Effects of Mugwort, Dried Orange Peel and Duching on Lipid Metabolism in Hyperlipidemia Rats (쑥 , 진피, 두충이 고지혈증 흰쥐의 체내 지질대사에 미치는 영향)

  • This study was performed to investigate the influence of dietray food from the three CHinese medicinal herbs(Mugwort, Dried organge peel and Duchung) on lipi d metabolism in rats. Male rats of Sprague-Dawley strain were divided into four dietary groups(Cellulose (CL), Mugwort(MW), Dried organge peel9OP), Duchung(DC)) after three weeks of hyperlipidemic dieting and were fed ad libitum each of the experimental diets for six weeks. The fiber sources of the experimental diets were prepared by drying and milling mugwort, orange peel, duchung and cellulose. Each of the Fiber sources was mixed into the diet to achieve 10g% levels of total dietary fiber. Levels of apparent fat absorption exhibited a significant decrease in the MW and DC groups. The OP group also showed a decreasing trend, but to a lesser extent. The levels of serum total lipid were not significantly different among the four experimental groups. Total serum cholesterol levels were significantly lower in the OP grop that in both the CM and MW group. Serum TG and LDL-cholesterol levels were significantly higher in the DL group than other groups. Serum HDL-cholesterol levels were significantly levels were significantly lower in the CL group than in the other groups. Total lipid and total cholesterol in liver were significantly higher in the CL group than in the other groups. Total lipid and total cholesterol in feces displayed a significantly increase in the MW and DC groups when compared with the CL group. The OP group also showed an increase compared to the CL group.

Antihyperglycemic and Antihyperlipidemic Effects of Fermented Rhynchosia nulubilis in Alloxan-induced Diabetic Rats

  • Alloxan administration in rats is used as a model for non-insulin dependent diabetes mellitus (NIDDM). NIDDM is a multifactorial disease, characterized by hyperglycemia and lipoprotein abnormalities. In this study, we evaluated the antihyperglycemic and antihyperlipidemic effects of fermented Rhynchosia nulubilis (FRN) through the regulation of glucose uptake in alloxan-induced rats. Fermented R. nulubilis was administered orally for 28 d at 500 mg/kg of body weight. Body weight and food intake were monitored every day. Biochemical parameters were quantified after 4 week. In the diabetic + FRN group, body weight increased significantly and blood glucose concentrations decreased when compared to those of the diabetic group. After 2 hr of administration, the oral glucose tolerance test (OGTT) indicated a significant reduction in the diabetic + FRN group compared to diabetic group. The diabetic + FRN group experienced a significant reduction in total cholesterol, triglycerides, low density lipoprotein, coronary risk factors, and malondialdehyde concentrations, with significantly increased high density lipoprotein compared to those of diabetic group. These results demonstrate that fermented R. nulubilis possesses potent antihyperglycemic and antihyperlipidemic activity in alloxan-induced diabetic rats.

Antihyperlipidemic Activity of Scutellaris baicalensis Georg., Coptidis japonica Makino and Rhei koreanum Nakai on Experimental Hyperlipidemia in Rats (고지혈증 랫트를 이용한 황금, 황련, 대황의 항고지혈 효과)

  • Active component was sought among three herb medicines, which are used in combination as a traditional medicine prescribed for patients with hyperlipidemia related diseases. Antihyperlipidemic effect of this remedy has previously been shown model by the authors on the animal model. Hyperlipidemia was induced on male Wistar rats by keeping them on high lipid diet for one week, as previously described by the authors. Blood lipid profile was verified on these rats by measuring total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL). Then, the diet was changed to normal. At the same time, methanol extracts of Scutellaris baicalensis Georg.(radix), Coptidis japonica Makino(rhizoma) or Rhei koreanum Nakai(rhizoma) were given on daily basis, and changes in the blood lipid profile were monitored for 4 weeks. Methanol extract of Scutellaris baicalensis Georg. significantly reduced the TC value, implying the in vivo antihyperlipidemic effect.

