The Effect of the Timing and Dose of Human Chorionic Gonadotropin on Oocyte Recovery, in Vitro Fertilization, and Preimplantation Development in Superovulation of Mouse (생쥐에서 과배란 유도시 인간융묘 성선자극 홀몬 투여 방법이 체외수정 및 배자의 체외성장에 미치는 영향에 관한 연구)

  • This study was carried out to investigate the effect of the timing and dose of human chorionic gonadotropin(hCG) on oocyte recovery, in vitro fertilization, and preimplantation development in superovulation of mouse. F1 hybrid($C57BL{\times}CBA$) mice were obtained and superovulation was induced in female mice by sequential intraperitoneal injection of PMSG and hCG. In the first series of experiments, mice received 5 IU of PMSG given intraperitoneally, and 48 hours later were injected 1 IU, 5 IU, or 10 IU of hCG respectively. In the second series of experiments, mice received 5 IU of PMSG given intraperitoneally and were injected 5IU of hCG 36, 48, or 60 hours later respectively. 1. When the mice received 5 IU of PMSG given intraperitoneally and 48 hours later were injected 1 ItT, 5 IU, or 10 IU of hCG respectively, there were no differences in the total number of the oocytes obtained from the three experimental groups. When the cultures were examined 48 hrs after the termination of insemination the proportion of unfragmented oocytes which had developed over two-cell stage was observer to be lowest in 10 IU hCG group. When the cultrues were examined 120 hour after termination of insemination the proportion of embryos which had developed to the blastocyst stage was observed to be significantly higher in 10IU hCG group than 5IU hCG group(p<0.05), but there was no difference between 10 IU hCG group and 1IU hCG group. 2. When the mice received 5 IU of PMSG and were injected 5 IU of hCG 36, 48, or 60 hours later respectively, there were no differences in the total number of oocytes obtained from the three experimental groups. When cultures were examined 48 hour after the termination of insemination the proportion of unfragmented oocytes which had developed over two-cell stage was observed to be significantly lower in 36 hour interval group than 48 hour interval and 60 hour interval group(p<0.05). When the cultures were examined 120 hour after termination of insemination the proportion of embryos which had developed to the blastocyst stage was found to be higher in 60 hour interval group than 36 interval or 48 hour interval group (P<0.05), and the proportion of hatching blastocyst was found to be higher in 60 hour interval group as well. In this study, it was concluded that the administration of adequate dose of hCG, and long (60 hour) PMSG-hCG interval were necessary in superovulation of mice($C57BL{\times}CBA$) in order to get a large number of oocytes which had an early oocytes which had an early embryonic developmental capability when fertilized in vitro, and especially it had better have been avoided to administer a large dose of hCG.

Vitamin A Concentration of Serum from Infertile Dairy Cows (불임우(난소기능감퇴유우) 혈청의 Vitamin A 함량)

  • The experiment was carried out to determine the concentration of vitamin A by Carr-Price reaction ot fifty six sera obtained from normal cows and fourty four sera from the cows with ovarian hypofunction. The results obtained in this work were summerized as follows: 1. The mean values of vitamin A concentration of sera obtained from the cows with ovarian hypofunction were 88 IU/100 ml in summer (grazing), and in winter 79 IU/100 ml (hay) and 173 IU/100 ml (trench silage). The mean values of vitamin A concentration of sera from normal cows were 212 IU/100 ml in summer (grazing), and in winter 113IU/100 ml (hay) and 338IU/100 ml (trench silage). The differences of the concentrations of vitamin A among three groups were statistically highly significant (p<0.01). 2. The differences of vitamin A concentrations between the hay group and the trench silage group in normal cows and in cows with ovarian hypofunction were statistically highly significant (p<0.01). 3. The concentrations of vitamin A in summer group (grazing) were higher than those of the hay group in winter in normal cows and in cows with ovarian hypofunction. 4. The concentrations of vitamin A in the trench silage group in winter were higher than those of summer group (grazing) in normal cows and in cows with ovarian hypofunction.

