Previously, we identified that the overexpression of IEX-1 induces apoptosis in ovarian cancer cells. Herein we report a new binding partner of IEX-1, CATHEPSIN B, as a lysosomal enzyme which contributes to the various apoptotic signaling in tumor cells. To investigate how IEX-1 regulates cellular survival and death event, we performed yeast two-hybrid screening of rat ovarian cDNA library using IEX-1 as the bait and found CATHEPSIN B. In the present study, CATHEPSIN B and IEX-1 proteins were overexpressed in 293T cells and their specific association was determined by immunoprecipitation and immunoblot analysis. In addition, the endogenous interaction between CATHEPSIN B and IEX-1 was confirmed in HeLa cells. The current finding of lysosomal CATHEPSIN B as the IEX-1-binding partner implies that IEX-1 may involve in lysosome-mediated apoptotic signaling pathways.
Apoptosis is a crucial mechanism for the proper regulation of homeostasis. BCL-2 family proteins are key molecules which control cellular survival and apoptosis. MCL-1 (myeloid cell leukemia-1) is a pro-survival member of BCL-2 family that promotes the survival of cells, and is highly expressed in diverse cancers including ovarian cancer, leukemia, and cervical cancer. Previously we identified IEX-1 (immediate early response gene X-1) as a binding partner of MCL-1. In the present study, we demonstrated that overexpression of IEX-1 induced apoptosis of ovarian cancer cells. Moreover, IEX-1 significantly attenuated the pro-survival function of MCL-1 in these cells. Also, IEX-1-induced cell death activity was able to be modulated by changes in the expression level of MCL-1. Thus, these results suggest that both IEX-1 and MCL-1 modulate each other's function controlling cellular survival and death and the inhibitory activity of IEX-1 toward MCL-1 may be applied for the development of chemotherapeutics.
[Purpose] The purpose of this study was to compare the excess post-exercise oxygen consumption (EPOC) between different types of exercises in women with normal weight obesity (NWO). [Methods] Nine university students with NWO having body mass index <25 kg/m2 and body fat percentage >30% participated in the study. First, continuous exercise (CEx) on an ergometer for 30 minutes at 60% of maximal oxygen consumption (VO2max) and interval exercise (IEx) at 80% VO2max for 2 minutes were performed. This was followed by exercise performed at 40% VO2max for 1 minute and at 80% VO2max for 3 minutes, performed 6 times repeatedly for a total of 26 minutes. The accumulation of short duration exercise (AEx) was performed for 3-bouts of 10 minutes each at 60% VO2max. [Results] The major findings were as follows: energy consumption during the exercises showed no significant difference between CEx, IEx, and AEx; EPOC was higher in IEx and AEx as compared to CEx for all dependent variables (e.g. total oxygen consumption, total calorie, summation of heart rate, and EPOC duration); and the lipid profile showed no significant difference. [Conclusions] Our study confirmed that when homogenizing the energy expenditure for various exercises in NWO individuals, EPOC was higher in IEx and AEx than in CEx. Therefore, IEx and AEx can be considered as effective exercise methods for increasing energy expenditure in NWO females.
The objective of this study was to compare the effects of a combined treatment consisting of an isoflavone diet and aerobic exercise on the Caspase 9 levels, and Bcl-2 levels during menopause, using 30 S.D ovariectomized rats as a model, over 12 weeks. The ovariectomized rats were divided into 4 experiment groups, with 8 rats in each group, as follows: (1) General diet group (GD, n=8), (2) Isoflavone diet group (ID, n=8), (3) General diet + Exercise group (GEX, n=8), (4) Isoflavone diet + Exercise group (IEX, n=8). The findings of this study were as follows: 1. Combined treatment consisting of an isoflavone diet and regular exercise resulted in a significant reduction in the expression of caspase-9 proteins (p<0.05), and a significant increase in the expression of Bcl-2 proteins (P<0.001). In addition, apoptotic cells were more decreased (p<0.001) in GEX and IEX group. This also suggests that cardiovascular disease in postmenopausal women might be prevented or inhibited to exert positive apoptotic inhibitory effects by increasing Bcl-2 protein expression within aortic tissues during vascular movement and decrease caspase-9 protein.
