기능성 위장관 장애에서 혈청 IL-$1\beta$, IL-2, IL-6의 변화 : 우울증 및 스트레스와의 상관성 (Serum Levels of IL-$1\beta$, IL-2 and IL-6 in Functional Gastrointestinal Disorders in Relation to Depression and Stressful Life Events)

  • 스트레스가 위장관 기능을 변화시키며 이에는 interleukin과 시상하부-뇌하수체-부신피질 축이 관여한 다고 알려져 있다. 한편 우울증이 기능성 위장관 장애 및 interleukin과 연관성을 지닌다. 본 연구에서는 IL-$1\beta$, IL-2, IL-6의 혈청 농도를 기능성 위장관 장애 환자에서 측정하고, 혈청 cortisol 및 우울증과 스트레스의 정도를 조사하여 기능성 위장관 장애에 있어서 interleukin의 역할과 우울증 및 스트레스와의 상관성을 살펴보고자 하였다. 방법: 기능성 위장관 장애의 진단을 위한 로마기준 II에 따라 진단된 환자 20명과 건강한 대조군 20명을 대상으로 하였다. 채취한 혈액에서 혈청을 분리한 다음 Enzyme-linked Immunosorbent Assay 방법으로 혈청 IL-$1\beta$, IL-2, IL-6 농도를 측정하고 Fluorescence Polarization Immunoassay 방법으로 혈청 cortisol 농도 를 측정하였다. 우울증상의 정도를 Hamilton Rating Scale for Depression을 이용하여 평가하고, Schedule of Recent Experience를 이용하여 최근 일년간의 스트레스 생활사건의 양을 측정하여 비교하였다. 결과는 다음과 같다. 결과: 1) 기능성 위장관 장애 환자들의 혈청 IL-$1\beta$, IL-2 농도는 대조군에 비해 유의하게 낮았으나 혈청 IL-6 농도는 차이가 없었다. 2) 기능성 위장관 장애 환자군의 혈청 cortisol 농도는 대조군에 비해 유의하게 높았고, 우울증상도 유의하게 더 심하였으며, 스트레스 생활사건의 양 또한 유의하게 더 많았다. 3) 기능성 위장관 장애에서 우울증상의 정도와 혈청 cortisol 간에는 유의한 정상관관계가 있었다. 정도는 IL-$1\beta$와 역상관하는 경향이 있고, IL-2와는 정상관하였다. 스트레스 생활사건의 양파 혈청 cortisol 농도는 IL-6 와 유의하게 정상관하였다 대조군을 포함한 전 집단에서 IL-$1\beta$는 우울 정도, 스트레스 생활사건의 양, 혈청 cortisol 등과 유의하게 역 상관하였다. IL-6와 의하게 정상관하였다. 결론: 기능성 위장관 장애는 우울증 및 스트레스와 연관되며, 우울증, 스트레스가 심할수록 혈청 cortisol이 높았다. 기능성 위장관 장애 환자들에서 IL-$1\beta$, IL-2가 감소되어 있었지만, IL-6는 변화가 없었다. 상대적으로 증가된 혈청 cortisol이 IL의 변화와 관련될 수 있다고 생각된다.

Baicalein이 Lipopolysaccharide에 의한 생쥐의 Neuroinflammation에 미치는 영향 (Effects of Baicalein on Neuroinflammation in Lipopolysaccharide-treated Mice)

