렛 근육세포 L6에서 둥굴레 추출물의 인슐린저항성 개선 (Attenuation of insulin resistance using steamed Polygonatum odoratum var pluriflorum extract in rat skeletal muscle cells L6 myoblast)

  • Objectives : The purpose of this study was to investigate inhibitory effects of steamed Polygonatum odoratum extract (POE) on insulin resistance in rat skeletal muscle cells, L6 cells.Methods : Polygonatum odoratum (P. odoratum) extract was extracted with ethyl acetate. Activity of α-glucosidase in POE was measured for blood glucose regulation. MTT assay was examined for cell toxicity. Western blot analysis for measurement of adiponectine, peroxisome proliferator-activated receptorγ (PPARγ), insulin receptor substrate (IRS), glucose transporter 4 (Glut-4) and phosphorylation of serine/threonine-specific protein kinase (Akt) expressions were performed. Akt signaling pathway were analyzed with LY294002, which is a specific PI3K/Akt inhibitor.Results : The results revealed that POE inhibited α-glucosidase activity. Treatment of POE in L6 cells inhibited the differentiation of L6 cells compared to those of vehicl control. Additionally, protein expressions of adiponectine, PPARγ, IRS and Glut-4 were significantly regulated compared to those of vehicle control (p < 0.05), respectively. Futhermore, phosphorylation of Akt was increased in L6 cells treated with POE compared to that of vehicle control (p < 0.05). pAkt expression was significantly accentuated with Akt inhibitor (LY294002).Conclusions : These results suggest that POE may have potential as a natural agent for prevention/improvement of diabetes, especially, regulation of blood glucose. Therefore, further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

이중 센서를 이용한 유도탄 광축 정렬 연구 (A Study on the Optical Axis Alignment of Missile using the Dual-Mode Sensor)

  • In this paper, the optical axis alignment(OAA) of an infrared sensor(IRS) using a microwave sensor(MWS) was presented as a method of an axis alignment to minimize the problems that could be caused by the misalignment of the two sensors in the missile including the dual-mode sensor. The azimuth(AZ) and elevation(EL) angles of the two targets used for each sensor test were calculated by using the transformation equation and the test results of the MWS and IRS, and then the proposed OAA was verified by comparing the angles. Furthermore, the validity of the proposed OAA was demonstrated by confirming the abnormality of the OAA through the test results of the electro optical head(EOH) of the IRS which was equipped with a tilt on the missile fuselage.

A modified index for damage detection of structures using improved reduction system method

  • The modal strain energy method is one of the efficient methods for detecting damage in the structures. Due to existing some limitations in real-world structures, sensors can only be located on a limited number of degrees of freedom (DOFs) of a structure. Therefore, the mode shape values in all DOFs of structures cannot be measured. In this paper, a modified modal strain energy based index (MMSEBI) is introduced to locate damaged elements of structures when a limited number of sensors are used. The proposed MMSEBI is based on the reconstruction of mode shapes using Improved Reduction System (IRS) method. Therefore, in the first step by employing IRS method, mode shapes in slave degrees of freedom are estimated by those of master degrees of freedom. In the second step, the proposed MMSEBI is used to located damage elements. In order to evaluate the efficiency of the proposed method, two numerical examples are considered under different damage patterns considering the measurement noise. Moreover, the universal threshold based on statistical hypothesis testing principles is applied to damage index values. The results show the effectiveness of the proposed MMSEBI for the structural damage localization when comparing with the available damage index named MESBI. The results demonstrate that the presented method can be used as a practical strategy for structural damage identification, especially when a limited number of sensors are installed on the structure. Finally, the combination of MMSEBI and IRS method can provide a reliable tool to identify the location of damage accurately.

