Isolation and Identification of Nonpolar Taxane Derivatives from the Plant Cell Culture of Taxus chinensis

  • Nonpolar taxoides extracted from a large-scale cell culture of Taxus chinensis were isolated through the normal and reverse phase column chromatographies, and their compounds were identified via NMR spectroscopy. The complete separation method was systematically established and described. In dichloromethane, dissolved paclitaxel and other taxoids with hexane were precipitated during the purification of paclitaxel from the plant cell culture of T. chinensis through a large-scale process while the relatively nonpolar taxane derivatives remained dissolved in the hexane phase. 13-Deoxy baccatin III (I), baccatin VI (II), taxchinin I (III), $2{\alpha}$, $5{\alpha}$, $10{\beta}$, $14{\beta}$-tetraacetoxy-4(20), 11-taxadiene(IV), 1-deoxy baccatinVI(V), and taxayuntin C (VI) were isolated through column chromatography and identified via NMR spectroscopy. Compounds I and IV were found to the major components, aside from paclitaxel, in the plant cell culture of T. chinensis. The concentrations of I and IV were compared with the that concentration of the paclitaxel in each of plant cell culture. The possible applications of compounds I, II, IV, and V were discussed.

XAD-16-[4-(2-thiazolylazo)]orcinol 킬레이트 수지에 의한 금속이온의 분리 및 농축에 관한 연구 (Studies on the Separation and Preconcentration of Metal Ions by XAD-16-[4-(2-thiazolylazo)] orcinol Chelating Resin)

  • XAD-16-[4-(2-thiazolylao)]orcinol (TAO)형 킬레이트 수지에 대한 U(VI), Tb(IV), Zr(IV), Cu(II), Pb(II), Ni(II), Zd(II), Cd(II) 및 Mn(II) 이온의 흡착 및 탈착 특성을 용리법으로 조사하였다. 각 금속이온의 총괄용량과 pH 변화에 따르는 혼합 금속이온의 분리능 및 금속이온의 최적 흡착 조건을 조사하기 위하여 금속이온의 흡착에 미치는 흐름속도, 완충용액의 농도에 대한 영향을 검토하였다. 킬레이트 수지에 의한 금속이온의 총괄 흡착 용량은 각각 0.35nmol U(VI)/g resin, 0.49nmol Th(VI)/g resin, 0.41nmol Cu(II)/g resin 및 0.31nmol Zr(IV)/g resin이었다. 그리고 pH 5.0에서 돌파점 용량과 총괄 용량으로부터 얻은 금속이온의 용리순서는 Th(IV)>Cu(II)>U(VI)>Zr(IV)>Pb(II)>Ni(II)>Zn(II)>Mn(II)>Cd(II)이었다. 흐름속도 0.28mL/min 및 pH 2~5범위에서 혼합 금속이온을 분리한 결과 pH가 증가함에 따라 이들 이온의 군분리가 가능하였다. 한편, $HNO_3$, HCl, $HClO_4$, $H_2SO_4$$Na_2CO_3$ 등의 탈착제에 의한 탈착특성을 조사한 결과 2M $HNO_3$는 Zr(IV)을 제외한 대부분의 금속이온들은에 대하여 높은 탈착효율을 나타냈으며, 1M $H_2SO_4$ 용액을 사용하면 Zr(IV)의 탈착 및 회수가 가능하였다. 또한 킬레이트 수지를 이용하여 미량의 U(VI) 이온이 함유된 인공 해수 시료를 용리시키고 회수한 결과 94%이상의 높은 회수율을 나타내었다. 아울러 XAD-16-TAO 킬레이트 수지는 희토류 금속의 혼합시료를 2M $HNO_3$ 용액으로 용리시키면 U(VI)의 선택적인 분리, 농축 및 회수에 유용함을 알 수 있었다.