  • Objects : Baicalein is a major bioactive flavonoid component of Scutellaria baicalensis Georgi that shows a wide range of biological activities, including neuroprotections and anti-inflammatory actions. Hence it is a potential therapeutic material for the treatment of neuroinflammation. In this study, we investigated the modulatory effect of baicalein on neuroinflammation. Method : Pro-inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 mRNA), COX-2 mRNA expression and microglial activation in the brain tissue is induced by systemic lipopolysaccharide (LPS) treatment in C57BL/6 mice. Baicalein was treated orally with 10, 20, and 30 mg/kg 1 hour prior to the LPS (3 mg/kg, i.p.) injection. TNF-${\alpha}$, IL-$1{\beta}$, IL-6 and COX-2 mRNA expression in the brain tissue was measured by the quantitative real-time polymerase chain reaction(PCR) method. Iba1 expression in the brain was measured by western blotting method. Microglia was observed with immunohistochemistry. Results : Baicalein 30 mg/kg significantly attenuated the expression of TNF-${\alpha}$, IL-$1{\beta}$, IL-6 and COX-2 mRNA in the brain tissue. Baicalein 20 mg/kg significantly attenuated the expression of IL-6 mRNA in the brain tissue. Baicalein 30 mg/kg significantly attenuated the expression of Iba1 protein expression in the brain tissue. Baicalein 30 mg/kg significantly decreased the number and cell size of microglia in the cerebral cortex and hypothalamic region and the area percentage of Iba1-expressed microglia in the hippocampus. Conclusion : These results demonstrated that baicalein attenuates LPS induced neuroinflammation in the mice via reduction of pro-inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6), COX-2 mRNA expression and microglial activation.

Lipopolysaccharide로 유발된 마우스 대식세포의 염증매개성 Cytokine 생성증가에 대한 참당귀 물추출물의 효능 연구 (Anti-inflammatory Effect of Angelicae Gigantis Radix Water Extract on LPS-stimulated Mouse Macrophages)

  • Objectives : The purpose of this study was to investigate the effects of Angelicae Gigantis Radix Water Extract(AG) on the production of proinflammatory mediators in RAW 264.7 cells stimulated with lipopolysaccharide(LPS). Method : RAW 264.7 cells were cotreated with AG(50 and 100 ug/mL) and lipopolysaccharide(LPS; 1 ug/mL) for 24 hours. After 24 hour treatment, using Bead-based multiplex cytokine assay, concentrations of various cytokines such as interleukin(IL)-6, IL-$1{\beta}$, IL-10, tumor necrosis factor-alpha(TNF-${\alpha}$), granulocyte colony-stimulating factor(G-CSF), granulocyte macrophage colony-stimulating factor(GM-CSF), interferon inducible protein-10(IP-10), leukemia inhibitory factor(LIF), lipopolysaccharide-induced chemokine(LIX), monocyte chemoattractant protein-1(MCP-1), macrophage colony-stimulating factor(M-CSF), macrophage inflammatory protein(MIP)-$1{\alpha}$, MIP-$1{\beta}$, MIP-2, Regulated on Activation, Normal T cell Expressed and Secreted(RANTES) and vascular endothelial growth factor(VEGF) were measured. Result : AG significantly inhibited LPS-induced production of TNF-${\alpha}$, MIP-$1{\alpha}$, G-CSF, RANTES, IL-10, and M-CSF from LPS-stimulated RAW 264.7 cells at the concentrations of 50 and 100 ug/mL. AG significantly inhibited LPS-induced production of MIP-$1{\beta}$, MIP-2, GM-CSF, and IL-6 from LPS-stimulated RAW 264.7 cells at the concentrations of 50 ug/mL. AG significantly inhibited LPS-induced production of VEGF from LPS-stimulated RAW 264.7 cells at the concentrations of 100 ug/mL. But AG did not show any significant effect on the production of MCP-1, LIF, LIX, IP-10 and IL-$1{\beta}$ from LPS-induced RAW 264.7 cells. Conclusion : These results suggest that AG has anti-inflammatory effect related with its inhibition of proinflammatory mediators such as TNF-${\alpha}$, MIP-$1{\alpha}$, G-CSF, RANTES, IL-10, MIP-$1{\beta}$, MIP-2, GM-CSF, IL-6, VEGF and M-CSF in LPS-induced macrophages.

