Conducting Polymer-Silica Composites for Immobilization of Enzymes

  • A new enzyme immobilization method based on hydrophobic interaction between supporting material and enzyme has been successfully developed. The efficacy of the new technique has been investigated by loading a horse radish peroxidase (HRP) enzyme on the surface of conducting polymer-silica composites and by measuring the enzyme activity and leaching property of HRP loaded within polymer-silica composites. The immobilized HRP enzyme showed activity profiles similar to that of free HRP in phosphate buffer (pH 6). Above all, HRP adsorbed on the polymer-silica composites has showed excellent stability over 10 days, compared to HRP adsorbed on the pristine silica. It is thought that with appropriate optimization works, the present method would be used as a cost-effective and facile route for the immobilization of biomolecules.

  • The direct chromatographic separation of enantiomers by chiral stationary phases (CSPs) has been extensively developed over the past two decades, and has now become the most popular method for the analytical and preparative separations of enantiomers. Polysaccharide derivatives coated onto silica gel, as CSPs, playa significantly important role in the enantioseparations of a wide range of chiral compounds using high-performance liquid chromatography (HPLC). Unfortunately, the strict solvent limitation of the mobile phases is the main defect in the method developments of these types of coated CSPs. Therefore, the immobilization of polysaccharide derivatives onto silica gel, via chemical bonding, to obtain a new generation of CSPs compatible with the universal solvents used in HPLC is increasingly important. In this article, our recent studies on the immobilization of polysaccharide derivatives onto the silica gel, as CSPs, through radical copolymerization with various vinyl monomers are reported. Polysaccharide derivatives, with low vinyl content, can be efficiently fixed onto silica gel with high chiral recognition.

Functionalized Poplar Powder as a Support Material for Immobilization of Enoate Reductase and a Cofactor Regeneration System

  • In this study, functionalized poplar powder (FPP) was used as a support material for the immobilization of enoate reductase (ER) and glucose-6-phosphate dehydrogenase (GDH) by covalent binding. Under optimal conditions, the immobilization efficiency of ER-FPP and GDH-FPP was 95.1% and 84.7%, and the activity recovery of ER and GDH was 47.5% and 37.8%, respectively. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) analysis indicated that FPP was a suitable carrier for enzyme immobilization. ER-FPP and GDH-FPP exhibit excellent thermal stabilities and superior reusability. Especially, ER-FPP and GDH-FPP enable the continuous conversion of 4-(4-Methoxyphenyl)-3-buten-2-one with $NAD^+$ recycling. While the immobilization strategies established here were simple and inexpensive, they exploited a new method for the immobilization and application of ER and its cofactor recycling system.

동전기 기법과 Zeolite를 이용한 중금속 오염토의 고정화에 관한 연구

  • This study applied EK method to remediate contaminated soil by heavy metal(Pb), tried to increase the efficiency of remediation using zeolite as enhanced EK remediation method to overcome the limit of traditional EK remediation method. Adsorption tests on Pb were conducted to apply the EK extraction and test conditions which contained the electrode reversal, operating time were established. After tests, lead-ions were transported into the specific position of zeolite by EK phenomena, then the immobilization of lead-ions at zeolite was developed. Based on these tests, the efficiency of remediation is different with its test conditions. In addition, the efficiency of remediation was improved dramatically by adding zeolite without electrode reversal and satisfied TCLP regulation of EPA in USA through the whole sample range. Finally, absorption and immobilization capacity of zeolite were proved its excellence and confirmed the possibility of application as enhanced EK remediation method.

  • Purpose : In this study we tested the hypothesis that eucommia ulmoides (EU) extract would improve disuse-induced muscle atrophy following hindlimb immobilization. Method : The hindlimb immobilization was performed with casting tape to keep the left ankle joint in a fully extended position. The rats in EU treated group were orally administrated with eucommia ulmoides water extract. After 2 weeks of immobilization, all animals were sacrificed, and the whole gastrocnemius muscles were dissected from both legs. The morphology of right and left gastrocnemius muscles in both EU and control groups were assessed by hematoxylin and eosin staining. Results : Eucommia ulmoides extract represented the significant protective effects against the reductions of the gastrocnemius muscles weight and average cross section area to compared with Control group. Conclusion : Eucommia ulmoides has protective effects against immobilization induced muscle atrophy.

