Effect of Helianthus tuberosus Juice Mixed with Dried Bitter Melon Juice on Hypoglycemic Function in Streptozotocin-Induced Diabetic Rats (당뇨유발 흰쥐에서 돼지감자 여주 혼합즙 음용에 의한 혈당저하 효과)

  • This study was conducted to develop Helianthus tuberosus (HT) juice mixed with dried bitter melon juice and assess its hypoglycemic effect in streptozotocin (STZ)-induced diabetic rats. HT juice mixed with 5.0% dried bitter melon juice was used in this study. Male Sprague-Dawley rats were divided into four groups (eight rats per group) and drunk each sample for 4 weeks: normal water [normal control (NC) group], STZ+normal water (STZ group), STZ+HT juice (HT group), STZ+HT juice mixed with 2.5% bitter melon juice (HT2.5 group), and STZ+HT juice mixed with 5.0% bitter melon juice (HT5.0 group). HT juice was diluted to 25% in distilled water and supplied to rats. Food intake, body weight gain, and food efficiency ratio were lower in the STZ group than in the NC group. HT, HT2.5, and HT5.0 groups showed higher parameters than the STZ groups. Water intakes were higher in the STZ group than in the NC group. After 3 weeks, HT, HT2.5, and HT5.0 groups showed lower parameters than the STZ group. After 1 week, blood glucose level of the STZ group ($476.7{\pm}22.8mg/dL$) was significantly higher than those of the HT group ($376.3{\pm}25.8mg/dL$), HT2.5 group ($405.2{\pm}35.1mg/dL$), and HT5.0 group ($342.8{\pm}29.7mg/dL$). After 4 weeks, blood glucose level of the STZ group were significantly higher than those of the HT, HT2.5, and HT5.0 group. Serum insulin levels of the HT group ($3.13{\pm}0.32ng/mL$), HT2.5 group ($3.40{\pm}0.23ng/mL$), and HT5.0 group ($3.48{\pm}0.43ng/mL$) were higher than that of the STZ group ($2.72{\pm}0.53ng/mL$). These results indicate that H. tuberosus juice mixed with dried bitter melon juice helps prevent or attenuate progression of diabetes in rats with STZ-induced diabetes.

Hypoglycemic Effect of the Giant Embryonic Rice Supplementation on Streptozotocin-induced Diabetic Rats (거대배아미 식이에 의한 Streptozotocin 유발 당뇨 흰쥐의 혈당강하 효과)

  • This study was performed to investigate the influence of giant embryo feeding on hypoglycemia in streptozotocin-induced diabetic rats. Streptozotocin -induced diabetic rats were fed four kinds of experimental diets such as corn starch diet as a control, polished rice diet, brown rice diet and giant embeyonic rice diet for 6 weeks. The body weight gain diabetic of control was signaficantly suppressed compared to that of the normal. The supplementation of giant embryo increased the surivival rate. Feeding of giant embryo appeared to have ameliorating effects on diabetic symptoms including features such as polyphasia, polyuria. Giant embryo shortened gastrointestinal transit time and increased total fecal weight, total fecal dry weight and fecal water content compared with diabetic control. Giant embryo showed fasting blood glucose lowering effect compared with diabetic control. The disaccharidase activities in proximal part of intestine such as maltase sucrase and lactase in giant embryo feeding groups were lower than diabetic control. The results of this study show that giant embryo supplementation may have a beneficial Veffect on the hypoglycemia may be useful in the diet therapy for diabetic.

Anti-oxidative activity of the herb mixture prescribed to induce blood glucose level and effect on the differentiation of 3T3-L1 fibroblast (혈당저하를 위해 처방된 천연물 복합제의 항산화 활성 및 3T3-L1 세포의 분화에 미치는 영향)

  • We prepared five different kinds of herb mixtures prescribed for hypoglycemic effect. And the physicochemical properties of their water extracts were assessed to identify functional materials. Yields were in the range $19.52{\sim}29.79%$. Total phenolics and flavonoid contents were $349.24{\sim}1,752.21\;mg%$ and $163.06{\sim}1,118.47\;mg%$, respectively, and herb mixtures No. 2, 3 and 5 showed particularly high levels greater than 1,000 mg%. Electron-donating ability was best in herb mixtures showing high levels of total phenolics and flavonoids. Nitrite-scavenging abilities were more than 70% in herb mixtures No. 2 and 5, and decreased as pH increased. Herb mixture extracts strongly inhibited differentiation of 3T3-L1 fibroblasts, with potencies ranked in the herb mixture order 5, 1, 4, 3, and 2. The five different kinds of herb mixtures prescribed for their hypoglycemic effects may be useful as functional food materials.