Expression of TNF-${\alpha}$ and IL-$1{\beta}$ in Splenic Dendritic Cells and Their Serum Levels in Mouse Sparganosis

  • Sparganosis is a tissue invading helminthiasis infecting intermediate hosts, including humans. Strong immune responses are expected to occur in early phases of infection. Thus, we investigated cytokine expressions in splenic dendritic cells and in sera after experimental infection of mice. In splenic dendritic cells, TNF-${\alpha}$ and IL-$1{\beta}$ expression peaked at week 1 and week 3 post -infection (PI), respectively, and also early phase (week 2 PI) depressed cytokine expression was noticed. Serum IL-$1{\beta}$ concentration increased significantly at week 2 PI and peaked at week 6 PI, and that of TNF-${\alpha}$ peaked at week 6 PI. These results showed that pro-inflammatory cytokines, TNF-${\alpha}$ and IL-$1{\beta}$, are chronologically regulated in mouse sparganosis.

Cytokine expression and localization during the development of glomerulosclerosis in FGS mice

  • To elucidate the mechanism of age-related development in FGS/NgaKIST mice with spontaneous glomerulosclerotic lesion, we examined expression and localization of various cytokine mRNA in the kidney in the progression of diseases. This mouse model is the first to develop spontanously occuring glomerosclerotic lesion in the kidney. In this study, we detected the up-regulation of local cytokine genes such as IL-1$\beta$, IL-2, IL-6, IL-10, TNF-$\alpha$, TGF-$\beta$, and IFN- $\gamma$ in the kidneys. In RT-PCR and Southern blot analysis, we detected gradual expressions of cytokine mRNA of IL-1$\beta$, IL-2, IL-6, IFN- $\gamma$, and TNF $\alpha$ mRNA during the course of disease. Other cytokines including IL -10 and TGF -$\beta$ were found to be appeared the slightly expressed level at 3 to 12 weeks before onset of inflammatory lesion but they are highly expressed at the end-stage of the disease accompaning high proteinurea and wasting. In situ RT-PCR, each cytokine mRNA were specifically localized in a variety of cells including mesangial, endothelial, parietal epithelial, tubular epithelial, arterial muscle cell, and infiltrated inflammatory cells. In addition, TNF - $\alpha$was detected moderately in the visceral and parietal epithelial cell, but weakly in endothelial and mesangial cells, whereas IL-1 $\beta$ and IL -6 were strong in mesangial regions. IL-6 and TNF- $\alpha$ was highly localized in the damaged proximal and collecting tubules. Especially, TGF -$\beta$ mRNA was highly found in mesangial cells within glomerulus and interstitium during the end-stage of this disease.. These results indicate that pro inflammatory cytokines such as IL-1 $\beta$, IL-2, IL-6, and TNF- $\alpha$ were gradually expressed from the early stage of this disease to the end-stage, and that IL-10 and TGF-$\beta$ may be important in the accumulation of extracellular matrix(ECM) within glomerulus and periglomerular fibrosis in the progression of this disease as well as tissue destruction in end-stage of this disease.

트레드밀 운동이 뼈관절염 흰쥐의 척수 내 IL-1β 발현에 미치는 영향 (The Effect of Treadmill Exercise on the Expression of IL-1β in the Spinal Cord in Osteoarthritis Rats)

  • PURPOSE: This study examined the effects of treadmill exercise of diverse intensities on the expression of IL-$1{\beta}$ (interleukine-$1{\beta}$) in the spinal cord in osteoarthritis rats. METHODS: The authors applied treadmill exercise of diverse intensity for 4 weeks to Sprague-Dawley rats to which intra-articular injection of monosodium iodoacetate(MIA, $3mg/50{\mu}l$, diluted in saline) was applied in the right knee joint to induce osteoarthritis. The four-week exercise was not applied to the control group(CG, n=15), while exercise of applicable intensity was applied to the low-intensity exercise group(LEG, n=15), moderate-intensity exercise group (MEG, n=15), and high-intensity exercise group(HEG, n=15) for four weeks. Observations were made of expression of IL-$1{\beta}$ in the spinal cord in osteoarthritis rats using western blot analysis. RESULTS: There were significant differences(p<.05) in the comparison of expression of IL-$1{\beta}$ in the spinal cord between the four groups involved. And the LEG and MEG had reduced expression of IL-$1{\beta}$ significantly than the CG(p<.05); in particular, the MEG showed the lowest expression. On the other hand, the HEG had more elevated expression of IL-$1{\beta}$ significantly than the CG(p<.05). CONCLUSION: As a result, factors that induce neuropathic pain such as IL-$1{\beta}$ are reduced; thus, the recovery of damaged neurons is improved and neuropathic pain is reduced. Further, when prescribing exercise to treat osteoarthritis patients, exercise of moderate intensity suitable for patients' physical conditions, rather than high intensity, maximizes the effects of this therapy.

