Objective: Since athletic performance is a most importance trait in horses, most research focused on physiological and physical studies of horse athletic abilities. In contrast, the molecular analysis as well as the regulatory pathway studies remain insufficient for evaluation and prediction of horse athletic abilities. In our previous study, we identified AXL receptor tyrosine kinase (AXL) gene which was expressed as alternative spliced isoforms in skeletal muscle during exercise. In the present study, we validated two AXL alternative splicing transcripts (named as AXLa for long form and AXLb for short form) in equine skeletal muscle to gain insight(s) into the role of each alternative transcript during exercise. Methods: We validated two isoforms of AXL transcripts in horse tissues by reverse transcriptase polymerase chain reaction (RT-PCR), and then cloned the transcripts to confirm the alternative locus and its sequences. Additionally, we examined the expression patterns of AXLa and AXLb transcripts in horse tissues by quantitative RT-PCR (qRT-PCR). Results: Both of AXLa and AXLb transcripts were expressed in horse skeletal muscle and the expression levels were significantly increased after exercise. The sequencing analysis showed that there was an alternative splicing event at exon 11 between AXLa and AXLb transcripts. 3-dimentional (3D) prediction of the alternative protein structures revealed that the structural distance of the connective region between fibronectin type 3 (FN3) and immunoglobin (Ig) domain was different between two alternative isoforms. Conclusion: It is assumed that the expression patterns of AXLa and AXLb transcripts would be involved in regulation of exercise-induced stress in horse muscle possibly through an $NF-{\kappa}B$ signaling pathway. Further study is necessary to uncover biological function(s) and significance of the alternative splicing isoforms in race horse skeletal muscle.
Objectives : This study was carried out for the purpose of knowing the effect from anti-arthma action of the abstraction from a extract of Paridis Rhizoma(EPR). In order to know what the effect of controlling an abstraction from Paridis Rhizoma. and about the expression of B cells and Ig E cells, mast cells it was necessary for it to be activated by ovalbumin. Methods : In order to know what the effect was on the organization of cytokine gene expression from The increase and divorce of the B cells and allergic acting by EPR, we found it necessary to examine the BALF. At the same time, as we examined the histamine release by ELISA method, we also examined the effect of EPR. Results : EPR at $100\;{\mu}g/ml$, the highest concentration examined did not have any cytotoxic effects on mLFCs. In FACS analysis, number of granulocyte/lymphocyte, $CD3e^+/CCR3^+,\;CD4^+\;and\;CD23^+/B220^+$ in asthma-induced lung cells were significantly decreased by EPR treatment compared to the control group. In RT-PCR analysis, mRNA expression for CCR3, eotaxin and histamine in asthma-induced lung cells, which was induced by rIL-3 plus rmIL-5 treatments, was significantly decreased by EPR treatment. In ELISA analysis, production levels of IL-4, IL-13 and histamine in asthma-induced lung cells, which were induced by rIL-3 plus rmIL-5 co-treatment, were significantly decreased by EPR treatment. EPR treatments significantly inhibited the proliferation of eosinohils prepared from asthma-induced mouse lung tissues compared to the non-EPR treated control cells. Immunohistochemical analysis revealed that EPR treatment significantly decreased the levels of eosipnphil activation compared to non-treated cells. Conclusion : The present data suggested that Paridis Rhizoma may have an effects on the inhibition of parameters associated with asthma responses in eosinpophils, and thus implicate the possibility for the clinical application of Paridis Rhizoma.
