This study is aimed to understand the effects of temperature, light and GA soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping hentgrass 'Penlinks', tall fescue 'Arid' and Kentucky hluegrass 'Nasaw'. These experiments are carried in vitro, and their results obtained are summarized as the followings ; The optimum temperature for seed germination of perennial ryegrass 'Dandy' was found to he ranged at 20~30$^{\circ}C$, and the optimum temperature for germination of creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw' seeds was found to be ranged at 20~25$^{\circ}C$, whereas the germination percentage under the below or above temperature of this range decreased. The germination of perennial ryegrass 'Dandy' and tall fescue 'Arid' seeds showed no effect on light, and while the germination percentage of creeping hentgrass 'Penlinks' increased more highly under the dark condition than under the light condition. The germination percentage decreased according to the higher temperature. The germination of Kentucky bluegrass 'Nasaw' seeds increased more highly under the dark condition than under the light condition at 20~25$^{\circ}C$, whereas the germination percentage under the dark condition at 15$^{\circ}C$ and 30$^{\circ}C$ decreased. In case of GA soaking treatment of seeds, all the cool season turfgrasses tested showed their own promoting effects on seed germination. The early germination of tall fescue 'Arid' was good without the soaking times. Creeping bentgrass 'Penlinks' and perennial ryegrass 'Dandy' had re-spectively the highest germination percentage at 10min and 1hr soaking treatment, and the longer the soaking time, the lesser the germination percentage. Kentucky bluegrass 'Nasaw' obtained the best results at 24has soaking treatment, and the longer the soaking time, the better the ger-mination percentage.

To find the optimal propagation condition for endangered Cypripedium macranthos Sw., also known as lady's slipper orchid, the effect of various organic additives on in vitro germination, protocorm formation and seedling growth was investigated during asymbiotic seed culture. When $100ml{\cdot}L^{-1}$ coconut water was added to the basal medium, the highest germination rate and protocorm formation rate were achieved, with 70.8% and 74.2% respectively. Supplementation of phloem sap from birch tree or maple tree also showed a facilitating effect to improve the germination and protocorm development. With $100ml{\cdot}L^{-1}$ birch sap or maple sap, both the germination and protocorm formation rates were roughly more than 65% and 68%. The roots and buds of the seedlings grew vigorously in the medium containing $100ml{\cdot}L^{-1}$ coconut water or phloem sap, in particular, their bud formation rates increased by more than 70%. Addition of banana powder and peptone could not create a more significantly favorable culture condition, and non-addition had the worst results. Our results demonstrated that proper organic amendments such as coconut water and phloem sap might be preferred to in vitro germination and the growth of seedlings developed from the protocorm of C. macranthos Sw. during asymbiotic seed culture.

We investigated the effects of sodium hypochlorite (NaOCl) and culture medium on embryo swelling and germination of Calanthe discolor Lindl., and established a method for determining the swelling and protocorm formation of C. discolor seeds via in vitro examination of immature seeds. Treatment of immature seeds with NaOCl greatly enhanced the extent of embryo swelling and protocorm formation of immature zygote embryos compared to seeds without NaOCl treatment. The effects of the culture media were also evaluated with regard to embryo swelling and protocorm formation of in vitro cultured seeds with and without NaOCl treatment. Additionally, the effects of white fluorescent light and red and blue LEDs lights on seedling growth in in vitro culture were examined. The most suitable condition for seedling growth after 12 weeks of culture was the red LEDs light with POM medium. These results show effective asymbiotic germination and growth of C. discolor.

The early germination of perennial ryegrass 'Dandy'. creeping bentgrass 'Penlinks' and tall fes- cue 'Arid' increased more highly at NAA (0.lmg /1) soaking treatment than at the control, and especially had the best results at l2hr. soaking treatment. An efficient and reproducible protocol for in vitro germination of seeds of Couroupita guianensis has been developed in the present study. Couroupita guianensis is a medicinally important tree and categorized as threatened plant species by the IUCN. The seeds were surface sterilized using 0.1% $HgCl_2$ and inoculated on MS (Murashige and Skoog) and Woody Plants (Llyod and McCown) medium. The seeds with partially removed seed coat responded better (100%) on half strength MS medium as compared to the full (51%), one-fourth strength MS (79%) and the WP medium. Half strength MS medium conjunct with $2.0mg{\cdot}L^{-1}$ indole-3 butyric acid (IBA) was found best among the different types of auxins and cytokinins tested for seed germination, as all the seeds germinated on this medium combination. The seedlings were successfully hardened in the greenhouse and restored in the field with 83% survival rate. The protocol can be used as conservation measure for large scale multiplication of this threatened forest tree species.
This study was conducted to investigate the effects of host tree species, temperature and humidity on ex vitro for seed germination in endangered species of Loranthus tanakae. In addition, we compared seed shapes between Loranthus tanakae and Viscum album that we could have easily shown in ex vitro condition. Seeds were germinated after one week inoculation and followed to develop radicles. Seed germination rates of Loranthus tanakae were 80~95% in most of the experimental conditions. The highest rate of holdfast penetrated to host plants was 72% in Populus alba var. pyramidalis among 13 different species tested at $20^{\circ}C$. Also the rates of their penetration were 57% in Morus bombycis, 55% in Acer palmatum and 42% in Castanea crenata at $20^{\circ}C$. Seeds were germinated under condition without irrigation and followed to withered in 12 weeks later. Stages of seed germination of the Loranthus tanakae were followed by radicle induction, holdfast development, haustorium formation and penetration in order in total period of 14 weeks.
Orchid seeds are nearly microscopic in size. Because of that, many fanciful theories were proposed for the origin of orchids. Almost 400 years separate the time when orchid seeds were seen for the first time and the development of a practical asymbiotic method for their germination. The seeds were first observed and drawn during the sixteenth century. Seedlings were first described and illustrated in 1804. The association between orchid and fungi was observed as early as 1824, while the requirement for mycorrhiza for seed germination was established in 1899. An asymbiotic method for orchid seed germination was developed in 1921. After Knudson's media B and C were formulated, orchids growing and hybridization became widespread. Hybrids which early growers may not have even imagined became possible.