Participation of nitric oxide pathways in interleukin 1$\beta$-induced mechanical allodynia in the orofacial area of rats

  • The purpose of the present study was to examine the role of peripheral nitric oxide (NO) pathways in the onset of interleukin (IL)-1$\beta$-induced mechanical allodynia in the orofacial area. Experiments were carried out on male Sprague-Dawley rats weighing 230-280 gm and surgical procedures were performed under pentobarbital sodium (40 mg/kg, i.p.). Under anesthesia, a polyethylene tube (PE10) was implanted into the subcutaneous area of one vibrissa pad, which enabled the injection of IL-1$\beta$ or other chemicals. We subcutaneously injected 50 ${\mu}L$ of IL-1$\beta$ into a vibrissa pad through the implanted polyethylene tube with a 100 ${\mu}L$ Hamilton syringe. After the administration of 0.01, 0.1, 1, or 10 pg of IL-1$\beta$, withdrawal behavioral responses were examined. The subcutaneous injection of saline had no effects on the air-puff thresholds. Following the subcutaneous injection of 0.01, 0.1, 1, or 10 pg of IL-1$\beta$, the threshold of air puffs decreased significantly to 12 $\pm$ 3, 7 $\pm$ 2, 5 $\pm$ 1, or 5 $\pm$ 1 psi, respectively, in a dose dependent manner. Pretreatment with L-NAME, a nitric oxide synthase (NOS) inhibitor, blocked IL-1$\beta$-induced mechanical allodynia. However, neither D-NAME, an inactive isomer of L-NAME, nor vehicle affected the IL-1$\beta$-induced mechanical allodynia. Subcutaneous injection of IL-1$\beta$ increased the number of c-fos-like immunoreactive neurons, whereas pretreatment with L-NAME decreased this number, in the trigeminal caudal nucleus. These results suggest that pro-inflammatory cytokines and NO are important contributors to the pathogenesis of persistent and exaggerated IL-1$\beta$-induced pain states. Based on these observations, peripheral application of NOS inhibitors may be of therapeutic value in treating pain disorders in the clinic.

도토리 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과 (Effects of Water Extract Acorn on Mouse Immune Cell Activation Ex Vivo)

  • Acorns have been used as a traditional remed as well as food source. However, few studies on their immunomodulating effects have been reported. In this study, the combined immunomodulative effect of a water extract of acorns was tested on seven to eight weeks old mice(balb/c). The mice were fed ad libitum on a chow diet, and a water extract of the plant mixture was orally administered every other day for four weeks at two different concentrations(50 and 500 mg/kg B.W.). The production of cytokine(IL-$1{\beta}$, IL-6, IL-2, IL-10, IFN-$\gamma$), secreted by macrophages stimulated with LPS or not, detected by ELISA assay using cytokine kit. After 48 h of incubation with mitogen(ConA or LPS) ex vivo study showed that cytokine (IL-$1{\beta}$, IL-6, IL-2, IL-10, IFN-$\gamma$) was detected in both of the 50 and 500 mg/kg B.W. supplementation groups with LPS stimulation. The results of this study may suggest that supplementation with acorn water extract increase immune function by regulating cytokine production capacity by activated macrophages.

