Pollinator and pollination mechanism of Impatiens furcillata (Balsaminaceae) in Korea

  • An effective pollinator was investigated based on visiting insects to confirm the pollination mechanism of Impatiens furcillata Hemsl. (cheo-jin-mul-bong-seon), an annual herb that is also a species endemic to Korea that has hardly been studied in relation to pollination ecology. The insects that visited the group of I. furcillata studied here consisted of four orders, 11 families, and 16 species; Hymenoptera had seven species (43.8%), Lepidoptera had four (25.0%), Diptera four (25.0%), and Hemiptera one (6.2%). Visiting insects were divided into those that took only nectar, those that took nectar and pollen, and those that took neither. Insects that are effective for pollination are judged considering the length and body type of their mouth parts, and Amegilla florea Smith (huin-jul-beol) is judged to be the most effective pollinator in the survey area. As a result of observing pollination behavior, when visiting a flower, A. florea, which extended its glossa, approached the front, landed on a wing petal of I. furcillata, crawled into the flower tube, and then backed up and reversed its steps, with pollen adhered to its back. The findings here present basic information about species biology related to both I. furcillata and A. florea.

A study of the major insect pest communities associated with Hibiscus syriacus (Columniferae, Malvaceae)

  • The study on the major insect pests community of Hibiscus syriacus. There were three orders, seven families, and thirteen species of insects harmful to H. syriacus. When the harmful insects eating H. syriacus were classified according to which part of the flower they were eating, insects eating leaves, which considered of nine species four families and one order, accounted for 69.23 %. Insects sucking juice from leaves and stems comprised 15.38 %, with two species two families and two orders, while insects eating seeds comprised 15.38 %, with two species two families and two orders. Aphis gossypii appeared once per H. syriacus from the fourth week of April to the second week of June, appearing the most at a rate of 69.05 per H. syriacus during the first week of May. Dichocrocis surusalis appeared at a rate of 0.05 per H. syriacus during the third week of April and regularly appeared from the first week of July to the second week of September. Nortacha derogata appeared at a rate of 0.1 per tree during the first week of May, after which it continuously appeared until the third week of October. Anomis privata appeared at a rate of 0.05 per tree during the first week of May and continuously occurred from the fourth week of June to the fourth week of October. Its occurrence was most frequent at 2.30 per tree during the third week of September.

First Report on Bacterial Heart Rot of Garlic Caused by Pseudomonas fluorescens in China

  • An unreported disease of garlic was observed in commercial fields in Jiangsu province, China. The symptoms started as water soaked lesions at the base of the leaves. Later, water-soaked areas developed on stems and spread to the internal tissues, followed by yellowing and necrosis along leaf edges and soft rot of the stems. The causal organism isolated from symptomatic plants was identified as Pseudomonas fluorescens based on its biochemical and physiological characteristics and confirmed by the cellular fatty acid composition and Biolog data as well as 168 rRNA gene sequence analysis. The bacterial isolates caused similar symptoms when inoculated onto garlic plants. In addition, leek and shallot were susceptible to the P. fluorescens pathogen. However, the P. fluorescens pathogen failed to cause any symptoms when it was inoculated onto 15 other plants. This is the first report of a bacterial disease of garlic caused by P. fluorescens in China.

Effect of Chitosan Solution on the Inhibition of Pseudomonas fluorescens Causing Bacterial Head Rot of Broccoli

  • The in vitro antibacterial properties of two kinds of chitosan solutions and their effect in protection of broccoli from bacterial head rot disease were evaluated. Results showed that the two kinds of chitosan solution at different concentrations exhibited strong antibacterial activity against Pseudomonas fluorescens. However, the antibacterial activity of chitosan A solution increased with the increase of chitosan concentration up to 0.10 mg/ml while the antibacterial activity of chitosan B solution increased with the increase of chitosan concentration up to 0.05 mg/ml. In addition, the antibacterial activity of chitosan A and chitosan B solution of 0.10 mg/ml increased with the incubation time within 12 h and 24 h, respectively. The disease incidence and the lesion diameter of broccoli inoculated with P. fluorescens were significantly reduced when plants were either pretreated or post-treated with six different combinations of chitosan solutions. Overall, the results indicated that the two kinds of chitosan solutions had a potential in controlling bacterial head rot of broccoli.

