Streptomyces 배양액에서의 Interleukin-1 유사물질의 동정 (Identification of Interleukin-l Like Material in Streptornyces Culture Supernatant)

  • We have identified a T cell-activating material in the culture supernatant of Streptomyces species. The factor in microbial culture supernatant (MCS) induced thymocyte proliferation in a does dependent fashion and it could be detected by immunoblot analysis using anti-interleukin-1(IL-1) antibody. The factor in MCS was slightly larger(about 21 kd) in its molecular weight than IL-1 on SDS-PAGE. When 125I-MCS was covalently coupled with homo-bifunctional cross-linking agent, disuccinimidyl-propionate to IL-1 receptor(IL-1R) on mouse thymoma cell(EL-4) and immunoprecipitated with anti-IL-1R antibody the molecular weight of this complex of 110 kd was observed.

소아의 미세변화형 신증후군 및 초점성 분절성 사구체 경화증 환아에서 혈청 및 요의 용해성 인터루킨-2수용체 (Serum and Urinary Levels of Soluble Interleukin-2 receptor in Childhood Minimal Change Nephrotic Syndrome and Focal Segmental Glomerulosclerosis)

  • 목 적 : 가능한 교란인자들, 즉 연령, 단백뇨, 스테로이드 사용 등의 영향이 배제된 조건에서 소아의 신증후군, 또는 그 중의 어떤 특성이 혈청이나 요의 용해성 인터루킨-2수용체 (sIL-2R)에 영향을 주는지를 알기 위해 이 연구를 시행하였다. 방 법 : 소아의 일차성 신증후군 중 임상적 혹은 병리소견으로 미세변화형 신증후군으로 진단되거나 병리소견상 초점성 분절성 사구체경화증으로 진단된 환아를 대상으로 이들을 연령 (0-l세, 2-4세, 5세 이상), 단백뇨 및 스테로이드 사용 여부 (PU+Tx-, PU+Tx+, PU-Tx+, PU-Tx-)로 구분하였다. 이들과 대조군의 혈청, 요에서 ELISA법으로 각각 sIL-2R를 정량하고, 요에서는 크레아티닌치도 측정하였다. 각 군의 혈청 sIL-2R치와 요 sIL-2R/크레아티닌 비를 계산하여 비교하였다. 결 과 : 혈청 sIL-2R는 환자와 대조군에서 도두 연령이 어릴수록 높았고, 신증후군에서 대조군보다 높지 않았다. 환자군 중에서 재발한 경우에는 높고 스테로이드 투여 시에는 낮은 경향을 보였다. 요 sIL-2R/크레아티닌 비는 특히 단백뇨가 있을 때 연령이 어릴수록 높았고 (P=0.01), 혈청 치와 마찬가지로 재발과 스테로이드의 영향을 받았다. 혈청 sIL-2R치와 요 sIL-2R/크레아티닌 비는 신 병리소견, 스테로이드 반응도에 따른 차이를 보이지 않았다 (P>0.05). 결 론 : 혈청 sIL-2R치는 연령에 따른 차이가 크고, 신증후군에서 대조군에 비해 높지 않았으나, 재발상태의 환자는 완해 상태의 환자보다 높았고, 스테로이드를 투여할 때에 낮았다. 요 sIL-2R/크레아티닌 비는 특히 단백뇨가 있을 때 혈청 sIL-2R치를 잘 반영하였다.

  • IY-80843, N[2-(2-methoxyphenyl)ethyl]-N'-[4-(lmidazole-4-yl)phenyl] formamidine, is a new potent $H_2-receptor$ anagonist. The potential secondary pharmacologic effects of this agent, on the central nervous and cardiovascular systems were studied. IY-80843 caused ptosis, suppression of locomotion, hypotehrmia, prolongation of sleeping time and hypotensive effects in mice, rats and dogs. These results suggest that IY-80843 affects the funcition of the central nervous and cardiovascular systems in a dose-dependent manner.