Expression Characteristics of Proteins of the Insulin-like Growth Factor Axis in Non-small Cell Lung Cancer Patients with Preexisting Type 2 Diabetes Mellitus

  • Background: Preexisting type 2 diabetes mellitus (T2DM) affects the prognosis and mortality of patients with some cancers. Insulin like growth factor (IGF) and insulin receptor (IR) signaling axes play important roles in both cancer and diabetes development. We aimed to explore the expression characteristics of proteins in IGF/IR axis in non-small cell lung cancer (NSCLC) cases with preexisting T2DM. Methods: Fifty-five NSCLC patients with preexisting T2DM were retrospectively included and matched by 55 NSCLC without diabetes at a 1:1 ratio. The expression of proteins in IGF/IR axis was detected by immunohistochemical staining. Clinicopathological data were collected to analyze their relationship with the protein expression. Results: Both IGF 1 receptor (IGF-1R) and insulin receptor substrate 2 (IRS-2) showed higher expression in the NSCLC with T2DM group, compared with those without T2DM. The high expression of IGF-1R and IRS-2 were found to be negatively associated with lymph node metastases and T staging in the T2DM group, respectively, and IRS-2 expression was also found more in the subgroup whose T2DM duration was more than 4 years. No difference was detected in the expression of IRS-1, IGF-1, IGF-2, IGFBP3, IR and mTOR between groups with or without T2DM. Conclusion: Our study found higher expression of IGF-1R and IRS-2 proteins in NSCLC patients with preexisting T2DM, and that there was an association with early stage NSCLC, which suggested that IGF signaling may play an important early event in development of NSCLC associated with diabetes.

  • 고해상도 위성 IRS-1C의 Panchromatic 영상 자료를 이용하여 한국 근해에서 선박의 수와 위치를 파악할 수 있는 가능성을 살펴보았다. Panchromatic 위성 영상에서 선박과 주변환경에 대한 위치, 크기, 모양, 그림자, 색조, 질감 및 모양의 공간배열, 높이와 깊이, 현황 및 연관성 등의 영상해석 요소를 활용하여 선박 모니터링의 논리적 접근기법을 연구하였다. 선박의 공간적 위치는 GCP가 없는 해상에서 영상의 중심위치 정보로부터 반경 35km내에서 지구좌표점으로 전환하였다. 선박의 크기는 선수에서 선미까지이나 선미에서 생기는 종파(following wave)와 추정 구분하여야 했다. 길이 100m를 기준으로 연근해 어선과 상선으로 구분하였다. 선박의 모양은 종파의 영향으로 영상에서 유선형이었다. 작은 구름의 경우 선박의 모양과 유사했으나, 해상에 반영된 구름의 그림자로 배와 구분할 수 있었다. 색조는 바다표면이 검은색을 띤 반면 소형선박은 밝은 흰색으로 나타났다. 바다의 거친 정도와 파도의 방향성 배열 등을 통해 파도와 선박을 구분하였다. 또한 쌍끌이 어선과 같은 조업방법이 어선의 어업별 종류를 영상에서 추정하는데 용이한 기초 연관 자료가 되었다.

Hesperidin Improves the IL-6-Mediated Hepatic Insulin Resistance in Hepa-1c1c7 Cells

  • Hesperidin (HES) is a bioflavonoid with antioxidant, anti-inflammatory and anti-diabetic properties. IL-6 is well known as a primary proinflammatory cytokine that contributes to impaired insulin signaling in liver. This study was to investigate whether HES improves IL-6-mediated impairment of insulin sensitivity in liver. Hepa-1c1c7 cells were pre-treated with 50 and $100{\mu}M$ HES in complete media for 1 h and then cultured in the presence or absence of IL-6 (20 ng/ml). These results demonstrated that HES restored IL-6-suppressed expression of IRS-1 protein, downregulated IL-6-increased expression of CRP and SOCS-3 mRNA, and inhibited LPS-induced production of IL-6 in Hepa-1c1c7 cells. These findings indicate that HES may ameliorate hepatic insulin resistance via improvement of IL-6-mediated impaired insulin signaling in hepatocytes.