  • Sera obtained from 20 milk cows and 20 Korean cattle in group feeding and from 20 Korean cattle from different farmhouses were analyzed for the determination of carotenoid and vitamin A by Carr-Price reaction. The result obtained were as follows: 1. Carotenoid contents of sera from milk cows and Korean cattle in group feeding, and from Korean cattle from different farmhouses were in winter $394.7{\mu}g/100ml$, $157.8{\mu}g/100ml$ and $187.9{\mu}g/100ml$, respectively, and were in summer $735.3{\mu}g/100ml$, $728.2{\mu}g/100ml$ and $760.3{\mu}g/100ml$, respectively. 2. Vitamin A contents of sera from milk cows and Korean cattle in group feeding and from Korean cattle from different farmhouses were in winter 101.2 IU/100 ml, 88.9 IU/100 ml and 48.4 IU/100 ml, respectively, and were in summer 212.0 IU/100 ml, 208.7 IU/100 ml and 208.3 IU/100 ml respectively.

Induction of superovulation in mature mice and rats using serum of spayed female dogs

  • The following experiments were designed to examine the effect of serum of spayed dogs on superovulation response in mice and rats. In Experiment 1, female mice at diestrus (n=30) were divided into three equal groups and superovulated with either administration of 5 IU pregnant mare serum gonadotropin (PMSG) or recombinant follicle stimulating hormone (rFSH) (reducing dose from 2.5 to 0.5 IU) and 5 IU human chorionic gonadotropin (hCG) administered 48h later. Serum of spayed dogs was administered intraperitoneally at a reduced dose from 0.1 to 0.025 mL in a 48 h period. In Experiment 2, female rats (n=30) at diestrus stage were divided into three equal groups. Superovulation was induced using either 30 IU PMSG, or a dose reduced from 5 to 1 IU rFSH and 25 IU hCG administered 48h later. Serum of spayed dogs was administered in a reduced dose from 0.6 to 0.1 mL in a 48 hour period. Female mice and rats were mated 24 h following hCG administration. On day 14 after mating, animals were euthanized and ovarian sections were fixed for histopathological evaluation and corpus luteum (CL) counting. No significant difference observed in mean (${\pm}SEM$) number of CLs between the PMSG group and the mice that received serum of spayed dog ($10.4{\pm}1.3$ vs $9.2{\pm}1.0$). Mean (${\pm}SEM$) number of CLs tended to be lower in rats that received serum of spayed dog than those of rats which received either PMSG or rFSH ($15.1{\pm}1.9$ vs $23.6{\pm}3.1$ and $23.1{\pm}2.9$, P=0.06, respectively). In conclusion, serum of spayed dogs is able to induce a superovulatory response in mice and rats.

Analysis of HBeAg and HBV DNA Detection in Hepatitis B Patients Treated with Antiviral Therapy (항 바이러스 치료중인 B형 간염환자에서 HBeAg 및 HBV DNA 검출에 관한 분석)

