Development of Bacillus thuringensis var. kurstaki NRD 6-Stu 1 by Site-Directed Mutagenesis (위치지정 변이기법을 이용한 Bacillus thuringiensis var. kurstaki NRD 6-Stu 1의 육성)

  • A cloned delta-endotoxin gene from Bacillus thuringiensis var. kurstaki NRD 6 was mutated at N-terminal toxic portion of the protoxin by site-directed mutagenesis. A Stu 1 restriction site was created in protoxin region of the mutant B.t.kurstaki NRD 6-Stu 1 which changed A to C in 177 region of the protoxin(silent mutation). Antigenic property of crystalline inclusion (delta-endotoxin) of the mutant was no difference to intact strain by the western blot analysis and its toxidity threshold value was also showed 0.015~0.030ng against Choristoneura femiferana-1 insect, similar with 0.01~0.024ng of parent strain, but stronger than that of B.t.kurstaki NRD 5 (500~1000ng) and B.t.kurstaki NRD 4(none toxic).

  • Lowland and upland rice were grown concurrently under the both lowland and upland conditions. Yield and its components such as one liter weight, 1, 000 grain weight, number of grain per panicle and number of panicle per hill were compared between varieties and between cultural conditions. Variations of rice quality were also observed about amylose content, alkali digestion value and protein content between varieties and cultural conditions. Most of lowland varieties showed high growing vigour and yielding ability in lowland culture, and they also showed high variations between the lowland and upland cultures compared to upland variety. Upland varieties were revealed higher adaptability in productivity to the lowland condition than upland condition.

  • Phylogenetic relationship and DNA polymorphism among local populations of the Asparagus cochinchinensis have been investigated based on nuclear ribosomal DNA ITS sequences and RAPD analysis in Korea. In result, two genetically distinct groups of local populations except Geoje were recognized by the phylogenetic tree both in rDNA-ITS and RAPD. One was called 'western coast group' that includes the Buan 1, 2 and Taean and the other was 'southern coast group' that includes Haenam, Yeosu and Namhae. Thus, the geographical relationship of Asparagus cochinchinensis was two well-typified clades. These results suggest that the geographical genetic variation of Asparagus cochinchinensis is closely connected with the slow and long period of propagation via the coast in Korean Peninsula.

Genetic variation and differentiation among populations of Euphorbia ebracteolata (Euphorbiaceae) in Korea (한국산 붉은대극(대극과) 집단의 유전적 변이와 분화)

  • Using starch gel electrophoresis, ten isozyme loci were examined in nine populations of Euphorbia ebracteolata in Korea. Populations of E. ebracteolata tend to have higher within-population levels of genetic variation (A = 2.2, P = 61.1, He = 0.165) along with low levels of genetic differentiation ($F_{ST}$ = 0.075) compared to previously reported Euphorbia species. The relatively high genetic variation in E. ebracteolata may be due to its long-lived regional distribution and to the outcrossed nature of the species. Low levels of genetic differentiation and the absence of unique alleles in E. ebracteolata populations suggest that Korean populations diverged recently. This study does not support the recognition of var. coreana and for. magna as separate taxa, and suggests that they should be combined with E. ebracteolata.

  • Genetic association study has been progressed in medicine along with advance in genetic technology. It focused on the individual differences in genotype due to errors occurring during DNA duplication, which can cause vulnerability to specific diseases. Polymorphism defines the varieties in phenotype due to those genetic variations. Polymorphism due to change in one DNA base sequence is called as a Single Nucleotide Polymorphism. In the near future, the evaluation of relative risk to specific disease according to SNP will be essential part of fundamental of medicine for the diagnosis, treatment and prevention. Dental caries and periodontal diseases has been first subject to genetic association study in dentistry and broaden out to other areas like bone formation and resorption. This article presents the current state of genetic association study and its application to dentistry.

