New record of three economic Hypnea species (Gigartinales) in Korea

  • Three economic marine algae that have been used as food and carrageenan sources were collected from Korea during a survey of marine algal flora. They share the generic features of Hypnea, and three major clades supported by the sectional features were confirmed in a phylogenetic tree based on rbcL sequences. The first species, which belongs to a species group corresponding to the sect. Spinuligerae, nests in the same clade with Hypnea yamadae in a genetic distance of 0%. It is morphologically characterized by an entangled base, subcompressed or subterete to terete axes, somewhat percurrent main axis, irregularly alternately branching with wide angle, and rarely hooked spinous branchlets. The second one is also referred to the sect. Spinuligerae and formed the same clade as Hypnea cenomyce. The genetic distance between both sequences was calculated as 0.0-0.1%, which is considered to be intraspecific. This species is distinct by somewhat entangled thallus at the basal part, percurrent axis, short spine-like branchlets densely covering the axis, and medullary lenticular thickenings. The third alga, which forms a species group corresponding to the sect. Pulvinatae, nests in the same clade as Hypnea nidulans (no intraspecific divergence). It shows occasionally epiphytic habitat rather than epilithic habitat of low mat-forming growth and percurrent erect main axes with dense lateral branchlets. Based on these morphological and molecular data, the three Korean species are identified as H. yamadae, H. cenomyce, and H. nidulans. This is the first record of the Hypnea species in Korea.

Molecular analyses and reproductive structure to verify the generic relationships of Hypnea and Calliblepharis (Cystocloniaceae, Gigartinales), with proposal of C. saidana comb. nov.

  • The genera Hypnea and Calliblepharis of the family Cystocloniaceae are discriminated by their female reproductive structure, especially in the formation of carposporangia and gonimoblasts. Hypnea saidana, once classified based on obsolete evidence, has not been studied phylogenetically using molecular analysis and detailed reproductive structure though it shares many morphologic features with the genus Calliblepharis. To provide better understanding of generic relationship of H. saidana with Hypnea and Calliblepharis, we carried out molecular analyses using the nuclear-encoded small subunit ribosomal DNA (SSU) and chloroplast-encoded large subunit of the RuBisCO (rbcL), and exact morphological observations focusing on the reproductive structures of wild specimens. Our molecular phylogeny showed that H. saidana is closely related to Calliblepharis, but distinct from the clade of Hypnea. Female reproductive structure of H. saidana characterized by upwardly developing chains of carposporangia, central reticulum of cell, and gonimoblast filaments not connected to the pericarp provides definite evidence to assign the taxonomic position of this species to Calliblepharis. Based on our combined molecular and morphological analyses, we have proposed Calliblepharis saidana comb. nov., expanding the distribution of Calliblepharis habitat from the eastern Atlantic South Africa, the northern Indian Ocean, Australasia, and Brazil to the western Pacific Ocean.

한국산 미기록 해조 잔가지가시우무 (돌가사리목, 열매가지과) (Hypnea esperi (Cystocloniaceae, Gigartinales): a newly recorded species of marine algae in Korea)

  • In the course of survey of marine algal flora in Korea, a red algal species was collected from Pohang located in the eastern coast of Korea. This shares features of Hypnea, and is characterized by more or less percurrent axis and numerous and short branchlets densely covering axis and branches with basal constrictions, and absence of lenticular thickenings in medullary cell walls. In a phylogenetic tree based on rbcL sequences, this species nests in the same clade as Hypnea esperi. Genetic distance between this Korean entity and H. esperi was in intraspecific range as 0.6%. This alga is identified as Hypnea esperi based on the morphological and molecular data. This is the first record of H. esperi in Korea.

