Intermittent chlorination shifts the marine biofilm population on reverse osmosis membranes

  • The influence of chlorine on marine bacterial communities was examined in this study. A non-chlorine-adapted marine bacterial community (NCAM) and a chlorine-adapted bacterial community (CAM, bacterial community treated with $0.2mg-Cl_2/L$ chlorine) were cultivated for 1 month. A distinct difference was observed between the NCAM and CAM, which shared only eight operational taxonomic units (OTUs), corresponding to 13.1% of the total number of identified OTUs. This result suggested that chlorine was responsible for the changes in the marine bacterial communities. Kordiimonas aquimaris was found to be a chlorine-resistant marine bacterium. The effect of intermittent chlorination on the two marine biofilm communities formed on the reverse osmosis (RO) membrane surface was investigated using various chlorine concentrations (0, 0.2, 0.4, 0.6 and 0.8 mg $Cl_2/L$). Although the average number of adherent marine bacteria on the RO membrane over a period of 7 weeks decreased with increasing chlorine concentration, disinfection efficiencies showed substantial fluctuations throughout the experiment. This is due to chlorine depletion that occurs during intermittent chlorination. These results suggest that intermittent chlorination is not an effective disinfection strategy to control biofilm formation.

Evolution and Identification of Thermo-Tolerant Hybrids in the Silkworm, Bombyx mori L.

  • Four thermo-tolerant lines of silkworm, Bombyx mori, (L.) viz., A HT, B HT (Chinese type) and F HT, G HT (Japanese type) were evolved by utilizing the breeding resource material (identified from initial screening at a temperature of 31 ${\pm} 1^{\circ}C$ and relative humidity 85 ${\pm}$ 5%) through conventional breeding. These tolerant lines were crossed with productive breeds and forty four hybrids were evaluated on eight economic traits by the Multiple Trait Evaluation Index Method. Ten hybrids were short-listed based on the average evaluation index value larger than 50 for eight economic traits studied. The identified ten hybrids recorded higher index values (> 50) for most of the traits studied. Single hybrid G ${\times}$ CSR12 indicated average index value larger than 50 for six traits viz., pupation number (58), cocoon weight (67), shell weight (65), average filament length (74), raw silk % (69), reelability % (51) except for shell ratio % (41). The standard deviation of the cocoons in the above hybrid was 8.41 in the hybrid cocoon length and width measurement. However, two selected hybrids viz., A ${\times}$ CSR5 and G ${\times}$ CSR13 recorded average index value larger than 50 for all the traits viz., pupation number (57, 60), cocoon weight (50, 54), shell weight (56, 57), shell ratio percentage (59, 53), average filament length (55, 60), raw silk percentage (63, 67) and reelability percentage (53, 53). The standard deviation of the cocoons in the two selected hybrids viz., A ${\times}$ CSR5 and G ${\times}$ CSR13 was 8.41 and 8.06 respectively in the cocoon length and width measurement.

A Comparison of the Designation Characteristics of Korean Scenic Sites Policies and National Park System in the United States (국내 명승 정책과 미국 국립공원 시스템의 지정 특성 비교)