Baicalin Improves the IL-6-Mediated Hepatic Insulin Resistance in Hepa-1c1c7 Cells

  • Baicalin has antioxidant, anti-inflammatory and anti-diabetic properties. IL-6 is a primary proinflammatory cytokine that contributes to impaired insulin signaling in liver. This study was carried out to investigate whether baicalin improves IL-6-mediated insulin resistance in liver. Hepa-1c1c7 cells were pre-treated with 50 and 100 ${\mu}M$ baicalin in complete media for 1 h and then cultured in the presence or absence of IL-6 (20 ng/ml). These results demonstrated that baicalin restored IL-6-suppressed expression of insulin receptor substrate (IRS)-1 protein, downregulated IL-6-increased gene expression of C-reactive protein (CRP) and suppressor of cytokine signaling (SOCS)-3, and inhibited LPS-induced production of IL-6 in Hepa-1c1c7 cells. These findings indicate that baicalin may ameliorate hepatic insulin resistance via improvement of IL-6-mediated impaired insulin signaling in hepatocytes.

A Small group of protostellar objects: L1251C

  • We present various observational results toward a small group of Young Stellar Objects (YSOs), L1251C. Observations by Spitzer Space Telescope legacy program "From Molecular Cores to Planet Forming Disks" (c2d; Evans et al. 2003) revealed that there are three YSOs within ~15" in L1251C: IRS1 (Class I), IRS2 (Class II), and IRS3 (Class II). In order to understand the molecular environment around these YSOs, we carried out the KVN single-dish observations in $HCO^+$ J=1-0, $H^{13}CO^+$ J=1-0, $N_2H^+$ J=1-0 and HCN J=1-0. $^{12}CO$ J=1-0 was also mapped in L1251C with the TRAO 14m telescope. Integrated intensity maps of high density tracers such as $H^{13}CO^+$ J=1-0, $N_2H^+$ J=1-0 and HCN J=1-0 show similar emission distributions, whose peaks are off the positions of YSOs. A compact $HCO^+$ J=1-0 outflow and an extended $^{12}CO$ J=1-0 outflow were observed, but their outflow axes are not cosistent ($HCO^+$: NW-SE, $^{12}CO$: EW). However, the highest velocity component of the $^{12}CO$ J=1-0 outflow shows similar morphology to the $HCO^+$ J=1-0 outflow, and ~ 23 % of $^{12}CO$ outflow momentum flux is loaded onto this high velocity component. Furthermore, continuum emission has been observed at 350, 450, 850 ${\mu}m$, and 1.3mm. With the KVN single dish, the 22 GHz $H_2O$ maser emission has been also monitored toward L1251C to find variations of the systemic velocity and intensity with time.

With the KVN single dish, the 22 GHz $H_2O$ maser emission has been also monitored toward L1251C to find variations of the systemic velocity and intensity with time.

Ginsenoside compound K reduces ischemia/reperfusion-induced neuronal apoptosis by inhibiting PTP1B-mediated IRS1 tyrosine dephosphorylation

  • Background: Ginsenoside compound K (CK) stimulated activation of the PI3K-Akt signaling is one of the major mechanisms in promoting cell survival after stroke. However, the underlying mediators remain poorly understood. This study aimed to explore the docking protein of ginsenoside CK mediating the neuroprotective effects. Materials and methods: Molecular docking, surface plasmon resonance, and cellular thermal shift assay were performed to explore ginsenoside CK interacting proteins. Neuroscreen-1 cells and middle cerebral artery occlusion (MCAO) model in rats were utilized as in-vitro and in-vivo models. Results: Ginsenoside CK interacted with recombinant human PTP1B protein and impaired its tyrosine phosphatase activity. Pathway and process enrichment analysis confirmed the involvement of PTP1B and its interacting proteins in PI3K-Akt signaling pathway. PTP1B overexpression reduced the tyrosine phosphorylation of insulin receptor substrate 1 (IRS1) after oxygen-glucose deprivation/reoxygenation (OGD/R) in neuroscreen-1 cells. These regulations were confirmed in the ipsilateral ischemic hemisphere of the rat brains after MCAO/R. Ginsenoside CK treatment reversed these alterations and attenuated neuronal apoptosis. Conclusion: Ginsenoside CK binds to PTP1B with a high affinity and inhibits PTP1B-mediated IRS1 tyrosine dephosphorylation. This novel mechanism helps explain the role of ginsenoside CK in activating the neuronal protective PI3K-Akt signaling pathway after ischemia-reperfusion injury.
