Diagnostic accuracy of a combination of salivary hemoglobin levels, self-report questionnaires, and age in periodontitis screening

  • Purpose: This study evaluated the predictive performance of a combination of self-report questionnaires, salivary hemoglobin levels, and age as a non-invasive screening method for periodontitis. Methods: The periodontitis status of 202 adults was examined using salivary hemoglobin levels, responses to 10 questions on a self-report questionnaire, and the Community Periodontal Index (CPI). The ability of those two variables and the combination thereof with age to predict the presence of CPI scores of 3-4 and 4 was assessed using logistic regression and receiver operating characteristic (ROC) curve analysis. Results: CPI scores of 3-4 and 4 were present among 79.7% and 46.5% of the sample, respectively. The area under the ROC curves (AUROCs) of salivary hemoglobin levels for predicting prevalence of CPI scores of 3-4 and 4 were 0.63 and 0.67, respectively (with sensitivity values of 71% and 60% and specificity values of 56% and 72%, respectively). Two distinct sets of five questions were associated with CPI scores of 3-4 and 4, with AUROCs of 0.73 and 0.71, sensitivity values of 76% and 66%, and specificity values of 63% and 69%. The combined model incorporating both variables and age showed the best predictive performance, with AUROCs of 0.78 and 0.76, sensitivity values of 71% and 65%, and specificity values of 68% and 77% for CPI scores of 3-4 and 4, respectively. Conclusions: The combination of salivary hemoglobin levels and self-report questionnaires was shown to be a valuable screening method for detecting periodontitis.

Correlation between Bone Maturation and Obese Degree in Childhood (소아기에 있어 비만도와 골성숙도의 상관성)

  • Objectives : It has been suggested that obesity in childhood may lead early menarche and rapid bony maturation. The purpose of this study is to investigate correlation between bone maturation and obese degree in childhood in Korea. Methods : Height, body weight, BMI, percent body fat were taken 138 patients. Ultrasonic scan was taken through calcaneous of the right foot to evaluate bone age. $DA({\Delta}age)$ was calculated from the difference of bone age and chronological age, and it was used in correlation with obese degree. Results : 1. The average age of patients was $11.91{\pm}2.86$ years old in boys, $11.44{\pm}2.27$ years old in girls. 2. The average BMI was $20.92{\pm}4.53kg/m2$ in boys, $19.76{\pm}4.05kg/m2in$ girls, and average percent body fat was $25.13{\pm}8.23%$ in boys, $27.66{\pm}5.95%$ in girls. 3. The average OI(osteo index) was 37.88 in boys, 36.64 in girls, and average bone age was $12.00{\pm}2.61$ years old in boys, $11.81{\pm}2.11$ years old in girls. 4. There was significant correlation between $DA({\Delta}age)$ and PBF in boys, but no significant correlation with BMI(p<0.05). 5. There was significant correlation between $DA({\Delta}age)$ and PBF, BMI in girls(p<0.01). 6. There was significant correlation between RA and OI(p<0.01). Conclusion : It is suggested that obese degree may have an impact on early bony maturation especially in case of girls.

Optimum Conditions for the Separation of Lecithin from Egg Yolk by Response Surface Methodology (반응표면 분석법을 이용한 계란 난황 레시틴 분리 최적조건)

  • The optimum conditions for the extraction of lecithin from egg yolk were determined using response surface methodology (RSM). On the basis of the results of preliminary experiments, the most effective values were selected. The effects of three independent variables, dilution ratio, solvent composition, and extraction temperature on the response of crude egg lecithin (g) were then determined. The optimum conditions for egg lecithin separation obtained using ridge analysis were 6.51, 95.83%, and $40.2^{\circ}C$ for the dilution ratio, solvent composition, and extraction temperature, respectively. Using the optimum conditions, 55.04% of crude lecithin in total phospholipid can be obtained from 100 g liquid egg yolk. The experimental values (56.21% crude lecithin in total phospholipid) agreed with the predicted values.

Microencapsulation of Caramel Flavor and Properties of Ready-to-drink Milk Beverages Supplemented with Coffee Containing These Microcapsules

  • This study aimed to extend the retention of flavor in coffee-containing milk beverage by microencapsulation. The core material was caramel flavor, and the primary and secondary coating materials were medium-chain triglyceride and maltodextrin, respectively. Polyglycerol polyricinoleate was used as the primary emulsifier, and the secondary emulsifier was polyoxyethylene sorbitan monolaurate. Response surface methodology was employed to determine optimum microencapsulation conditions, and headspace solid-phase microextraction was used to detect the caramel flavor during storage. The microencapsulation yield of the caramel flavor increased as the ratio of primary to secondary coating material increased. The optimum ratio of core to primary coating material for the water-in-oil (W/O) phase was 1:9, and that of the W/O phase to the secondary coating material was also 1:9. Microencapsulation yield was observed to be approximately 93.43%. In case of in vitro release behavior, the release rate of the capsules in the simulated gastric environment was feeble; however, the release rate in the simulated intestinal environment rapidly increased within 30 min, and nearly 70% of the core material was released within 120 min. The caramel flavor-supplemented beverage sample exhibited an exponential degradation in its flavor components. However, microcapsules containing flavor samples showed sustained flavor release compared to caramel flavor-filled samples under higher storage temperatures. In conclusion, the addition of coffee flavor microcapsules to coffee-containing milk beverages effectively extended the retention of the coffee flavor during the storage period.