This study was conducted to evaluate the performance of Italian ryegrass cultivars for salt tolerance under in vitro condition. Italian ryegrass cultivars such as Greenfarm, Florida80, Kowinearly, and Hwasan101 were tested for their tolerance to various sodium chloride levels (0, 50, 150, 250, and 350 mM). The seed germination, growth, and activities of antioxidant enzymes were investigated under salt treatment. Physiological traits such as seed germination percentage, germination period, shoot and root length, and dry weight were suppressed under entire salt stress conditions. The results indicated that the highest germination percentage and shoot and root length were recorded at normal conditions. Increased sodium chloride levels caused a significant reduction in the seed germination and growth rate. Among the four tested cultivars, Italian ryegrass 'Hwasan101' could be considered as salt tolerant owing to its higher germination percentage, better seedling growth and antioxidant activities under salinity stress, whereas Greenfarm cultivar was more sensitive. The selection of Italian ryegrass cultivars for greater tolerance to saline environment would allow greater productivity from large saline lands.

This study suggested suitable soil textures that is proper to propagate native plants to manage and restore barren land in DMZ. Germination tests were conducted for 16 native herbaceous plants growing in the DMZ border area in accordance with FAO-BI (Biodiversity International) standards, and the germination rate and T50 in vitro were investigated. In order to examine the germination characteristics according to the soil textures, we used gravel, bed and mixed soil and investigated the germination characteristics under ordinary room temperature conditions in the greenhouse. As a result, it was observed that the germination rate in the greenhouse was significantly decreased compared to the germination rate in vitro of the species advertised due to soil textures. T50 between the in vitro and each soil texture showed significant differences whereas T50 between soil textures alone did not in all species advertised. The germination rate in vitro of Aster koraiensis, Dendranthema zawadskii var. latilobum, Hosta clausa, and Hosta minor there was no significant difference compared to ordinary room temperature conditions. In addition, as the germination rate is demonstrated more than 70%, which is relatively higher than other species advertised, it is considered to have strong environmentally adaptable. On the other hand, considering that the 6 species of Leontopodium coreanum, Plantago major, Potentilla chinensis, Sedum kamtschaticum, Sedum latiovalifolium, and Veronica kiusiana demonstrated less than 50% of germination rate in vitro, it is expected to be difficult to propagate without pre-treatment. In order to use these 6 species as restoration material plants, it needs to be considered to pre-treat to improve germination rate, or to enhance the vitality of seeds by improving the seed gathering period and storage method.
A series of experiments were conducted to suggest efficient in vitro germination conditions to improve germination rate and aseptic seedling production in chicory and garland chrysanthemum. For this purpose, various levels of NaOCl concentration and soaking treatment time combinations were tasted. Fifteen minutes of soaking treatment with 5% NaOCl solution significantly improved germination and seedling formation percentages and prevented contamination in both chicory 'Precole' and 'Chiavari' and garland chrysanthemum 'Okiku 3'. There was no significant difference in germination response between MS and 1/2 MS medium compositions, but germination and seedling formation were delayed as sucrose concentration increased. Although petri-dish among various culture containers gave rise to improved germination responses in chicory 'Precole', there was no significant difference in germination responses among culture container types in chicory 'Chiavari' and garland chrysanthemum 'Okiku 3'. The ultrasonic treatment stimulated germination and seedling formation in both chicory cultivars but there was no effect in garland chrysanthemum 'Okiku 3'. The results suggested 15 minutes with 5% NaOCl for seed sterilization, 1/2 MS medium with no sucrose and petri-dish as proper medium composition and culture container, and 120 minutes with 5% NaOCl solution of ultrasonic treatment as promoting method for in vitro germination in chicory and garland chrysanthemum.