  • This study examined the definition and major values, the designated procedures and types, and the designation trend in Korean scenic sites and national parks in the United States. Based on this, the analysis of the characteristics of the designation of the two natural heritages. The results are as follows; First, Scenic Sites has characteristics of complex heritage that includes academic, historical, and humanities values on the basis of landscape. As a natural heritage based on public nature, the U.S. National Park aims to contribute to the people's natural heritage and satisfy both ecological and historical values through the protection of the landscape. Second, the designation of a scenic sites are decided through deliberation by the Cultural Heritage Committee after the request of the owner, manager, or local government or by the authority of the head of the Cultural Heritage Administration. The designated survey is divided into basic resource surveys and resource surveys by type. Since the initial designation of the Sogeumgang Mountain in Cheonghakdong, Myeongju in 1970, the number of designated scenic sites was low until the 2000s, but the number of designated scenic sites has increased rapidly since 2006 due to the policy to promote the scenic site, and the proportion of natural and historical and cultural scenic sites has been balanced. The designation of the U.S. national park is decided by the Congress or the president, and the National Park Service makes a series of decisions on whether to conduct a special resource study of provisional resources through a preliminary inspection survey, whether to satisfy the criteria for designation of national parks based on the results of special resource research, and to prioritize them. The U.S. National Parks have been expanded not only by Congress but also by the president's empowerment to designate them as national monuments. With the integrated operation of the National Park Service, the number of designated cases increased as the national park included the heritage sites under the control of various ministries. In addition, a number of historical areas were designated by the enactment of the Historical Site Act, and recreational areas were designated to provide leisure space and classified and managed in a total of 18 units. Third, the comparison of the designation characteristics of the two heritage properties confirmed that the designation of natural heritage with complex value, the classification of types according to complementary designation system and resource characteristics, the establishment of the competent ministry and the balancing of the heritage according to the designation policy. The two heritages had the characteristics of complex natural heritages that met ecological, historical and academic values at the same time based on landscape and public nature. In addition, both countries have identified a system for deliberating the designation of heritage through a basic resource survey and an in-depth designation survey, and classified each type according to the characteristics of the resource. In addition, the policies for promoting scenic sites in Korea and the integrated operation of the National Park Service in the U.S. influenced the designated aspects of the two heritage sites, balancing natural heritage with historical and cultural heritage. Fourth, the resource types and conservation management methods of Scenic site and National Park were largely related. The natural areas of the U.S. National Park include types of natural monuments in Korea as major resources, and have characteristics similar to natural scenic sites. In addition, historical resources were similar to the criteria for designation of historical and cultural scenic sites in terms of landscape, and the aspects of war and celebrity-related relics were related to the types of historic sites. In terms of conservation management, the natural area of the U.S. national park has a way of keeping the original ecosystem intact, but the Korean natural heritage protection system is likely to be useful for focusing on the resource of viscosity. Meanwhile, historical resources include historical sites and historical and cultural scenic sites in the traditional era, but historical relics in the U.S. National Parks have set a time limit to modern times for war history and celebrity-related relics, and the active provision of entertainment programs based on existing resources was derived as a difference.

Genome-wide Copy Number Variation in a Korean Native Chicken Breed (한국 토종닭의 전장 유전체 복제수변이(CNV) 발굴)

  • Copy number variation (CNV) is a form of structural variation that shows various numbers of copies in segments of the DNA. It has been shown to account for phenotypic variations in human diseases and agricultural production traits. Currently, most of chicken breeds in the poultry industry are based on European-origin breeds that have been mostly provided from several international breeding companies. Therefore, National Institute of Animal Science, RDA has been trying to restore and improve Korean native chicken breeds (12 lines of 5 breeds) for about 20 years. Thanks to the recent advance of sequencing technologies, genome-wide CNV can be accessed in the higher resolution throughout the genome of species of interest. However, there is no systematic study available to dissect the CNV in the native chicken breed in Korea. Here, we report genome-wide copy number variations identified from a genome of Korean native chicken (Line L) by comparing between the chicken reference sequence assembly (Gallus gallus) and a de novo sequencing assembly of the Korean native chicken (Line L). Throughout all twenty eight chicken autosomes, we identified a total of 501 CNVs; defined as gain and loss of duplication and deletion respectively. Furthermore, we performed gene ontology (GO) analysis for the putative CNVs using DAVID, leading to 68 GO terms clustered independently. Of the clustered GO terms, genes related to transcription and gene regulation were mainly detected. This study provides useful genomic resource to investigate potential biological implications of CNVs with traits of interest in the Korean native chicken.

