The Effects of Tofu (Soybean curd) Containing Mushrooms on the Immune Activities (버섯첨가 두부의 면역활성에 미치는 영향)

  • This research was carried out to evaluate the value of tofu containing mushroom as a immunomodulator. Tofu was made using $CaSO_4{\cdot}2H_2O$ or Lactobacillus extract as a coagulant after adding powder of fruit bodies or mycelia of Letino edodes and Lepista nuda to soybean milk. Proximate compositions of tofu and tofu containing mushroom were analyzed. Levels of interferon ${\gamma}$ (IFN-${\gamma}$), interleukin 4 (IL-4) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in culture media of lymphocytes collected from mouse spleens after being injected with mushroom, regular tofu, or tofu made with mushroom were measured by sandwich ELISA. In addition, concentrations of IgG1, IgG2a and IgE in plasma or lymphocyte culture media were analyzed. Crude protein, crude lipid and crude ash were decreased in tofu containing mushroom but phosphorus was increased significantly. IFN-${\gamma}$ concentration was significantly decreased in mice injected with fruit body or tofu alone. IL-4 level was decreased significantly in mice injected with tofu containing fruit body of L. edodes. However, TNF-${\alpha}$ was increased in mice injected with tofu containing fruit body of L. edodes. Plasma levels of IgG1 were increased in almost all groups, while there was no significant change in IgG2a levels among treated mice groups. Concentrations of IgG1 and IgG2a were increased significantly in lymphocyte culture media of mice injected with tofu containing mushroom. Plasma levels of IgE level was significantly increased in mice injected with tofu or fruit body of L. edodes, but not in mice treated with tofu containing mushroom. These results showed that tofu with mushroom affected immune activities, and it seems valuable to consider developing the mixture of tofu and L. edodes as an immunomodulator.

  • An experiment was conducted to establish a large scale production method of anti-serum against chicken IgM and to profile the developmental changes of serum IgM levels during the feeding period(from hatching to 7 weeks of age) in broiler chicks. Blood samples were taken from Hubbard chicken at the age of hatching, three days of age, and weekly thereafter till to 7 weeks of age. The pure IgM was isolated from ammonium sulfate treated chicken serum by both sephadex G-200 and sepharose CL-6B chromatography. The breaking-through peak containing IgM appeared from the fraction 26 to 28. These fractions consisted mainly of IgM when tested by anti-chicken IgM(Nordic, Netherlands). Immunized with the heavy chain of this purified IgM, the rabbit immune sera(anti-chicken IgM) were formed a reaction only with the purified chicken IgM. The quantitative assay of serum IgM were carried by RID method. The optimal time for diffusion was 14 hours and the coefficient of determination($R^{2}$) for regression equation of standard curve was 0.992. It was observed that IgM concentrations were the highest at hatching(3.23 mg /mL), after that decreased gradually. From 2 to 5 weeks of age the levels unchanged(2.0 ~ 2.3mg /mL), and gradually decreased to 7 weeks of age(1.3 mg /mL).

The Solubilization Behavior of DOPE-Immunoliposomes with Immunoglobulin G(IgG) by Added Bile Salts (Immunoglobulin G(IgG)를 함유한 DOPE 리포솜의 제조와 담즙산염에 의한 용해 특성)

  • The effects of bile salts (BS) on the stability of dioleoylphosphatidylethanolamine (DOPE) liposomes were investigated, observing apparent absorbance of vacant liposomes and calcein release from entrapped liposomes. Unilamellar liposomes were prepared by using a small quantity of palmitoly-immunoglobulin G(IgG) ($2.5{\times}10^{-4}$ mo1/lipid mol) to stabilize the bilayer phase of the unsaturated DOPE which by itself does not form stable liposomes. The destabilization of PE immunoliposomes by papain, clearly demonstrates that the IgG is essential for stabilization of PE bilayer. Approximately 4% of the entrapped calcein was released from the PE liposomes after 1 hr from liposome formation. Calcein release and absorbance of liposomes depended on the BS/lipid ratio because of the solubilization of lipid molecule in bilayer and the formation of mixed micelles. At very low BS concentrations, the incorporation of BS induced BS/lipid aggregates in the outer vesicles monolayer, while high BS concentrations, mixed micelles were formed. Chelate and its conjugates as $3{\alpha},\;7{\alpha},\;12{\alpha}-trihydroxy$ BS induce the concentration of the $3{\alpha}$, $12{\alpha}-dihydroxy$ BS at half-maximal solubilization of immunoliposomes to approximately 2.5-, or 5-fold. Conjugation of BS with glycine or taurine slightly enhanced their capacities to perturb membranes.