  • 해수 중 U(VI) 등과 같은 미량 금속이온의 선택적인 분리, 농축 및 회수를 위하여 XAD-16-[2-(2-thiazolylazo)-p-cresol](TAC)형 킬레이트 수지에 대한 몇 가지 금속이온의 흡착 및 탈착 특성을 용리법으로 조사하였다. 금속이온의 흡착에 미치는 흐름속도, pH 및 완충 용액의 농도에 대한 영향을 조사하여 흡착 최적 조건을 결정하였다. 킬레이트 수지에 대한 금속이온의 총괄 흡착 용량을 조사한 결과 각각 0.41mmol U(VI)/g resin, 0.55mmol Th(IV)/g resin, 0.43mmol Cu(II)/g resin 및 0.32mmol Zr(IV) /g resin이었다. pH 5.0에서 돌파점 용량과 총괄 용량으로부터 얻은 금속이온의 용리 순서는 Th(IV)>Cu(II)>U(VI)>Zr(IV)>Pb(II)>Ni(II)>Zn(II)>Cd(II)>Mn(II)이었다. $HNO_3$, HCl, $HClO_4$, $H_2SO_4$$Na_2CO_3$ 등의 탈착제에 의한 탈착특성을 조사한 결과 Zr(IV)을 제외한 대부분의 금속이온들에 대하여 2M $HNO_3$이 탈착효율이 높게 나타났으며, 1M $H_2SO_4$ 용액을 사용하면 Zr(IV)의 탈착 및 회수가 가능하였다. 또한 킬레이트수지를 이용하여 미량의 U(VI) 이온이 함유된 인공 해수 시료를 용리시키고 회수한 결과 96% 이상의 높은 회수율을 나타내었다.

  • 무기시료 중에 게르마늄의 DPP에 의한 미량 분석법이 연구되었다. 1, 2, 3-트리하이드 록시벤젠이 함유된 과염소산 용액 중에 게르마늄(IV)의 환원 피크가 -0.45V(기준전극 Ag/AgCl)에서 나타났고, 이 피크의 높이는 농도에 직선성을 보였다. 게르마늄 분석시에 감도 및 정밀도에 영향을 주는 인자들이 조사되었고, 최적 조건하에서 검출한계는 1ng/ml였다. 무기물 시료들은 potassium pyrosulfate를 사용한 용융법으로 분해하였다. 게르마늄 검출시 Se(IV), Pb(II), As(III)의 간섭에 대한 조사가 있었고, 이들 간섭은 10M HCl용액 중에서 $CCl_4$로 추출함으로써 해결할 수 있었다. 무기물 시료들 (Pb bf. dust, Cu bf. dust, 편마암, Cu anode slime)중의 게르마늄 함량은 위의 방법으로 정량되었다.

고온 화력 P91강 재열증기배관의 건전성 제고 방안 (Schemes to enhance the integrity of P91 steel reheat steam pipe of a high-temperature thermal plant)

  • A number of so-called 'Type IV' cracking was reported to occur at the welded joints of the P91 steel or P92 steel reheat steam piping systems in Korean supercritical thermal power plants. The reheat steam piping systems are subjected to severe thermal and pressure loading conditions of coolant higher than 570℃ and 4MPa, respectively. In this study, piping analyses and design evaluations were conducted for the piping system of a specific thermal plant in Korea and suggestions were made how structural integrity could be improved so that type IV cracks at the welded joints could be prevented. Integrity evaluations were conducted as per ASME B31.1 code with implicit consideration of creep effects which was used in original design of the piping system and as per nuclear-grade RCC-MRx code with explicit consideration of creep effects. Comparisons were made between the evaluation results from the two design rules. Another approach with modification or reduction of the redundant supports in the piping systems was investigated as a tool to mitigate thermal stresses which should essentially contribute to prevention of Type IV cracking without major modification of the existing piping systems. In addition, a post weld heat treatment method and repair weld method which could improve integrity of the welded joint of P91 steel were investigated.