[Purpose] This pilot study aimed to develop a regression model to estimate the excess post-exercise oxygen consumption (EPOC) of Korean adults using various easy-to-measure dependent variables. [Methods] The EPOC and dependent variables for its estimation (e.g., sex, age, height, weight, body mass index, fat-free mass [FFM], fat mass, % body fat, and heart rate_sum [HR_sum]) were measured in 75 healthy adults (31 males, 44 females). Statistical analysis was performed to develop an EPOC estimation regression model using the stepwise regression method. [Results] We confirmed that FFM and HR_sum were important variables in the EPOC regression models of various exercise types. The explanatory power and standard errors of estimates (SEE) for EPOC of each exercise type were as follows: the continuous exercise (CEx) regression model was 86.3% (R2) and 85.9% (adjusted R2), and the mean SEE was 11.73 kcal, interval exercise (IEx) regression model was 83.1% (R2) and 82.6% (adjusted R2), while the mean SEE was 13.68 kcal, and the accumulation of short-duration exercise (AEx) regression models was 91.3% (R2) and 91.0% (adjusted R2), while the mean SEE was 27.71 kcal. There was no significant difference between the measured EPOC using a metabolic gas analyzer and the predicted EPOC for each exercise type. [Conclusion] This pilot study developed a regression model to estimate EPOC in healthy Korean adults. The regression model was as follows: CEx = -37.128 + 1.003 × (FFM) + 0.016 × (HR_sum), IEx = -49.265 + 1.442 × (FFM) + 0.013 × (HR_sum), and AEx = -100.942 + 2.209 × (FFM) + 0.020 × (HR_sum).

Gas-Phase reactions of methyl and ethyl nitrites with anionic nucleophiles of SH-, F- and OH- are investigated theoretically at the MP2/6-311+G* level. The SN2 processes are all highly exothermic and proceed with a typ-icaI double-weIl reaction coordinate profile. The elimination reactions of methyl nitrite with SH- and F- are double-well energy surface processes,with stabilizedproduct complexes of NO-...H2S and NO-...HF, pro-ceeding by an E1 cb-like E2 mechanism. The $\beta-elimination$ of ethyl nitrite is an E2 type process. The $\alpha-elimi-nation$ reactions of methyl and ethyl nitrites with OH- have triple-well energy profiles of Elcb pathway with an $\alpha-carbanion$ intermediate which is stabilized bythe vicinal $nc\alpha-{\sigma}*o-N$ charge transfer interactions. CompIex-ation ofmethyl carbanion with HF seems to provide a stable intermediate within a triple-well energy profile of El cb channel in the reaction of F- with methyl nitrite.
Purpose: This study was to identify the persuasive effects of exercise promotion advertisements for obesity prevention according to the source types( ordinary people, experts, celebrity endorsers) and advertising message types(slices of life, testimonials). Method: Gender, height, body weight, BMI, attitude toward obesity(Aob) and exercise(Aex), and intention to exercise(Iex) were collected from 626 elementary school students in a pretest. After 2 months, six advertisements type attached to a questionnaire were provided for 20 minutes and then Aob, Aex, lex, source credibility and attitude toward advertisements(Aad) were collected in posttest. Result: 1) In posttest the lex of 6 the groups increased significantly compared with that of the pretest, 2) Source credibility of the Ordinary+Testimonial group was lower than the Celebrity+Testimonial, Celebrity+Slice of life, Ordinary+Slice of life, and Expert+Testimonial groups, Aad of the Celebrity+Testimonial group was higher than the Ordinary+ Testimonial group. 3) The Main effect and interaction effect of source types and advertising message types were significant in source credibility and Aad, Conclusion: Persuasive effects of exercise promotion advertisements in elementary school students was found to be the most effective in Celebrity+Testimonial. This study suggests that selection of health education advertisements according to demographic characteristics is important to promote persuasive effects.