Fermented Ginseng Attenuates Hepatic Lipid Accumulation and Hyperglycemia through AMPK Activation

  • Fermented ginseng (FG) is an ethanol extract of ginseng radix processed with $\beta$-galactosidase. It was hypothesized that FG may exert anti-hyperlipidemic and anti-diabetic activities through modulating AMP-activated protein kinase (AMPK) in HepG2 human hepatoma cells. In this study, we showed that AMPK phosphorylation was stimulated by FG. These effects were abolished by pretreatment with an AMPK inhibitor, compound C. In addition, FG regulated the expression of genes associated with lipogenesis and lipolysis, thus causing suppression of hepatic triglyceride accumulation. In vivo study using db/db mice, FG reduced fasting plasma glucose, HbAlc, and insulin resistance index, when compared to diabetic control. FG also increased the phospho-AMPK and glucose transporter 4 (GLUT4) expressions in liver and skeletal muscle, respectively. In liver, expressions of lipogenic gene were decreased whereas expressions of lipolytic genes were induced, when compared to diabetic control. Taken together, we may suggest that FG ameliorates hyperglycemia and hyperlipidemia through activation of AMPK and could be developed as a health functional food or therapeutic agent for type 2 diabetic patients.

Is the relationship between periodontitis and hyperlipidemia mediated by lipoprotein-associated inflammatory mediators?

  • Purpose: The aim of this study was to evaluate the serum levels of oxidized low-density lipoprotein (oxLDL), anti-oxLDL, and myeloperoxidase (MPO) in hyperlipidemic patients with periodontal disease. Methods: This study included 123 patients with hyperlipidemia categorized based on metabolic control as mild to moderate (H1) (n=66) or poor (H2) (n=57), as well as systemically healthy controls (C) (n=68). Serum levels of lipids, oxLDL, anti-oxLDL, and MPO were evaluated, along with clinical periodontal parameters. Results: The percentage of bleeding on probing (BOP%) and the clinical attachment level were significantly higher in the H2 group than in the C group. Patients with hyperlipidemia had a relatively high risk of developing periodontal disease. The oxLDL and anti-oxLDL levels were higher in H2 patients with periodontitis than in the control or H1 patients with periodontitis. In the H1 and H2 groups, the ratio of total cholesterol to high-density lipoprotein was significantly correlated with gingival index, BOP%, and oxLDL levels. Conclusions: Our findings indicate that the lipoprotein-associated inflammatory mediators of oxLDL, anti-oxLDL, and MPO may play an important role in the relationship between periodontal disease and hyperlipidemia.

Effects of Coix lachryma-jobi var. mayuen stapf. Pharmacopuncture on Plasma lipid composition and glucose in rat fed high fat diet. (율무 약침이 고지방식이 급여 흰쥐의 혈액 내 지질구성 및 Glucose량에 미치는 영향)

  • Objective : To investigate the effects of Yullmoo (Coix lachryma-jobi var. mayuen stapf.) Pharmacopuncture on plasma lipid composition and glucose concentration in hyperlipidemic rats. Methods : ${\beta}-lipoprotein$ free fatty acids, triglyceride and total cholesterol levels in Yullmoo (Coix lachryma-jobi var. mayuen stapf.) Pharmacopuncture groups were compared with those in the control group. Results : Concentration of ${\beta}-lipoprotein$ showed no significantly different in all treatment groups. Concentration of free fatty acids, triglyceride and total cholesterol in plasma were decreased in the Yullmoo (Coix lachryma-jobi var. mayuen stapf.) Pharmacopuncture groups. In the Yullmoo (Coix lachryma-jobi val. mayuen stapf.) Pharmacopuncture groups, plasma LDL-cholesterol showed a lower value and HDL-cholesterol showed a higher value than those of the control group. Contents of glucose were decreased in the Yullmoo (Coix lachryma-jobi val. mayuen stapf.) Pharmacopuncture groups. Conclusions : The results suggest that Yullmoo (Coix lach.yma-jobi var. mayuen stapf.) Pharmacopuncture may have an impact on lipid metabolism to potentially prevent obesity and enhance treatment effect.