Morphology and Molecular Phylogeny of Hypnea flexicaulis(Gigartinales, Rhodophyta) from Korea

  • Morphology and molecular phylogeny of a red algal species, Hypnea flexicaulis that is recently described from Japan, were investigated based on 23 collections from Korea (21), Taiwan (1), and the Philippines (1). Hypnea flexicaulis has percurrent axes with flexuous, antler-like branches which have wide branching angles, and abaxially curved ultimate branchlets. In order to study DNA divergence and phylogenetic relationships of the species, we determined plastid rbcL and mitochondrial cox1 sequences from the 23 collections. All 21 specimens from five different locations in Korea were almost identical to H. flexicaulis from Japan in rbcL sequences. Although there was a difference of three to five base pairs (bp) between samples from Korea and the Philippines or between the Philippines and Taiwan, Bayesian analyses of the rbcL data showed that all specimens from Korea, Japan, the Philippines, and Taiwan were strongly monophyletic. However, it is interesting that specimens from the Philippines differed by 31-34 base pairs in mitochondrial cox1 gene from those of materials from Korea and Taiwan, which differed by one to seven bp in rbcL between them. Although H. boergesenii is different from H. flexicaulis in having many antler-like branchlets, both appeared as sisters in all analyses of the rbcL data. This is the first report of H. flexicaulis from Korea based on morphology, rbcL, and cox1 gene sequences.

해조류를 이용한 Pb 및 Cu의 흡착 (Biosorption of Pb and Cu by Marine Algae)

  • Biosorption of Pb and Cu was evaluated for 23 species of marine algae collected from a Korean coast. Among a variety of species for biosorbent potential, Hypnea charoides showed the highest capacity for Pb. An adsorption equilibrium was reached in about 2 hr for Pb and 30 min for Cu. The uptake capacity was 192.8 mg Pb/g biomass and 256 mg Cu/g biomass, respectively. The adsorption parameters for Pb and Cu were determined according to Langmuir model. With an increase in pH value, more negative sites are becoming avaliable for adsorption of pH and Cu, thus the removal of Pb and Cu increases at alkaline conditions. The selectivity of mixture solution shows the uptake order of Pb>Cu>Cr>Cd. When Ca concentration increases in Pb solution, Pb was selectively adsorbed.

한국산 해조의 주해 (Notes on Marine Algal Species from Korea)

  • 해양 생물은 중요한 생물자원으로 인식되고 있다. 특히, 해조류 자원은 식용, 유용 유기물질 추출, 바이오연료 생산, 탄소 흡수 등과 같은 많은 잠재적인 이용 가능성을 갖는다. 한국산 자생생물 조사의 일환으로 Halymenia latifolia P.L. Crouan et H.M. Crouan ex Kutzing, Grateloupia livida (Harvey) Yamada and Hypnea boergesenii Tanaka의 3종이 동해 연안에서 채집되었다. 본 3종에 대한 형태학 및 분류학적 주해가 본 연구에서 제시된다. 이 중에서 Halymenia latifolia는 한국산 미기록 종으로 여기서 처음으로 보고된다.

Effect of Rhodophyta extracts on in vitro ruminal fermentation characteristics, methanogenesis and microbial populations

  • Objective: Due to the threat of global warming, the livestock industry is increasingly interested in exploring how feed additives may reduce anthropogenic greenhouse gas emissions, especially from ruminants. This study investigated the effect of Rhodophyta supplemented bovine diets on in vitro rumen fermentation and rumen microbial diversity. Methods: Cannulated Holstein cows were used as rumen fluid donors. Rumen fluid:buffer (1:2; 15 mL) solution was incubated for up to 72 h in six treatments: a control (timothy hay only), along with substrates containing 5% extracts from five Rhodophyta species (Grateloupia lanceolata [Okamura] Kawaguchi, Hypnea japonica Tanaka, Pterocladia capillacea [Gmelin] Bornet, Chondria crassicaulis Harvey, or Gelidium amansii [Lam.] Lamouroux). Results: Compared with control, Rhodophyta extracts increased cumulative gas production after 24 and 72 h (p = 0.0297 and p = 0.0047). The extracts reduced methane emission at 12 and 24 h (p<0.05). In particular, real-time polymerase chain reaction analysis indicated that at 24 h, ciliate-associated methanogens, Ruminococcus albus and Ruminococcus flavefaciens decreased at 24 h (p = 0.0002, p<0.0001, and p<0.0001), while Fibrobacter succinogenes (F. succinogenes) increased (p = 0.0004). Additionally, Rhodophyta extracts improved acetate concentration at 12 and 24 h (p = 0.0766 and p = 0.0132), as well as acetate/propionate (A/P) ratio at 6 and 12 h (p = 0.0106 and p = 0.0278). Conclusion: Rhodophyta extracts are a viable additive that can improve ruminant growth performance (higher total gas production, lower A/P ratio) and methane abatement (less ciliateassociated methanogens, Ruminococcus albus and Ruminococcus flavefaciens and more F. succinogenes.

