Using immunohistochemical technique, three endocrine cell types were observed in the GIT of the Korean salamander, Hynobius leechii. BPP-immunoreactive cells were distributed from the pyloric region to the small intestine. A moderated number of somatostatin-immunoreactive cells were restricted to the pyloric region. 5-HT-immunoreactive cells were demonstrated throughout the GIT, reaching a peak in the pyloric region. The presence of BPP- and 5-HT-immunoreactive cells are identified for the first time in Hynobius leechii.
한국산 도롱뇽(Hynobius leechii) 유생의 다리 발생 과정을 관찰하고 잠재적 돌연변이 유발물질인 제-methyl-N'-nitro-N-nitrosoguanidine(MNNG)과 잠재적 기형 유발물질인 retinol palmitate가 다리 발생에 미치는 영향을 알아보았다. 정상적인 개체에서 됫다리의 발생 단계는 외부와 내부의 형태적 특징을 기준으로 하여 7단계로 나누어 볼 수 있었으며 전연골세포의 밀집에 따른 골격형성은 다리 발생아의 근위부에서 시작하여 원위부로 진행됨을 확인할 수 있었다. 도롱뇽 유생을 MNNG(10 rpm)나 retinol palmitate(37.5 lpm)로 처리한 경우, 공통적으로 다리 원위부의 골격 형성이 억제되었으며, 특히 이러한 골격 형성 억제 현상은 처리시기가 이를수록 보다 심하게 나타났다. 이러한 결과는 발생을 저해하는 물질에 대한 미분화 상태의 세포들의 반응성이 발생과정 중 특정 단계에 국한됨을 시사하는 것으로 해석된다.
This study was conducted to establish a Habitat Suitability index (HSI) based on literature research and field surveys on ecology and habitat of 'Hynobius leechii'. And this study will be used as basic data for qualitative evaluation of habitat environment. The survey sites were divided into natural habitats close to the prototype habitat and artificial restoration areas where Hynobius leechii was monitored. So the types of habitats were diversified. Hynobius leechii is a vulnerable species to climate change because it is affected by the microhabitat and has low mobility. HSI variables of Hynobius leechii were extracted through domestic and overseas literature, and standards were extracted from literature research and field survey. The standards were presented as a value of the physical allowable category in consideration of realization. To verify the study, an in-depth consultation was conducted by amphibians experts. HSI variables of Hynobius leechii were included 9 variables such as Overstory canopy cover(%), Understory cover(%), Water-pH, Soil-pH, Soil relative humidity(%), Leaf litter depth(cm), Rock substrates (%), Type of Coarse woody, Distance from Street or Pollutant(m).
Polyacrylamide gel electrophoresis was used to investigate the patterns of LDH and MDH isozymes in the embryo and adult of amphibia; Rana nigromaculata, Rana plancyi chosenica and Hynobius leechii. Rana nigromaculata is considered to be heterozygous for the gene specifying the "B" subunit of LDH, and Hynobius leechii to be heterozygous for the gene specifying the "A" subunit of LDH. The LDH isozyme paatern of embryos of the above three species is characterized by a gradual increase in the activity of LDH-5 (muscular form)during development. Two or three molecular forms of MDH is present steadily from early embryos and in adult. Of the MDH isozymes, the more cathodic one (MDH-m) appears weakly in early developing stages, but increases slowly in the activity as the embryo develops.the embryo develops.
A series of no-choice olfactory response tests using water pre-conditioned with females, with intact and surgically removed ventral glands, at various reproductive states were conducted to determine whether male Hynobius leechii discriminates among females in different reproductive states based on chemical cues. Similarly, ventral gland extracts were tested, and ventral glands were examined histologically. Males’ responses to putative odors of females in four (ovulating, ovulated, ovipositing, and oviposited) reproductive states were independently measured by: i) the latency time to initiate male behavioral response, ii) the arrival time at a fixed point of putative odor source, and iii) the staying time close to the odor point source. Male salamanders showed significant olfactory responses to recently ovulated and ovipositing female odors by quickly arriving at odor sources and staying longer at the origin of the source, but the olfactory responses to the earlier staged ovulating females and the later stage of already oviposited females were not different from controls. Olfactory responses of test males to water preconditioned by intact females or females with ventral glands excised were not different. In addition, ventral gland extracts did not induce significant olfactory responses of test males although the lumens of alveoli in ventral glands of oviposited females were smaller than those of ovulated females. These results indicate that male H. leechii recognizes ovulated and ovipositing females based on chemical cues released but not from the ventral glands.