한국인 건선 환자에서의 IL-1B (-511, +3954)와 IL-1RN 유전자의 다양성 조사 (Investigation of IL-1B (-511, +3954) and IL-1RN Gene Polymorphisms in Korean Psoriasis Patients)

  • Background: Psoriasis is an inflammatory skin disorder that is characterized by a marked proliferation of keratinocytes, vascular dilation and leukocyte infiltration. Cytokines play important roles in the pathogenesis of inflammatory disorders. An overexpression of proinflammatory cytokines was characterized in psoriasis plaque. Among these cytokines, IL-$1{\beta}$ is major pro-inflammatory cytokine synthesized during the infection and inflammatory process. The IL-1 receptor antagonist (IL-1Ra) competes for the same IL-1 receptor for $IL-1{\alpha}$ and $-1{\beta}$, which prevents activation of the target cells. Three single nucleotide polymorphisms (SNPs) in the IL-$1{\beta}$ gene have been reported at position -31, -511 and +3954. Within the IL-1Ra gene (IL-1RN), there is a variable number of tandem repeats (VNTR) of an 86 bp length in intron 2. These polymorphisms related to cytokine production and associated with various diseases. Methods: We investigated the polymorphisms of IL-1B (promoter -511 and +3954) and IL-1RN on 114 psoriasis patients and 311 healthy normal controls in Korean. We performed PCR-RFLP on single nucleotide polymorphisms (SNPs) of IL-1B (promoter -511 and +3954) and fragment analysis on IL-1RN 86 bp VNTR polymorphism. Results: The frequency of IL-1B $-511^*1$ allele (patients vs. controls; 50.0% vs. 42.3%, RR=1.4) was significantly increased and IL-1B $-511^*2$ allele (patients vs. controls; 50.0% vs. 57.7%, RR=0.7) decreased in psoriasis patients compared to normal controls. We also analyzed the IL-1B -511 polymorphism according to patients' characters (age of onset, sex and family history). The IL-1B -511 alleles were significantly associated in patients with male and family history than health normal controls. There were no significant associations of IL-1B +3954 and IL-1RN polymorphisms with psoriasis patients. Conclusion: These results suggest that the polymorphism of IL-1B -511 could be genetic susceptibility to psoriasis in Koreans.

혈관평활근세포에서 glycated albumin에 의한 interleukin-6 증가에 관여하는 인자에 대한 연구 (Glycated Serum Albumin Induces Interleukin-6 Expression in Vascular Smooth Muscle Cells)

  • Glycate화된 단백질이 혈관질환의 발생에 관여하는지 알아보기 위하여 glycated albumin (GA)이 혈관평활근 세포에서 인터루킨-6 발현에 영향을 주는지 조사하고 또한 그 기전을 구명하였다. GA에 노출된 혈관평활근세포에서 인터루킨-6 transcript가 증가하고, 인터루킨-6 단백질의 분비가 증가하고, 또한 인터루킨-6 유전자의 promoter가 활성화되었다. GA에 의한 인터루킨-6 유전자의 promoter 활성화는 dominant negative 형태의 Toll-like receptor (TLR)-4와 myeloid differentiation factor 88 (Myd88)에 의하여 크게 감소되었지만, dominant negative 형태의 TLR-2와 TIR-domain-containing adapter-inducing interferon-$\beta$ (TRIF)의 영향을 받지 않았다. 그리고 Extrcellular signal-related kinase (ERK) 억제 물질들은 GA에 의한 인터루킨-6의 분비 및 인터루킨-6 유전자 promoter 활성화를 억제하였다. 그리고 인터루킨-6 유전자의 promoter의 NF-${\kappa}B$-binding sequence에 변이는 GA에 의한 인터루킨-6 유전자의 promoter 활성화 억제하였다. 이러한 결과는 혈관평활근세포에서 GA에 의한 인터루킨-6 유전자 활성화에 TLR-4와 ERK 및 NF-${\kappa}B$가 관여함을 의미한다.

