Changes of Immuno-Activity in Yogurt Prepared with Immunized Milk Containning Anti-Helicobacter pylori Antibody (Anti-Helicobacter pylori 항체를 함유한 면역우유로 제조한 요구르트의 면역활성의 변화)

  • The utilization possibility of immunized milk containing anti-Helicobacter pylori antibody to manufacture of yogurt was evaluated. The pH and titratable acidity of immunized milk changed significantly after incubation for 6 hours at $37^{\circ}C$ and thereafter did not change. The number of lactic acid bacteria reached $10^9\;CFU/mL$ after incubation for 6 hours at $37^{\circ}C$ and maintained the same number thereafter. The IgG content of heat treated immunized milk and yogurt maintained 97% and 93.5% compared with non heat treated immunized milk, respectively. The pH, titratable acidity and lactic acid bacteria of yogurt made of immunized milk were not changed apparently during storage for 21 days at $2^{\circ}C$ and $4^{\circ}C$, respectively. The IgG content of yogurt did not decrease significantly during storage for 14 days at $2^{\circ}C$, $4^{\circ}C$, and $10^{\circ}C$ but rapidly decreased after storage for 14 days at the same conditions, respectively.

  • This study was performed to investigate the effect of oral administration of GSRE against the asthma. Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. Concentrations of IL-4, IL-5 in BALF and splenocyte were assessed by ELISA, IgG and IgE from serum were calculated by same method. Concentration of IL-4 in splenocyte was significantly decreased in GSRE group compared with control group. Concentrations of IL-5 from BALF and splenocyte were significantly decreased in GSRE group compared with control group, respectively. Level of IgE in serum was significantly decreased in GSRE group compared with control group, but not IgG. We found that the effect of GSRE extract in asthma was implicated in reductions of IL-4, IL-5 released from Th2 cell, and decreses of IgE, from plasma cell. These findings suggest that GSRE extract can produce anti-asthmatic effect, which may play a role in allergen-induced asthma therapy.

Identification of Functionally Different Rat IgE in RBL-2H3 Exocytosis

  • Background: IgE is closely related to the development of allergies. However, the poor relationship between the specific IgE level and the severity of allergic diseases suggests the possibility of functionally different IgE isoforms. With this in mind, rat basophilic leukemia (RBL)-2H3 activation was analyzed with each type of rat IgE for two parameters, exocytosis and IL-4 mRNA production. RBL-2H3 has been well documented in the rat mucosal mast cell line. Methods: RBL-2H3 cells sensitized with each kind of rat IgE was activated by cross-linking FcRI with B5 (monoclonal anti-rat IgE mouse IgG antibodies). The RBL-2H3 exocytosis was measured by analyzing the ${\beta}$-hexosaminidase level, and the level of IL-4 mRNA synthesis was analyzed using semiquantitative RT-PCR. Rat IgE, which was produced by a parasite infection (REP), was prepared using either Paragonimus westermani metacercariae (REP-PW) or Anisakis simplex third stage larvae (REP-AS). A rat IgE prototype of IR162 was prepared by a peritoneal injection of immunocytoma. Results: The level of exocytosis showed a linear relationship with the rat IgE concentration when REP-PW or REP-AS was applied. However, it exhibited a biphasic response with IR162. In addition, the time course of heating at $56^{\circ}C$ illustrated the similarity between REP-PW and REP-AS, which differed from that of IR162. In contrast, the level of IL-4 mRNA synthesis in the RBL-2H3 cells with IR162 was comparable to that of either REP-PW or REP-AS. Conclusion: These results suggest that functionally different rat IgE isoforms exists in RBL-2H3 exocytosis.

