all four doses of PAE (5, 10, 20, and 40 ${\mu}g/ml$) had no significant cytotoxicity during the entire experimental period. PAE increased the production of inducible nitric oxide synthase (iNOS) and nitric oxide (NO), and mRNA expressions and protein levels of pro-inflammatory cytokines(tumor necrotic factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$ and IL-6) in the same cells. These immune-stimulating activities of PAE were found to be caused by the stimulation of $NF{\kappa}B$ signal and phosphorylation of MAP kinases (p38, ERK and JNK).

Glycoproteins Contained within Soamsan, a Traditional Oriental Medicine, are the Main Class of Active Ingredients Responsible for the Medicine-induced Immune Stimulation

  • In our previous study, Soamsan, a traditional Oriental medicine, was shown to enhance the induction of antigen-specific immune responses, and it was speculated that the enhancing activity might be closely associated with glycoproteins contained within the medicine. To elucidate this speculation, protein samples from each component, used in the preparation of Soamsan, were obtained and their immune stimulating activities were tested with mouse splenocytes. All the samples markedly enhanced the lymphocyte proliferation and cytokine secretion by the mouse splenocytes. In particular, the enhancement was significantly higher with the protein sample treatments than with those of the original crude sample. Furthermore, the pronase E- and $NaIO_4$-mediated inhibition of splenocyte-stimulation activity of the protein samples clearly supported that glycoproteins are the main class of active ingredients responsible for the lymphocyte stimulating activity of the samples. Consequently, our findings suggest that glycoproteins might have a pivotal role in Soamsan-mediated immune modulation, although the in vivo effect of the glycoproteins should be further elucidated.

Probiotic and Antioxidant Properties of Novel Lactobacillus brevis KCCM 12203P Isolated from Kimchi and Evaluation of Immune-Stimulating Activities of Its Heat-Killed Cells in RAW 264.7 Cells

  • The purpose of this study was to determine the probiotic properties of Lactobacillus brevis KCCM 12203P isolated from the Korean traditional food kimchi and to evaluate the antioxidative activity and immune-stimulating potential of its heat-killed cells to improve their bio-functional activities. Lactobacillus rhamnosus GG, which is a representative commercial probiotic, was used as a comparative sample. Regarding probiotic properties, L. brevis KCCM 12203P was resistant to 0.3% pepsin with a pH of 2.5 for 3 h and 0.3% oxgall solution for 24 h, having approximately a 99% survival rate. It also showed strong adhesion activity (6.84%) onto HT-29 cells and did not produce β-glucuronidase but produced high quantities of leucine arylamidase, valine arylamidase, β-galactosidase, and N-acetyl-β-glucosaminidase. For antioxidant activity, it appeared that viable cells had higher radical scavenging activity in the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, while in the 2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay, heat-killed cells had higher antioxidant activity. Additionally, L. brevis KCCM 12203P showed higher lipid oxidation inhibition ability than L. rhamnosus GG; however, there was no significant difference (p < 0.05) between heat-killed cells and control cells. Furthermore, heat-killed L. brevis KCCM 12203P activated RAW 264.7 macrophage cells without cytotoxicity at a concentration lower than 108 CFU/ml and promoted higher gene expression levels of inducible nitric oxide synthase, interleukin-1β, and interleukin-6 than L. rhamnosus GG. These results suggest that novel L. brevis KCCM 12203P could be used as a probiotic or applied to functional food processing and pharmaceutical fields for immunocompromised people.

The Chemical Characteristics and Immune-Modulating Activity of Polysaccharides Isolated from Cold-Brew Coffee

  • To elucidate new biological ingredients in cold-brew coffee extracted with cold water, crude polysaccharide (CCP-0) was isolated by ethanol precipitation, and its immune-stimulating activities were assayed. CCP-0 mainly comprised galactose (53.6%), mannose (15.7%), arabinose (11.9%), and uronic acid (12.4%), suggesting that it might exist as a mixture of galactomannan and arabinogalactan. CCP-0 significantly increased cell proliferation on both murine peritoneal macrophages and splenocytes in a dose dependent manner. CCP-0 also significantly augmented nitric oxide and reactive oxygen species production by murine peritoneal macrophages. In addition, macrophages stimulated by CCP-0 enhanced production of various cytokines such as tumor necrosis factor-${\alpha}$, interleukin (IL)-6, and IL-12. In an in vitro assay for intestinal immune-modulating activity, CCP-0 showed higher bone-marrow cell-proliferation activity through Peyer's patch cells at $100{\mu}g/mL$ than the negative control. These results suggest that CCP-0 may potentially enhance macrophage functions and the intestinal immune system.

Immunomodulatory Actions of Lysophosphatidylcholine

  • Lysophosphatidylcholine (LPC) is an endogenous phospholipid. LPC has various stimulating or modulating activities on immune cells, including lymphocytes, monocytes/macrophages and neutrophils. Studies generally revealed pro-inflammatory activities of LPC, but accumulating evidence indicates that LPC has also anti-inflammatory actions. Here we summarize immunomodulatory actions of LPC.

  • In this study, we investigated the immunopotent activities of lepidan, a water soluble proteoglycan from Lentinus lepideus. To examine whether lepidan may affect nonspecific immune responses, we determined the effect on the production of nitric oxide (NO). Lepidan effectively increased the NO production in IFN-${\gamma}$ and LPS triggered RAW 264.7 cells. To further demonstrate the evidence that lepidan activates various immune cells, we determined the alkaline phosphatase activity, plaque-forming cell number and secretion of interleukine-4 (IL-4) and granulocyte/macrophage-colony stimulating factor (GM-CSF) after lepidan treatment in murine splenocytes. The results showed that lepidan increased alkaline phosphatase activity and the number of plaque-forming cells, which indicates that lepidan can lead B lymphocytes to late stage of differentiation. Also, when the splenocytes were cultured with lepidan for 48 hr, the level of IL-4 and GM-CSF in the supernatant dramatically increased. Taken together, these data suggest that lepidan is a biological response modulator that is able to activate immune responses.

  • $\beta$-l, 3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1, 3-glucans linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immunopotentiating activities. In this study, we tested the immunophamacological activities of insoluble $\beta$-1, 3-glucan from Agrobacterium sp. R259 KCTC 10197BP and confirmed the following activities: (1) IFN-${\gamma}$ production in PBMCs in the presence or in the absence of PHA, LPS, IL-18, and IL-12; (2) the induction of various cytokines in the spleen and thymus; (3) the adjuvant effect on the antibody production; (4) the cytotoxic and antitumor effects on cell lines and ICR mice. These results strongly suggest that $\beta$-1, 3-glucan from Agrobacterium sp. R259 KCTC 10197BP possesses various immunopharmacologica1 activities.

  • Aloe vera L.(Aloe barbadensis Miller) has been used for many centuries for various medical, cosmetic and neutraceutical purposes. The gel of A. vera was reported to exert numerous biological activities including wound healing, immunomodulatory, anti-cancer, anti-inflammatory, anti-bacterial, anti-viral, anti-diabetic, and anti-psoriasis activities. Acemannan, found predominantly in the inner leaf gel of A. vera, has been identified as the main active ingredient exerting diverse biological activities. In this review, we summarized the recent findings on the immunomodulatory activities of acemannan. Studies that used purified acemannan demonstrate that acemannan exerts immune-stimulating, anti-cancer, anti-viral, and hematopoiesis-increasing activities. In addition, it is being clear that most of these activities of acemannan are mediated primarily by activating professional antigen presenting cells such as macrophages and dendritic cells.