Effects of Passive Transfer Status on Growth Performance in Buffalo Calves

  • The objective of the study was to evaluate the effect of passive transfer status, determined by measuring serum immunoglobulin (Ig) concentration 24 hours after parturition, on growth performance in buffalo calves allowed to nurse the dam during the first month of life. Serum Ig concentration 24 hours after birth ranged from 28.1 to 35.9 mg/ml, birth weight ranged from 29 to 41 kg, body weight 30 days after birth ranged from 48.5 to 62.9 kg. The Average Daily Gain (ADG) from birth to day 30 ranged from 448 to 1,089 g/d. Significant linear associations were detected between serum Ig concentration 24 hours after birth and day-30 weight (p< 0.05; $R^2$ = 0.31) and between serum Ig concentration 24 hours after birth and ADG from birth to day 30 (p<0.001; $R^2$ = 0.72). Results indicated that passive transfer status was a significant source of variation in growth performance when buffalo calves nursed the dam. Maximizing passive transfer of immunity by allowing calves to nurse the dam can increase growth performance during the first month of life.

Antibody response to pneumococcal vaccination in children with chronic or recurrent rhinosinusitis

  • Purpose: Although chronic and recurrent rhinosinusitis is prevalent in children, little is known about its causes. Here, we investigated the humoral immunity in children with chronic or recurrent rhinosinusitis. Methods: We examined 16 children attending the outpatient clinic at the CHA Bundang Medical Center including 11 boys and 5 girls, aged 3-11 years (mean age, 5.6 years), who had rhinosinusitis for >3 months or >3 times per year. The complete blood count with differential and total serum concentrations of Immunoglobulin (Ig) E, IgA, IgD, IgM, IgG, and IgG subclasses ($IgG_1$, $IgG_2$, $IgG_3$, and $IgG_4$) of all children were measured. All subjects received 23-polysaccharide pneumococcal vaccination (PPV), and the levels of antibodies to 5 serologic types (4, 6B, 14, 18C, and 23F) of pneumococcal capsular polysaccharide antigens were measured before and after vaccination. Post-PPV antibody titers ${\geq}0.35{\mu}g/mL$ or with a ${\geq}4$-fold increase were considered as positive responses. Results: The titers of IgG, IgA, IgD, and IgM were within normal range in all 16 children, whereas the total IgE concentration was higher than normal in 2 children. $IgG_1$ deficiency was observed in 1 patient and $IgG_3$ deficiency in 3. After PPV, 1 patient failed to respond to all 5 serologic types, 2 failed to respond to 4 serologic types, and 2 failed to respond to 3 serologic types. Conclusion: Clinicians should consider the evaluation of humoral immune functions in children with chronic or recurrent rhinosinusitis who do not respond to prolonged antibiotic treatment.

Immunoliposomes Carrying Plasmid DNA : Preparation and Characterization

  • The objective of this study was to characterize immunoliposomes carrying plasmid DNA with optimal encapsulation efficiency and antibody density. Plasmid DNA was encapsulated by the freezing/thawing method into liposomes composed of POPC (1-palmitoyl-2-oleoyl-sn-glycerol- 3-phosphocholine), DDAB (didodecyl dimethyl ammonium bromide), DSPE-PEG 2000 (distearoyl phosphatidyl ethanolamine polyethylene glycol 2000) and DSPE-PEG 2000-maleimide. The liposomes carrying plasmid DNA were extruded through two stacked polycarbonate filters, of different pore size, to control the liposome size. Then, rat IgG molecules were conjugated to the liposomes. The immunoliposomes containing plasmid DNA were separated from the free plasmid DNA and unconjugated IgG by Sepharose CL-4B column chromatography. The DNA amount encapsulated was affected by DDAB (cationic lipid) concentration, the initial amount of plasmid DNA between 10 ${\mu}g$ and 200 ${\mu}g$, the total lipid amount and plasmid DNA size, but not significantly by liposome size. By varying the ratio of DSPE-PEG 2000-maleimide to IgG, the number of IgG molecules per liposome was changed significantly.

Fractionated Aged Black Garlic Extracts Enhance Growth of Anti-My-10 Hybridoma Cells and Production of IgG1 Antibody

  • Aged black garlic (ABG) was extracted with 20% ethanol and water (crude extracts) and fractionated into three categories (>10, 3-10, and <3 kDa). The effect of crude extract supplements on anti-My-10 hybridoma cell growth and IgG1 antibody production was investigated in suspension culture with a chemically defined protein-free medium. We observed that supplementation of ABG to the cell culture medium stimulated anti-My-10 hybridoma cell growth and production of IgG1 antibody, particularly with fractionated ABG of low molecular weight. The stimulation depended upon the concentration and the size of the fractionated ABG. We also found that the growth-promoting activity was not correlated with high antibody production. These results suggest that fractionated ABG is a novel and promising alternative as an animal cell culture supplement.