  • Purpose Hepatitis B virus (hepatitis B virus, HBV) infection is a worldwide major public health problem and it is known as a major cause of chronic hepatitis, liver cirrhosis and liver cancer. And serologic tests of hepatitis B virus is essential for diagnosing and treating these diseases. In addition, with the development of molecular diagnostics, the detection of HBV DNA in serum diagnoses HBV infection and is recognized as an important indicator for the antiviral agent treatment response assessment. We performed HBeAg assay using Immunoradiometric assay (IRMA) and Chemiluminescent Microparticle Immunoassay (CMIA) in hepatitis B patients treated with antiviral agents. The detection rate of HBV DNA in serum was measured and compared by RT-PCR (Real Time - Polymerase Chain Reaction) method Materials and Methods HBeAg serum examination and HBV DNA quantification test were conducted on 270 hepatitis B patients undergoing anti-virus treatment after diagnosis of hepatitis B virus infection. Two serologic tests (IRMA, CMIA) with different detection principles were applied for the HBeAg serum test. Serum HBV DNA was quantitatively measured by real-time polymerase chain reaction (RT-PCR) using the Abbott m2000 System. Results The detection rate of HBeAg was 24.1% (65/270) for IRMA and 82.2% (222/270) for CMIA. Detection rate of serum HBV DNA by real-time RT-PCR is 29.3% (79/270). The measured amount of serum HBV DNA concentration is $4.8{\times}10^7{\pm}1.9{\times}10^8IU/mL$($mean{\pm}SD$). The minimum value is 16IU/mL, the maximum value is $1.0{\times}10^9IU/mL$, and the reference value for quantitative detection limit is 15IU/mL. The detection rates and concentrations of HBV DNA by group according to the results of HBeAg serological (IRMA, CMIA)tests were as follows. 1) Group I (IRMA negative, CMIA positive, N = 169), HBV DNA detection rate of 17.7% (30/169), $6.8{\times}10^5{\pm}1.9{\times}10^6IU/mL$ 2) Group II (IRMA positive, CMIA positive, N = 53), HBV DNA detection rate 62.3% (33/53), $1.1{\times}10^8{\pm}2.8{\times}10^8IU/mL$ 3) Group III (IRMA negative, CMIA negative, N = 36), HBV DNA detection rate 36.1% (13/36), $3.0{\times}10^5{\pm}1.1{\times}10^6IU/mL$ 4) Group IV(IRMA positive, CMIA negative, N = 12), HBV DNA detection rate 25% (3/12), $1.3{\times}10^3{\pm}1.1{\times}10^3IU/mL$ Conclusion HBeAg detection rate according to the serological test showed a large difference. This difference is considered for a number of reasons such as characteristics of the Ab used for assay kit and epitope, HBV of genotype. Detection rate and the concentration of the group-specific HBV DNA classified serologic results confirmed the high detection rate and the concentration in Group II (IRMA-positive, CMIA positive, N = 53).

Comparison of serum isoenzyme levels of CPK and LDH in patients ungergoing thoracic operations (흉부수술 환자에서의 CPK 와 LDH I soenzyme 의 변화)

  • A analysis of CPK & LDH Isoenzyme was done on the consecutive patients undergoing thoracic operations from July 1982 to October 1982 in the Department of Thoracic and Cardiovascular Surgery, Capital Armed Forces General Hospital. Eighteen patients were analysed by three groups, such as open heart surgery [group A], major thoracic operation [group B] , minor thoracic operation group [group C]. In all patients serial determination of total level and Isoenzyme of CPK, LDH wad done on preoperative operative and up to 8th post-operative day, The results obtained are as follows. 1. The average value of serum CPK before the operation was 61 IU/L. The value of serum CPK was increased following the operation mainly MM portion and reached to the maximal level of 536107 IU/L in A group 1200191 IU/L in B group, 306150 IU/L in C group on the first postoperative day. The enzyme activity was gradually decreased thereafter and returned to the normal range on the 3rd or 4th day after the operation. 2. The average value of serum LDH before the operation was found to be 83 IU/L. The value was increased during the operation and reached to the maximal level of 481108 IU/L in group A, 14827 I U/L in group B, 10035 IU/L in group C on the second day after the operation. The enzyme activity was gradually decreased thereafter and returned to the normal range on the seventh day after the operation. The enzyme activity was dependent to the duration of operation, severity of muscle damage, type of thoracotomy, effect of extracorporeal circulation, state of disease.

Studies on Embryo Transfer in Rabbit I. Ovarian response to PMSG and hCG administration (가토의 수정란이식에 관한 연구 I. PMSG와 HCG투여에 따른 난소반응)

  • As a preliminary experiment to establish the process of embryo transfer in rabbit, present sutdies were carried out with 75 mature Japanese of ovary to pregnant mare's serum gonadotropin(PMSG) and human chorionic gonadotropin(hCG) and collection rate of embryos at various times after hCG injection. Female rabbits were superovulated using 50∼100IU hCG or 75∼100IU PMSG and 50∼751IU hCG injected 83hrs apart. The results obtained were as follows: 1. The average number of growth follicles obtained from all of rabbits treated with hCG or PMSG-hCG was 28.1. PMSG-hCG treatment group (30.9) was clearly increased more than hCG treatment group (16.7). 2. In ovulation score, PMSG-hCG treatment group (21.0) was increased more than hCG treatment group (7.9), showing the same trends in the growth of follicles. 3. The ovulation rate per follicles developed was higher in the rabbits treated with 100 IU PMSG and 75 IU hCG (18.9%) than that from the other groups. 4. The oviduct score (72.9%) was inclined to higher than that from uteri (57.1%) in score of embryo collection.

