한국인 아동에서의 IL-1 유전자 다변성 연구 (The IL-1 Gene polymorphisms in Korean Children)

  • The severe form of chronic periodontitis(CP) has been reported to be strongly associated with the presence of allele 2 of composite IL-1B(+3954) and IL-1A(+4845) genetic polymorphisms(genotype positive). However, other studies have reported conflicting findings. These might have resulted from differences in ethnic background and disease entities. The aim of this study was to determine the distribution of IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN(VNTR) genetic polymorphisms in children as a future Korean population. The study population consisted of 92 children from the Dept. of Pediatric Dentistry, Chonnam National University Hospital. Genomic DNA was obtained from buccal swab. The IL-1A(+4845), IL-1B(+3954), and IL-1B(-511) genes were genotyped by amplifying the polymorphic region using multiplex polymerase chain reaction(PCR), followed by restriction enzyme digestion and gel electrophoresis. IL-1 RN(VNTR) polymorphism were then evaluated by PCR amplification and fragment size analysis in agarose gel. The allele 2 frequency was 41.3%, 4.3%, 47.8%, and 9.9% for IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN respectively. The frequency of genotype with allele 2 carriage for IL-1A(+4845), IL-1B(+3954), IL-1B(-511), and IL-1 RN was 77.1%, 7.6%, 63.0%, and 15.2% respectively. The allele 2 frequency in IL-1B(+3954) was significantly higher in female than in male population(p<0.05). The negative association was shown between the presence of allele 2 in IL-1B(-511) and in IL-1B(+3954), and the carriage rate of IL-1B(+3954) allele 2 tended to lower in IL-1B(-511) allele 2(P=0.056). Only 7.3% of children carried the composite genotype of IL-1A(+4845) and IL-1B(+3954). These results suggest that the polymorphism of IL-1B(+3954) and the positive composite genotype was relatively rare in Korean population.

Effect of scaling and root planing on the expression of anti-inflammatory cytokines (IL-4, IL-9, IL-10, and IL-13) in the gingival crevicular fluid of electronic cigarette users and non-smokers with moderate chronic periodontitis

  • Purpose: The aim of this cross-sectional study was to investigate the effect of scaling and root planing (SRP) on the expression of anti-inflammatory cytokines (interleukin [IL]-4, IL-9, IL-10, and IL-13) in the gingival crevicular fluid (GCF) of electronic cigarette users and non-smokers with moderate chronic periodontitis (CP). Methods: Electronic cigarette users and non-smokers with CP were included in the study. Full-mouth plaque and gingival indices, probing depth (PD), clinical attachment loss (CAL), and marginal bone loss (MBL) were assessed. The GCF was collected, and its volume and levels of IL-4, IL-9, IL-10, and IL-13 were assessed. These parameters were evaluated at baseline and 3 months after SRP. The sample size was estimated, and comparisons between groups were performed. P<0.05 was considered to indicate statistical significance. Results: Thirty-six electronic cigarette users (47.7±5.8 years old) and 35 non-smokers (46.5±3.4 years old) with CP were included. At baseline, there were no differences in plaque index (PI), PD, CAL, MBL, and GCF IL-4, IL-9, IL-10, and IL-13 between electronic cigarette users and nonsmokers. At the 3-month follow-up, there were no significant differences in PI, gingival index (GI), PD, CAL, and MBL in electronic cigarette users compared to baseline, while there were significant reductions in PI, GI, and PD among non-smokers. At the 3-month follow-up, GCF IL-4, IL-9, IL-10, and IL-13 levels were significantly elevated in both groups (P<0.05) compared to baseline. The increases in GCF IL-4, IL-9, IL-10, and IL-13 levels were significantly higher in non-smokers (P<0.05) than in electronic cigarette users at the 3-month follow-up. Conclusions: Levels of GCF IL-4, IL-9, IL-10, and IL-13 increased after SRP in electronic cigarette users and non-smokers with CP; however, the anti-inflammatory effect of SRP was more profound in non-smokers than in electronic cigarette users.