Study on Cosmeceutical Activities from Fermented Ginseng Berry Extracts (진생베리 발효추출물의 화장품 약리활성)

  • Ginseng berry contains a large amount of Ginsenoside Re and has anti-inflammatory, anticancer, hypoglycemic and whitening effects. In this study, Rhizopus Oligosporus strain was used to establish ginseng berry fermentation process and cosmetic pharmacological activity of ginseng berry fermented product was analyzed.. The electron donating ability of ginseng berry extract by fermentation shown 81% at 1,000 ㎍/mL concentration. The ABTS+ radical scavenging ability of shown 100.2% at 1,000 ㎍/mL concentration. The tyrosinase inhibitory effect which is related to skin-whitening, was 57% at the concentration of 1,000 ㎍/mL. The elastase inhibitory effect which is related to skin-wrinkle, was 47% at 1,000 ㎍/mL concentration. Also, the collagenase inhibition effect was 33% at 1,000 ㎍/mL concentration. From these results, ginseng berry extracts by fermentation is considered to have anti-inflammatory, anti-wrinkle effect and whitening effect. Therefor, ginseng berry fermented product is expected to be very useful as an anti-inflammatory and anti-aging cosmetic raw material.

Effect of Sea Tangle on Proliferation of Splenocytes from Normal and Diabetic Mice (다시마섭취가 정상과 당뇨쥐의 비장세포 증식에 미치는 영향)

  • To investigate the effect of sea tangle on immune function in normal and diabetic states, 10-week old ICR mice were feed control(C) and sea tangle(5) diets containing 5%(w/w) cellulose and 13.6%(w/w) dry sea tangle for 4 weeks. After 4 weeks, three quarters of mice(CD and SD) were made diabetic by intramuscular injection of streptozotocin(150mg/kg bw). On the 4th day after diabetes was apparent by urinary glucose, one third of diabetic mire(CDG and SDG) were treated with glipizide(20mg/kg bw) and the other third(CDM and SDM) with metformin (500mg/kg bw) orally. Spleen weights of diabetic mice with no hypoglycemic drug treatment appeared to be higher in the sea tangle group(SD) than in control(CD), but were not different when drugs were administered. Data on splenocyte proliferation stimulated by lipopolysaccaride from Salmonella abortus equi(0.l$\mu\textrm{g}$/ml) showed that sea tangle increased mitogen response in normal mice(C group vs S group) and appeared to have the same effect in diabetic mice with or without drug treatment. Splenocyte proliferation induced by concanavalin A(0.1$\mu\textrm{g}$/ml) also showed similar results, although there were not statistically significant. Concentration of interleukin-2(IL-2) released from splenocytes of the S group seemed higher than from the C group, but the IL-2 concentrations were not different among six diabetic groups. Results of fatty acid compositions of splenocyte phospholipids showed that diabetes reduced arachidonic acid/linoleic acid ratios and that sea tangle intake and glipizide treatments increased contents of polyunsaturated fatty acids. It is concluded that dietary sea tangle has a positive effect on splenocyte proliferation under normal condition and could have the same effect under diabetic conditions. The Wound Healing Effect of Hydnocarpi Semen Extract on Ulcer in Diabetic Mice

  • The extract from Hydnocarpi Semen (HS) has been used to treat leprosy and its anti-inflammatory activity has been reported. However, the effect of HS on the treatment of diabetic or peripheral ulcer is not well known. We therefore examined its wound healing effects on ulcer area in diabetic mice. GC and GC/MS analysis with the total extract of HS show that the main constituents of the extract are chaulmoogric acid, hydnocarpic acid, and gorlic acid. Whereas HS showed wound healing effect in diabetic ulcer, there was no hypoglycemic effect in diabetic mice. The treatment of HS extract significantly decreased the level of total WBC and neutrophils in mice compared to control mice. Cutting ulcer was induced by the round-shaped punch on the backside of diabetic mice and the extract of HS was given orally or topically. The wound area score significantly decreased after treatment of HS at dose of 50 mg/kg. The treatment of HS also induced the activation of macrophages and increased the production of IL-12 and TNF-$\alpha$ in macrophages, indicating that the wound healing by HS extract is associated with the inflammatory effect via the activation of macrophages. Our results suggest that HS extract can be a new therapeutic candidate for treatment of diabetic ulcer.

The effect of Ginkgo biloba Extract (GB) on Glucose Uptake in L6 Rat Skeletal Muscle Cells (L6 근육세포에서 은행잎 추출물의 당 흡수효과)

  • Objectives: Evidences suggests that Ginkgo biloba, a widely used traditional medicine, shows a hypoglycemic effect. Thus, we investigatd the effect of G. biloba extract (GB) on glucose uptake in L6 rat skeletal muscle cells. Method : Effect of GB on glucose uptake and phosphatidylinositol (PI) 3-kinase activity were assessed using Glucose uptake assay and PI 3-kinase assay, respectively. Also, AMP-activated protein kinase (AMPK), p38 mitogen activated protein kinase (p38 MAPK) expression were identified by Western blot. Results : Glucose uptake assay revealed that GB increased glucose uptake about 2.5-fold compared to thecontrol. GB stimulated the activity of PI 3-kinase which is a major switch element on the glucose uptake pathway. About a 6.5-fold increase in activity of PI 3-kinase was observed with GB. We then assessed the activity of AMPK, another regulatory molecule on the glucose uptake pathway. The result was that GB increased the phosphorylation level of both AMPK ${\alpha}$l and ${\alpha}$2. The activity of p38 MAPK, a downstream mediator of AMPK, was also increased by CB. Conclusion : These results suggest that GB may stimulate glucose uptake through both PI 3-kinase and AMPK mediated pathways in L6 skeletal muscle cells thereby contributing to glucose homeostasis.