Ginsenoside Rf inhibits cyclooxygenase-2 induction via peroxisome proliferator-activated receptor gamma in A549 cells

  • Background: Ginsenoside Rf is a ginseng saponin found only in Panax ginseng that affects lipid metabolism. It also has neuroprotective and antiinflammatory properties. We previously showed that Korean Red Ginseng (KRG) inhibited the expression of cyclooxygenase-2 (COX-2) by hypoxia via peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$). The aim of the current study was to evaluate the possibility of ginsenoside Rf as an active ingredient of KRG in the inhibition of hypoxia-induced COX-2 via $PPAR{\gamma}$. Methods: The effects of ginsenoside Rf on the upregulation of COX-2 by hypoxia and its antimigration effects were evaluated in A549 cells. Docking of ginsenoside Rf was performed with the $PPAR{\gamma}$ structure using Surflex-Dock in Sybyl-X 2.1.1. Results: $PPAR{\gamma}$ protein levels and peroxisome proliferator response element promoter activities were promoted by ginsenoside Rf. Inhibition of COX-2 expression by ginsenoside Rf was blocked by the $PPAR{\gamma}-specific$ inhibitor, T0070907. The $PPAR{\gamma}$ inhibitor also blocked the ability of ginsenoside Rf to suppress cell migration under hypoxia. The docking simulation results indicate that ginsenoside Rf binds to the active site of $PPAR{\gamma}$. Conclusions: Our results demonstrate that ginsenoside Rf inhibits hypoxia induced-COX-2 expression and cellular migration, which are dependent on $PPAR{\gamma}$ activation. These results suggest that ginsenoside Rf has an antiinflammatory effect under hypoxic conditions. Moreover, docking analysis of ginsenoside Rf into the active site of $PPAR{\gamma}$ suggests that the compound binds to $PPAR{\gamma}$ in a position similar to that of known agonists.

Analyzing Growth Reactions of Herbaceous Plants for Irrigation Management

  • Background and objective: The purpose of this study was to provide guidelines for irrigation management by analyzing the effects of soil moisture on the growth characteristics of herbaceous plants in green infrastructure. Methods: In a rain shelter greenhouse, the growth performance of nine species of experimental plants was assessed under different soil moisture contents (20%, 15%, 10%, 5%, and 1%) for about 5 months to analyze plant growth characteristics due to soil humidity. Methods to determine plant growth conditions include surveying growth conditions of the crowns, stems, leaves, flowers and fruits on the aerial part and surveying growth conditions of the roots in the underground part. Results: The results showed that Mukdenia rossii and Astilbe rubra grew well at 15% moisture content with irrigation intervals of 10 and 13 days, respectively. Soil moisture content of 10% with irrigation intervals of 13 and 17 days was appropriate for Sedum kamtschaticum and Pachysandra terminalis. Similarly, Aquilegia japonica and Liriope platyphylla grew well at 15% moisture content with irrigation intervals of 10 and 17 days. However, Ligularia stenocephala grew well-developed stems and roots at 1% soil moisture content and an irrigation interval of 25 days, while the optimum conditions for Lythrum anceps were 5% moisture content and an irrigation interval of 8 days. Conclusion: Although a limited number of experimental plants were used in this study, this study could propose an appropriate irrigation cycle for planting on artificial soil substrates. Based on these results, it is possible to plan suitable planting designs considered irrigation cycles.