흰털오가피의 면역활성 증진 효과를 알아보기 위하여 선발된 수컷 마우스를 대상으로 하여 발효유에 흰털오가피 (뿌리 : 잎 : 줄기)와 더덕 열수추출건조물을 (5 : 2 : 1.5) : 1.5 비율로 혼합하여 투여한 그룹 1 mg/mL(A), 3 mg/mL(B), 9 mg/mL(C) 3그룹으로 나누어 임상적용 경로인 경구투여를 선택하여 발효유를 각각 3 mL/kg씩 위내로 직접 투여하였다. 대조군은 발효유만 먹인 그룹(D)과 식염수만 먹인 그룹(E)을 두었다. 흰털오가피의 함량이 ICR계 수컷 마우스의 면역기능에 미치는 영향에 대해 알아본 결과 모든 그룹에서 마우스의 증체량과 체중 증가는 유의성 있는 차이를 보이지 않았고, 7주와 10주령에 안락사 시킨 마우스의 각 장기 무게에서도 간, 신장, 심장, 폐, 고환의 무게에서는 유의성 있는 차이를 보이지 않았다. 비장계수는 7주령 C군과 10주령 B군에서 유의성(p<0.05)있는 증가가 있었으며, 식이 지속기간이 길어질수록 증가된 반면, 흉선계수는 모든 군에서 유의성 있는 증가가 관찰되지 않았다. 항체생산 세포수는 흰털오가피 투여군이 대조군에 비해 증가하였으며, 7주령에 비해 10주령이 감소하는 경향을 보였다. 항원에 대한 항체 생성량을 알아보기 위하여 혈구응집반응을 실시한 결과 흰털오가피 투여군이 대조군에 비하여 증가하는 경향을 보였고 10주령 C군에서 유의성 있는 차이를 보였다. 면역글로블린 농도는 흰털오가피 투여군에서 증가하였고, 7 및 10주령 C군에서 유의성이 있었다.
본 연구에서는 유전자 변형 옥수수(GM 옥수수)에 특이한 단크론성 항체를 개발하고 이에 대한 특성을 확인하는 연구를 수행하고자 하였다. 먼저 형질전환 대장균으로부터 PAT 단백질을 발현시킬 수 있는 시스템을 확립하였고, 재조합 PAT 단백질을 대량 생산하여 항원으로 사용하였다. 준비된 항원을 면역한 결과 재조합 PAT 단백질의 항원성은 매우 높은 것으로 확인되었으며, 세포융합과 클로닝을 통해 12 종의 hybridoma를 확립하였고 western blot 결과 10 종의 hybridoma가 재조합 PAT 단백질과 강한 반응성을 나타내었다. 10종의 hybridoma가 생산하는 항체가 실제 GM 옥수수에 반응하는지를 추가의 western blot으로 분석한 결과 2종의 단크론성 항체(PATmAb-7 and PATmAb-12)가 재조합 PAT 단백질뿐만 아니라 실제 GM 옥수수 중 PAT와 반응하는 것으로 확인되었다. 항체를 대량 생산하고 정제한 후 2종의 항체는 SDS-PAGE 상에서 대표적인 항체의 분리패턴(heavy와 light chain)을 나타내었고, 전형적인 $IgG_1$과 ${\kappa}$ type으로 확인되었다. 정제된 단크론성 항체는 특성을 조사한 결과 다른 GMO에서 발현될 수 있는 재조합 단백질과 non-GM 옥수수 추출물에는 반응성이 없고 PAT 단백질에만 특이적으로 반응하는 것을 확인할 수 있었다. PATmAb-7 를 이용한 간접효소면역분석법의 검출한계는 0.3 ng/mL 수준으로 기준의 유전자변형 콩 면역분석법과 비교했을 때 높은 민감도를 나타내었다. 이상의 결과로 볼 때 개발된 2종의 항체(PATmAb-7 and PATmAb-12)는 GM 옥수수에서 발현되는 PAT 단백질에 특이적으로 반응하는 항체로 확인되었고, 2종의 항체를 이용한 면역분석법과 바이오센서의 개발 가능성을 제시할 수 있었다.
