A Study on the Wild Exochorda serratifolia for Landscape Horticuitural Cultivation (야생 가침박탈(Exochorda serratifolia)의 조경원예화에 관한 연구)

  • Exochorda serratifolia, broad-leaved shrub has beautiful flowers and is resistant to cold, shade and decease. New this native plant is considered to be worth being exploited as the outstanding plant for landscaping and horticulture. So this study was executed to utilize Exochorda serratifolia as the planting material for landscaping and horticulture through the survey of its habitat environment and the experimennt of its seed physiology and germination, vegetative propagation, culture and utilization, etc.. The results are as follows ; 1. The color of the flower is white, blooming in the early and middle of May and the seeds ripen late in Sep.. 2. The elevation, elevation, gradient and direction of the native habitat were 250m, 20-25$^{\circ}$and northern side respectively. 3. The soil pH of the natural habitat was 5.3 and soil fertility was poor. 4. Exochorda serratifolia appeared as indicator within P. densiflora community and its neighboring species were Q. mongolica, Q. dentata, R. mucronulatum, L. obtusiloba, E. oxyphillus, C. heterolphylla, var thunbergii, etc.. 5. The optimum temperature for seed germination was found at 20$^{\circ}C$ and the longer the stratification period at 5$^{\circ}C$ was, the lower the germination rate was. 6. The treat merits of GA and Kinetin increased the seed germination rate, especially under the dark condition but their high concentrations decreased the seed germination rate. 7. The rooted rate of the greenwood cutting was the highest at IBA 100ppm plot of vermiculite bed but its high concentration decreased the rooted rate conspicuously. 8. In the tissue culture, the each 1.0 ppd plot of NAA, Kinetin and NAA 1.0ppm + Kinetin showed the best growth. And the mixture of NAA 1.0ppm and Kinetin showed better growth than the single treatment of NAA or Kinetin did. 9. Transplanted Exochorda serratifolia showed healthy growth with shaded environmental condition(42.1% light intensity), therefore they can be cultivated as a shade tolerant landscape plant. 10. It was considered that Exochorda serratifolia was applicable to group planting at shade places or under trees in parks, homes, etc..

ANTIOXIDATIVE ACTIVITIES OF SOME DIETARY FIBERS DETERMINED BY AN NIR EMISSION SPECTROSCOPY

  • Constituents of several .representative seaweeds, such as wakame Undaria pinnatifida; hijikia Hizikia fusifome; and kombu Laminaria japonica, were found to have fairly large reaction rates determined by quenching experiments of emission spectra in the near-infrared region (1max: 1270 nm) from singlet oxygen (102). Emission spectra of singlet oxygen generated from an aqueous solution of Rose Bengal under irradiation with a green laser (330 nm) were measured by a near-infrared (NIR) emission spectrometer constructed in our laboratory. The quenching experiments were as follows: Intensities of emission spectra were measured in the absence (I0) and in the presence of the seaweed constituents (I): Ratios of I0/I were plotted against every concentration of the quenchers (Stern-Volmer plots) which gives a straight line. The slope of each line gives a kqt value which gives a quenching constant kq value (an antioxidative constant against singlet oxygen) when the t value (half-life time of singlet oxygen in the solvent used) was given. The determined reaction rates are between 103-105 (g/l)-ls-1; the larger ones are as large as that of ascorbic acid, 8.4 ${\times}$ 104 (g/1)-ls-1. Most of these seaweed constituents also showed antioxidative activity against auto-oxidation and superoxide as well as their immunological enhancing activity. These results suggest a possibility that dietary fibers which are supposed to prevent the large-intestine cancer by their physical properties may prevent the cancer, at least in parts, by their chemical, antioxidative activity.