The ultrastructure of spermatozoa in Hynobius leechii was examined with transmission electron microscope and compared with those of other urodeles and anurans. The perforatorium and the tail show most of the common feature of urodeles. However, there were several ultrastructural characteristics in an acrosome, nucleus, neck, axial rod and mitochondria related to the nucleus. The acrosome was trifoliate in transverse sections and the perforatorium consisted of two different concentric parts with a fine sharp point in a subacrosomal lumen. The nucleus consisted of two different regions of chromatin area and nuclear ridge. The nuclear ridge was composed of several bundles of five to ten minute tubular subunits of 19 nm diameter in this species, while in higher urodeles it was well developed in multi layers. The protoplasmic bead was separated by a cytoplasmic canal except the connection with only the distal portion of the nucleus. The neck was a short cylinder and contained pericentriolar material with transverse striations. In Hynobius the ring was not elongated to the tail and the mitochondria were distributed only in the protoplasmic bead around the nucleus as in Cryptobranchus, while in higher groups of urodeles it elongates to the length of the middle piece and the mitochondria follow the ring. The ring elongation may be related to the distribution of mitochondria. Hynobiidae and Cryptobranchidae are closely related based on structure of neck, the axial rod and location of mitochondria, although they are different in the composition of the axial rod.
Although there are many studies of the effect of climate change on the breeding phenology and community diversity of amphibians, the studies of variations in reproductive population size of individual species according to climate change are still lacking. We examined the effect of climate change on the reproductive population size of Rana huanrenensis and Hynobius leechii, which bred in mountain valleys, by surveying the reproductive population of the two species between 2005 and 2012 and analyzing the correlation between the variation of the outdoor population and the surrounding climate change factors, obtained from a meteorological observatory located at 5.6 km from the study site. The size of the reproductive population of the two species commonly fluctuated with aan pproximately 3.5-year cycle. That of H. leechii, in particular, decreased significantly over eight years. The air temperature tended to more closely relate with the reproductive population size of R. huanrenensis as was the case of the precipitation with that of H. leechii. The yearly mean highest temperature and spring mean temperature variation consistently decreased over the eight years, and the latter was related with the significantly decreased size of H. leechii reproductive population. These results showed that recent climate change directly could affect the reproductive population size of amphibians, particularly H. leechii, which breeds in mountain valleys.
The effects of retinoic acid (RA) on the regenerating limbs of Korean newt (Hynobius leechii) larvae have been studied. Intraperitoneal injection of RA at 4 days post-amputation caused the proximalization of regenerate structures in the proximodistal (PD) axis of the limbs amputated through either the distal zeugopodium or the distal stylopodium. The mean level of proximalization (MLP) increased as a dose-dependent manner, and the MLP was also dependent on the level of amputation at a given dose of RA. At the dose of 150 $\mu$ g/g body wt., MLP's in either amputation level reached close to a peak value, and an increasing number of inhibition of regeneration or duplication in the transverse plane occurred at the dose of 200 $\mu$ g/ g body wt. and above. The changes induced by RA treatment suggest that positional values in the cells of the regenerating limbs have been modified in the three cardinal axes including PD and transverse axes in a graded mode. Furthermore, from the comparison of data in two different amputation levels, it can be speculated that the sensitivity of cells to RA effect along the PD axis might not be linear.
To determine the age structure of a Hynobius leechii breeding population and analyze relationships between the order of entrance to breeding ponds and physical parameters and age, we studied a wild population of the species in the Research Forests of Kangwon National University in Chuncheon, Kangwon, South Korea from March 16 to April 13, 2005. The age of breeding males ranged one to nine years old and that of females ranged from three to nine years old. The asymptotic sizes of males and females were 6.36 and 6.51 cm, respectively, and the growth coefficients of males and females were 0.71 and 0.81, respectively. The snout-vent length (SVL), head length, and body mass of males were all positively correlated with their age, but female age did not show a significant relationship with any physical parameter. The tail depth, body mass, and condition factors (SVL/body mass $\times$ 100) of both males and females were negatively related with the order of entrance to the breeding pond. The head width and SVL of males were also negatively correlated with the order of entrance, but the SVL of females was positively related with the order of entrance. These results suggest that physical parameters are more important determinants of breeding migration patterns than age. We discuss which of two hypotheses, the mate opportunity hypothesis and the susceptibility hypothesis, is better able to explain the order of entrance to breeding ponds for male and female H. leechii.