두시 하태독법의 IL-4 활성 조절이 D. farinae 유도 아토피유사피부염 발병 조절에 미치는 효과 (The Effect of Douchi Hataedock Treatment for Dermatophagoides Farinae-Induced Atopic Dermatitis-like Skin Lesions by Controlling IL-4 Activity)

  • Objectives Hataedock method is a Korean medical therapy which removes fetal toxin by orally administering herbal decoction to neonates. This study was to observe skin damage and anti-inflammatory effect via regulating IL-4 activity in NC/Nga mice which were induced atopic dermatitis (AD)-like skin lesion by Dermatophagoides (D.) farinae after applying Douchi Hataedock method. Methods NC/Nga mice with 3 weeks of gestational age were used. Each 10 mice were allocated to the control group (Ctrl), the AD-induced group (AE), and the group which induced AD after administering Douchi extract (GT). After 4 weeks from administering Douchi extract to the mice, the primary AD was induced by applying D. farinae extract 6 times per week for 3 weeks and then the secondary AD was induced by the same method after 1 week from the primary AD induction. To identify the skin damage and anti-inflammatory effect, we observed LxR, IL-4, Fc ${\varepsilon}$ receptor, substance P, and $NF-{\kappa}B$. Results The GT group showed alleviation of skin injury and decrease in capillary angiogenesis. Stratum corneum damage, epithelial cell hyperplasia, lymphocyte infiltration, and capillary distribution relatively decreased in the GT group. LxR-positive reaction in the GT group were increased by 53% than that of the AE group. IL-4 production, $Fc{\varepsilon}$ receptor activity, and substance P-positive reaction in the GT group were decreased by 82%, 42%, and 82% respectively compare to those of the AE group. $NF-{\kappa}B$-positive reaction in the GT group were decreased by 15% compare to that of the AE group. Conclusions Hataedock method with Douchi extract alleviated AD via reducing inflammatory cytokines secreted at the early stage of AD. Thus, Douchi Hataedock method has a beneficial effect for the prevention and treatment of AD.

결핵성 늑막삼출과 비결핵성 늑막삼출에서의 가용성 Interleukin-2 수용체의 농도 (Soluble Interleukin-2 Receptor(sIL-2R) Levels in Patients Tuberculous Pleurisy VS Nontuberculous Pleurisy)

  • 연구목적 : 혈청 sIL-2R 농도는 T세포의 면역활성화가 관여되는 육아종성질환, 장기이식, 자가면역질환에서, 또한 혈액종양 및 림프세망내 피계종양등에서 증가 한다고 알려져있다. 최근 결핵환자에서도 질병의 활성화 정도에 따라 혈청 및 흉강 삼출액에서 sIL-2R 농도의 증가를 보고하고 있다. 임상에서 결핵성 흉강삼출과 악성 흉강삼출의 감별이 어려운 경우를 경험하게 되는데 흉강삼출액에서 sIL-2R 농도 측정이 이들 질환의 감별에 도움이 될수 있는지 알아보기 위해 늑막조직검사에서 결핵성으로 확인된 12명의 결핵성 흉강삼출액환자와 32명의 비결핵성 흉강 삼출액환자를 대상으로 혈청 및 흉강삼출액에서 sIL-2R 농도를 측정하였다. 방법 : 대상환자의 늑막삼출액을 원심분리하여 얻은 상층액과 동시에 채취한 혈청을 $-70^{\circ}C$에 보관한 후 검사를 시행하였다. 가용성 IL-2수용체 농도 측정은 Cellfree(r) IL-2R test kit(T-cell sciences Inc., Cambridge, MA, USA)를 이용하였다. 측정방법을 간단히 설명하면 IL-2R 분자의 두가지 항원 결정기(epitope)에 대한 단일 항체를 이용한 샌드위치 ELISA 검사로 측정하였다. 결과 : 1) 늑막삼출액의 sIL-2R 농도는 비결핵성 늑막삼출환자군보다 결핵성 늑막삼출환자군에서 높았다(p<0.005). 2) 늑막삼출액에서 감별진단 기준을 sIL-2R 농도 5,000u/ml로 할 경우 결핵성 늑막삼출환자군을 악성 늑막삼출환자군과 비교시 민감도는 84.6%, 특이도는 99.9%였다. 3) 혈청 sIL-2R 농도는 결핵성 늑막삼출환자군에서 세균성 늑막 삼출환자군보다 유의한 증가를 보였으나(p<0.05), 악성 늑막 삼출환자군 및 늑막여출액환자군과는 유의한 차이가 없었다(p>0.05). 4) 결핵성 늑막삼출환자군에서 sIL-2R 농도는 혈청에서보다 늑막삼출액에서 더 높았다(p<0.005). 결론 : 결핵성 늑막삼출에서 늑막삼출액내의 sIL-2R 농도는 감별기준 5,000 u/ml로 사용할 경우 결핵성 늑막삼출과 비결핵성 늑막삼출의 감별진단에 도움이 되리라 생각된다.

