신령버섯(Agaricus blazei Murill) 균사체내외 다당체의 면역활성효과 (Immune Enhancing Effects of Intracellular and Extracellular Polysaccharides Extracted from Mycelial Cultivate of Agaricus blazei Murill)

  • 신령버섯(Agaricus blazei Murill)의 액체 배양으로 다당체의 균사체외 분비를 유도하였으며, 버섯 균사체의 새포내 다당체와 세포외 분비 다당체의 면역증진활성을 in vitro 시험으로 비교하였다. 부분 정제된 세포내 다당체와 세포외 다당체의 총당 함량은 각각 85.6%와 95.3%였으며, ${\beta}$-glucan 함량은 각각 67.9%와 88.1%로 측정되었다. 면역활성 실험에는 시료의 닥 함량을 동일하게 맞추어 사용하였다. In vitro에서 균사체내외 다당체는 대식세포 주인 RAW 264.7을 활성화시켜 nitric oxide (NO) 생성을 농도 의존적으로 증가시켰으며, 각각 최대 53.9%, 53.1%의 비슷한 증가활성을 나타내었다. 또 균사채내외 다당체는 모두 RAW 264.7을 활성화시켜 염중성 cytokine류인 interleukin (IL)-$1{\beta}$, IL-6, tumor necrosis factor (TNF)-${\alpha}$의 생성을 증가시켰으며, 이때 3종의 cytokine 모두에서, 세포내 다당체에 비해 세포외 다당체를 처리했을때 저농도에서 높은 증가률을 나타내었다. 두 다당체는 in vitro 상에서 비장세포를 증식시키는 효과를 나타내었으며, 세포내 다당체가 농도 의존적으로 증식효과를 보인데 반해 세포외 다당체는 저농도에서 증식이 높았고, $250\;{\mu}g/ml$ 농도 이상에서는 더 높아지지 않았다. 두 다당체 모두 암세포인 B16F0 melanoma에 대한 직접적인 세포독성 효과는 나타내지 않았다. 신령버섯 균사체 배양으로 생성된 세포내외 다당체는 in vitro에서 모두 면역활성을 증가시키는 것으로 나타났으며 전반적으로 그 활성은 세포내 다당체보다 세포외 다당체가 우수한 것으로 판단되었다.

발아 전후 보리 당단백질 추출물의 영양성분 및 면역 활성 변화 (Changes in the Nutritional Components and Immune-enhancing Effect of Glycoprotein Extract from Pre- and Post-germinated Barley Seeds)

  • 발아 전후 보리 당단백질 추출물에 대한 영양성분 변화와 RAW 264.7 세포에 처리 후 생성하는 산화질소 II와 사이토카인(인터류킨-6, 종양괴사인자-${\alpha}$) 양을 측정하여 면역 증진 효능을 예측하였다. 보리의 발아가 진행됨에 따라 당단백질 추출물의 단백질과 지방질 함량은 감소하고 총당 함량은 증가하는 추세를 확인하였다. 주요 중성당은 아라비노스, 포도당, 자일로스이며 발아함에 따라 6탄당인 포도당은 감소한 반면, 5탄당인 아라비노스와 자일로스는 증가하였다. 아미노산 함량은 GEB에 비해 24-GEGB와 48-GEGB에서 각각 1.03, 1.24배 증가하였다. 면역 활성을 측정하기 위하여 RAW 264.7 세포에 24-GEGB와 48-GEGB를 처리한 결과 GEB에 비해 더 높은 산화질소 II와 사이토카인(인터류킨-6, 종양괴사인자-${\alpha}$)을 생성하였다. 결론적으로, 보리 종자가 발아하면 종자 내 단백질, 지방질 및 당질 등의 영양성분 조성이 변하고, 그 결과 발아보리 당단백질 추출물은 외부 침입에 대해 1차적으로 방어하는 대식세포를 자극하여 세포에 독성을 미치지 않는 범위의 면역매개물질 생성하여 면역 증진 효과를 기대할 수 있는 것으로 판단된다.