  • Flagellin, a major structural protein of the flagellum found in all motile bacteria, activates the TLR5- or NLRC4 inflammasome-dependent signaling pathway to induce innate immune responses. Flagellin can also serve as a specific antigen for the adaptive immune system and stimulate anti-flagellin antibody responses. Failure to recognize commensal-derived flagellin in TLR5-deficient mice leads to the reduction in anti-flagellin IgA antibodies at steady state and causes microbial dysbiosis and mucosal barrier breach by flagellated bacteria to promote chronic intestinal inflammation. Despite the important role of anti-flagellin antibodies in maintaining the intestinal homeostasis, regulatory mechanisms underlying the flagellin-specific antibody responses are not well understood. In this study, we show that flagellin induces interferon-β (IFN-β) production and subsequently activates type I IFN receptor signaling in a TLR5- and MyD88-dependent manner in vitro and in vivo. Internalization of TLR5 from the plasma membrane to the acidic environment of endolysosomes was required for the production of IFN-β, but not for other pro-inflammatory cytokines. In addition, we found that anti-flagellin IgG2c and IgA responses were severely impaired in interferon-alpha receptor 1 (IFNAR1)-deficient mice, suggesting that IFN-β produced by the flagellin stimulation regulates anti-flagellin antibody class switching. Our findings shed a new light on the regulation of flagellin-mediated immune activation and may help find new strategies to promote the intestinal health and develop mucosal vaccines.

  • Artemisinin, a new antimalarial to treat patients infected with strains of Plasmodium jalciparum, derived from the plant Artemisia annua Linn, has immunopharmacologic actions such as enhence the PHA -induced lymphocyte transformation rate, increased the weight of spleen but reduced the weight of thymus, reduced phagocytic function of peritoneal macrophage, remarkably reduced the level of serum IgG and hemolysin fonning capacity (sentitized with SRBC), inhibited the activity of Ts cells of donor mice by supraoptimal immunuization(SOI), but enhenced activity of Ts cells of recipient mice by SOI. These results suggested that Ts cells may be the target cells of artemisinin. To the serum complement C3 level of plasmodium berghei-infeted mice, artemisinin (i. m,) could remarkly increase it. The artemisinin also obviously reduced the prostaglandin E(PGE) in the mouse hind paw swelling induced by carrageenin. Numerous studies have demonstrated that pharmacologic doses of PGE attenuate the development of immunocomplex nephritis. Some autologous immune mechanisms may be invoolved In the pathogensis of some types of glomurulonephritis. Glomerular abnormalities can be induced in animals by variety of immunological manipulations. The resulting disorder has many clinical and pathogical similarities to the disease in human. Our purpose was therefore to test the ability of the artemisinin to lessen the severity of rabbit IgG accelerated nephrotoxic serum glomerulonephritis in mice model. Mice which had treated with rabbit IgG and NTS, administrated with saline, showed Significant inceases of urinary protein, cholesterol level, and decrease of serum albumin in NS group. On the contrary, By i.g. adminstration of artemisinin at dose of 12.5, 25 and 50 mg/kg for 14 days after NTS injection, shown that artemisinin inhibited the nephritic changes in some parameters by means of urinary protein(p<0.05, p<0.01) and serum choleterol(p<0.05, p<0.01) and albumin (p<0.05, p<0.01), blood urea nitrogen (p<0.05, p<0.01), serum albumin(p<0.05, p<0.01); Cyclophosphamide(i.p. 10mg/kg for 14d) had almost same effect as the artemisinin had. Morphological studies shown that The picture of kidney from the mouse with NTS-nephritis accerated with rabbit IgG, treated with i.g. saline as the control, the mesangiocapillary were enlarged and proliferated; There were inflammatory cells infiltrating around the glomeruli; The ethelial cell were proliferated in the wall of Bowman's capsule. Histopatholological picture of kidney from the NTS-nephritis accerated with rabbit IgG mouse treated with i.p. 10mg/kg cyclophosphamide as the positive control. No siginicant histopathological evidence were found. Treaded with i.p. 12.5mg/kg artemisinine, the picture shown that mesangiocapillary were lightly proliferated; There were inflammatory cells infiltrating around the glomeruli; Treaded with i.p. 25mg/kg artemisinine, The picture shown that the mesangiocapillary were lightly proliferated; Treaded with i.p. 50mg/kg artemisinine, The picture shown that both the mesangiocapillary proliferated and the inflammatory cells infiltrating around the glomeruli are less than treated with saline, 12.5 and 25 mg/kg artemisinine. On the basis of these studies we conclude that the artemisinin can relieve pathological change caused by NTS-nephritis aacerated with rabbit IgG.

