A Comparison of the Ability of Fungal Internal Transcribed Spacers and D1/D2 Domain Regions to Accurately Identify Candida glabrata Clinical Isolates Using Sequence Analysis

  • Candida glabrata is the second most prevalent causative agent for candidiasis following C. albicans. The opportunistic yeast, C. glabrata, is able to cause the critical bloodstream infections in hospitalized patients. Conventional identification methods for yeasts are often time consuming and labor intensive. Therefore, recent studies on sequence-based identification have been conducted. Recently, sequencing the D1/D2 domain of the large subunit ribosomal RNA gene and the internal transcribed spacers (ITS) 1 and ITS2 regions of the ribosomal DNA has proven useful for DNA-based identification of most species of fungi. In the present study, therefore, fungal ITS and D1/D2 domain regions were targeted and analyzed by DNA sequencing for the accurate identification of C. glabrata clinical isolates. A total of 102 C. glabrata clinical isolates from various clinical samples including bloodstream, catheterized urine, bile and other body fluids were used in the study. The results of the DNA sequence analysis showed that the mean standard deviation of species identity percent score between ITS and D1/D2 domain regions was $97.8%{\pm}2.9$ and $99.7%{\pm}0.46$, respectively. These results revealed that the D1/D2 domain region might be a better target for identifying C. glabrata clinical isolates based on DNA sequences than the ITS1 and ITS2 regions. However, in order to evaluate the usefulness of D1/D2 domain region for species identification of all Candida species, other Candida species such as C. albicans, C. tropicalis, C. dubliniensis, and C. krusei should be verified in further studies additionally.

Variation of nuclear ribosomal ITS sequences of Polygonum section Persicaria (Polygonaceae) in Korea (한국산 여뀌속 Persicaria절(마디풀과)의 핵 리보오솜 ITS 염기서열 변이)

  • We examined nrDNA ITS sequences from 16 taxa of Polygonum sect. Persicaria(Polygonaceae) in Korea to infer relationships among the taxa within the section. A neighbor-joining tree obtained from the analysis of the ITS sequences suggest that the ITS region was useful inferring the phylogenetic relationships among the taxa. The neighbor-joining tree indicates that P.amphibium is clearly separated from the other Korean taxa. The tree also reveals the presence of five major groups in the Korean taxa of the section; 1) P. lapathifolium var. lapathifolium, 2) P. persicaria and P. viscoferum, 3) P. orientale and P. viscosum, 4) P. japonicum and 5) a group including the remaining taxa. these relationships depicted on the ITS tree are largely congruent with those inferred from morphological and anatomical characters.

The Role of Substituents of ar-Turmerone for its Anticancer Activity

  • For the evaluation of the role of substituents of ar-turmerone for its anticancer activity, ar-turmerone (1a) and its analogs like 2-methyl-6-(4'-methyphenyl)-2-octen-4-one (1b), 2-methyl-6-phenyl-2-hepten-4-one (1c), 2-methyl-6-phenyl-2-octen-4-one (1d) and 2 methyl-6-(trans-4'-methylcyclohexyl)-2-hepten-4-one (1e) were preparedd and their cytotoxic activities against $L_{1210}$ cell were determined. Omission of methyl group at para-position dose not variate the cytotoxicity of ar-turmerone. Elongation of alkyl group at 6-position decreases $ED_{50}$ value. Saturation of aromatic ring of ar-turmerone markedly decreases the cytotoxicity. Therefore the smaller size of alkyl group at 6-position and aromatic ring of ar-turmerone should be essential for exhibiting its anticancer activity.

Determination of false positives in PCR diagnostics based on the internal transcribed spacer (ITS) of Gyrodactylus salaris using RFLP (RFLP를 이용한 Gyrodactylus salaris의 internal transcribed spacer(ITS) PCR 위양성 판별)

  • The World Organization for Animal Health (WOAH) recommends two protocols (ITS and COI) for conventional PCR of G. salaris diagnosis. However, ITS PCR protocol may yield false-positive results, leading to unnecessary countermeasures. It's difficult to distinguish between G. salaris and false-positive by similar amplicon size of PCR, since the amplicon size of ITS PCR in G. salaris and false-positive was 1,300 and 1,187 bp, respectively. The nucleotide sequences of ITS false-positive in rainbow trout is 99.7% identical to previously reported host genome sequences of rainbow trout (Oncorhynchus mykiss) and 95.3 to 89.1% identical to those of other salmonid fish species. To reduce false-positive PCR band, PCR was performed by the different annealing temperature, but PCR bands were still detected. In RFLP analysis by HaeIII, the PCR product of G. salaris was digested into four bands of 512, 399, 234 and 154 bp, while the false-positive was digested into seven bands of 297, 263, 242, 144, 93, 80 and 68 bp. In the RFLP patterns digested by HindIII, G. salaris showed two bands of 659 and 640 bp, while false-positive had one fragment of 1,187 bp without any digestion. Therefore, the RFLP method of ITS PCR with HaeIII and HindIII can be used for differentiation between G. salaris and false-positive. These results might provide important information on the improvement of PCR diagnostic method of G. salaris.

  • Over its 30-year lifetime, UKIRT has been the world's leading telescope devoted exclusively to observations in the infrared. Its outstanding success over this period is attributable to a number of factors, foremost among which are the size (3.6m) and high quality of the mirror, the excellent site at the summit of Mauna Kea, and the deployment of unique, world-beating instrumentation. In recent years, as the UK has re-directed its investment in astronomical facilities towards ESO, UKIRT has focussed on wide-field imaging in the near infrared, and we are currently conducting a definitive programme known as the UKIRT Infrared Deep Sky Survey (UKIDSS). In this presentation I will describe UKIRT's current capabilities, its evolving operation, opportunities for Korean participation, and possibilities for its future development.