  • 산업의 고도화가 진행됨에 따라 화학원료의 사용이 증가하고 있고 독성을 가진 화학물질이 하천으로 유입되는 사고가 빈번하게 발생하고 있다. 수환경으로 유입되는 유해화학물질은 주로 무색무취의 물질들로 사고가 발생하더라도 초기 발견이 어려워 어류폐사를 유발하거나 취수시설에서 용수로 취수되는 경우가 발생하기 때문에 이에 대한 대응책 마련이 필수적이다. 하천에 유입된 오염물질의 거동을 신속하게 예측하기 위해 1차원 오염물질 추적 모형이 활용되는데, Fickian 이송-분산 모형(Fickian Advection-dispersion equation model; FADE)이 주로 사용되고 있다. 하지만 FADE는 오염물질이 하천 저장대에서 지체되는 현상을 반영하지 못하기 때문에 농도곡선의 왜곡도를 구현하지 못하는 단점을 가지고 있다. 따라서 본 연구에서는 하천저장대모형(River Storage Model; RSM)을 개발하고 이를 국가하천인 감천에 적용하였다. 본 연구에서 개발한 RSM은 분산계수, 본류대 면적, 저장대 면적, 저장대 교환계수의 네 가지 매개변수를 통해서 하천의 물질 저장 및 교환 특성를 구현한 non-Fickian 모형으로서, 생화학반응, 휘발, 흡·탈착항을 추가하여 유해화학물질의 혼합 거동을 정확하게 모의할 수 있도록 개발하였다. 저장대 모형의 매개변수를 산정하기 위해서 하천 유량과 지형자료를 기반으로 HEC-RAS를 모의하여 계산된 수리특성을 입력변수로 사용하였다. 저수기, 평수기, 풍수기 유량을 기준으로 세 경우의 시나리오 모의를 수행하였는데, 5ton의 톨루엔이 김천산업단지에서 감천으로 유입된 경우 약 20km 하류에 위치한 취수장에서 톨루엔의 농도변화를 예측했다. 보존성 물질에 대한 모의 결과, 풍수기의 경우 저수기에 비해 유속이 크기 때문에 취수장에서 20.56시간 먼저 기준농도에 도달하고, 7.21시간 더 짧게 머무는 것으로 나타났다. 유해화학물질의 반응특성에 대한 민감도 분석을 시행한 결과, 생화학적 반감기가 18.98시간보다 길고, 옥탄올-물 분배계수가 2.267 이하인 물질은 생분해 및 흡·탈착 반응에 둔감한 것으로 나타났다. 1m 수심 기준 0.114m/s 이하 유속에서의 하천 수리조건에서는 화학물질의 휘발성을 무시할 수 있는 것으로 밝혀졌다.

Biological properties of fermented milk with fortified whey protein

  • As a byproduct obtained from cheese manufacture, whey protein was developed as a functional food that contains multi-functional proteins. In this study, the biochemical activity of fermented milk prepared by fortifying whey protein with excellent physiological activity was investigated. Immunoglobulin (IgG) content was higher in 10% fortified whey protein fermented milk than in the control. The viable cell counts were 20% higher in the fermented milk with 10% fortified whey protein than in the control group. The antibacterial effect of 10% fortified whey protein fermented milk compared to the control group was shown to be effective against four pathogenic microorganisms, Escherichia coli (KCTC1039), Pseudomonas aeruginosa 530, Salmonela Typhimurium (KCTC3216), and Staphylococcus aureus (KCTC1621). The antioxidant effect by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities wasincreased two-fold in 10% fortified whey protein fermented milk compared to the control. The 10% fortified whey protein fermented milk inhibited the expression of the inflammatory cytokines (interleukin [IL]-6, tumor necrosis factor [TNF]-α, and induced nitric oxide synthase [iNOS]) in a concentration-dependent manner. In a piglets feeding test, the weight gain with 10% fortified whey protein fermented milk was increased by 18% compared to the control group, and no diarrhea symptoms appeared. Our results clearly demonstrated that 10% fortified whey protein fermented milk could be a useful functional ingredient for improving health.

Assessment of Displacement and Axial Force of Earth Retaining Wall at Each Excavation Step Using Direct Algorithm Back Analysis (직접알고리즘 역해석 기법을 이용한 굴착단계별 흙막이 가시설 변위 및 축력의 적정성 평가)

  • In this study, direct algorithm-based back analysis was utilized to perform back analysis on two actual earth retaining wall fields, which was then compared with genetic algorithm-based method to evaluate the suitability of the back analysis. Additionally, in order to propose effective utilization methods of the program, the measurement data, as the input for the back analysis, was varied for each excavation step, and the applicability of the back analysis results(displacement, axial force) was examined. The research findings indicate that both direct algorithm and genetic algorithm show high applicability; however, the optimization for this program is better predicted by the direct algorithm. Moreover, in order to effectively use the back analysis program employing the direct algorithm, it was evaluated that relatively accurate prediction of the earth retaining wall behavior could be achieved by inputting measurement data from the 7th excavation step for fields with final excavation steps ranging from 8 to 11.

Cytotoxic Effects of Decursin from Angelica gigas Nakai in Human Cancer Cells (당귀로부터 정제한 Decursin의 인체암세포주에 대한 세포독성)

  • Anticarcinogenic-active compound was isolated and purified from Angelica gigas Nakai. The compound was identified as decursin ($C_{19}H_{20}O_5$; molecular weight 328) by mass, IR spectrophotometry $^1H-NMR$ and $^{13}C-NMR$. The proliferation decreased in a dose dependant fashion in the MCF-7 cells treated with decursin for 24 hours over the concentration of $20{\mu}g/mL$. The $IC_{50}$ value of the decursin treatment for 24 hours were 31.04, 33.60, 27,24, $20.45{\mu}g/mL$ in the SW480, 293, HepG2 and MCF-7 cells, respectively, The growth inhibitory effect was stronger in the MCF-7 cells compared to other cells including 293 of human normal cells. The chromatin condensation, apoptotic body formation and DNA fragmentation were examined in the cells treated with decursin. These results suggest that decursin from Angelica gigas Nakai inhibited the growth through apoptosis in MCF-7 cells.