Effects of Poncirus trifoliata Pharmacopuncture on plasma lipid composition and concentration of $TNF-{\alpha}$ and IL-6 in rats fed high fat diet (지각(枳穀) 약침이 고지방 식이 급여 흰쥐의 혈액 내 지질구성과 $TNF-{\alpha}$ 및 IL-6농도에 미치는 영향)

  • Objective : To investigate the effects of Poncirus trifoliata Pharmacopuncture on lowering lipid and contents of serum tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ and interleukin-6 (IL-6) in hyperlipidemic rats. Methods : Triglyceride, total cholesterol, $TNF-{\alpha}$, and IL-6 levels in Poncirus trifoliata Pharmacopuncture groups were compared with those in the control group. Results : Concentration of triglyceride and total cholesterol in plasma was decreased in the Poncirus trifoliata Pharmacopuncture groups. In Poncirus trifoliata Pharmacopuncre groups, plasma LDL-cholesterol showed a lower value and HDL-cholesterol showed a higher value than those of the control group. Contents of $TNF-{\alpha}$ was decreased in the Poncirus trifoliata Pharmacopuncture groups. Contents of IL-6, however, were not significantly different. Conclusions : The results suggest that Poncirus trifoliata Pharmacopuncture may have an impact on lipid metabolism to potentially prevent development of diabetes mellitus and accompanying cerebrovascular diseases.

Depression and Dietary Factors Related to Hyperlipidemia in Urban Living Elderly Female from Low Income Group (도시 저소득층 여자노인의 고지혈증과 관련된 우울정도와 식생활 요인에 관한 연구)

  • We conducted a case-control study to examine the relationship of depression and dietary related factors with the hyperlipidemia for urban living elderly women from low income group. The case group consisted of 45 elderly females with hyperlipidemia (serum cholesterol $\geq$ 240mg/dl or serum TG $\geq$ 250mg/dl and the control group of 95 age matched elderly women with serum cholesterol levels less than 240mg/dl and serum TG less than 250mg/dl. In a univariate analysis, vitamin C intake, the number of family members living with the subject, and their depression scores were significantly higher in the hyperlipidemic group than in the control group. In the logistic regression analysis, the vitamin C intake ($\geq$75% Korean RDA), the number of family members living with the subject ($\geq$ 1), depression scores ($\geq$7), BMI ($\geq$27), and subscapular skinfold thickness ($\geq$18mm) were associated with significantly higher (p<0.05) risks of hyperlipidemia in the elderly women. However after adjustment for other covariables, the depression scores (Odds Ratio 2.48 for depression score$\geq$7;95%CI:1.10-5.60) and subscapular skinfold thicknesses (Odds Ratio 5.69 for SBT$\geq$18mm, 95%CI:1.87-17.32) were the significant risk factors associated with hyperlipidemia in the elderly women.

Effects of Cinnamomi Cortex on Hypercholesterolemia Induced by Estradiol Valerate in Rats (육계(肉桂)가 Estradiol valerate에 의하여 유발된 고콜레스테롤 혈증에 미치는 영향)

  • This study was designed to investigate the effects of Cinnamomi Cortex extract (CC) on hypercholesterolemia induced by Estradiol valerate (EV) in female rats. We investigated the effects of CC on Changes in body weights for 5 weeks. In addition, we examined the effects on fasting blood glucose (FBS) and tested oral glucose tolerance test after oral administration of CC for 5 weeks. We also investigated the effects on levels of total cholesterol, HDL-cholesterol and LDL-cholesterol in serum. Treatment with CC restored weight gain rates suppressed by EV significantly. CC did not affect serum glucose level and glucose tolerance. Levels of total cholesterol and triglyceride were lowered by administration of CC compared to EV treated control respectively. These result suggest that CC can lower serum cholesterol level then prevent weight loss induced by EV. We also suggest the possibility as anti-hyperlipidemic agent of CC.