Study on Antioxidative Activity of Plant Extracts in Fish Oil (식물 추출물이 어유의 산화 안정성에 미치는 영향)

  • Extract from forty four species of natural plants in Korea were obtained. Each extinct (1,000 ppm) was added into fish oil and their antioxidative activities were screened by rancimat, From the results, seven species including Brassica juncea, Solanum tuberosum, Capsella bursa, Ulums davidiana, Broussonetia kazinoki, Oenanthe javanica, and Rhus chinensis were selected due to their relatively prolonged induction periods. When the concentration (100 - 1,000 ppm) of the selected extract was considered, the most prolonged induction period was observed from Solanum tuberosum throughout the studied concentrations, suggesting that its extinct showed the strongest antioxidative activity against fish oil. In addition, peroxide value (POV) and thiobarbituric acid reactive substance (TBARS) value were considered at $60^{\circ}C$ for 5 days. Among the selected extract, extract from Solanum tuberosum showed the lout POV while the extract from Brassica juncea var showed the least TBARS values after 5 days. Also, the free radical scavenging effect against DPPH radical were studied with the selected extract, showing that the extract from Rhus chinensis was found to be the most effective.

Ginsenosides Production through in vitro Culture of Adventitous Roots Induced from Panax ginseng "Chunpoong" (인삼 천풍의 부정근 배양을 통한 Ginsenosides 생산)

  • We have investigated the optimal conditions for the growth and ginsenoside production in adventitious root of “Chunpoong” (Panax ginseng C.A. Meyer). Ginseng adventitous roots induced from the embryo of “Chunpoong” were cultured in various plant media supplemented with several growth hormones. The best growth rate and high ginsenoside contents were obtained in B5 medium supplemented with IBA (2 mg/L) and kinetin (0.1 mg/L). The supplement of 2.5 mM KH$_2$PO$_4$ was good for high growth rate of the adventitious roots, but the accumulation of ginsenosides was increased by reducing the KH$_2$PO$_4$ concentration to 1.25 mM. We have established the effective liquid culture system for the optimal growth and ginsenoside production of “Chunpoong” adventitious roots.

Callus formation and multiple shoot induction of Hovenia dulcis Thunb. (헛개나무의 캘러스 형성 및 multiple shoot 유기)

  • Loaves, stems, cotyledons, and roots of Hovenia dulcis Thunb grown in test tube were cultured on media containing different concentrations of single or combined growth regulators. In MS media containing 2mg/ι BA, the shoot formation rate was 95.5% and it was the highest frequency of shoot formation. MS media showed most efficiency in the shoot formation at 0.01mg/ι TDZ for the callus formation, but the color of callus changed to brown at a higher concentration of TDZ. Callus formation was 89.% at 0.5mg/ 2.4-D, but IAA, IBA, and NAA were not effective on the formation of callus. Calli were formed only on wound area when IAA, IBA, and NAA were added into MS media. Combined growth regulators (BA + auxin) were more effective in roots and nodes than leaves and cotyledons on the formation of shoot. More than 97% of shoot formation was obtained on MS media containing BA and auxin. For the production of multiple shoot, nodes of Hovenia dulcis were used and effect of growth regulators on the formation of multiple shoot was evaluated on MS media. Highest shoots (5.3) of Hovenia dulcis were induced on MS media supplied with 0.1mg/ι BA and 0.1mg/ι NAA, and an average of 6.4 shoots per explant were obtained in 1/2 MS media containing same concentration and growth regulators. An average of 7 shoots per explant after 4 weeks of culture from nodes of Hovenia dulcis was produced on a woody plant medium(WPM) containing 0.1mg/ι BA and 0.1mg/ι NAA. Shoot length was 6.0 cm in average.

Shoot-tip culture for massive production of radish foundation seeds (무 원종 대량생산을 위한 경정배양)