Preventive Effect of Hwangryunhaedok-tang on Inflammatory Responses in PHA-stimulated Peripheral Blood Mononuclear Cells from Cerebral Infarction Patients

  • Objectives: Hwangryunhaedok-tang (HRHDT), a prescription composed of four herbs, has been wi dely used in Oriental Medicine for the treatment of cerebral infarction. However, the mechanisms by which the herbal formula affects on the production of pro- and anti-inflammatory cytokines in cerebral infarction patients remain unknown yet. Methods: The levels of pro- and anti-inflammatory cytokines, including tumor necrosis factor (TNF)-a, interleukin (IL)-1b, and IL-6, IL-10, and TGF-${\beta}1$ were determined in peripheral blood mononuclear cells (PBMCs) from cerebral infarction patients under our experimental conditions. Results: The secretory levels of pro- and anti-inflammatory cytokines, including tumor necrosis factor (TNF)-a, interleukin (IL)-1b, and IL-6, and IL-10 were significantly increased in phytohemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMCs) from cerebral infarction patients. However, pretreatment with HRHDT significantly inhibited the secretion of pro- and anti-inflammatory in PBMCs. Also, HRHDT induced a significant increase of transforming growth factor (TGF)-b1 in PBMCs. Conclusions: These data indicate that HRHDT may be beneficial in the suppression of inflammatory processes of cerebral infarct through suppression of TNF-$\alpha$, IL-$1{\beta}$, IL-6, and IL-10 and induction of TGF-${\beta}1$.

The Effects of Pulsatilla Koreana NAKAI Pharmacopuncture Therapy on the Chunchu (ST25) for Dextran Sulfate Sodium-Induced Colitis in Mice

  • Background: This research was performed to investigate the effects of Pulsatilla Koreana NAKAI pharmacopuncture (PPA) therapy on intestinal disease in rats with dextran sulfate sodium (DSS)-induced colitis. Methods: The subjects were divided into five groups : A control group, saline group, pharmacopuncture group PPA1 ($0.2mg/1kg/40{\mu}{\ell}$), pharmacopuncture group PPA2 ($0.5mg/1kg/40{\mu}{\ell}$), and pharmacopuncture group PPA 3($1mg/1kg/40{\mu}{\ell}$). The experimental model of colitis was induced by infection of dextran sulfate sodium (DSS) for eighteen days. After colitis was induced, PPA therapy was practiced on the Chunchu (ST25) once every two days for a total six times. Thereafter Disease Activity Index (DAI), colon length, damage to the colonic mucosa, body weight, IL-6, IL-10, $IL-1{\beta}$, $IFN-{\gamma}$, $TNF-{\alpha}$, $TGF-{\beta}1$, IL-23 and IL-17 were measured. Results: The results were as follows. 1. DAI was significantly decreased in the PPA groups. 2. Colon length was significantly increased in the PPA groups. 3. Damage of colonic mucosa was observed less in the PPA groups. 4. Body weight was significantly increased in the saline group and the PPA groups. 5. The PPA2 group showed a significant decrease in the intensity of IL-6, $IL-1{\beta}$, $IFN-{\gamma}$ and $TNF-{\alpha}$ levels and the mean of IL-23. 6. The PPA3 group showed a significant increase in the intensity of IL-10 and $TGF-{\beta}1$ levels. 7. No significant differences were shown in the mean of IL-17. Conclusion: These results suggest that PPA therapy on Chunchu (ST25) can be used as an effective treatment for inflammatory bowel disease.

백화사설초(白花蛇舌草)가 ovalbumin으로 유도된 천식동물모델에서 Eosinophil의 수, IgE 및 IL-4에 미치는 영향 (The Effects of Oldenlandiae Diffusae Herba Extract on Eosinophil, IgE and IL-4 in Experimental Asthma induced by Ovalbumin)

  • Objective : This study was performed to investigate the effect of oral administration of Oldenlandiae Diffusae Herba (ODH) on eosinophil, immunoglobulin-E and interleukin-4 in the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. And, it was observed the change of the concentrations of IL-4 in BALF and splenocyte, IgE in serum by ELISA. Results : 1. The number of eosinophil in BALF was significantly decreased in ODH group copared with control group. 2. Concentration of IL-4 in serum, BALF and splenocyte was significantly decreased in ODH group compared with control group, respectively. 3. Level of IgE in serum was significantly decreased in ODH group compared with control group. Conclusion : We found that the effect of ODH extract in asthma was implicated in reductions of IL-4 released from Th2 cell, and decreases of IgE from plasma cell. These findings suggest that ODH extract can produce anti-asthmatic effect, which may playa role in allergen-induced asthma therapy.