Changes in the serum immunoglobulin levels and viral antibody titers of colostrum-conferred Korean native calves during the first 12 weeks postpartum (초유를 섭취한 한우 송아지의 출생후 12주 동안의 혈청 면역글로불린과 각종 바이러스 항체가의 변화)

  • The changes in serum total protein and immunoglobulin levels, and BVD, IBR and PI-3 viral neutralizing antibody titers in colostrum-conferred Korean native calves during the first 12 weeks postpartum were studied, and the results obtained were summerized as follows: The Mean concentration of total protein, total immunoglobulin, IgG, IgM and IgA in sera of 9 calves at birth were $3.8{\pm}0.5g/dl$, $0.27{\pm}0.15mg/ml$, $0.06{\pm}0.08mg/ml$, $0.21{\pm}0.11mg/ml$, and extremely low concentration, respectively. Serum total protein level reached a maximum at 20 hours after birth, total immunoglobulin, IgG, and IgM levels at 24 hours, and IgA level at 28 hours, respectively. Serum IgA level reached a minimum at 4 weeks old, IgM level at 5 weeks, total immunoglobulin level at 8 weeks, and IgG level at 10 weeks, respectively. After then those levels had begun to increase, but total protein level was still decreasing at 12 weeks old. The half-lives of IgG, IgM, and IgA were 21.1 days, 4.0 days, and 2.6 days-respectively. In 10 Korean native cows immediately after parturition, serum neutralizing antibody titers specific to BVD, IBR and PI-3 virus were $8.7{\pm}1.5{\log}_2$, $5.7{\pm}1.2{\log}_2$, and $6.8{\pm}1.01{\log}_2$, respectively. And colostral neutralizing antibody titers against BVD, IBR, and PI-3 virus were $10.1{\pm}1.4{\log}_2$, $6.8{\pm}1.3{\log}_2$ and $7.8{\pm}1.7{\log}_2$, respectively. Before suckling the colostrum, SN antibody titers against BVD, IBR, and PI-3 virus were undetectable from all of 9 Korean native calves. Nevertheless SN antibody titer against BVD virus reached a maximum level ($9.2{\pm}0.6{\log}_2$) at 24 hours after birth, that against IBR virus ($6.1{\pm}1.0{\log}_2$) at 20 hours after birth, and that against PI-3 virus ($6.8{\pm}0.9{\log}_2$) at 32 hours after birth, respectively. In 12 weeks old calves, the SN antibodies against BVD and IBR virus were still decreasing, but that against PI-3 virus reached a minimum at 10 weeks, and increased after 12 weeks of age. The half-lives of SN antibodies against BVD, PI-3 and IBR, virus were 16.0 days, 22.6 days, and 25.5 days, respectively.

Changes in the Blood Components Caused by Water Intake (물 섭취에 의한 혈액 성분 변화에 관한 연구)

  • Although water is an essential component of the human body and is involved in many physiological processes, the effect of a steady and sufficient water intake on blood components has not well elucidated. Therefore, we investigated the changes in hematological parameters, high-sensitivity C-reactive protein (hs-CRP), and immunoglobulin G (IgG) after water intake in 13 healthy adults. They were divided into two groups: The control group (N=4), which consumed water ad libitum, and the experimental group (N=9), which consumed 2 L of water per day. Two weeks later, blood cell counts, hematocrit, and hemoglobin content had increased in the experimental group, although not significantly (p>0.05); however, there was a significant increase in the mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) (p<0.05; and p<0.01, respectively), and a significant decrease in the mean platelet volume (MPV) (p<0.05). Of the immunologic parameters, a non-significant decrease in the concentration of hs-CRP, an indicator of cardiovascular disease risk, was observed (p>0.05). However, there was a dramatic and significant increase in the concentration of IgG (p<0.05). In conclusion, a steady and sufficient water intake may contribute to alleviate anemia by increasing hemoglobin. Additionally, it may decrease the risk of cardiovascular disease by decreasing platelet activation and concentration of hs-CRP. Furthermore, a steady intake of water may improve immune function by increasing the concentration of the components of humoral immunity.

Changes in Compositions of Holstein Colostrum during Lactation Period (Holstein 젖소 초유의 성분 변화에 관한 연구)

  • To investigate the compositions of Holstein colostrums, samples were collected at 12 hour-interval after 12hrs postpartum. Milk protein, milk fat, SNF and total solid content of the colostum rapidly decreased from 12 hours to 48 hours after calving whereas lactose was the lowest at 12 hours after calving. Ash content was not shown to changes during lactation periods. Immunoglobulin G(IgG) concentration was also significantly(p<0.05) high in both primiparous and multiparous colostrum collected at 12 and 24 hours after calving. IgG concentration of primiparous and multiparous colostrums at 12 hours after calving was 44mg/ml and 44.27mg/ml, respectively. There was no apparent difference in IgG level between primipara and multipara. Fatty acid composition of colostral lipid was not shown to changes during lactation period. However, lauric acid, myristic acid and total saturated, fatty acid were slightly decreased in multiparous colostral lipid. Capric acid, myristic acid and palmitoleic acid composition in primiparous colostral lipid were slightly higher than those of multiparous colostral lipid throughout all lactation periods.
