  • We studied the effects of 2 different dosages of high-molecular-weight poly-${\gamma}$-glutamic acid (hm ${\gamma}$-PGA) derived from Bacillus subtilis chungkookjang on lipid metabolism in a high-fructose diet-induced hypertriglyceridemic animal model. For 4 weeks, rats were fed either AIN-93 diet (normal control, NC; n = 10) or modified AIN-93 diet in which cornstarch was substituted with 63% fructose (n = 30) to induce hypertriglyceridemia. After 4 weeks, the hypertriglyceridemic rats were treated with daily oral doses of 0 mg (hypertriglyceridemic control, HC), 2.5 mg (hypertriglyceridemic, low hm ${\gamma}$-PGA, HL), or 5 $mg{\cdot}kg{\cdot}bw^{-1}{\cdot}d^{-1}$ (hypertriglyceridemic, high hm ${\gamma}$-PGA, HH) hm ${\gamma}$-PGA for 4 weeks. The HL and HH groups exhibited significantly lower levels of serum triglyceride, total cholesterol, LDL cholesterol, and free fatty acids than the HC group. The administration of hm ${\gamma}$-PGA reduced serum ALT and AST levels. The activities of lipogenic enzymes such as hepatic malic enzyme and glucose-6-phosphate dehydrogenase as well as glucose-6-phosphate dehydrogenase mRNA expression were significantly decreased by hm ${\gamma}$-PGA administration (p < 0.05). These results indicate that hm ${\gamma}$-PGA has an anti-hypertriglyceridemic effect in high-fructose diet-induced hypertriglyceridemic rats.

  • The effect of Red ginseng saponin on plasma lipid levels in Wistar rats fed on a diet high in cholesterol and triglyceride was determined. A dose of Red ginseng-crude saponin (150 mg/kg/day) was administered orally for 4 weeks to Wistar rats fed on a diet containing 2% cholesterol and 10% olive oil. Plasma levels of total cholesterol, HDL-cholesterol and triglyceride were measured and lipoproteins were analyzed by using electrophoretic technique. Red ginseng saponin showed no significant changes of HDL-cholesterol level but it lowered plasma levels of total cholesterol and elevate those of triglyceride intensively.

  • This study intended to compare the hypolipidemic effects among six experimental groups fed by three different dietary carbohydrates on hyperlipodemic rats. Sixty experimental animals were divided into 6 groups, SB, ST, SP and CB, CT, CP after production of hyperlipidemia fed by SB diet on Sprague-Dawley rats for 4 weeks. Rats were fed by six experimental diets for eight weeks. Hyjperlipidemic rats showed three times higher in plasma TG level not in cholesterol content compare to control group fed by stock diet. Two different dietary carbohydrates seem to be effective in body weight gain and fat cumulation as weight of epididymal fat pad. In comparison of S and C groups, C fed groups showed lowering effect in plasma TG and total lipid contents, but among S fed groups, ST and SP group showed lower than SB in this respects. Dietary carbohydrates seem to be more effective than fat in plasma lipid contents. When we compare among three different fat groups, only T groups with different carbohydrates increased in peroxisomal lipid oxidation and decreased in lipogenic enzyme activites. As same token, sucrose fed group with three fat sources seem to increase activiteies enzyme activities. In epididymal fat pad and Heart, SP and CP effect more in LPL activites than other groups. In conclusion, we can recommed to consume polysaccharides rather than disaccharide and n-3 fatty acids such as perilla and tuna oils to alleviate hypertringlycemic condition.

  • The triglyceride-lowering effect of probiotics Lactobacillus plantarum KY1032 and Lactobacillus curvatus HY7601 were investigated. Male SD Wistar rats were randomly divided into three groups and fed high-fat diet (HFD), HFD and probiotics (5 × 109 CFU/day of L. plantarum KY1032 and 5 × 109 CFU/day of L. curvatus HY7601), or normal diet for 6 weeks. Probiotic treatment significantly lowered the elevated plasma triglyceride and increased plasma free fatty acid, glycerol, and plasma apolipoprotein A-V (ApoA-V) levels. The probiotic-treated group showed elevated hepatic mRNA expression of PPARα, bile acid receptor (FXR), and ApoA-V. These results demonstrate that L. plantarum KY1032 and L. curvatus HY7601 lower triglycerides in hypertriglyceridemic rats by upregulating ApoA-V, PPARα, and FXR.

  • Anti-hyperlipidemic effect of red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE), which are used frequently in China and Korea as herbal medicines to treat arteriosclerosis, were investigated. Treatments of RG and CF significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. Ginsenoside Rg3 and ursolic acid, the main constituents of RG and CF, respectively, also reduced TG and TC levels in hyperlipidemic mice. RG and CF significantly lowered the high blood TG and TC levels and body and epididymal mass weights induced by long-term feeding of a high-fat diet and increased the high-fat diet-induced decrease in blood HDL cholesterol levels. RG and Rg3 reduced the blood TC levels more than CF and ursolic acid. However, blood TG level were reduced by CF and ursolic acid more than RG and Rg3. RG, CF, and their constituents also inhibited pancreatic lipase and HMG-CoA reductase activities. The most potent inhibitor was Rg3. These findings suggest that RG and CF may be suitable for the therapies of hypercholesterolemia and triglyceridemia, respectively.

  • Hypolipidemic effect of Insamsansa-eum (ISE, Ren-Shen-Shan-Zah-Yin in Chinese) consisted of Red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE) is used frequently in China and Korea as a herbal medicine to treat arteriosclerosis, was investigated. Treatment of ISE significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. ISE significantly lowered the high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by long-term feeding of a high-fat diet and increased blood HDL cholesterol level. ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

  • This study was designed to investigate the effects of Coriandrum Sativum L. on lipid metabolism in rats with hypertriglyceridemia. Also, we compared the effects of the leaf, seed and root of Coriandrum Sativum L. on lipid metabolism in rats with hypertriglyceridemia. Hypertriglyceridemia was induced in 32 male Sprague-Dawley rats weighting 108$\pm$13 g through the feeding of high-fat diets containing 20% fat and 5% cellulose for ten days. The rats were divided into four groups as follows : control (C), leaf (L), seed (S) and root (R) groups. For the five-week experimental period, the C group was fed the above diet an the L, S and R groups were fed Coriandrum Sativum L. diets containing 5% dry leaf, seed and root of Coriandrum Sativum L., respectively. Intake of diet and weight gain were significantly lower in the C group than in the L, S and R groups. But there was no significant difference among the L, S and R groups. Because of weight differences among the groups, all obtained data was adjusted to weight. The level of plasma total cholesterol was not significantly different among the four groups. But after adjusting to weight differences, the level of plasma triglyceride was significantly higher in the C groups than in the L and S groups. These results suggest that dietary Coriandrum Sativum L. may increase appetite and have an inhibitory effect on the lipid metabolism of rats with hypertriglyceridemia. Also, those effects may be partly different (leaf, root and seed) from those of Coriandrum Sativum L.