광 전달 쐐기를 이용한 중합 방법이 레진 충전물의 미세 누출과 미세 경도에 미치는 효과 (THE INFLUENCE OF CURING WITH LIGHT TRANSMITTING WEDGE INSERTED INTO THE COMPOSITE RESIN BODY ON THE DEGREE OF MICROLEAKAGE AND MICROHARDNESS)

  • The purpose of this study was to test the hypothesis that the more thorough cure of lightactivated composite resin could be obtained if a new way of delivering the light source deep through the resin body is developed when compared to the conventional bulk cure or incremental cure. Using cylindrical resin blocks and natural teeth, various curing conditions were tested for their effects on the degree of microleakage and microhardness. Data were analyzed statistically using One-Way ANOVA and Scheff's Multiple Range test. The results of the present study were as follows: 1. The increment of reduction in microhardness with depth was shown to be higher in group I and II than in group II and V. 2. At the level of lower inner surface and the bottom surface, significantly lower microhardness values were obtained in group I and II when compared to group III, IV and V. Group IV showed the lowest value among three groups(III, IV, V) representing the only statistically significant difference from group V(p<05). 3. Differences between groups in the degree of microleakage detected were shown to be statistically significant (p<05) with the exception of group IV and V at both occlusal and gingival margins. 4. Based upon the above-mentioned results, commonly cited recommendation of incremental curing is strongly supported and the light-transmitting wedge insertion method can be considered as a effective and feasible clinical procedure for the better curing of composite resin. However, more studies under a variety of conditions should be completed before this method is applied actually to the clinical setting.

Rietveld법에 의한 $Nd_3Ba_5Co_4O_{15}$상의 정밀화 (Structure Refinement of $Nd_3Ba_5Co_4O_{15}$ Phase by Rietveld Method)

  • Nd2O3, BaCo3, Co3O4를 출발물질로 하여 새로운 Nd3Ba5Co4O15상을 고상반응법에 의하여 합성하였다. 반응 온도는 1200℃ 로 하였고 열처리 중 반복적인 혼합을 실시하였다. Nd3Ba5Co4O15상의 결정구조는 분말 X-선회절법에 의한 데이터를 Rietveld법을 이용하여 정밀화하였다. 출발 모델은 Nd3.43Ba4.42Co2.23Al1.77O15의 결정구조를 따랐고 공간군 P63mc(186), 격자상수 a=11.636(5) Å, c=6.846(3) Å. 최종 신뢰도 R값은 Rwp=0.097, Rp=0.068로 나타났다. 이 상의 결정구조는 CoVICoIV3O15 클러스터로 구성되어 있고 이 클러스터는 CoVI 육면체가 3개의 CoIV 사면체와 corner sharing하는 형태를 보인다.

Performance evaluation study of a commercially available smart patient-controlled analgesia pump with the microbalance method and an infusion analyzer

  • Background: Patient-controlled analgesia (PCA) has been widely used as an effective medical treatment for pain and for postoperative analgesia. However, improper dose errors in intravenous (IV) administration of narcotic analgesics from a PCA infusion pump can cause patient harm. Furthermore, opioid overdose is considered one of the highest risk factors for patients receiving pain medications. Therefore, accurate delivery of opioid analgesics is a critical function of PCA infusion pumps. Methods: We designed a microbalance method that consisted of a closed acrylic chamber containing a layer and an oil layer with an electronic balance. A commercially available infusion analyzer (IDA-5, Fluke Co., Everett, WA, USA) was used to measure the accuracy of the infusion flow rate from a commercially available smart PCA infusion pump (PS-1000, UNIMEDICS, Co., Ltd., Seoul, Korea) and compared with the results of the microbalance method. We evaluated the uncertainty of the flow rate measurement using the ISO guide (GUM:1995 part3). The battery life, delay time of the occlusion alarm, and bolus function of the PCA pump were also tested. Results: The microbalance method was good in the short-term 2 h measurement, and IDA-5 was good in the long-term 24 h measurement. The two measurement systems can complement each other in the case of the measurement time. Regarding battery performance, PS-1000 lasted approximately 5 days in a 1 ml/hr flow rate condition without recharging the battery. The occlusion pressure alarm delays of PS-1000 satisfied the conventional alarm threshold of occlusion pressure (300-800 mmHg). Average accuracy bolus volume was measured as 63%, 95%, and 98.5% with 0.1 ml, 1 ml, and 2 ml bolus volume presets, respectively. A 1 ml/hr flow rate measurement was evaluated as 2.08% of expanded uncertainty, with a 95% confidence level. Conclusion: PS-1000 showed a flow accuracy to be within the infusion pump standard, which is ± 5% of flow accuracy. Occlusion alarm of PS-1000 was quickly transmitted, resulting in better safety for patients receiving IV infusion of opioids. PS-1000 is sufficient for a portable smart PCA infusion pump.