Anti-inflammatory Effects of Allium victorialis Extract in Lipopolysaccharide Exposed Rats and Raw 264.7 Cells

  • This study examined the inflammatory reaction effects of Allium victorialis var. platyphyllum in vivo at the time of a lipopolysaccharide (LPS) shock in rats, and in vitro in cultured Raw 264.7 cells, with the aim of facilitating the development of a new anti-inflammatory medicine. Plasma concentrations of interleukin (IL)-$1{\beta}$, IL-6, tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$), and IL-10 in rats peaked 5 h after LPS treatment in all experimental groups, with those of IL-$1{\beta}$, IL-6, and TNF-${\alpha}$ being significantly lower in all animals treated with A. victorialis than in the control group at that time point. Conversely, the plasma concentration of IL-10 was higher in the rats treated with 300 mg/kg A. victorialis extract than in the control group at both 2 and 5 h after LPS treatment. Concentrations of IL-$1{\beta}$ and IL-6 in the liver of rats treated with A. victorialis extract were significantly lower than those of the saline-treated control group. However, the liver concentrations of TNF-${\alpha}$ and IL-10 did not vary significantly between the four animal groups. Similarly, concentrations of IL-$1{\beta}$, IL-6, and TNF-${\alpha}$ obtained from cultured Raw 264.7 macrophages were lower in all of the A.-victorialis-extract-treated groups than in the control group. Although the concentration of IL-10 in the A.-victorialis-extract-treated groups tended to be greater than in the control group, the differences between groups were not statistically significant. Together the findings of this study suggest that A. victorialis var. platyphyllum contains functional substances that are involved in inflammatory reactions.

A Study on Bone Formation & Osteoporosis by Taeyoungion-Jahage Extracts

  • 생쥐의 calvarial osteoblast세포를 분리배양하여 gelatinase생성여부를 골흡수과정에서의 역할을 규명하기 위하여 SDS-PAGE-zymography분석을 한 결과 progelatinase-A를 항속적으로 합성하고 있음을 확인하였다. 생쥐의 osteoblasts를 골재흡수 약물인 PTH, $1,25(OH)_2D_3$, 단핵구배양액 (MCM) 그리고 IL-1으로 자극시키면 gelatinase생산을 촉진하여 콜라겐분해가 증가되었으나, indomethacin과 dexamethasone은 생쥐의 osteoblastic세포의 collagenolysis를 저해하였다. 한편, 골재흡수에 IL-1을 생쥐태아 유래의 장골조직배양 (fetal mouse long bone organ culture)에 처리하자 IL-1 은 골재흡수를 촉진하였다. 더우기, $IL-1{\alpha}$의 농도의존성에 대한 indomethacin과 dexametasone의 영향을 검토한 결과 직선형의 비례커브로 영향을 미쳤다. 이러한 골대사의 지견을 바탕으로 대영전-자하거의 열수추출물의 시험관내 독성검사에서 $1-200\;{\mu}g/ml$의 농도에서는 독성이 없었으며, 또한, $300\;{\mu}g/ml$ 농도에서도 생쥐의 calvarial골에는 독성이 없었다. 대영전-자하거 extract는 PTH (2 units/ml), MCM (5%, v/v), $rhIL-1{\alpha}$ (1 ng/ml) $1,25(OH)_2D_3$ (10 ng/ml)처리에 대해서 그리고 $IL-1{\alpha}$$IL-1{\beta}$-유발 collagenolysis에 대해서도 보호효과가 있었다. 대영전-자하거extract을 1시간동안 전처리와 후처리에서 콜라겐분해에 약간의 보호활성이 있었으며 $IL-1{\alpha}$$IL-1{\beta}$에 의해 유발되는 콜라겐분해에 보호활성이 보였다. 1시간동안 전처리는 콜라겐분해를 감소시키며, 대영전-자하거 extract는 gelatinase효소를 저해하였으며 PTH, $1,25(OH)_2D_3$, $IL-1{\beta}$$IL-1{\alpha}$로 유발된 효소활성화가 저해되었다. 즉, 대영전-자하거 extracts는 $IL-1{\alpha}-$$IL-1{\beta}$에 의해 촉진되는 골재흡수에 효과적이었으며, 비스테로이드성 항염증제제 (indomethacin 과 dexamethasone)에 의한 골재흡수방지 효과와 유사하였다. 이러한 결과는 대영전-자하거extract가 골다공증치료에 효과적임을 나타내는 것이다.