음식폐기물 건조사료를 이용한 곤충의 생육특성 연구 (Study on Growth Characteristics of Insects using a Food waste-Derived Dry Feed)

  • 본 연구는 음식폐기물 건조사료를 곤충의 먹이(사료)로 공급하여 대량 증식을 위한 특성을 연구하였다. 특히 아메리카동애등에(Hermetia illucens)의 경우 음식폐기물 건조사료 함량에 따른 유충 증식량을 평가한 결과, 발육 초기에는 호프박과 일반사료를 음식폐기물 건조사료와 함께 혼합하여 공급한 경우 증체량이 증가하였으나, 발육이 서서히 진행되면서 종령시기에는 음식폐기물 건조사료 100%와 혼합사료간 유충의 중량에 큰 차이가 없었다. 집파리 유충은 음식폐기물 건조사료를 먹이로 100% 공급할 경우 발육이 부진하여 비정상적으로 용화되었으며, 정상적인 사육을 위해서는 음식폐기물 건조사료 함량을 20~30%로 낮게 공급하는 것이 효율적이다. 아메리카왕거저리의 유충은 사료의 종류에 크게 영향을 받지 않고 발육하는 것으로 나타났으며, 음식폐기물 건조사료에서 사망하는 개체가 조사되지 않아 먹이(사료)로 공급해도 생육을 저해하지 않았다.

  • 국내 유용 곤충의 70% 에탄올 및 물 추출물의 혈전용해 효과를 확인하기 위하여 fibrin plate를 이용하여 혈전용해능을 측정하였고, 그 분해산물을 SDS-PAGE를 이용하여 분리하였다. 그 결과 5종 (방울벌레 유충, 방울벌레 성충, 장수풍뎅이 성충, 꽃무지 유충, 꿀벌 유충)의 물 추출물에서 plasmin과 비교하여, 높은 혈전 용해 능을 나타내었고, 분해 패턴 또한 plasmin과 비슷하거나 더 강한 단백질 분해능을 나타내어 물 추출물에서 높은 혈전 용해능을 확인할 수 있었다.

소리곤충을 이용한 체험학습프로그램의 학습효과 (Effects of experience-based learning program using singing insects)

  • 본 연구에서는 소리곤충인 왕귀뚜라미를 이용한 체험학습프로그램을 개발하여 초등학생에게 적용함으로써 학습성취도의 변화를 알아보고자 하였다. 연구의 대상은 서대문자연사박물관에서 운영 중인 교육프로그램을 신청한 초등학교 2-3학년생 123명이었다. 총 7회에 걸쳐 동일한 검사지를 통해 사전 및 사후 학습 성취도 검사를 실시하였다. 그 결과 학습 성취도는 사전검사보다 사후검사결과에서 평균점수가 높게 나왔으며, 대응표본 t-test 결과 통계적으로도 유의한 것으로 나타났다. 따라서 소리곤충을 이용한 체험학습프로그램의 적용은 학생들의 학습 성취도를 향상 시키는데 매우 효과적임을 확인할 수 있었다. 이를 통해 교육현장에서는 다양한 형태의 체험학습프로그램의 개발과 적용이 가능해져 보다 체계적인 프로그램이 적극적으로 활용될 수 있을 것으로 기대된다.