Effect of Garlic Diets on the Changes of Serum Cholesterol Glucose Level and Coagulation Time in Rats (마늘첨가식이(添加食餌)가 흰쥐의 혈청(血淸) 콜레스테롤, 글루코오스의 함량(含量) 및 혈액응고시간(血液凝固時間)에 미치는 영향(影響))

  • Effects of garlic on hypocholesterolemia, anticoagulation and hypoglycemia were studied in the present experiments using male rats. The results were summarized as follows. 1. The supplementation of $2{\sim}4%$ garlic to 2% cholesterol diets did not affect food intake and weight gain in male rats. 2. Rat's groups fed the diets supplemented with $2{\sim}3%$ garlic (C.D.E.F) to 2% cholesterol diet (B) decreased serum total cholesterol levels by $16{\sim}32%$, triglyceride levels by $18.6{\sim}26.8%$ and ${\beta}/{\alpha}-lipoportein$ rations by $42{\sim}58%$, but increased HDL-cholesterol levels by $29{\sim}65%$ as compared to B group, and so the author assumes that garlic supplementation exerts hypocholesterolemic effect on cholesterol - fed rats because of the increase of HDL and HDL - cholesterol levels. 3. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (C.D.E.F) to 2% cholesterol plus 0.25% bile salt diet (B) increased whole blood coagulation times, prothrombin times and fibrinolytic activities, but decreased plasma fibrinogen levels as compared to B group, and so the author assumes that garlic supplementation exerts anticoagulative effect because of the inhibition of fibrinogen synthesis in male rat's liver. 4. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (B.C.D.E) to standard diet (A) decreased serum glucose levels by $1{\sim}24%$, but increased serum insulin concentrations by $0.5{\sim}3.0$ times as compared to A group, and so the author assumes that garlic supplementation exerts hypoglycemic effect because of the increase of serum insulin levels by stimulation pancreatic secretion of insulin from ${\beta}-cells$ in the islets of Langerhans.

A Study on the Blood Sugar Improvement Effect and Safety of Galgunhwangryunhwanggum-tang for Type 2 Diabetes without Complications: A Systemic Review and Meta-Analysis (합병증을 동반하지 않은 2형 당뇨병에 대한 갈근황금황련탕의 혈당개선 효과와 안전성 연구 : 체계적 문헌고찰과 메타분석)

  • Objectives: The purpose of this study is to assess the antidiabetic effect and safety of Galgunhwangryunhwanggum-tang for type 2 diabetes without complications by analyzing related research. Methods: For a systematic review and meta-analysis, we searched for the antidiabetic effect and safety of Galgunhwangryunhwanggum-tang for type 2 diabetes without complications in 10 databases up to September 2021. Only randomized controlled trials were chosen. Results: In the treatment effectiveness analysis and meta-analysis, Galgunhwangryunhwanggum-tang had significant improvement effects on fasting plasma glucose level, 2-hour postprandial glucose level, glycated hemoglobin, fasting insulin, and homeostasis model assessment for insulin resistance compared to the control group when treated in parallel with oral glycemic drugs. Conclusion: Galgunhwangryunhwanggum-tang is effective in improving blood sugar and insulin resistance in type 2 diabetes patients without complications and can especially be considered in parallel treatment with oral hypoglycemic drugs. A large-scale randomized controlled clinical trial is required to complement the limitations presented in this study in the future.

Protein Tyrosine Phosphatase 1B inhibitory Activity of Anthraquinones and Stilbenes

  • Protein tyrosine phosphatase 1B (PTP1B) is emerging as a potential therapeutic target for the treatment of type-2 diabetes and obesity. To search for new types of PTP1B inhibitors, we have undertaken in vitro enzyme assay for some anthraquinones and stilbenes isolated from plants. Of the anthraquinones tested, physcion (1), 1-O-methylemodin (2), and emodin (3) showed high activities, with $IC_{50}$ values of 7.6, 7.0, and $3.8{\mu}g/mL$, respectively, while the anthraquinone glycosides, physcion-8-O-${\beta}$-D-glucopyranoside (4) and emodin-8- O-${\beta}$-D-glucopyranoside (5), were less active than their aglycones. All the stilbenens (6 - 15) slightly inhibited PTP1B activity at high concentration of $30{\mu}g/mL$. Our findings suggest that the hypoglycemic effect of anthraquinones may be associated with their PTP1B inhibitory activity.