  • Chitosan, a deacetylated derivative of chitin, is one of the abundant resources, and its biological properties such as antibactierial activity, hypocholesterolemic activity, and hypertensive action are remarked. However, increasing attention has recently been yen to converting chitosan to its oligosaccharides because they possess various functional properties like antitumor activity, immune-enhancing effects, enhancing protective effects against infection with some pathogens in mice, antifungal activity, and antimicrobial activity (Jeon et al., 2001).   • Vaccine is a protective clinical measure capable of persuading immune system against infectious agents. Vaccine can be categorized as live attenuated and inactivated. Live attenuated vaccines activate immunity similar to natural infection by replicating living organisms whereas inactivated vaccines are either whole cell vaccines, eliciting immune response by killed organisms,or subunit vaccines, stimulating immunity by non-replicating sub cellular parts. The components of vaccine play a critical role in deciding the immune response mediated by the vaccine. The innate immune responds against the antigen component. Adjuvants represent an importantcomponent of vaccine for enhancing the immunogenicity of the antigens. Subunit vaccines with isolated fractions of killed and recombinant antigens are mostly co-administered with adjuvants. The delivery system of the vaccine is another essential component to ensurethat vaccine is delivered to the right target with right dosage form. Furthermore, vaccine delivery system ensures that the desired immune response is achieved by manipulating the optimal interaction of vaccine and adjuvantwith the immune cell. The aforementioned components along with routes of administration of vaccine are the key elements of a successful vaccination procedure. Vaccines can be administered either orally or by parenteral routes. Many groups had made remarkable efforts for the development of new vaccine and delivery system. The emergence of new vaccine delivery system may lead to pursue the immunization goals with better clinical practices.

Immune-enhancing effect of Acanthopanax Koreanum and its component, Eleutheroside E on the protein-energy malnourished C57bl/6 mice

  • Acanthopanax Koreanum stem (AK) has been used in Korea as a tonic and sedative as well as a drug with ginseng like activities. The purpose of our present study was to investigate the effects of AK extract (AKE) and Eleutheroside E, major component of AKE on an exacerbated immune function through utilization of protein-energy malnutrition (PEM) diet by using forced swimming test (FST). The immobility time were significantly decreased in the AKE or Eleutheroside E-administrated group compared with the control group on the FST (P < 0.05). The level of blood parameters were not changed significantly. PEM-induced weight loss of mice was reduced by oral administration of 500 mg/kg AKE. AKE oral administration improved the nutritional status such as the food efficiency ratio and the adrenal gland weight. AKE treatment significantly increased the production of interferon (IFN)-$\gamma$ compared with unstimulated splenocytes but not interleukin (IL)-4. Eleutheroside E also significantly increased the IFN-$\gamma$ production but not IL-2 and IL-4 in T cell line, MOLT-4 cells. These results suggest that AKE and Eleutheroside E may influence to immune-enhancing through increasing the physical endurance capacity and immune cell activation.

Evaluation of the Effects of Euglena gracilis on Enhancing Immune Responses in RAW264.7 Cells and a Cyclophosphamide-Induced Mouse Model

  • In this study we evaluated the immune-enhancing effects of β-glucan, the main component of Euglena gracilis (Euglena), and Euglena on inflammatory factor expression in RAW264.7 macrophages and ICR mice with cyclophosphamide-induced immunosuppression. Macrophages were treated with β-glucan or Euglena for 48 h. The β-glucan and Euglena groups exhibited higher levels of inducible nitric oxide synthase, nitric oxide, and tumor necrosis factor (TNF)-α than the control (vehicle alone) group. Animals were fed saline and β-glucan (400 mg/kg body weight (B.W.)) or Euglena (400 or 800 mg/kg B.W.) for 19 days, and on days 17-19, cyclophosphamide (CCP, 80 mg/kg B.W.) was administered to induce immunosuppression in the ICR mouse model. CCP reduced the body weight, spleen index, and cytokine expression of the mice. To measure cytokine and receptor expression, splenocytes were treated with concanavalin A (ConA) or lipopolysaccharide (LPS) as a mitogen for 24 h. In vivo, ConA stimulation significantly upregulated the expression of interferon (IFN)-γ, interleukin (IL)-10, IL-12 receptor β1, IL-1β, and IL-2 in splenocytes from the β-glucan- or Euglena-treated groups compared with those in the splenocytes from the CCP-treated group; LPS stimulation increased the levels of the cytokines TNF-α, IL-1β, and IL-6 in splenocytes from the β-glucan- or Euglena- treated groups compared with those from the CCP-treated group, but most of these differences were not significant. These results demonstrate the effect of Euglena in ameliorating macrophages and immunosuppression in CCP-treated mice. Thus, Euglena has the potential to enhance macrophage- and splenocyte- mediated immune-stimulating responses.