Improvement of Abiotic Stress Resilience for Stable Rice Production

  • Recently, stable crop production is threatened by the effects of climate change. In particular, it is difficult to consistently maintain agricultural policies due to large price fluctuations depending on the difference in total domestic rice production from year to year. For stable rice production amid changes in the crop growing environment, development of varieties with improved disease resistance and abiotic stress stability is becoming more important. In here, drought and cold tolerant trait have been studied. First, for the development of drought tolerant varieties, we analyzed which agricultural traits are mainly affected by domestic drought conditions. As a result, it was observed that drought caused by the lack of water during transplanting season inhibits the development of the number of tiller and reduces the yield. 'Samgang' was selected as a useful genetic resource with strong drought tolerant and stable tiller number development even under drought conditions by phenotype screening. Three of drought tolerant QTLs were identified using doubled haploid (DH) population derived from a cross between Nacdong and Samgang, a drought sensitive and a tolerant, respectively. Among these QTLs, when qVDT2 and qVDTl1 were integrated, it was investigated that the tiller number development was relatively stable in the rainfed paddy field conditions. It is known that the high-yielding Tongil-type cultivars are severely affected by cold stress throughout the entire growth stage. In this study, we established conditions that can test the cold tolerance phenotype with alternate temperature to treat low temperatures in indoor growth conditions similar to those in field conditions at seedling stage. Three cold tolerant QTLs were explored using population derived from a cross between Hanareum2 (cold sensitive variety, Tongil-type) and Unkwang (cold tolerant variety, Japonica). Among these QTLs, qSCT12 showed strong cold tolerant phenotype, and when all of three QTLs were integrated, it was investigated that cold tolerant score was relatively similar to its donor parent, Unkwang, in our experimental conditions. We are performing that development of new variety with improved cold tolerant through the introduction of these QTLs.

Isolation and Identification of Lactic Acid Bacteria from Spent Mushroom Substrate for Silage Making and Determination of Optimal Medium Conditions for Growth

  • This study was conducted to isolate and identify the lactic acid bacteria (LAB) from spent mushroom substrates (SMS) for the effective anaerobic fermentation to utilize SMS as an animal feed and to determine the optimal medium conditions for their growth. At first, a total of 23 strains were isolated from the ensiled SMS based on the LAB counts and pH tested. Then, a total of 16 strains which rapidly produce lactate and decreased the pH, were selected for a screening test. The optical density (OD), pH, and yellow clear zone were tested for the selected 16 strains. Among the strains, KU5 strain had wider yellow clear zone and lower pH and KU13 strain had higher OD at 24 hr of incubation and wider yellow clear zone compared to other strains and control strain (Lactobacillus plantarum KCCM 12116). Accordingly, KU5 and KU13 strains were finally selected. The KU5 and KU13 were identified as Lactobacillus plantarum by the 16S rRNA sequencing. The KU5 strain was named as Lactobacillus plantarum KU5, and the KU13 strain was named as Lactobacillus plantarum KU13. Lactobacillus plantarum KU5 and Lactobacillus plantarum KU13 were registered at the National Center for Biotechnology Information (NCBI). Access number of Lactobacillus plantarum KU5 was HQ542227 and that of Lactobacillus plantarum KU13 was HQ542228. The optimal medium conditions for growth of KU5 and KU13 were soybean meal 2% and formulated feed 2%, respectively.

A whole genome sequence association study of muscle fiber traits in a White Duroc×Erhualian F2 resource population