Characteristics and Structure of Benthic Algal Community in Pohang New Port Area

  • Community structure and biodiversity of benthic marine algae were studied in the rocky shore of Phohang New Port, Yeoungil Bay. A total of 79 species of marine algae including 8 Chlorophyta, 23 Phaeophyta and 48 Rhodophyta are listed. The dominant algal species were Corallina pilulifera in all seasons, and Sargassum homeri in winter. Lomentaria catenata, Lomentaria hakodatensis, Grateloupia laceolata, Sargassum thunbergii, Chondria crassicaulis, Ulva pertusa, and Hypnea charoides were subdominant at different seasons. The algal biomass per unit area of the benthic algal community in Pohang New Port under construction was 14506 - 398.93 g dry wt m-2, which means that its annually averaged value is 273.45 g dry wt m-2 . The seasonal change in algal biomass showed highs in winter and spring, and lows in summer and autumn. The algal species diversity (H') based on algal biomass was 2.07 annually in average, with 2.42 in winter, 2.65 in spring, 1.79 in summer and 1.43 in autumn. It was high in winter and spring seasons and low in summer and autumn seasons. It is caused by that spring and winter are the profitable growth time for algae and species components in this study were dominated by the annual or seasonal rather than perennial plants.

경남 대송리산 홍조 16종의 과포자 및 사분포자의 형성시기 (Formation Periods of Carpospores and Tetraspores of 16 Red Algae in Daesongri, Kyungsangnam-do, Korea)

  • 본 연구는 홍조 16종에 대해서 경남 양산군 대송리의 자연개체군을 대상으로 사분포자체 및 과포자체의 출현시기를 파악하였다. 그 결과 G. furcata가 춘$\cdot$하계에 L. intermedia, L. undulata, R. intricata는 하계에, C. dasyphylla, H. charoides는 추계에, C. affinis, P lanceolata, C. ocellatus, A. polyneurum, G. flabelliformis, G. intermedia, C. parvula, C. Paniculatum는 하계와 추계에 사분포자 및 과포자를 형성하여 뚜렷한 계절성을 보였다. 조사대상 16종 중에서 G. flabelliformis, G. intermedia, R. intricate에서는 과포자상(carposporic phase)만 관찰되었고, P. telfairiae, H. charoides, C. parvula, C. Paniculatum에서는 사분포자상(tetrasporic phase)만이 관찰되었다. 이 중 G. flabelliformis는 고수온역인 고리 원자력 발전소취수구에서는 사분포자상만이, 또 C. paniculatum는 과포자상만이 형성되어 자연개체군의 생활사는 불안정하며, 또 수온에 의해 지역적으로 제한됨을 시사하였다. 또한 과포자상 및 사분포자상을 갖는 종 중에서 C. affinis는 과포자상이 우세한 종이며, G. furcata, C. ocellaltus, C. dasyphylla, S. latiuscula는 사분포자상이 우세하였다. 또 C. affinis, C. ocellatus, A. polyneurum, C. dasyphylla, L. intermedia, L. undulata에서는 과포자상의 출현후 1개월 이내에 사분포자상이 출현하는 것으로 나타났다. 생식세포의 형성시기와 생체량의 변동은 생식세포를 형성하는 시점에서 생체량이 극대화되고, 생식세포를 방출하면서 생체량이 점차 감소하는 것으로 나타났다.