  • Purpose : This study was performed for find out temporal spatial parameter of the gait according to age. Method : Four groups of healthy people were allocated randomly in this study : group I(little child, 15), group II(child, 18) and group III(young people, 17), group(elder people, 16). This study was performed from 01 December to 31 December in 2007. Results : The results were as follows : 1. The swing phase was the longest group II, group IV was the shortest. Each groups, there was significant difference in swing phase(p<.05). 2. The stance phase was the longest group IV, group II was the shortest. Each groups, there was significant difference in stance phase(p<.05). 3. The single support time was the longest group II, group IV was the shortest. Each groups, there was significant difference in single support time(p<.05). 4. The double support time was the longest group IV, group II was the shortest. Each groups, there was significant difference in double support time(p<.05). 5. The gait velocity was the fastest group II, group I was the slowest. Each groups, there was significant difference in gait velocity(p<.05). 6. The toe in/out was very increased group IV, group I was very decreased. Each groups, there was significant difference in toe in/out(p<.05). 7. The cadence was the highest group I, group IV was the lowest. Each groups, there was significant difference in cadence(p<.05). 8. The step length was the longest group III, group I was the shortest. Each groups, there was significant difference in step length(p<.05). 9. The step length asymmetry ratio was the highest group IV, group III was the lowest. Each groups, there was no significant difference in step length asymmetry ratio(p>.05). 10. The single support time asymmetry ratio was the highest group I, group IV was the lowest. Each groups, there was no significant difference in single support time asymmetry ratio (p>.05). 11. The FAP was the highest group III, group I was the lowest. Each groups, there was significant difference in FAP(p<.05).

자궁적출술 환자의 진통제 투여시작 시기에 따른 수술 후 통증호소 비교(IV-Patient Controlled Analgesia를 이용하여) (Comparative Study of Postoperative Analgesic Effect of IV-PCA According to Timing of Infusion in Patients with Total Abdominal Hysterectomy)

  • Purpose: This study was designed to verify preemptive effects of intravenous patient-controlled analgesia (IV-PCA) infusion on postoperative pain in women having a total abdominal hysterectomy. Method: The research design was a nonequivalent control group post test only design. The participants in this study were 50 women who were scheduled for a total abdominal hysterectomy at a University affiliated Hospital in Suwon, Korea. The subjects were divided into two groups. For the experimental group, IV-PCA infusion was started before the skin incision and for the control group. IV-PCA infusion was started after the skin was closed. Each group was evaluated in terms of pain score by the visual analogue scale (VAS) and the number of times they pushed the button for IV-PCA at postoperative hours 1, 2, 3, 6, 12, and 24. The data were collected from July 1 to December 10, 2001. Collected data was analyzed by SPSS/PC + program. Result: 1. There was no difference between the two groups, over six points for the number of times the control button for IV-PCA was used. Group differences and interaction effect were not significant. 2. There was no significant difference in pain scores between the two groups, over seven time points. A significant interaction effect was observed between groups and measurement Points in time. 3. There was a significant difference in the requirements for additional analgesia between the two groups, 32% of the control group received additional analgesia. Conclusion: Preemptive analgesics administration may have a better effect in relieving postoperative pain than the usual analgesic treatment which is started after surgery.