  • Shoot-tip culture was used to produce clonal plants of radish stock seeds. Using 6-benzyladenine (BA), the largest number of RA2 line multi-shoots were formed with an average of 14.67 shoots on 1.33 µM BA in early seedlings and 11.33 shoots on 1.78 µM BA in juvenile seedlings. The largest number of RA4 line multi-shoots were formed with an average of 11.67 shoots on 2.22 µM BA in early seedlings and 13.67 shoots on 1.33 µM BA in juvenile seedlings. There was little difference in the significance level by BA concentration in both lines. Using Thidiazuron (TDZ), the number of RA2 line multi-shoots increased with increasing TDZ concentration, forming the largest number of multi-shoots in 0.45 µM TDZ (7.0 and 3.0 multi-shoots for early and juvenile seedlings, respectively), but few multi-shoots were formed from TDZ 2.25 and 4.5 µM. RA4 line produced almost no multi-shoots in early seedlings, and 3.7 multi-shoots were produced in 0.23 and 0.45 µM TDZ in juvenile seedlings, but not at higher concentrations. Analysis of the tissue culture seedlings grown by cultivating the generated multi-shoots with Radish Foundation seeds using SSR marker revealed a weak pattern of mutation in the generated tissue culture seedlings, but there was no mutant. In addition, in terms of root roots, both RA2 and RA4 lines generally had the best rooting, number of roots, and degree of root development in 4.9 µM indol-3- butyric acid (IBA).

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants.   • aA series of experiments were conducted to investigate the effect of plant growth regulator treatments on the growth and lateral root formation in soybean sprouts in order to establish the effective method of producing root-less or short-rooted soybean sprouts with larger diameter in the hypocotyl. Major results can be summarized as follows. 1. Soybean sprouts showed fairly uniform elongation rate from 3 to g days after imbibition with daily increase of 3.8cm. The speed of elongation of hypocotyl was reduced whereas that of root accelerated 7 days after imbibition. Lateral roots began to emerge fairly evenly from 5 to 9 days after imbibition with a daily increase of 4.4. 2. Auxins(IAA, IBA, NAA, 2,4-D) inhibited hypocotyl elongation and formation of lateral roots and increased hypocotyl diameter without influencing root length and hook diameter at higher concentrations. The dry weight of cotyledon was increased significantly as compared to that of hypocotyl and root. Among the tested auxins, 2, 4-D was the most effective. 3. BA and 4PU-30 significantly reduced elongation of hypocotyl and root and resulted in the biggest diameter of hypocotyl when treated at higher concentrations. The lowest effective concentration of BA to prevent the formation of larval gal roots was 12.5ppm. The formation of lateral roots could be completely prevented by BA and 4PU-30 treatment but kinetin, zeatin, zeatin riboside resulted in many lateral roots and increased thickness of soybean sprouts with little influence. Cotyledon deformation was found in soybean sprouts treated by 4PU-30. 4. 2, 4-D was the most effective for increasing the hypocotyl diameter while 4PU-30 was the most effective for reducing no. of lateral roots. 5. It can be concluded that among the plant growth regulators tested, BA was effective in reducing root length and increasing hypocotyl diameter. BA 12.5 ppm or 15 ppm may thus be the more practical for production of soybean sprouts. 6. ABA showed no significant effect of growth parameter, however ABA 25 ppm inhibited only no of lateral roots with little influence on the growth of seedling. 7. Ethephon inhibited the elongation of hypocotyl and root and increased hypocotyl diameter at higher concentrations. 8. The combined effect of cytokinins and ethephon was very similar to result of BA treatment alone. As the ethephon concentration increased, hypocotyl diameter and dry weight of cotyledon tended to increase.

  • Thuja koraiensis Nak. is a short and creeping evergreen shrub which reaches about 3 m in height and only occurs in the northeast China and in high mountains over the Korea. It's designated as a rare and endangered tree species in Korea and DD (data deficient) in Red List Category & Criteria of IUCN. This study was carried out to develop the propagation technique by cutting for conservation of genetic resources of T. koraiensis. The rooting responses of branch cuttings, obtained from hard (May) and semi-hard wood shots (August) to three plant growth regulators (PGRs), namely, IAA, IBA, and NAA applied at various concentrations (0, 100, 500, 1000, 2000, and 3000 mg/l) were examined in sand and mixed soil media. Percentage of rooting showed significant difference between cutting time, among kinds and among concentration of PGRs. The optimum cutting time was April to May in hardwood cutting. The application of IAA 1000 mg/l and NAA 500mg/l were effective in callus formation and rooting of cutting. Relatively, rooting of cutting of the control taken in May was above 93%.