소나무를 가해(加害)하는 천공성해충(穿孔性害蟲)에 관(關)한 연구(硏究) (Boring Insects of the Pinus densiflora S. et Z. in Korea)

  • 강원도 춘천 홍천의 소나무 건전림과 평창 정선의 솔잎혹파리 피해지역(충영형성율 70%) 임내에 식이목을 설치하고, 1999년 4월부터 8월까지 천공성해충의 분포 및 개체수를 조사한 결과 총 12과 35종이 조사되었으며, 이 가운데는 한국미기록종 5종과 천적 2과 5종이 채집되었다. 천공성해충의 종에 따른 수목의 가해선호부위를 조사한 결과 소나무의 수간하부에는 왕바구미(Siphalinus gigas)외 3종, 수간중부에는 깨다시수염하늘소(Monachamus sutor)외 7종, 수간상부에는 이비나무좀(Orthotamicus suturalis)외 1종으로 가해부위에 따라 종이 다르게 나타났으며, 소나무줄나무좀(Hylurgops interstitialis)은 수간전체부위에서 조사되었다. 천공성해충의 가해습성은 가문비왕나무좀(Xyleborus validius) 및 하늘소과(Cerambycidae)는 변재부, 솔곰보바구미(Hybitelus haroldi) 왕바구미(Siphalinus gigas)는 심재부, 노랑무늬 솔바구미(Pissodes nitidius) 노랑점바구미(P. abscurus) 솔흰점박이바구미(Shirahoshizo insidiosus) 및 나무좀과(Scolytidae)는 인피부(靭皮部)를 가해하는 것으로 조사되었다.

Molecular Characterization of Burkholderia cepacia Complex Isolates Causing Bacterial Fruit Rot of Apricot

  • The Burkholderia cepacia complex isolates causing bacterial fruit rot of apricot were characterized by speciesspecific PCR tests, recA-HaeIII restriction fragment length polymorphism (RFLP) assays, rep-PCR genomic fingerprinting, recA gene sequencing, and multilocus sequence typing (MLST) analysis. Results indicated that the isolates Bca 0901 and Bca 0902 gave positive amplifications with primers specific for B. vietnamiensis while the two bacterial isolates showed different recA-RFLP and rep-PCR profiles from those of B. vietnamiensis strains. In addition, the two bacterial isolates had a higher proteolytic activity compared with that of the non-pathogenic B. vietnamiensis strains while no cblA and esmR marker genes were detected for the two bacterial isolates and B. vietnamiensis strains. The two bacterial isolates were identified as Burkholderia seminalis based on recA gene sequence analysis and MLST analysis. Overall, this is the first characterization of B. seminalis that cause bacterial fruit rot of apricot.

Complete Nucleotide Sequence and Organization of the Mitogenome of the Red-Spotted Apollo Butterfly, Parnassius bremeri (Lepidoptera: Papilionidae) and Comparison with Other Lepidopteran Insects

  • The 15,389-bp long complete mitogenome of the endangered red-spotted apollo butterfly, Parnassius bremeri (Lepidoptera: Papilionidae) was determined in this study. The start codon for the COI gene in insects has been extensively discussed, and has long remained a matter of some controversy. Herein, we propose that the CGA (arginine) sequence functions as the start codon for the COI gene in lepidopteran insects, on the basis of complete mitogenome sequences of lepidopteran insects, including P. bremeri, as well as additional sequences of the COI start region from a diverse taxonomic range of lepidopteran species (a total of 53 species from 15 families). In our extensive search for a tRNA-like structure in the A+T-rich region, one $tRNA^{Trp}$-like sequence and one $tRNA^{Leu}(UUR)$-like sequence were detected in the P. bremeri A+T-rich region, and one or more tRNA-like structures were detected in the A+T-rich region of the majority of other sequenced lepidopteran insects, thereby indicating that such features occur frequently in the lepidopteran mitogenomes. Phylogenetic analysis using the concatenated 13 amino acid sequences and nucleotide sequences of PCGs of the four macrolepidopteran superfamilies together with the Tortricoidea and Pyraloidea resulted in the successful recovery of a monophyly of Papilionoidea and a monophyly of Bombycoidea. However, the Geometroidea were unexpectedly identified as a sister group of the Bombycoidea, rather than the Papilionoidea.