Background: The prevalence of allergic inflammatory diseases such as atopic dermatitis (AD), asthma, and allergic rhinitis worldwide has increased and complete recovery is difficult. Korean Red Ginseng, which is the heat-processed root of Panax ginseng Meyer, is widely and frequently used as a traditional medicine in East Asia. In this study, we investigated whether Korean Red Ginseng water extract (RGE) regulates the expression of proinflammatory cytokines and chemokines via the mitogen-activated protein kinases (MAPKs)/nuclear factor kappa B ($NF-{\kappa}B$) pathway in allergic inflammation. Methods: Compound 48/80-induced anaphylactic shock and 1-fluoro-2,4-dinitrobenzene (DNFB)-induced AD-like skin lesion mice models were used to investigate the antiallergic effects of RGE. Human keratinocytes (HaCaT cells) and human mast cells (HMC-1) were also used to clarify the effects of RGE on the expression of proinflammatory cytokines and chemokines. Results: Anaphylactic shock and DNFB-induced AD-like skin lesions were attenuated by RGE administration through reduction of serum immunoglobulin E (IgE) and interleukin (IL)-6 levels in mouse models. RGE also reduced the production of proinflammatory cytokines including $IL-1{\beta}$, IL-6, and IL-8, and expression of chemokines such as IL-8, thymus and activation-regulated chemokine (TARC), and macrophage-derived chemokine (MDC) in HaCaT cells. Additionally, RGE decreased the release of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), $IL-1{\beta}$, IL-6, and IL-8 as well as expressions of chemokines including macro-phage inflammatory protein $(MIP)-1{\alpha}$, $MIP-1{\beta}$, regulated on activation, normal T cell expressed and secreted (RANTES), monocyte chemoattractant protein (MCP)-1, and IL-8 in HMC-1 cells. Furthermore, our data demonstrated that these inhibitory effects occurred through blockage of the MAPK and $NF-{\kappa}B$ pathway. Conclusion: RGE may be a useful therapeutic agent for the treatment of allergic inflammatory diseases such as AD-like dermatitis.

The prime-boost vaccination with DNA vaccine and recombinant viral vector has emerged as an effective prophylactic strategy to control infectious diseases. Here, we compared the protective immunities induced by multiple alternating immunizations with DNA vaccine (pCIgB) and replication-incompetent adenovirus (Ad-gB) expressing glycoprotein gB of pseudorabies virus (PrV). The platform of pCIgB-prime and Ad-gB-boost induced the most effective immune responses and provided protection against virulent PrV infection. However, priming with pCIgB prior to vaccinating animals by the DNA vaccine-prime and Ad-boost protocol provided neither effective immune responses nor protection against PrV. Similarly, boosting with Ad-gB following immunization with DNA vaccine-prime and Ad-boost showed no significant responses. Moreover, whereas the administration of Ad-gB for primary immunization induced Th2-type-biased immunity, priming with pCIgB induced Th1-type-biased immunity, as judged by the production of PrV-specific IgG isotypes and cytokine IFN-$\gamma$. These results indicate that the order and injection frequency of vaccine vehicles used for heterologous prime-boost vaccination affect the magnitude and nature of the immunity. Therefore, our demonstration implies that the prime-boost protocol should be carefully considered and selected to induce the desired immune responses.
