간장조직의 활성산소 및 그 제거효소에 미치는 누에(Bombyx mori L.) 분말의 영향 (Effects of Silkworm (Bombyx mori L.) Powder on Oxygen Radicals and Their Scavenger Enzymes in Liver of SD Rats)

  • This study was designed to investigate the effects of silkworm (Bombyx mori L.) powder (SWP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley(SD) male rats(160$\pm$10g) were fed basic diet (control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

  • This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.

Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group. Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group, but significant difference between GSHPx activities in liver cytosol could be not obtained.   • This study was designed to investigate the effect of silk fibroin (Mw 500)power (SFP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a considerable decreases (5.8% and 8.4%, 3.7% and 11.1%, respectively) in liver motochondria and micorsomes of SFP-2.5 and SFP-5.0 groups compared with control group, and $O_2$radical level was remarkably decreased about 15% and 20% in liver cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

  • This study was designed to investigate the effect of silkworm powder on oxygen radicals and their scavenger enzymes in brain membrances of SD rats. Hydroxyl radical (OH) levels resulted in a considerable decreases in brain mitochondria fraction. Superoxide radical (O$_2$) levels were a slightly decreased in brain cytosol fraction. Lipid peroxide (LPO) and Oxidized protein (OP) levels were significantly decreased in brain mitochondria and microsomes fraction. Mn-superoxide dismutase (SOD) activity was remarkably increased in the mitochondria fraction. Cu and Zn-SOD activities were effectively increased in brain cytosol fraction. GSHPx activity was considerably increased in brain cytosol fraction. These results suggest that anti-aging effect of silkworm plays an effective role in attenuating an oxidative stress and increasing a scravenger enzyme activity in brain membranes.

Superoxide radical (O2) levels were a slightly decreased (2.0% and 9.1%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide(LPO) levels were significantly decreased (12.9% and 21.9%, 13.2% and 22.5%, respectively) in brain mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (16.7% and 15.7%, respectively) in brain microsomes of SFP-2.5 and SFP-5.0 group compared with control group, but significantly difference between in brain mitochondria of these two groups could not be obtained. Mn-SOD activities were remarkably increased (11.2% and 24.2%, respectively) in mitochodria of SFP-2.5 and SFP-5.0 groups. CuZn-SOD activities were effectively increased (7.7% and 19.6%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups, but significant difference between control and SFP-2.5 groups could be not obtained. GSHPx activities were considerably increased (5.3% and 11.7%, respectively) in brain cytosol of SFP-2.0 and SFP-5.0 groups compared with control group. There results suggest that anti-aging effect of silk fibroin may play an effective learning and memory role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in brain membranes.

  • 간장중의 활성산소 및 제거효소의 활성에 미치는 영향을 평가하기 위하여 미역의 건조분말을 $10{\%},\;20{\%},\;40{\%}$-첨가 조제한 미역국수 ($10{\%},\;20{\%},\;40{\%}\;FBA-noodle$)를 4주동안 SD계 흰쥐에 투여하여 미역국수의 생리작용을 평가하였다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 mitochondria의 ${\cdot}OH$ 생성은 대조그룹 대비 각각 $20{\%},\;25{\%},\;35{\%}$의 현저한 OH 생성 억제효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 microsome의 ${\cdot}OH$ 생성도 대조그룹 대비 $12{\~}20{\%}$의 유의적인 ${\cdot}OH$ 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 microsome의 $H_2O_2$의 생성은 유의적인 억제효과를 인정할 수 없었지만, cytosol중의 $O_2^({\cdot}-)$ 생성은 $20{\%},\;40{\%}$ FBA-noodle에서 $10{\%}$$O_2^({\cdot}-)$의 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 간장 mitochondria의 Mn-SOD 활성은 대조그룹 대비 $10{\~}15{\%}$의 Mn-SOD 활성의 증가효과가 인정되었다. 간장 microsome의 Mn-SOD 활성은 $10{\%} 및 20{\%}$ FBA-noodle 투여그룹은 Mn-SOD 활성의 유의적인 증가 효과를 인정할 수 없었지만, $40{\%}$ FBA-noodle 투여그룹은 $12{\%}$의 유의적인 Mn-SOD 활성 증가효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 간장 cytosol의 Cu, Zn-SOD 활성은 대조그룹 대비 $10{\~}20{\%}$의 Cu, Zn-SOD 활성의 유의적인 증가효과가 인정되었다. 한편 간장 cytosol의 글루타치온 퍼옥시다아제 (GSHPx) 활성을 평가하여 보면 $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 cytosol의 GSHPx 활성은 대조그룹 대비 $20{\~}40{\%}$의 매우 효과적인 GSHPx활성의 증가효과가 인정되었다. $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 mitochondria의 LPO의 함량을 비교하여 보면 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%},\;40{\%}$ FBA-noodle은 $10{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 microsome의 LPO의 함량은 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%} 및 40{\%}$ FBA-noodle은 약 $10{\~}12{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다.

유용버섯 Lentinus tuber-regium이 산화적 스트레스 및 방어체계에 미치는 영향 (Effects of Edible Lentinus tuber-regium on Oxidative Stress and Defense System in Serum of SD Rats)

  • SD계 랫트를 사용하여 6주 동안 탄수화물 대용으로 50% 및 100%의 Lentinus tuber-regium(LTR)-첨가 투여그룹의 산화적 스트레스 및 방어체계에 미치는 영향을 평가하였다. LTR-50 및 LTR-100 투여그룹은 대조그룹 대비 21.7% 및 16.4%의 매우 유의적인 ${\cdot}OH$ 라디칼의 생성 억제효과가 인정되었고, 이들 두 투여그룹은 대조그룹 대비 약 10%의 $H_2O_2$의 생성 억제효과 및 $6{\sim}10%$ 정도의 바람직한 NO의 생성 억제효과가 인정되었다. 그렇지만 이들 LTR 투여에 의하여 ${O_2}^-$ 라디칼의 생성 억제효과는 인정할 수 없었다. 이들 활성산소의 공격에 의한 산화적 스트레스로서 LPO의 생성은 LTR 투여에 의하여 대조그룹 대비 $6{\sim}12%$의 유의적인 LPO의 생성 억제효과가 인정되었고, OP의 생성은 이들 LTR의 부여에 의하여 대조그룹 대비 5.0% 및 12.8%의 생성 억제효과가 인정되었다. 이들 LTR-50, LTR-100 투여그룹은 대조그룹에 비해 $15{\sim}50%$의 매우 유의적인 SOD의 활성 증가효과가 인정되었고, 또한 이들 LTR투여그룹의 GSHPx은 대조그룹 대비 $10{\sim}25%$의 매우 유의적인 GSHPx의 활성 증가효과가 인정되었다. 한편 CAT의 활성에 미치는 LTR투여의 영향도 대조그룹 대비 $60{\sim}90%$의 매우 유의적인 CAT의 활성 증가효과가 인정되었다. 이상의 결과에서 탄수화물 대용으로 사용된 유용버섯 LTR의 투여는 ${\cdot}OH$ 라디칼, $H_2O_2$ 및 NO의 생성 및 그로 인한 LPO 및 OP 같은 산화적 스트레스를 효과적으로 억제할 수 있을 뿐만 아니라 SOD, GSHPx 및 CAT 같은 방어효소의 활성을 매우 효과적으로 증가할 수 있기 때문에 노화를 매우 효과적으로 억제할 수 있을 것으로 기대된다.