  • In this study, variation of electrochemical parameters and characteristics of lead immobilization due to phosphoric acid injection in soil were studied during electrokinetic remediation of lead contaminated soil. TCLP result showed about 100% of soil was satisfied TCLP regulation criteria. And injected ion from cathode reservoir by ionmigration was proportionate to concentration of phosphoric acid and elapsed time. Therefore, when removal is infeasible or not cost-effective, in situ immobilization method would be more effective.

  • We present a method for spatially selective immobilization of functional materials, such as proteins and nanoparticles, onto pre-activated silicon surfaces by electrochemical reaction. Carboxymethylbenzendiazonium (CMBD) cations, being adsorbable on silicon surfaces through electrochemically reductive deposition, is used as an anchor molecule to prepare the pre-activated silicon surfaces. It is demonstrated that the use of BD reaction is very efficient for the selective immobilization because the functional materials are immobilized exclusively onto the pre-adsorbed CMBD region. The method is applied to immobilize gold nanoparticles on the selected nanowire of the nanowire array.

Experimental Study on the Antidepressant Effect of Sam-Jeong-Hwan (삼정환(三精九)의 항우울 효과에 대한 실험적 연구)

  • Objective: The purpose of this study was to investigate the protective effects of Sam-Jeong-Hwan(SJH) on the animal model of depression induced immobilization stress. Method: The subject were divided into 4 groups(l. normal 2. saline solution administered during immobilization stress treatment 3. SJH of 100mg/kg administered 4. BKJ of 400mg/kg administered). Immobilization stress was treated for 1 hours on day. During 2 days of immobilization stress treatment, they were executed forced swimming test, passive avoidance test, elevated plus maze test. Corticosterone and ACTH in blood were measured. Results: In forced swimming test, SJH of 400mg/kg group showed decreased immobilization. In passive avoidance test, SJH of 400mg/kg group showed increased learning execution. In EPM test, SJH of 400mg/kg group showed decreased anxiety. In locomotor activity test, SJH groups showed significantly increased locomotor activity. Stress group showed significantly increase in serum level of corticosterone, SJH of 400mg/kg group showed decreased serum level of corticosterone. Stress group showed significantly increase in serum level of ACTH, SJH of 400mg/kg group showed decreased serum level of ACTH. Conclusion: These results suggest that Sam-Jeong-Hwan(SJH) is effective in the treatment of depression.

The Change of the Mechanoreceptors of Injured Achilles Tendon According to the Immobilization Periods (아킬레스건 손상 후 고정기간에 따른 기계적 수용기의 변화)

  • Purpose: This study was designed to determine the quantitative changes of the numbers of the mechanoreceptors in the experimentally tenotomized Achilles tendon of rabbits as compared with short-term immobilization and long-term immobilization. Materials and Methods: 14 white rabbits were used. After tenotomizing the right Achilles tendon, the subjects were divided into 2 groups according to the periods of immobilizaton. The left side of each Achilles tendon of the rabbits were used as controls. The tendons were stained with a modified gold-chloride method. Results: The number of mechanoreceptor was significantly decreased in the tenotomized Achilles tendon group than the control group (p<0.01). There was no statistical difference between the two experimental groups in the numbers of the mechanoreceptors (p>0.01). Conclusion: The injured Achilles tendons may more vulnerable to injury because of the decreased numbers of mechanoreceptors, but no difference between the periods of immobilization. It may suggest that post-operative immobilization period may not affect on the outcome of operative treatment from the viewpoint of mechanoreceptors.

Development of Supporting Materials with Curdlan and Activated carbon for Microbial Immobiliaztion (Curdlan과 활성탄을 이용한 미생물 고정화 담체개발)

  • The microbial immobilization media with curdlan and activated carbon which has great immobilization capacity has been developed. Characteristics of porosity and mechanical strength of this support media are dependent on manufacturing method. The support media showed the best cell immobilization performance when the ratio of curdlan and activated carbon was 30 g/L to 6 g/L in this study. The immobilization of iron-oxidizing bacteria on the supporting particles was photographed with a scanning electron microscope. Since cell concentration on the surface of supporting particle increased with the reaction time, the iron oxidation rate also increased.