The purpose of this study is to examine the effect of herbal medicine complex (HMC) containing Camellia sinensis L., Duchoesna chrysantha, Houttuynia cordata Thunberg, Poncirus trifoliata Rafinesque on skin inflammation and atopic dermatitis. First, we examined the anti-inflammatory effect of HMC in TNF-α induced human keratinocytes (HaCaT cell). Real-time PCR and western blotting were performed to evaluate the expression of inflammatory cytokines (e.g., iNOS, COX-2, IL-6, IL-8) mRNA and protein. Four-weeks old male NC/Nga mice were treated with 1% 2,4-dinitrochlorobenzene (DNCB) solution and used as an atopic dermatitis mice model. And, HMC (200 mg/kg or 400 mg/kg) was administered directly into the stomach of mice for 4 weeks, and blood or serum analysis, tissue staining were performed after oral gavage. As a result HMC inhibited the mRNA expression of iNOS, COX-2, IL-6, and IL-8, which had been increased by TNF-α in HaCaT cells. In addition, the protein expression was also significantly suppressed in the same way as the mRNA expression results. The in vivo experiment results showed that, HMC administration reduced thickening of the epidermis and infiltration of eosinophil into the skin stratum basale compared to DNCB treatment. In addition, HMC administration significantly reduced the inflammatory cytokines (IL-4, IL-5, IL-6, and IL-13) production and immunocyte (white blood cell, lymphocyte, neutrophil, and eosinophil) count compared to DNCB treatment. Moreover, the serum IgE and histamine level was decreased by HMC administration. These results suggest that HMC can be used as effective herbal medicine extract for skin inflammation and atopic dermatitis. And this study may contribute to the development of the herbal medicine-based drug for the treatment of skin inflammation and atopic dermatitis.
본 연구에서는 일본식으로 제조된 상업용 간장과 우리나라 전통방식으로 제조된 재래식 간장으로부터 다당을 분리하여 RAW 264.7 대식세포주를 이용한 면역 증진활성을 비교하였다. 간장 유래 다당, CSP-0 및 KTSP-0는 RAW 264.7 세포주에 대해 모든 농도에서 세포 독성을 나타내지 않았다. 또한 재래식 간장 유래 다당인 KTSP-0는 상업용 간장 유래 다당 CSP-0보다 대식세포에 의한 NO 및 ROS의 생산을 농도의 존적으로 증가시켰으며, KTSP-0를 $1,000{\mu}g/m{\ell}$ 처리하였을 시 가장 높은 생산능을 나타내었다. 간장 유래 다당의 RAW 264.7 세포로부터 면역반응에 중요한 cytokine인 IL-6와 TNF-${\alpha}$의 cytokine mRNA의 발현량과 해당 단백질의 생산을 각각 real-time PCR과 ELISA로 확인한 결과, CSP-0는 IL-6와 TNF-${\alpha}$ mRNA의 발현 및 생산에 유의적인 영향을 미치지 않았지만, KTSP-0는 농도의존적으로 IL-6와 TNF-${\alpha}$ mRNA 발현 및 생산을 증가시키는 것을 확인하였으며, $1,000{\mu}g/m{\ell}$ 처리 시 최대 활성을 보였다. 한편, 대식세포의 탐식작용에서 중요한 역할을 하는 Fc 수용체의 발현 증가를 RT-PCR로 확인한 결과, CSP-0는 FcR I, II의 발현에 모두 영향을 미치지 않았지만, KTSP-0는 FcR I의 발현을 선택적으로 증가시킴이 확인되어 항원에 결합한 IgG와 강하게 결합하여 탐식작용을 촉진시킬 것으로 예상되었다. 본 연구를 통해 상업용 간장에서 분리한 다당, CSP-0보다 우리나라 전통 재래식 간장에서 분리한 다당, KTSP-0가 대식세포를 활성화하여 높은 면역 증진 효과를 나타내는 것으로 결론 지을 수 있었다.