High Resolution Melting Curve Assay for Detecting rs12979860 IL28B Polymorphisms Involved in Response of Iranian Patients to Chronic Hepatitis C Treatment

  • Background: A recent genome-wide association study (GWAS) on patients with chronic hepatitis C (CHC) treated with peginterferon and ribavirin (pegIFN-${\alpha}$/RBV) identified a single nucleotide polymorphism (SNP) on chromosome 19 (rs12979860) which was strongly associated with a sustained virological response (SVR). The aim of this study was twofold: to study the relationship between IL28B rs12979860 and sustained virological response (SVR) to pegIFN-${\alpha}$/RVB therapy among CHC patients and to detect the rs12979860 polymorphism by high resolution melting curve (HRM) assay as a simple, fast, sensitive, and inexpensive method. Materials and Methods: The study examined outcomes in 100 patients with chronic hepatitis C in 2 provinces of Iran from December 2011 to June 2013. Two methods were applied to detect IL28B polymorphisms: PCR-sequencing as a gold standard method and HRM as a simple, fast, sensitive, and inexpensive method. Results: The frequencies of IL28B rs12979860 CC, CT, and TT alleles in chronic hepatitis C genotype 1a patients were 10% (10/100), 35% (35/100), and 6% (6/100) and in genotype 3a were 13% (13/100), 31% (31/100), and 5% (5/100), respectively. In genotype 3a infected patients, rs12979860 (CC and CT alleles) and in genotype 1a infected patients (CC allele) were significantly associated with a sustained virological response (SVR). The SVR rates for CC, CT and TT (IL28B rs12979860) were 18%, 34% and 4%, respectively. Multiple logistic regression analysis identified two independent factors that were significantly associated with SVR: IL-28B genotype (rs 12979860 CC vs TT and CT; odds ratio [ORs], 7.86 and 4.084, respectively), and HCV subtype 1a (OR, 7.46). In the present study, an association between SVR rates and IL28B polymorphisms was observed. Conclusions: The HRM assay described herein is rapid, inexpensive, sensitive and accurate for detecting rs12979860 alleles in CHC patients. This method can be readily adopted by any molecular diagnostic laboratory with HRM capability and will be clinically beneficial in predicting treatment response in HCV genotype 1 and 3 infected patients. In addition, it was demonstrated that CC and CT alleles in HCV-3a and the CC allele in HCV-1a were significantly associated with response to pegIFN-${\alpha}$/RBV treatment. The present results may help identify subjects for whom the therapy might be successful.

괴화 추출물이 대식세포에서의 nitric oxide와 interleukin-6의 생성에 미치는 영향 (Suppression of nitric oxide and interleukin-6 production by methanol extract of Sophorae Flos in macrophage cells)

  • Both nitric oxide (NO) and interleukin-6 (IL-6) have been thought to have a role in the pathogenesis of inflammatory periodontal disease as it does in other inflammatory diseases, and the inhibitors of NO and IL-6 production have been considered as potential anti-inflammatory agents. In this study, we evaluated methanol extract of Sophorae Flos for inhibition of NO and IL-6 production in Prevotella intermedia LPS-induced mouse macrophages RAW264.7 cells. Dried Sopharae Flos was sliced, and extracted with 100% methanol. LPS from p. intermedia ATCC 25611 was prepared by the standard hot phenol-water method. NO production was assayed by measuring the accumulation of nitrite in culture supematants and IL-6 was measured using mouse IL-6 ELISA kit. Western blot analysis of iNOS and analysis of reverse transcription (RT)-PCR products were carried out. The methanol extract of Sophorae Flos concentration-dependently reduced the production of NO and the expression of iNOS protein and mRNA in RAw264.7 cells treated with P. intermedia LPS. Sophorae Flos also suppressed IL-6 production and the expression of IL-6 mRNA in RAw264.7 cells stimulated by P. intermedia LPS. The inhibition of NO and IL-6 production by Sophorae Flos may be useful in the therapy of inflammatory diseases such as periodontitis. This hypothesis, however, remains to be tested.