Mucosal Immune Response and Adjuvant Activity of Genetically Fused Escherichia coli Heat-Labile Toxin B Subunit

  • Although the E. coli heat-labile enterotoxin B subunit (LTB) is known to be a potent mucosal adjuvant towards co-administrated unrelated antigens and immunoregulator in T-helper 1-type-mediated autoimmune diseases, a more efficient and useful LTB is still required for prospective vaccine adjuvants. To determine whether a novel chimeric LTB subunit would produce an enhanced mucosal adjuvant activity and immune response, a number of LTB subunits were genetically fused with chimeric proteins using the epitope genes of the envelope glycoprotein E2 (gp51-54) from the classical swine fever virus (CSFV). It was found that the total serum immunoglobulin (Ig) levels of BALB/c mice orally immunized with chimeric proteins containing an N-terminal linked LTB subunit (LE1, LE2, and LE3) were higher than those of mice immunized with LTB, E2 epitope, and chimeric proteins that contained a C-terminal linked LTB subunit. In particular, immunization with LE1 markedly increased both the total serum Ig and fecal IgA level compared to immunization with LTB or the E2 epitope. Accordingly, the current results demonstrated that the LTB subunit in a chimeric protein exhibited a strong mucosal adjuvant effect as a carrier molecule, while the chimeric protein containing the LTB subunit stimulated the mucosal immune system by mediating the induction of antigen-specific serum Ig and mucosal IgA. Consequently, an LE1-mediated mucosal response may contribute to the development of effective antidiarrhea vaccine adjuvants.

Anti-allergic Effects of Gagam-YangGyeokSan on RBL-2H3 Mast Cells and OVA/alum Sensitized Mice (가감양격산(加減凉膈散)이 RBL-2H3 비만세포와 OVA/alum에 감작된 생쥐에 미치는 항알레르기 효과)

  • Objectives: Gagamyanggyeoksan (G-YGS) has been used to suppress allergic reaction, however, the cellular target of G-YGS and its mode of action remain unclear. The present study was designed to investigate the effect of extracted G-YGS on the PMA and lonomycin (PI)-induced activation of RBL-2H3. Methods: For this investigation, We examined IL-4, IL-13 mRNA expression by Real-Time PCR, IL-4, IL-13 production by ELISA analysis and manifestations of GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting, OVA-specific IgE, IL-4, IL-13 by mouse be sensitive to OVA. Results: Here we showed that treatment of RBL-2H3 mast cells with G-YGS, suppressed PI-induced production of Th2 cytokines including IL-4 and IL-13 in a dose dependent manner. The mRNA expression of IL-4 were completely abolished by G-YGS at the concentration of $100{\mu}g/ml$. Data from a stable cell lines consistently expressing IL-4. And the mRNA expression of IL-13 were abolished by G-YGS at the $200{\mu}g/ml$. But there is no difference between the $50{\mu}g/ml$, the $100{\mu}g/ml$ and the comparison. Results from the western blot analysis of transcription factors involving IL-4 and IL-13 expression indicated that it prominently decreased the expression of mast cell specific transcricption factors including GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65 but not c-Fos. And G-YGS suppressed IgE, IL-4, IL-13 in mouse be sensitive to OVA. Conclusions We suggested the anti-allergic activities of G-YGS might be mediated by down-regulation of Th2 cytokines such as IL-4 and IL-13 through the regulation of transcription factors as GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65.

Effects of coffee intake on airway hypersensitivity and immunomodulation: an in vivo murine study