본 연구는 브로일러 사료 내 항생물질을 대체하기 위한 천연항균성장촉진제로서 국산 돼지감자로부터 추출, 제조한 미세캡슐화, 이눌로프리바이오틱스의 첨가에 따른 성장능력, 혈액 면역물질 및 맹장 미생물 변화를 조사하였다. 로스계통(Ross 308)의 성감별을 실시한 수컷 브로일러 240수를 4처리구$\times$3반복(반복 펜 당 20수)으로 완전임의 배치하였다. 브로일러는 기본사료(T1: 대조구), 항생제(T2: Chlorotetra cycline, CTC 0.1%), 이눌로프리바이오틱스 300ppm(T3), 이눌로프리바이오틱스 450ppm(T4) 함유사료를 각각 35일 동안 섭취하였다. 브로일러의 체중 및 도체율은 이눌로프리바이오틱스를 섭취한 처리구(T3, T4)가 대조구 (T1) 및 항생제(T2) 함유사료를 섭취한 처리구에 비해서 높았으며 통계적인 유의차(P<0.05)가 인정되었다. 브로일러의 도체중에 대한 비율로서 나타낸 닭 가슴살 및 다리살의 무게 비율은 T3, T4가 T1, T2에 비해서 높았으며 통계적인 유의차(P<0.05)가 인정되었다. 복강지방 무게는 T3, T4가 T2, T1에 비해서 유의하게(P<0.05) 감소경향을 나타냈다. 혈액 면역물질 IgG는 T3, T4가 T1, T2와 비교할 때 유의하게(P<0.05) 증가하였다. 생체중에 대한 비율로서 나타낸 면역기관 흉선, F낭의 무게 비율은 T3, T4가 T1, T2에 비해서 유의하게 (P<0.05) 높은 경향을 나타냈다. 장내 유익한 bifidobacteria와 Lactobacillus는 T3, T4가 T1, T2에 비해서 높았으나 유해한 E. coli와 Salmonella는 오히려 낮게 나타났으며 통계적인 유의차(P<0.05)가 인정되었다. 본 연구에서 나타난 중요한 점은 유기축산 사료용 항생제 대체 물질로써 이눌로프리바이오틱스 300ppm 수준을 브로일러 사료에 첨가 급여해 줌으로서 브로일러의 생산성을 크게 향상시킬 수 있다는 사실이었다.
연구배경: 외인성 천식은 TH2세포에 의한 매개체의 주요 원인중 하나임이 밝혀지고 있다. 연구자 등은 증상 악화로 내원한 기관지천식 환자와 만성기관지염 환자 사이에 T 림프구 아형의 변화와 싸어토카인 (cytokine)들의 변화에 차이가 있는지 연구하고자 하였다. 방 법: 기관지 천식으로 치료중인 환자 중에서 천식 악화로 내원한 외인성 천식 15예와 만성기관지염 환자 12예, 그리고 정상인 5예를 대상으로 하였다. 환자의 병력과 임상소견, 피부반응검사, 그리고 특이 IgE 측정을 시행하고 단일항체인 CD62L를 이용하여 flow cytometer로 림프구아형을 분석하고 ELISA kit(Quantikine IL-4, IFN-$\gamma$)를 이용하여 IL-4, IFN-$\gamma$을 측정하였다. 결 과: CD4+ T 림프구는 기관지천식환자에서 $40{\pm}7.2%$ 만성기관지염 환자 $43{\pm}19.8%$, 정상인에서 $41{\pm}14%$로 기관지 천식환자와 다른 군간에 의미있는 차이는 없었다(p=0.49, p=0.75). CD62L(L-selectin) 양성 T 림프구의 세포 백분율은 기관지천식환자(n=7) 에서 $24.8{\pm}23.6%$였고, 만성기관지염환자(n=5) $17.0{\pm}16.9%$, 정상인(n=5) $16.7{\pm}16.4%$으로 기관지천식환자와 다른 군간에 의미있는 차이는 없었다(p=0.32, p=0.22). 혈청 IL-4 의 활성도는 기관지천식환자에서 $3.6{\pm}0.9pg/ml$ 만성기관지염 환자 $2.0{\pm}1.2pg/ml$, 정상인에서 $0.7{\pm}1.1pg/ml$로 기관지 천식환자에서 만성기관지염 환자군에 비하여 증가되어 있었으며 (p=0.02) 정상인보다 증가되어 있었다(p=0.006)(Fig. 4). 혈청 INF-$\gamma$의 활성도는 증가되지 않았다. 결 론: 결론적으로 CD62L 양성 T 림프구의 세포 백분율은 기관지천식환자에서 증가되어 있지 않았으며 혈청 IL-4의 활성도는 기관지천식환자에서 증가되어 있었다.

허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.

ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