Study on the Freezing of Bovine Embryos I. Ovarian response to the administration of gonadotrophins (우 수정란의 동결보존에 관한 연구 I. 성선자극홀몬과 $PGF_{2\alpha}$의 투여에 따른 난소반응)

  • This study was carried out with 46 cows to investigate the ovarian response to the administration of PMSG, PGF2$\alpha$ and hCG, and the developmental stage of embryos recovered. Superovulation was induced by the injection of 2,000IU to 3,000IU PMSG on the days of 7-13 of the estrus cycle followed 48 hours later by the injection of 22.5mg PGF2$\alpha$. Of 29 cows treated with 3,000IU PMSG and 22.5mg PGF2$\alpha$ 18 cows were given 2,000IU hCG at the onset and 7 after artificial insemination. The results obtained were summarized as follows; 1. The number of developed follicles per cow after an injection of 2,000, 2,500, 3,000IU of PMSG and 3,000 PMSG-2,000IU hCG in combination with 22.5mg PGF2$\alpha$ were 12.6, 19.6, 21.5 and 29.3, respectively. This result indicated that the no. of developed follicles per cow was increased according to the increase of PMSG dosage and the combination with hCG injection. 2. The highest number of ovulation was 17.1 in cows treated with PMSG-hCG and the number of matured corpus luteum was increase as the dosage of PMSG was higher. 3. Ovulation rate from cows treated with 2,500IU PMSG was 71.0% and this reulst was higher than the average of ovulation rate (59.3%). 4. Average recovery rate was 36.8%(232/631), and the number of ova per cow was 5.0. 5. Of 232 recovered embryos, the number of morulae and blastocysts were 76 (32.8%) and 83 (35.8%), respectively. 6. 28.4% of total recovered embryos was abonormal morphologically.

Effectof of Hormonal Treatment of Induced Maturation and Ovulation in the Spotted Halibut, Verapser variegatus (호르몬 처리에 의한 범가자미, Verasper variegatus 의 성숙과 배란유도)

  • Hormone induction in maturation and ovulation was carried out with spotted halibut (Verasper variegatus) which normally does not spawn spontaneously in captivity. Prior to spawning females were treated with human chorionic gonadotropin(HCG 265-678 IU/kg BM) 17 ${\alpha}$-hydroxy 20-${\beta}$-dihydroprogesterone (17 ${\alpha}$ 20${\beta}$ OHP 0.5-1.0${\mu}$g/kg BW) and luteinizing hormone-releasing hormone analogue pellet (LHRHa pellet 63-81 ${\mu}$g/kg BW) In all the trials HCG injection succeeded in inducing ovulation at 265-300 IU/kg BW. HCG at 620 or 678 IU/kg BW was ineffective in inducing a good rate of ovulation. Final maturation and ovulation were not stimulated by treatment of 17 ${\alpha}$ 20${\beta}$OHP and LHRHa pellet. These results suggest that HCG injection (around 300 IU/kg BW) is more effective compared with LHRHa pellet implantation.

Enzymatic Characteristics of a Highly Thermostable β-(1-4)-Glucanase from Fervidobacterium islandicum AW-1 (KCTC 4680)

  • A highly thermostable ${\beta}-(1-4)-glucanase$ (NA23_08975) gene (fig) from Fervidobacterium islandicum AW-1, a native-feather degrading thermophilic eubacterium, was cloned and expressed in Escherichia coli. The recombinant FiG (rFiG) protein showed strong activity toward ${\beta}-{\small{D}}-glucan$ from barley (367.0 IU/mg), galactomannan (174.0 IU/mg), and 4-nitrophenyl-cellobioside (66.1 IU/mg), but relatively weak activity was observed with hydroxyethyl cellulose (5.3 IU/mg), carboxymethyl cellulose (2.4 IU/mg), and xylan from oat spelt (1.4 IU/mg). rFiG exhibited optimal activity at $90^{\circ}C$ and pH 5.0. In addition, this enzyme was extremely thermostable, showing a half-life of 113 h at $85^{\circ}C$. These results indicate that rFiG could be used for hydrolysis of cellulosic and hemicellulosic biomass substrates for biofuel production.