Immune Enhancement Effects of Neutral Lipids, Glycolipids, Phospholipids from Halocynthia aurantium Tunic on RAW264.7 Macrophages

  • Fractionated lipids of Halocynthia aurantium (Pyuridae) have been demonstrated to possess anti-inflammatory properties. However, their modulatory properties have not been reported yet. Thus, the objective of this study was to determine immune enhancing effects of fractionated lipids from H. aurantium tunic on macrophage cells. The tunic of H. aurantium was used to isolate total lipids, which were then subsequently separated into neutral lipids, glycolipids, and phospholipids. RAW264.7 cells were stimulated with different concentrations (0.5, 1.0, 2.0, and 4.0%) of each fractionated lipid. Cytotoxicity, production of NO, expression levels of immune-associated genes, and signaling pathways were then determined. Neutral lipids and glycolipids significantly stimulated NO and PGE2 production and expression levels of IL-1β, IL-6, TNF-α, and COX-2 in a dose-dependent manner, while phospholipids ineffectively induced NO production and mRNA expression. Furthermore, it was found that both neutral lipids and glycolipids increased NF-κB p-65, p38, ERK1/2, and JNK phosphorylation, suggesting that these lipids might enhance immunity by activating NF-κB and MAPK signaling pathways. In addition, H. aurantium lipids-induced TNF-α expression was decreased by blocking MAPK or NF-κB signaling pathways. Phagocytic activity of RAW 264.7 cells was also significantly enhanced by neutral lipids and glycolipids. These results suggest that neutral lipids and glycolipids from H. aurantium tunic have potential as immune-enhancing materials.

Glycoproteins Contained within Soamsan, a Traditional Oriental Medicine, are the Main Class of Active Ingredients Responsible for the Medicine-induced Immune Stimulation

  • In our previous study, Soamsan, a traditional Oriental medicine, was shown to enhance the induction of antigen-specific immune responses, and it was speculated that the enhancing activity might be closely associated with glycoproteins contained within the medicine. To elucidate this speculation, protein samples from each component, used in the preparation of Soamsan, were obtained and their immune stimulating activities were tested with mouse splenocytes. All the samples markedly enhanced the lymphocyte proliferation and cytokine secretion by the mouse splenocytes. In particular, the enhancement was significantly higher with the protein sample treatments than with those of the original crude sample. Furthermore, the pronase E- and $NaIO_4$-mediated inhibition of splenocyte-stimulation activity of the protein samples clearly supported that glycoproteins are the main class of active ingredients responsible for the lymphocyte stimulating activity of the samples. Consequently, our findings suggest that glycoproteins might have a pivotal role in Soamsan-mediated immune modulation, although the in vivo effect of the glycoproteins should be further elucidated.

수면증진 프로그램이 노인의 수면과 면역반응에 미치는 효과 (Effects of Sleep Promoting Program on Sleep and Immune Response in Elderly)

  • Purpose: This study was to examine the effects of sleep promoting program on sleep and the immune response in the elderly with insomnia. Methods: The study was designed as a quasi-experimental, nonequivalent control group pre-post test design. Thirty two elders who suffered from insomnia (16 experimental and 16 control subjects) were selected among the elders who had been enrolled in two churches. The subjects in the experimental group participated in a sleep promoting program which was composed of sleep hygiene education and progressive muscle relaxation for 4 weeks. Data were analyzed using the SPSS/WIN. Results: The experimental group showed higher sleep score than that of the control group (t=7.86, p=.00). The experimental group showed higher sleep satisfaction score than that of the control group (t=7.61, p=.00). The sleep promoting program was not effective in enhancing immune response. Conclusion: The sleep promoting program increased the sleep score, sleep-satisfaction score and B cell in immune response of elderly people suffering from insomnia. Therefore, sleep promoting program can be applied as an effective nursing intervention to promote sleep quality and sleep satisfaction.

바이오피드백을 이용한 점진적 근육이완이 중년여성의 지각된 스트레스, 스트레스반응, 면역반응 및 갱년기 증상에 미치는 효과 (Effect of Progressive Muscle Relaxation using Biofeedback on Perceived Stress, Stress Response, Immune Response and Climacteric Symptoms of Middle-Aged Women)

  • Purpose: This study was aimed to evaluate the effect of progressive muscle relaxation training using biofeedback on perceived stress, stress response, immune response and climacteric symptoms, Method: This was a crossover, pre-post test design, The study subjects are 36 middle-aged women who were selected at 2 public health centers, The independent variable was Biofeedback training for 4 weeks, twice a week and home training for 4 weeks, Dependent variables were perceived stress, stress response, immune response, and climacteric symptoms measured with Hildtch's scale (1996), Result: Progressive muscle relaxation training using biofeedback was not effective in reducing perceived stress, but it was shown to be effective in reducing physiological stress responses such as pulse rate and EMG, Though blood pressure and skin conductance were repeatedly down, and skin temperature slowly increased, there were no statistically significant differences. Progressive muscle relaxation training using biofeedback was not effective in reducing serum cortisol, enhancing immune responses, or decreasing climacteric symptoms. Conclusion: The findings point to a pressing need for further, well-controlled and designed research with consideration in selection of subjects and instruments, frequency of measurements, the sampling method, and intervention modalities.