New Variety of Cordyceps gunnii (Berk.) Berk. and Its Paecilomyces Anamorph (Cordycepes gunnii의 신변종과 이 균의 무성세대 Paceliomyces)

  • In the present paper, a new variety of Cordyceps gunnii (Berk.) Berk. is reported. It differs from C. gunnii by its much smaller secondary ascospores, (2.0-) 3.3 $(-4.3){\times}1.0{\sim}1.8\;{\mu}m$, as well as thinner asci and cap of the asci. Its anamorph is also reported as a new variety of Paecilomyces gunnii Liang, the ansmorph of C. gunnii. Its differ from P. gunnii by its conidia, mostly subglobose and much bigger, (2.2-) 4.7 $(-11.0){\times}(1.8-)\;2.6\;(-3.3)\;{\mu}m$. The type specimen and dry type culture are deposited at Research Center for Entomogenous Fungi, Anhui Agriculture University, China.

Receptor for Advanced Glycation Endproducts (RAGE), Its Ligands, and Soluble RAGE: Potential Biomarkers for Diagnosis and Therapeutic Targets for Human Renal Diseases

  • Receptor for advanced glycation endproducts (RAGE) is a multi-ligand receptor that is able to bind several different ligands, including advanced glycation endproducts, high-mobility group protein (B)1 (HMGB1), S-100 calcium-binding protein, amyloid-${\beta}$-protein, Mac-1, and phosphatidylserine. Its interaction is engaged in critical cellular processes, such as inflammation, proliferation, apoptosis, autophagy, and migration, and dysregulation of RAGE and its ligands leads to the development of numerous human diseases. In this review, we summarize the signaling pathways regulated by RAGE and its ligands identified up to date and demonstrate the effects of hyper-activation of RAGE signals on human diseases, focused mainly on renal disorders. Finally, we propose that RAGE and its ligands are the potential targets for the diagnosis, monitoring, and treatment of numerous renal diseases.

Phylogenetic Analysis of Korean Native Aster Plants Based on Internal Transcribed Spacer (ITS) Sequences (ITS 염기서열을 이용한 한국산 참취속 식물의 유연관계분석)

  • This study was carried out to decide ITS (internal transcribed spacer) sequence of some Korean native $Aster$ species and to resolve their relationship among Korean native $Aster$, including $Kalimeris$, $Gymnaster$, $Heteropappus$ genus separated from $Aster$ in a previously study based on the pappus length. We registered 11 ITS sequences of $Aster$ species including $A.$ $glehni$ to GenBank and those sequences were used for the cluster analysis with $Kalimeris$ species. The size of ITS1 was varied from 248 to 256 bp, while ITS2 was varied from 220 to 222 bp. The G + C content of the ITS region ranged from 49.4 to 53.5%. Pairwise comparison results showed that the substitution rate of ITS1 and ITS2 region was 9% and 10%, respectively. $Kalimeris$ sensu strict substitution rate was lower than that of $Aster$ sensu strict species. The strict consensus parsimonious cluster analysis showed $A.$ $tripolium$ is the first branching from the clade and the next is $A.$ $scaber$. The $Kalimeris$ species except for the $A.$ $hispidus$ were grouped into the same clade with high bootstrap value (91%) within $Aster$. $Gymnaster$ and $Heteropappus$ that has been classified by morphological characters were also grouped into broad sense $Aster$ clade. These results implied these three genera could be merged together into $Aster$ based on the ITS sequences.

GIS Interoperability Issues for ITS Services : Map Datum and Location Referencing

  • Two GIS and/or OGIS issues for ITS interoperability have been proposed and reviewed with some implications to Korean setting. They are location referencing and ITS map datum. The former must support ITS communication and data sharing. Therefore, an introduction of location referencing and other related issues have been addressed along with the Oak Ridge National Lab.'s (ORNL) location referencing scheme. The latter, proposed by ORNL, is a planned network of anchor points across the nation, that could potentially serve as a positional reference for ITS application (Gottsegen, 1997). It is composed of a set of nodes and links in a standard non-plannar network at a coarse scale for the entire nation for referencing purposes. To provide case of real time traffic information and to guarantee the seamless interoperability, we do need to develop the core ITS map datum as a national infrastructure, and the location referencing scheme should also be either developed or borrowed and localized to meet the domestic needs. Some institutional issues are also addressed along with the future research agenda.

Synthesis and Antitumor Activity of Phthalimide-Based Polymers Containing Camptothecin

  • The objective of this study was to develop a polymeric drug delivery system for camptothecin (CPT), capable of improving its therapeutic index and reducing its side effects. A monomeric conjugate, 3,6-endo-methylene-1,2,3,6-tetrahydrophthalimidoethanoylcamptothecin in (ETECPT) between CPT and 3,6-endo-methylene-1,2,3,6-tetrahydrophthalimidoethanoic acid was synthesized. Its homo-and copolymer with acrylic acid (AA) were prepared by photopolymerization using 2,2-dimethoxy-2-phenylacetophenone (DMP) as a photoinitiator. The monomer and its polymers were characterized by IR, $^1$H- and $^{13}$ C-NMR spectra. The ETECPT content in poly(ETECPT-co-AA) obtained by elemental analysis was 82 wt%. The number-average molecular weights of the polymers determined by gel permeation chromatography were as follows: M$_{n}$ = 11,400 for poly(ETECPT), M$_{n}$ = 17,900 for poly(ETECPT-co-AA). The $IC_{50}$/ values of ETECPT and its polymers against cancer cells were much larger than that of CPT. Our results from the in vivo antitumor activity indicated that all polymers show high antitumor activity than CPT at a dose of 100 mg/kg./kg.