Chronic cold stress-induced myocardial injury: effects on oxidative stress, inflammation and pyroptosis

  • Background: Hypothermia is a crucial environmental factor that elevates the risk of cardiovascular disease, but the underlying effect is unclear. Objectives: This study examined the role of cold stress (CS) in cardiac injury and its underlying mechanisms. Methods: In this study, a chronic CS-induced myocardial injury model was used; mice were subjected to chronic CS (4℃) for three hours per day for three weeks. Results: CS could result in myocardial injury by inducing the levels of heat shock proteins 70 (HSP70), enhancing the generation of creatine phosphokinase-isoenzyme (CKMB) and malondialdehyde (MDA), increasing the contents of tumor necrosis factor-α (TNF-α), high mobility group box 1 (HMGB1) interleukin1b (IL-1β), IL-18, IL-6, and triggering the depletion of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH). Multiple signaling pathways were activated by cold exposure, including pyroptosis-associated NOD-like receptor 3 (NLRP3)-regulated caspase-1-dependent/Gasdermin D (GSDMD), inflammation-related toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)-mediated nuclear factor kappa B (NF-κB), and mitogen-activated protein kinase (MAPK), as well as oxidative stressinvolved thioredoxin-1/thioredoxin-interacting protein (Txnip) signaling pathways, which play a pivotal role in myocardial injury resulting from hypothermia. Conclusions: These findings provide new insights into the increased risk of cardiovascular disease at extremely low temperatures.

JQ1, a BET inhibitor, controls TLR4-induced IL-10 production in regulatory B cells by BRD4-NF-κB axis

  • Regulatory B cells, also well-known as IL-10-producing B cells, play a role in the suppression of inflammatory responses. However, the epigenetic modulation of regulatory B cells is largely unknown. Recent studies showed that the bromodomain and extra-terminal domain (BET) protein inhibitor JQ1 controls the expression of various genes involving cell proliferation and cell cycle. However, the role of BET proteins on development of regulatory B cells is not reported. In this study, JQ1 potently suppressed IL-10 expression and secretion in murine splenic and peritoneal B cells. While bromodomain-containing protein 4 (BRD4) was associated with $NF-{\kappa}B$ on IL-10 promoter region by LPS stimulation, JQ1 interfered the interaction of BRD4 with $NF-{\kappa}B$ on IL-10 promoter. In summary, BRD4 is essential for toll like receptor 4 (TLR4)-mediated IL-10 expression, suggesting JQ1 could be a potential candidate in regulating IL-10-producing regulatory B cells in cancer.

  • This work was performed to investigate the effects of brazilin in vitro on mitrogen-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant-induced proliferation, ConA-induced TCGF release and responsiveness to recombinant IL-2 using splenocytes from C57BL/6 female mice. Brazilin (29-80 ng/ml) caused a noticeable increase in TCGF production of splenocytes, but did not affect responsivness to recombinant IL-2, the expression of ConA-induced high affinity IL-2 receptor and mitogen-induced proliferation of splenocytes.