  • Objective: Muscle fiber types, numbers and area are crucial aspects associated with meat production and quality. However, there are few studies of pig muscle fibre traits in terms of the detection power, false discovery rate and confidence interval precision of whole-genome quantitative trait loci (QTL). We had previously performed genome scanning for muscle fibre traits using 183 microsatellites and detected 8 significant QTLs in a White Duroc×Erhualian F2 population. The confidence intervals of these QTLs ranged between 11 and 127 centimorgan (cM), which contained hundreds of genes and hampered the identification of QTLs. A whole-genome sequence imputation of the population was used for fine mapping in this study. Methods: A whole-genome sequences association study was performed in the F2 population. Genotyping was performed for 1,020 individuals (19 F0, 68 F1, and 933 F2). The whole-genome variants were imputed and 21,624,800 single nucleotide polymorphisms (SNPs) were identified and examined for associations to 11 longissimus dorsi muscle fiber traits. Results: A total of 3,201 significant SNPs comprising 7 novel QTLs showing associations with the relative area of fiber type I (I_RA), the fiber number per square centimeter (FN) and the total fiber number (TFN). Moreover, one QTL on pig chromosome 14 was found to affect both FN and TFN. Furthermore, four plausible candidate genes associated with FN (kinase non-catalytic C-lobe domain containing [KNDC1]), TFN (KNDC1), and I_RA (solute carrier family 36 member 4, contactin associated protein like 5, and glutamate metabotropic receptor 8) were identified. Conclusion: An efficient and powerful imputation-based association approach was utilized to identify genes potentially associated with muscle fiber traits. These identified genes and SNPs could be explored to improve meat production and quality via marker-assisted selection in pigs.

A Study on Financial Management Practices of Rural Housewives (농촌 주부의 재무관리 행동에 관한 연구 -도시근교 농촌을 대상으로-)

  • The first objective of this study is to examine the finanacial management practices of rural housewives. The second objective is to determine the effects of sociodemographic characteristics on financial management practices. The Deacon & Firebaugh's model of family resource management framework and previous studies were used to determine the effects, regarding a9e, income, number of family, education level, and farming/nonfarming as independent/input variables, and monthly saving and managerial behavior index as dependent/throughput variables. A sample of 179 rural housewives aged less than 60 was selected from Shihung-Si Gyonggi-Do Province. Results showed that rural housewives more frequently did keeping bills, making purchase-list and verifying purchase need than recording where money spent making financial plan, and evaluating spending. With regard to household debt use, 60% of the sample had debt the most borrower used debt for farming, 73% of borrowers paid for their debt behind the schedule, and the main source of borrowing was NACF (NongHyup). Using installment credit, the rural housewives mainly bought cosmetics. 25% of the sample had credit cards. Average debt was 6, 070, 000 won, and 81% of annual income. In terms of saving, 85% of the sample saved, and 23% of the sample lived with the money after save. The main reasons of saving were for education and marriage of children and emergency. The main saving institutions were NACF and NLCF (ChukHyup). The regression showed that income was negatively associated with monthly saving, and age was identified as the positive determinants of managerial behavior index.

  • The increasing penetration of distributed energy resources on the distribution networks have brought a number of technical impacts where voltage and thermal variations have been identified as the dominant effects. Active network management in distribution networks aims to integrate distributed energy resources with flexible network management so that distributed energy resources are organized to make better use of existing capacity and infrastructure. This paper propose active solutions which aims to solve the voltage and thermal issues in a distributed manner utilizing a collaborative approach. The proposed algorithms have been fully tested on a distribution network with distributed generation units.

Analysis of Human Resource Status in Internal Accounting Control Systems in the Distribution and Service Industries

  • Purpose: We sought to investigate the operation status of internal accounting control systems in the distribution and service industries and verify whether investing in internal accounting personnel significantly influences the quality of the companies' financial reporting. Research design, data and methodology: We identified descriptive statistics and performed a univariate analysis. Furthermore, a regression analysis verified the impact of investing in internal accounting personnel on the absolute value of discretionary accruals, a proxy for financial reporting quality. Results: Distribution and service companies allocated a significantly higher number of accountants to their internal accounting control systems than companies in other industries. We also confirmed that the absolute value of discretionary accruals significantly decreased as the average working experience of internal accounting personnel in distribution and service companies, in months, increased. Conclusions: In this study, we confirmed that distribution and service companies are devoting efforts to establishing internal accounting control systems and found that financial reporting quality is more effectively controlled as companies allocate more experienced personnel in their internal accounting control system.