승정원일기(承政院日記)와 조선정조시대(朝鮮正禮時代)의 천문관측기록(天文觀測記錄) (ON THE ASTRONOMICAL RECORDS MADE IN THE DAYS OF KING JEONGJO OF YI DYNASTY)

  • Astronomical records in the days of King Jeongjo of Yi dynasty, 1777-1800, are examined for solar and lunar eclipses, meteors and comets. The books used for this investigation are Sung Jeong Won Il Gi, which are the original daily records written in the court. The records found in these Sung Jeong Won Il Gi are tabulated according to the four astronomical phenomena above, and they were cross checked with the records in the book, Jung Bo Mun Heon Bi Go. Although Jung Bo Mun Heon Bi Go has most frequently been used as the original records to those who work for the ancient Korean records, the inaccuracy and the deficiency are found.

  • 본 논문에서는 SNP데이터를 이용하여 간경화에 대한 감수성을 예측하기 위해 의사결정 트리를 이용하였다. 데이터는 간경화 환자와 정상환자 총 116명의 데이터를 사용하였으며, Feature 값으로는 간질환과 밀접한 연관성을 갖는 28개의 SNP데이터를 사용하였다. 실험방법은 각각의 SNP에 대하여 의사결정트리로 분류율을 측정한 후 가장 높은 분류율을 가지는 SNP부터 조합해 나가는 방식으로 C4.5 의사결정트리를 이용 leave-one-out cross validation으로 간경화와 정상을 구분하는 정확도를 측정하였다. 실험결과 간 질환 관련 SNP중 IL1RN-S130S, IRNGR2-Q64R, IL-10(-592), IL1B_S35S 4개의 SNP조합에서 65.52%의 정확도를 얻을 수 있었다.

Elevated Serum IL-17A but not IL-6 in Glioma Versus Meningioma and Schwannoma

  • Background: There is a Th1/Th2 cytokine imbalance and expression of IL-17 in patients with brain tumours. We aimed to compare the levels of IL-17A and IL-6 in sera of glioma, meningioma and schwannoma patients as well as in healthy individuals. Materials and Methods: IL-17A and IL-6 levels were measured in sera of 38 glioma, 24 meningioma and 18 schwannoma patients for comparison with 26 healthy controls by commercial ELISA assays. Results: We observed an increase in the IL-17A in 30% of glioma patients while only 4% and 5.5% of meningioma and schwannoma patients and none of the healthy controls showed elevated IL-17A in their sera ($0.29{\pm}0.54$, $0.03{\pm}0.15$ and $0.16{\pm}0.68$ vs. $0.00{\pm}0.00pg/ml$; p=0.01, p=0.01 and p=0.001, respectively). There was also a significant decrease in the level of IL-6 in glioma patients compared to healthy controls ($2.34{\pm}4.35$ vs. $4.67{\pm}4.32pg/ml$; p=0.01). There was a direct correlation between the level of IL-17A and age in glioma patients (p=0.005). Glioma patients over 30 years of age had higher IL-17A and lower IL-6 in their sera compared to the young patients. In addition, a non-significant grade-specific inverse trend between IL-17A and IL-6 was observed in glioma patients, where high-grade gliomas had higher IL-17A and lower IL-6. Conclusions: Our data suggest a Th17 mediated inflammatory response in the pathogenesis of glioma. Moreover, tuning of IL-6 and IL-17A inflammatory cytokines occurs during progression of glioma. IL-17A may be a potential biomarker and/or immunotherapeutic target in glioma cases.