A Simple Stereo Matching Algorithm using PBIL and its Alternative (PBIL을 이용한 소형 스테레오 정합 및 대안 알고리즘)

  • A simple stereo matching algorithm using population-based incremental learning(PBIL) is proposed in this paper to decrease the general problem of genetic algorithms, such as memory consumption and inefficiency of search. PBIL is a variation of genetic algorithms using stochastic search and competitive teaming based on a probability vector. The structure of PBIL is simpler than that of other genetic algorithm families, such as serial and parallel ones, due to the use of a probability vector. The PBIL strategy is simplified and adapted for stereo matching circumstances. Thus, gene pool, chromosome crossover, and gene mutation we removed, while the evolution rule, that fitter chromosomes should have higher survival probabilities, is preserved. As a result, memory space is decreased, matching rules are simplified and computation cost is reduced. In addition, a scheme controlling the distance of neighbors for disparity smoothness is inserted to obtain a wide-area consistency of disparities, like a result of coarse-to-fine matchers. Because of this scheme, the proposed algorithm can produce a stable disparity map with a small fixed-size window. Finally, an alterative version of the proposed algorithm without using probability vector is also presented for simpler set-ups.

Quantity and Quality of Seed Protein and Its Regional Variation in Barley (보리 단백질의 특성과 지역간 변이)

  • To breed good quality and high yielding varieties, and to evaluate quantity and quality of seed protein in barley, protein content of 139 cultivars, amino acid (AA) composition of 6 cultivars were analyzed. Total protein contents of cultivars tested showed variation from 12.0 to 15.7% in covered barley, 9.2 to 15.4% in hulless barley and 11.4 to 15.9% in malting barley. Varietal mean protein contents of covered barley were higher than any other barley. Barley grains harvested from northern area showed higher protein contents than those from southern area. Relationship between protein content and heading date showed negative correlation in covered barley and no significantly different in malting and hulless barley. Total amino acid (AA) showed variation from 9.51 to 11.47%, also essential amino acid (EAA) was 2.81 to 3.25%. Comparatively, percent of EAA was higher in hulless barley, but lower in covered barley. First limiting EAA was lysine, and other limiting EAAs were threonine and methionine in 6 barley cultivars tested.

Variation of nuclear ribosomal ITS sequences of Polygonum section Persicaria (Polygonaceae) in Korea (한국산 여뀌속 Persicaria절(마디풀과)의 핵 리보오솜 ITS 염기서열 변이)

  • We examined nrDNA ITS sequences from 16 taxa of Polygonum sect. Persicaria(Polygonaceae) in Korea to infer relationships among the taxa within the section. A neighbor-joining tree obtained from the analysis of the ITS sequences suggest that the ITS region was useful inferring the phylogenetic relationships among the taxa. The neighbor-joining tree indicates that P.amphibium is clearly separated from the other Korean taxa. The tree also reveals the presence of five major groups in the Korean taxa of the section; 1) P. lapathifolium var. lapathifolium, 2) P. persicaria and P. viscoferum, 3) P. orientale and P. viscosum, 4) P. japonicum and 5) a group including the remaining taxa. these relationships depicted on the ITS tree are largely congruent with those inferred from morphological and anatomical characters.

Genetic Relationships of Coprinus spp. on the Basis of Sequences in ITS II Region (먹물버섯류(Coprinus spp.)의 ITS II 영역 염기서열에 의한 유연관계 분석)

  • The internal transcribed spacer II regions (ITS II) of the ribosomal DNA gene repeat from Coprinus spp. were amplified using polymerase chain reaction (PCR) and sequenced. Sequences from 11 species including Coprinus comatus, C. atramentarius, C. micaceus, C. lagopus, C. cinereus, C. rhizophorus, C. flocculosus, C. radians, and C. echinosporus were compared. The spacer regions of them were $253{\sim}275$ nucleotide in length and partially contained 5.8S and 25S. The reciprocal homologies of each ITS II sequence among these strains were in the range of $50.6{\sim}100%$. According to the analysis of ITS II sequences, Coprinus spp. were classified into three clusters. Cluster I consisted of Coprinus lagopus, C. cinereus, C. echinosporus, C. rhizophorus, C. niveus, and C. atramentarius. Cluster II comprised C. micaceus, C. flocculosus, C. radians, and C. disseminatus. On the other hand C. comatus is in Cluster III even though this species is belonging to the section Coprinus in morphological aspect. These results suggest that Coprinus comatus, which was considered as a type species of the genus Coprinus in morphological classification, gives a doubt of monophyletic evolution and is assumed to be paraphyletic or polyphyletic.