  • BACKGROUND/OBJECTIVES: Coffee is a complex chemical mixture, with caffeine being the most well-known bioactive substance. The immunomodulatory and anti-inflammatory properties of coffee and caffeine impact health in various aspects, including the respiratory system. The objective is to investigate the effects of coffee and caffeine on airway hyperresponsiveness and allergic reactions, as well as to analyze and compare associated cytokine profiles. MATERIALS/METHODS: BALB/c mice were intraperitoneally sensitized with ovalbumin (OVA) and given OVA inhalation to induce airway hypersensitivity. Two weeks after sensitization, they were intragastrically gavaged with coffee or caffeine, both containing 0.3125 mg caffeine, daily for 4 weeks. Control mice were fed with double-distilled water. Serum OVA-specific antibody levels were measured beforehand and 5 weeks after the first gavage. Airway hyperresponsiveness was detected by whole body plethysmography after gavage. Cytokine levels of bronchoalveolar lavage and cultured splenocytes were analyzed. RESULTS: Coffee effectively suppressed T helper 2-mediated specific antibody response. Airway responsiveness was reduced in mice treated with either coffee or caffeine. Compared to the control, coffee significantly reduced OVA-specific immunoglobulin (Ig) G, IgG1 and IgE antibody responses (P < 0.05). Caffeine also attenuated specific IgG and IgG1 levels, though IgE level was unaffected. Coffee significantly reduced interleukin (IL)-4 and increased IL-10 concentration in spleen cells and bronchoalveolar lavage fluid (P < 0.05). CONCLUSIONS: Coffee effectively attenuated airway hyperresponsiveness and systemic allergic responses induced by OVA food allergen in mice. As a complex composition of bioactive substances, coffee displayed enhanced immunomodulatory and anti-inflammatory effects than caffeine.

Effect of Lonicerae Flos-Skin on Contact Hypersensitivity induced by Repeat Elicitation of DNCB (금은화 화장수가 DNCB로 유발된 접촉성피부염에 미치는 영향)

  • Objectives : Lonicerae Flos has been known as a useful plant with anti-inflammatory, antioxidative and immunosuppressive activity. To evaluate anti-inflammatory effect of Lonicerae Flos, we treated Lonicerae Flos-skin in animal model system induced contact hypersensitivity. Methods : Contact hypersensitivity, a local inflammatory response of the skin, was induced by spreading the right ear of BALB/c mouse with 1% DNCB. Lonicerae Flos-skin was prepared by dissolving 1% water extract of Lonicerae Flos in skin vehicle and treated everyday for 2 weeks on the right ear. Results : Lonicerae Flos-skin significantly reduced a mouse ear thickness swelled by 1% of DNCB treatment compared with skin vehicle-treated control group. Lonicerae Flos-skin also reduced IgG and IgE in serum obtained from blood of 1% DNCB-treated mouse. Conclusion : These results showed that Lonicerae Flos-skin could be used as a pharmaceutical material with antiinflammatory effects by reducing IgG and IgE in contact hypersensitivity mouse model by DNCB.

Immunopathology of Spleen following 7,12-Dimethylbenz[a]anthracene Treatment in BALB/C Mice (마우스의 비장에 미치는 7,12-Dimethylbenz[a]anthracene의 면역병리학적 연구)

  • This study was carried out to investigate the immunopathological effects of 7,12-Dimethylbenz[a]anthracene(DMBA) on spleen in mice. DMBA was administered subcutaneously to BALB/C mice by interscapular single injection of 50 or 100${\mu}g/g$ of body weight. Each DMBA treatment group and additional corn oil control group of mice were studied on day 1,3,7,14 and 21 following the injection of DMBA. DMBA treatment resulted in marked decrease in weights and cellularity of spleen. Spleen weights showed the greatest decrease at 14days after 50${\mu}g/g$ DMBA treatment, and at 21days after 100${\mu}g/g$ DMBA treatment. Spleen cellularity was similarly deceased in comparison with spleen weights. Spleen showed morphologically no typical changes throughout the experiment after 50${\mu}g/g$ DMBA treatment. Following the treatment of 100${\mu}g/g$ DMBA the spleen showed severe fibrosis, hemosiderin precipitation, and megakaryocytes decrease in red pulp at 14 days, while hemopoietic function was partly restored in addition to the appearance of a few megakaryocytes at 21 days. In spleen sections treated with antibodies to IgM or Thy1.2, lymphocytes strongly stained with IgM antibody were infiltrated around the central artery within the white pulp, and T-lymphocytes of periarterial lymphatic sheath (PALS) were diminished and destructed in sections treated with Thy1.2 antibody, at 14 days after the treatment of 100${\mu}g/g$ DMBA. By the electron microscopy phagocytic epithelial cells or macrophages were remarkably increased in spleen at 14and 21days following the treatment of 100${\mu}g/g$ DMBA.
